The oral absorption and disposition of itraconazole were studied in rats, rabbits and dogs. Serum levels of itraconazole and its active metabolite, hydroxyitraconazole, were determined by a validated HPLC method. The absorption of itraconazole was relatively rapid in rats and dogs but was slower in rabbits. The terminal elimination half-life ($T_{1/2,{\lambda}z}$), time to the peak concentration ($T_{max}$), dose and weight normalized area under the curve (AUC) and the peak concentration ($C_{max}$) of itraconazole found in the dog were comparable to those reported in humans. As in humans, the metabolite to parent drug AUC ratios in rats and dogs were greater than unity but was less in rabbits. The dog appears to be an appropriate animal model while the rat, not the rabbit, may be used as an alternative animal model in predicting the oral absorption of itraconazole in humans.

A total of 135 F2 finishing pigs (65 barrows and 69 gilts) from resource population (Large White${\times}$Meishan) were slaughtered at about 87.8 kg BW. Contents of inosine acid (IMP) and carnine (HR) in muscle were assayed by HLPC and genetic parameters for IMP content and HR content were estimated using full sibs model. There was significant sex effect on IMP content(p<0.05), $3.561{\pm}0.077mg/g$ for gilt and $3.287{\pm}0.085mg/g$ for barrow. Heritability estimates for IMP and HR content were 0.127 and 0.357, respectively. The phenotypic correlation between IMP content and HR was 0.335, pH (A) 0.024, water lose rate (WLR) -0.069, intramuscular fat (IMF) -0.214, average marbling score (MARB) -0.143, average backfat measurements (AVBF) -0.084 and average color value (CV) -0.156, respectively. The result indicated that inosine acid content in meat might be retained or slightly improved by reducing backfat depth in pig breeding.
The crystal structure of N-(2'-chloro-4'-vitrophenyl)-5-chlorosalicylamide( Niclosamide ) monohydrate has been determined from 1976 sig- nificant independent reflections collected on an automated CAD4 diffractometer using graphitemonochromated Mo-Ka radiation. The crystal is monoclinic, space group P211c, with unit cell dimensions, a=11.331 (3), b=16.964(2), c=7.347(4)A , P =98.20(3)° and Z=4 at T=293k. The structure was solved by direct method using seminvariants of ggg Parity group and refined by the full-matrix least-square method, resulting model with reliability factor, R=0.046. The feature of the molecule show planar structure in parallel to the ab crystal plane.
In this paper we Study a time-optimal model of pursuit in which the players move on a plane with bounded velocities. This game is supposed to be a nonzero-sum group pursuit game. The main point of the work is to construct and compare cooperative and non-cooperative solutions in the game and make a conclusion about cooperation possibility in differential pursuit games. We consider all possible cooperations of the players in the game. For that purpose for every game $\Gamma(x_0,y_0,z_0)$ we construct the corresponding game in characteristic function form $\Gamma_v(x_0,y_0,z_0)$. We show that in this game there exists the nonempty core for any initial positions of the players. The core can take four various forms depending on initial positions of the players. We study how the core changes when the game is proceeding. For the original agreement (an imputation from the original core) to remain in force at each current instant t it is necessary for the core to be time-consistent. Nonemptiness of the core in any current subgame constructing along a cooperative trajectory and its time-consistency are shown. Finally, we discuss advantages and disadvantages of choosing this or that imputation from the core.
Feedwater heaters of many nuclear power plants have recently experienced wall thinning damage, which will increase as operating time progresses. As it is judged that the wall thinning damages have generated due to local fluid behavior around the impingement baffle installed in downstream of the high pressure turbine extraction steam line to avoid colliding directly with the tubes, numerical analyses using PHOENICS code were performed for two models with original clogged impingement baffle and modified multi-hole impingement baffle. To identify the relation between wall thinning and fluid behavior, the local velocity components in x-, y-, and z-directions based on the numerical analysis for the model with the clogged impingement baffle were compared with the wall thickness data by ultrasonic test. From the comparison of the numerical analysis results and the wall thickness data, the local velocity component only in the y-direction, and not in the x- and z-direction, was analogous to the wall thinning configuration. From the result of the numerical analysis for the modified impingement baffle to mitigate the shell wall thinning, it was identified that the shell wall thinning may be controlled by the reduction of the local velocity in the y-direction.

