Immuno-regulatory Activities of Various Fractions from Ehpedrae Sinica STAPF, Rubus Coreanus Miq. and Angelica gigas Nakai Extracts with Ultrasonification (초음파 병행 추출을 이용한 마황과 복분자, 당귀 분획물의 면역활성 조절 효과)

  • This study was performed to examine immuno-regulatory activities of Ehpedrae Sinica STAPF, Rubus Coreanus Miq. and Angelica gigas Nakai extracts in conbination with ultrasonification. The extract yields of plants were the highest in the extraction system of $60^{\circ}C$ and 40 kHz of ultrasonification. The immune cell growth ratio of human immune B and T cells was increased compared to other fractions by the water fraction of the plants at $60^{\circ}C$ and 40 kHz. The water fractions of the plants at $60^{\circ}C$ and 40 kHz increased the specific secretion of IL-6 and $TNF-{\alpha}$ of human immune B and T cells compared to other fractions of the plants. The water fraction of Ehpedrae Sinica STAPF among the plants was observed to show the highest specific secretion of IL-6 and $TNF-{\alpha}$. Also, NK-92 MI cells growth was increased in adding the water fractions of the plants at $60^{\circ}C$ and 40 kHz. The water fraction of Ehpedrae Sinica STAPF among the plants showed the highest in NK-92 MI cell growth ratio. The differentiation activity of the HL-60 cells significantly increased in adding the water fraction of Ehpedrae Sinica STAPF compared to other fractions of the plants. These results suggest that the water fractions of the plants in extraction system of temperature $60^{\circ}C$ and ultrasonification 40 kHz have marked useful immuno-stimulatory activities.

Antimutagenic Effects and Compounds Identified from Hexane Fraction of Persimmon Leaves (감잎 핵산획분의 항돌연변이 효과와 항돌연변이 물질의 GC-MS를 이용한 동정)

  • Methanol extract of dried persimmon leaves was fractionated to hexane, chloroform, ethyl acetate, butanol, and aqueous tractions. Hexane, butanol, and aqueous fractions had high yields of extracts. Hexane fraction among these fractions showed the highest inhibition rate on the mutagenicities of aflatoxin (AFB$_1$), dimethyl-amino-bi-phenyl (DMAB), N-methyl-N'-nitro-N-nitrosoguanidine (MNNG), and 4-nitroquinoline-1-oxide (4-NQO) in Salmonella typhimurium TA100. Hexane fraction was further fractionated into eight fractions by silica gel column c-hromatography and thin layer chromatography (TLC). The fraction 5 on TLC exhibited the highest antimutagenic activity on AFB$_1$, DMAB, and MNNC. 1'-oxocannabinol, 3B-acetoxy-17-methyl-5a-18 (13-17) abeoardrost-13-one, 4-methoxy-2'6'-dinitro-3, 5-di-t-butylbiphenyl, 8, 9-dihydro-5, 6-dimethoxy-dibenz [c, h]isoquino [2, 1, 8-1 ma]carbazole-11, 16-dione were tentatively identified from this antimutagenic fraction by GC-MS.

  • Poly(ethylene 2,6-naphthalate), PEN, is a relatively well-known polymer used for engineering purposes. Naphthalene ring provides rigidity to the polymer backbone, thus, it elevated the glass transition temperature and enhanced mechanical properties. The structure and properties of PEN affect a processing conditions severely, and the high-thermal stability have been had a poor thermal processibility. Hence, the basic mechanism of solvent drawing, is very much the same as that of thermal drawing from glassy state since both involve the inducement of segmental mobility. The former achieves the goal by use of chemical energy, and the latter does so by use of thermal energy. Generally, the sorption of the solvent by the polymer has a plasticizing effect, and leads to a lowering of the glass transition temperature, $T_g$. In this paper, the dynamic viscoelasticity behavior in liquid-drawing process of an unoriented amorphous PEN films were investigated using Rheovibron. The results are as follows : (1) For the drawing in silicone oil, the drawing below $T_g$. had $\alpha{2}$-dispersion due to an inhomogeneous taut structure. (2) For the drawing in water, the inhomogeneous taut structure reduced by the effect of plasticization even below $T_g$. (3) For the drawing in butanol, the only aliphatic segment in PEN have some molecular mobility but the mobility of the aromatic segment having naphthalene ring is nearly impossible. (4) For the drawing in dioxane/water mixing solvent, the solvent effect is complementary each other and accordingly the entire molecular conformation have stable state. (5) For the drawing in dioxane/butanol mixing solvent, the inhomogeneity of the taut structure and the aromatic segment increase with increasing the temperature and this tendencies correspond with that of the draw ratio.

