Black Rice (Oryza sativa L. var. japonica) Hydrolyzed Peptides Induce Expression of Hyaluronan Synthase 2 Gene in HaCaT Keratinocytes

  • Black rice (Oryza sativa L. var. japonica) has been used in folk medicine in Asia. To understand the effects of black rice hydrolyzed peptides (BRP) from germinated black rice, we assessed the expression levels of about 20,000 transcripts in BRP-treated HaCaT keratinocytes using human 1A oligo microarray analysis. As a result, the BRP treatment showed a differential expression ratio of more than 2-fold: 745 were activated and 1,011 were repressed. One of the most interesting findings was a 2-fold increase in hyaluronan synthase 2 (HAS2) gene expression by BRP. Semiquantitative RT-PCR showed that BRP increased HAS2 mRNA in dose-dependent manners. ELISA showed that BRP effectively increased hyaluronan (HA) production in HaCaT keratinocytes.

Effective 3-D FEM for large-scale high temperature superconducting racetrack coil

  • In various types of large-scale electrical applications, the number of coil turns in such machines is usually large. Electromagnetic simulation of large-scale superconducting coils (tens to hundreds of turns) is indispensable in the design process of superconducting electrical equipment. However, due to the large scale of the coil and the large aspect ratio of super-conducting material layer in HTS coated conductor, it is usually difficult or even unable to perform 3-D transient electromagnetic simulation. This paper introduces an effective 3-D electromagnetic simulation method for large-scale HTS coated conductor coil based on T-A formulation. The simulation and experimental results show that the 3-D model based on the T-A formulation using homogeneous strategy is more accurate than the traditional 2-D models. The memory usage is not sensitive to the number of turns and this model will be even more superior as the number of turns becomes larger.

  • This research was study the accuracy of three-dimensional stereotactic breast biopsy, using a core Needle Biopsy and to assess the accuracy of Stereotactic biopsy and Sono guided biopsy. Using Stereotactic QC phantom to measure the accuracy of the 3D sterotactic machine. CT Scan and equipment obtained in the measured X, Y, Z and compares the accuracy of the length. Using Agar power phantom compare the accuracy of the 3D sterotactic machine and 2D ultrasound machine. Z axis measured by the equipment to compare the accuracy and reliability. Check the accuracy by using visual inspection and Specimen Medical application phantom. The accuracy of the 3D sterotactic machine measured by Stereotactic QC phantom was 100%. Accuracy as compared to CT, all of X, Y, Z axis is p > 0.05. The accuracy of the two devices was 100% as measured by Agar powder phantom. There was no difference between t he t wo d evices as C T and p > 0.05. 3D sterotactic machine of the ICC was 0.954, 2D ultrasound machine was 0.785. 2D ultrasound machine was different according to the inspector. Medical application phantom experiments in 3D sterotactic machine could not find the Sliced boneless ham. 2D ultrasound machine has not been able to find a small chalk powder group. The reproducibility of the three-dimensional stereotactic breast biopsy was better than effect of Sono guided biopsy.

TNF$\beta$ Induces Cytotoxicity of Antibody-Activated CD$4^+$T-lymphocytes Against Herpes Virus-Infected Target Cells

  • We have extended our previous work that cross-linking CD4 molecules using specific MAb induced antigen nonspecific, MHC unrestricted killing of virally infected target cells by CD$4^+$We have extended our previous work that cross-linking CD$4^+$ molecules using specific MAb induced antigen nonspecific, MHC unrestricted killing of virally infected target cells by CD$4^+$ T cells. The killing activity of antibody activated CD$4^+$T cells was completely blocked by herbimycin A, a protein tyrosine kinase (PTK) inhibitor, but not by bisindolylamaleimide, a protein kinase C (PKC) inhibitor. Herbimycin A treated human or bovine peripheral blood CD$4^+$T cells lacked PTK activity and failed to kill virally infected target cells even after cross-linking of CD4 molecules. The CD$4^+$cross-linking failed to induce effector cell proliferation or the transcription of TNF${\beta}$ Upregulation of TNF${\beta}$ was induced by incubating the antibody activated effector cells with BHV-1 infected D17 target cells for 10 h. Anti-TNF${\beta}$ antibody partially abolished (13-44%) the direct effector cell-mediated antiviral cytotoxicity. However, this antibody neutralized 70 to 100% of antiviral activity of effector and target cell culture supernatants against BHV-1 infected D17 cells. The inhibition level of the antiviral activity by the antibody was dependent on the effector and target cell ratio. These results support the hypothesis that increased p$56^ICK enzyme activity in effector cells transduces a signal critical for effector cell recognition of viral glycoproteins expressed on the target cells. Following target cell recognition, lytic cytokines known to participate in target cell killing were produced. A better understanding of the killing activity displayed by CD$4^+$T lymphocytes following surface receptor cross-linking will provide insight into the mechanisms of cytotoxic activity directed toward virally-infected cells.T cells. Changes in the thyroid hormone profiles in children with nephrotic syndrome