The present study has extended OpenSees, which is an open-source software framework DOS program for developing applications to idealize geotechnical and structural problems, for the static analysis of axial load capacity and settlement of single piles in MS Windows environment. The Windows version of OpenSees as improved by this study has enhanced the DOS version from a general purpose software program to a special purpose program for driven and bored pile analysis with additional features of pre-processing and post-processing and a user friendly graphical interface. The method used in the load capacity analysis is the numerical methods based on load transfer functions combined with finite elements. The use of empirical nonlinear T-z and Q-z load transfer curves to model soil-pile interaction in skin friction and end bearing, respectively, has been shown to capture the nonlinear soil-pile response under settlement due to load. Validation studies have shown the static load capacity and settlement predictions implemented in this study are in fair agreement with reference data from the static loading tests.
The electronic spectra of enzyme-model organocobalt(Ⅲ) complexes containing completely or partially conjugated macrocyclic ligands were measured in various solvents to investigate the solvent effect on the charge transfer band for the axial cobalt-carbon bond by the extent of conjugation in the equatorial macrocyclic ring; completely conjugated, $CH_3CoL,\;C_6H_5CoL,\;CNCoL,\;CH_3CoL',\;CNCoL'$, partially conjugated $CH_3(py)Co(DH)_2, CH_3CoL"$, unconjugated dienes, $[CH_3Co(1,4-CT)](ClO_4)_2$, and open ring, $CH_3Co(salen)$. The position of the charge transfer band which corresponds to the cobalt-carbon bond was shifted to a shorter wavelength as the polarity of solvent increased and the transition energy $(E_T)$ had a linear relationship with solvent polarity parameter, Z-value, only in the case of completely conjugated system. However, the linear correlation between $E_T$and Z was not observed for partially conjugated and open ring systems.

Background: Dendritic cells (DC) are professional antigen-presenting cells in the immune system and can induce T cell response against virus infections, microbial pathogens, and tumors. Therefore, immunization using DC loaded with tumor-associated antigens (TAAs) is a powerful method of inducing anti-tumor immunity. For induction of effective anti-tumor immunity, antigens should be efficiently introduced into DC and presented on MHC class I molecules at high levels to activate antigen-specific $CD8^+$ T cells. We have been exploring methods for loading exogenous antigens into APC with high efficiency of Ag presentation. In this study, we tested the effect of the cationic liposome (Lipofectin) for transferring and loading exogenous model antigen (OVA protein) into BM-DC. Methods: Bone marrow-derived DC (EM-DC) were incubated with OVA-Lipofectin complexes and then co-cultured with B3Z cells. B3Z activation, which is expressed as the amount of ${\beta}$-galactosidase induced by TCR stimulation, was determined by an enzymatic assay using ${\beta}$-gal assay system. C57BL/6 mice were immunized with OVA-pulsed DC to monitor the in vivo vaccination effect. After vaccination, mice were inoculated with EG7-OVA tumor cells. Results: BM-DC pulsed with OVA-Lipofectin complexes showed more efficient presentation of OVA-peptide on MHC class I molecules than soluble OVA-pulsed DC. OVA-Lipofectin complexes-pulsed DC pretreated with an inhibitor of MHC class I-mediated antigen presentation, brefeldin A, showed reduced ability in presenting OVA peptide on their surface MHC class I molecules. Finally, immunization of OVA-Lipofectin complexes-pulsed DC protected mice against subsequent tumor challenge. Conclusion: Our data provide evidence that antigen-loading into DC using Lipofectin can promote MHC class I- restricted antigen presentation. Therefore, antigen-loading into DC using Lipofectin can be one of several useful tools for achieving efficient induction of antigen-specific immunity in DC-based immunotherapy.
The Tongcheng pig breed is a famous Chinese indigenous breed. The Ministry of Agriculture of China has filed it as 1 of 19 national key conservation breeds selected from more than 100 Chinese indigenous pig breeds in 2000. In order to improve the reproductive performance, it has been intensively selected to increase the litter size for about 10 years. The population randomly sampled from conservation nucleus of eight families in the Tongcheng pigs was genotyped for identification of their estrogen receptor locus polymorphisms with the PCR-RFLPs method. Only AB heterozygotes and BB homozygotes were detected, and $X^2$ test demonstrated that the locus was in disequilibrium at a significant level (p<0.05). In the present paper, the litter sizes in different parities were regarded as different traits. Holistic status of other unspecific and unidentified genes was estimated by using the statistical methods. Coefficients of kurtosis and skewness showed that the litter size still presented segregating characteristic in the 2nd, 5th, 7th, 8th and 9th parities. Analysis of homogeneity of variance between families confirmed the results for the 5th, 7th and 8th parities. The heritability of litter size for the 1st to 10th parities was estimated with paternal half-sib model and individual estimated breeding values (EBVs) were evaluated by a single trait animal model as well. We found that the averages of EBVs for litter size in each parity did not differ significantly between genotypes, despite the significant difference for original phenotype records in the 3rd, 4th and 5th parities (p<0.05 or p<0.01). The results may be explained by the deduction that the polymorphisms of ESR locus are no longer the important genetic base of litter size variation when the frequency of allele B accumulated in the experience of selection procedure, and further conferring that there exist special genes associated with litter size in the recent Tongcheng pigs population can be made.