4-(Tert-butyl)-2,6-bis(1-phenylethyl)phenol induces pro-apoptotic activity

  • Previously, we found that KTH-13 isolated from the butanol fraction of Cordyceps bassiana (Cb-BF) displayed anti-cancer activity. To improve its antiproliferative activity and production yield, we employed a total synthetic approach and derivatized KTH-13 to obtain chemical analogs. In this study, one KTH-13 derivative, 4-(tert-butyl)-2,6-bis(1-phenylethyl)phenol (KTH-13-t-Bu), was selected to test its anti-cancer activity. KTH-13-t-Bu diminished the proliferation of C6 glioma, MDA-MB-231, LoVo, and HCT-15 cells. KTH-13-t-Bu induced morphological changes in C6 glioma cells in a dose-dependent manner. KTH-13-t-Bu also increased the level of early apoptotic cells stained with annexin V-FITC. Furthermore, KTH-13-t-Bu increased the levels of cleaved caspase-3 and -9. In contrast, KTH-13-t-Bu upregulated the levels of pro- and cleaved forms of caspase-3, -8, and -9 and Bcl- 2. Phospho-STAT3, phospho-Src, and phospho-AKT levels were also diminished by KTH13-t-Bu treatment. Therefore, these results strongly suggest that KTH-13-t-Bu can be considered a novel anti-cancer drug displaying pro-apoptotic activity.

Studies on the Effect of the Extract of Eugenia Flos on Gastritis and Gastric Lesion (급만성 위염 및 위손상에 관한 정향 추출물의 효과)

  • This study was perfonned to investigate effectiveness eness on the gastritis and gastric lesion with the methanol extract of the flower buds of Eugenio caryophyllata. The extract was fractionated with hexane, chIorofonn, ethyl acetate, butanol, followed by bioassay Oil antigastritis. The ethyl acetate and the buthanol fraction reduced significantly HCI.ethanol induced gastric lesion at the dose of 165 and 215 mg/kg, p.o., respectively. These results may indicate that remarkably.effective are ethyl acetate and butanol fractions in HCI-ethanol induced gastric lesion. Howeever, the fractions didn't exhibit any inhibition of gastric secretion and acid output. The buthanol fraction reduced significantly the acetic acid induced ulcer at a daily dose of 215 mg/Kg, p.o., given for 10 days. These result showed considerable inhibit of acetic acid induced ulcer without inhibition of indomethacin induced gastric lesion. The methanol extract showed low acute toxicity with minimum lethal dose of more than 3000 mg/kg, p.o. in mice. In conclusion, Eugenia F10s exhibited antigastric activity which might be attributable to inhibition of gastric secretion. It is indicated that activie component may be present in the buthanol fraction.

Anti-proliferation effect of Gyulyupsanbyonbang extracts on MCF-7 cells (귤엽산변방(橘葉散變方)이 유방암 세포주 MCF-7 생장 억제에 미치는 영향)

  • Purpose : This investigation was undertaken to evaluate the anti-proliferation, in hexane, chloroform, ethyl acetate, butanol and water fraction from extract of Gyulyupsanbyonbang(GYSB) using MCF-7 human breast cancer cells. Methods : GYSB was added to distilled water(1500ml) and was boiled then filtered. The residue was suspended in distilled water and extracted with hexane, chloroform, ethyl acetate, butanol and water. MCF-7 cells were cultured in RPMI1640 complex badge, NIH3T3 was cultered in 37$^{\circ}$C, 5% moisture incubator of carbon dioxide with Dulbecco's Modified Eagle Medium(DMEM) supplemented with 10% fetal bovine serum and antibiotics. Cell cytotoxicity test about cancer cell was measured used MTT assay. Results: When it synthesizes a result, hexane and butanol fraction had shown anti-proliferation effect and safety together, and those anti-proliferation effect operating selectively appeared. Ethyl acetate fraction had anti-proliferation effect however, it was not selective. The Chloroform and water soluble fraction did not almost appear anti-proliferation effect. Conclusion : I can conclude that GYSB have anti-proliferation effect and safety together on MCF-7 cells. It suggest that GYSB may be useful for brest cancer patients.