  • Purpose: We compared thyroid hormone profiles in children with nephrotic syndrome (NS) during the nephrotic phase and after remission. Methods: This study included 31 pediatric NS patients. The thyroid hormone profiles included serum levels of triiodothyronine (T3), thyroxine (T4), thyroid-stimulating hormone (TSH), and free T4. Results: Of the 31 patients, 16 (51.6%) showed abnormal thyroid hormone profiles: 6 had overt hypothyroidism, 8 had subclinical hypothyroidism, and 2 had low T3 syndrome. The mean serum T3, T4, and free T4 levels in the nephrotic phase and after remission were $82.37{\pm}23.64$ and $117.88{\pm}29.49ng/dL$, $5.47{\pm}1.14$ and $7.91{\pm}1.56{\mu}g/dL$, and $1.02{\pm}0.26$ and $1.38{\pm}0.23ng/dL$, respectively; the levels were significantly lower in the NS nephrotic phase (P=0.0007, P<0.0001, and P=0.0002). The mean serum TSH levels during the nephrotic phase and after remission were $8.05{\pm}3.53$ and $4.08{\pm}2.05{\mu}IU/mL$, respectively; they were significantly higher in the nephrotic phase (P=0.0005). The urinary protein/creatinine ratio during the nephrotic phase was significantly correlated with serum T3, T4, and free T4 levels (r=-0.5995, P=0.0032; r=-0.5797, P=0.0047; r=-0.5513, P=0.0078) as well as with TSH levels (r=0.5022, P=0.0172). A significant correlation was found between serum albumin and serum T3 levels during the nephrotic phase (r=0.5385, P=0.0018) but not between serum albumin and T4, TSH, or free T4 levels. These significant correlations all disappeared after remission. Conclusion: Abnormal thyroid hormone profile findings were observed in 51.6% of pediatric patients with NS. Thyroid hormone levels normalized after remission, regardless of levothyroxine therapy.

Effects of Substitution of Fermented King Mushroom By-Products Diet on the Growth Performance, Carcass Traits and Economics of Fattening Pigs

  • This study was conducted to investigate the effects of fermented king oyster mushroom by-products diet (FMBD) on the growth performance, blood characteristics, and carcass traits of fattening pigs and its economics. The fermented diet mainly contained 40.0% king oyster mushroom (Pleurotus eryngii) by-products, 20.0% corn, 28.0% soybean meal, 0.1% supplemental probiotics and 0.08% cellulase. The mixed ingredients were fermented for 1 d at $37^{\circ}C$ followed by 9 d at room temperature, after which they were dried at $45^{\circ}C$ for 3 d. Pigs (n=96) were divided into eight heads per pen, four diet treatments and three replications. The basal diet (C) was substituted with 20% (T1), 50% (T2) and 80% (T3) FMBD. The concentration of crude protein (CP) was significantly higher (p<0.05) at the end of the fermentation period. The average daily gain (ADG) and feed efficiency were significantly lower (p<0.05) in T3 than C. Additionally, carcass grade was significantly better (p<0.05) in all treatments than C and the ratio of high carcass grade was higher. Although substitution of FMBD decreased growth performance, it improved carcass grade and decreased the feed cost of fattening pigs. Therefore, it was expected that the increase in the utilization ratio of FMBD will reduce the cost of animal production.

Effects of Partial Replacement of Corn Grain and Soybean Meal with Agricultural By-Product Feeds on In Vitro Rumen Fermentation Characteristics and Optimum Levels of Mixing Ratio (농산부산물을 이용한 In Vitro 반추위발효 특성 및 적정 배합수준을 통한 옥수수 및 대두박 대체 효과)

  • This study was conducted to determine the effects of partial replacement of corn grain and soybean meal with agricultural by-product feeds on in vitro rumen fermentation characteristics and optimum levels of mixing ratio. The agricultural by-products to examine the effectiveness of the partial replacement of concentrate were wheat bran, corn gluten feed, bakery waste, soybean curd, rice bran, green kernel rice, soybean hull, distillers' grain, and mushroom substrate. In the first experiment, in vitro ruminal fermentation characteristics of feedstuffs were evaluated at 0, 3, 6, 12, 24, and 48 hours after incubation. In the second experiment, fermentation characteristics were investigated with green kernel rice and soybean curd which replaced corn grain or soybean meal. Feed were formulated with 40% corn grain + 20% soybean meal (T1), 40% corn grain + 17.5% soybean meal + 2.5% soybean curd (T2), 25% corn grain + 20% soybean meal + 15% green kernel rice (T3), and 30% corn grain + 15% soybean meal + 6% green kernel rice + 9% soybean curd (T4), respectively, with forage source of 10% alfalfa hay, 20% timothy hay, and 10% corn silage as fed-basis. In 24 and 48 hour cultivations, T4 showed significantly lower pH compared to T1, whereas in 3 and 24 hour cultivations, T4 showed significantly higher DM degradation compared to T1. In addition, the gas production of T3 was also higher than T1 (p<0.05). Overall results of the present experiments indicated that green kernel rice and soybean curd as agricultural by-products have the possibility of partial replacements of corn grain and soybean meal.

The Physical Properties of Super Bulky Yarn According to Textured Condition (Super Bulky Yarn의 사가공 조건에 따른 물성변화)

  • In this study, physical properties were studied by using latent stretching yarn in order to develop the texturing yarn technique for super bulky yarn, which is better in bulkiness and handle than natural wool and also adds property of synthetic fiber to natural wool. In order to obtain textured conditions by analysing basic properties for manufacturing DTY yarn with super bulky property, DTY 50d/12 after spinning latent yarn spined POY 80d/12 was obtained under the two conditions of (i) false twist(T/M) level 3 in DTY texturing and (ii) draw ratio level 4 in draw texturing. For DTY texturing yarn, Elongation rate increased as the heat treatment time and temperatures increased. In addition, shrinkage became higher as false twist was higher, so that elongation rate became lower. When annealing became longer in time and higher in temperature, initial modulus increased. In addition, as the count of false twist increased, the initial modulus showed higher values. For draw texturing yarn, under the conditions of heat temperature 180 and heating time 30 minutes, shrinkage rate in draw ratio 1.55 and 1.6 draw ratio was 7%, and that in 1.65 and 1.7 draw ratio was 8.5%. High draw ratio samples' tenacity was much influenced by heating time and temperature, but low draw ratio samples' tenacity was influenced not by treated time, but by treated temperature.

Real Time Scale Measurement of Inorganic Phosphate Release by Fluorophore Labeled Phosphate Binding Protein (형광단이 붙어 있는 인산결합 단백질에 의한 인산 배출의 실시간 측정)

  • Fluorescence change of coumarin labeled phosphate binding protein (PBP-MDCC) was monitored to measure the amount of released inorganic phosphate ($P_{i}$) during nucleoside triphosphate (NTP) hydrolysis reaction. After purification of PBP-MDCC, fluorescence emission spectra showed that fluorescence responded linearly to $P_{i}$ up to about 0.7 molar ratio of $P_{i}$ to protein. The correlation of fluorescence signal and $P_{i}$ standard was measured to obtain [$P_{i}$] - fluorescence intensity standard curve on the stopped-flow instrument. When T7 bacteriophage helicase, double-stranded DNA unwinding enzyme using dTTP hydrolysis as an energy source, reacted with dTTP, the change of fluorescence was able to be converted to the amount of released $P_{i}$ by the $P_{i}$ standard curve. $P_{i}$ release results showed that single-stranded Ml3 DNA stimulated dTTP hydrolysis reaction several folds by T7 helicase. Instead of end point assay in NTP hydrolysis reaction, real time $P_{i}$-release assay by PBP-MDCC was proven to be very easy and convenient to measure released $P_{i}$.

Channel Estimation in E-OCR for T-DMB System (T-DMB 시스템을 위한 E-OCR에서의 채널추정)

  • This paper proposes noise reduction methods to improve the channel estimation performance in Equalization On-Channel Repeater (E-OCR) for Terrestrial-Digital Multimedia Broadcasting (T-DMB) system. In the E-OCR for T-DMB system, the channel estimation is performed by the pilot signal extraction and the channel coefficient estimation. Since the performance of the channel estimation highly depends on the estimated channel coefficients whose accuracy is associated with the received signal-to-noise ratio (SNR), it is important to reduce noise existing at the estimated channel coefficients. To reduce such noise components, various noise reduction methods of the estimated channel coefficients based on the M -point weighted moving average are proposed. Computer simulations show that the proposed methods improve the estimation performance by more than 2 to 3 dB in terms of the symbol error rate after equalization.