Enzymatic Glycosylation of Acrylic Acid and Methacrylic Acid (아크릴산과 메타크릴산의 효소적 배당화)

  • Glyco-acrylate and methacrylate were synthesized by lipase-catalyzed glycosylation of acrylic acid, methacrylic acid and their vinyl esters with $\beta$-methyl fructoside and glycerol in t-butanol as a reaction medium. At the optimum conditions for enzymatic glycosylation of acrylic acid and vinyl methacrylate, we attained up to 80% conversion for glyco-acrylate from acrylic acid and 90% conversion for glyco-methacrylate from vinyl methacrylate. The polymerizable glyco-acrylates and methacrylate have biomedical application as hydrophilic monomers and hydration modifiers to be use for hydrogel contact lens formulation.

The study on collagen sysnthesis and collagenase inhibition assay in natural plants

  • Type I (collagen) and procollagen are reduced in aged human skin. This reduction could result from increased degration by metalloproteinases and from reduced procollagen synthesis and skin collagenase is required for initiation of the degration of type I collagen. In the present study, we study on assay the collagen and collagenase in natural plants using the fibroblast human skin cell. We select the 15 kind of plants used to herbal and 4 kind of fraction(by methylene chloride, ethyl acetate, n-butanol, water). Among these extract, the ethyl acetate fraction from benincasa hispida/prunus persica, trichosanthes kiriowii, trogopterus xanthipes and methylene chloride fractions from benincasa hispida/prunus persica, torilis japonica and n-butanol fraction from cnidium officinale, chrysanthemum sibiricum were selected for further experiments as they exhibited distinctive amount of collagen compared to other natural extracts. These extracts were again subjected to collagenase assay test. Benincasa hispida/prunus persica extract was shown to have exellent collagen synthesis activity from result of the collagen assay test and the other extract was shown to have over 130% of collagen synthesis activity. But, in the study of collagenase assay test just only trogopterus xanthipes extract was shown to have collagenase inhibition.

  • Objecgtive : The aim of present study was to investigate inhibition effect of Whakijogyung-Tang(WJT) on the tumor cell lines. This study estimated the cytotoxicity of WJT about viability of S-180 and NlH3T3. Methods : The cytotoxicity of WJT about viability of cells were tested using a colorimetric tetrazoliun assay(MTT assay) Results and Conclusion : 1. Water extract of WJT had $IC_{50}$ of 863 ${\mu}g/ml$ in S-180 cell lines, but cytotoxicity of NIH3T3 was not significant difference compare with S-180. 2. n-Hexane fraction of WJT had similar cytotoxicity between S-180 and NIH3T3, but that could not have $IC_{50}$ in S-180 cell lines. 3. Ethyl acetate fraction of WJT had low degree cytotoxicity both S-180 and NIH3T3 cell lines. 4. Significantly, Butanol fraction of WJT had differenct citotoxicity between S-180 and NIH3T3. 5. $H_2O_2$ fraction of WJT had no cytotoxicity both S-180 and NIH3T3.

Conservation of Excavated Lacquer-wares for using artificially water-soaked Lacquer-wares (인공수침 칠기를 이용한 고대칠기 보존연구)

  • Among the treatment results of test samples of the antique lacquer-ware, the treatment with PEG#3,350 40% solution displayed excellent effect with low shrinkage ratio; in weight gain the treatment with Sucrose 19%+Glycerin 1%(t-butanol 5% in water) solution showed consistent increase. However during the impregnation process of Sucrose, the weight of the testing samples decreased by dehydration because the inner part of the test samples and the treatment solution showed concentration gradient. Therefore, we concluded longer impregnation period should be necessary to prevent dehydration. Since both higher and lower molecular weight treatment chemicals could penetrate into the wood of the lacquer-ware, air drying and conditioning after impregnation treatment with high concentration chemicals would be possible, as well as vacuum freeze-drying.
