Genomewide Profiling of Rapamycin Sensitivity in Saccharomyces cerevisiae on Synthetic Medium

  • The target of rapamycin (TOR) signaling pathway is a conserved pathway that regulates eukaryotic cell growth in response to environmental cues. Chemical genomic approaches that profile rapamycin sensitivity of yeast deletion strains have given insights into the function of TOR signaling pathway. In the present study, we analyzed the rapamycin sensitivity of yeast deletion library strains on synthetic medium. As a result, we identified 130 strains that are hypersensitive or resistant to rapamycin compared with wild-type cells. Among them, 36 genes are newly identified to be related to rapamycin sensitivity. Moreover, we found 16 strains that show alteration in rapamycin sensitivity between complex and synthetic media. We suggest that these genes may be involved in part of TOR signaling activities that is differentially regulated by media composition.

  • Three-component coupling of thiourea/urea, various structurally diverse aromatic aldehydes and ammonium acetate is catalyzed by activated fly ash in dry media under microwave irradiation to give 6-aryl-1,2,4,5-tetrazinan- 3-thiones/ones in good yields. The structure of 6-aryl-1,2,4,5-tetrazinan-3-thiones/ones have been elucidated on the basis of their melting points, elemental analysis, MS, IR, 1H NMR, D2O exchange, 13C NMR and two dimensional NMR spectral studies including Homonuclear Correlation (HOMOCOR) and Heteronuclear Single Quantum Correlation (HSQC) spectra.

Development of a New Process for PVA Degradation in Desize Wastewater by lmmobilized Symbiotic Bacteria (고정화 공생 Bacteria를 이용한 호발폐수 중 PVA분해 신공정 개발)

  • A new process was developed to degrade PVA(polyvinyl alcohol) in desize wastewater. Two symbiotic bacteria of Pseudomonas strain G5Y and PW were immobilized on the media by adsorption. A natural zeolite was chosen as the best media considering cell adhesion capacity, sedimentation rate, and material cost. PVA and COD removal efficiencies of this system for synthetic wastewater were 84% and 85% at the retention time of 6 hr, when the volumetric loading rate was PVA 8g/L·day and COD 8g/L·day, and cell density was 19,775 mg/L. In case of desire wastewater, they were 78% and 72% at the retention time of 6 hr, respectively, when the volumetric loading rate was PVA 8g/L·day and COD 13.2g/L·day, and cell density was 32,899mg/L. In case of desize wastewater, PVA and COD removal efficiencies were lower than synthetic wastewater, but cell density of the desize wastewater was lower than that of the synthetic wastewater, because there were insufficiency of necessary nutrition and variety of desize materials in the desize wastewater. A pilot test was successfully performed showing 88% and 82%, PVA and COD removal efficiencies at the retention time of 24 hr, when volumetric loading rate were 4.7 PVA g/L·day and COD 6.9g/L·day, and cell density was 12,,324 mg/L.

Seeded Crystal Growth onto Enamel Mineral and Synthetic Hydroxyapatite in Dilute Supersaturated Solutions Containing Low Concentrations of Fluoride (불소농도가 Seeded Enamel Mineral과 합성 Hyproxyapatite에 Crystal 성장에 미치는 영향)

  • The present study was undertaken to investigate the crystal growth onto enamel mineral and synthetic hydroxyapatite seeds in media resembling the enamel fluid composition. Effects of fluoride at low concentrations on the precipitation were also examined in a benchtop crystal growth model adopting a miniaturized reaction column. X-ray diffraction and Fourier transform infrared spectroscopy(FTIR), as well as chemical analyses, were employed for characterization of both seed materials before and after experimentation. Remarkable findings were that (1) both biological and synthetic seeds at the same total surface areas yielded rather similar precipitation rates at all levels of fluoride concentration in solution and (2) the precipitation rate was accelerated in a manner depending on fluoride concentrations in media. FTIR differential analysis disclosed that the precipitating phase was characterized as poorly crystallized apatite, which incorporated subtle carbonate. Most of the fluoride ions in soution were readily incorporated into crystals. The overall results support the view that the seeded crystal growth model is of value to gain insight into the mechanism of enamel crystal growth under fluoride regimens.

Proposal for License Plate Recognition Using Synthetic Data and Vehicle Type Recognition System (가상 데이터를 활용한 번호판 문자 인식 및 차종 인식 시스템 제안)

  • In this paper, a vehicle type recognition system using deep learning and a license plate recognition system are proposed. In the existing system, the number plate area extraction through image processing and the character recognition method using DNN were used. These systems have the problem of declining recognition rates as the environment changes. Therefore, the proposed system used the one-stage object detection method YOLO v3, focusing on real-time detection and decreasing accuracy due to environmental changes, enabling real-time vehicle type and license plate character recognition with one RGB camera. Training data consists of actual data for vehicle type recognition and license plate area detection, and synthetic data for license plate character recognition. The accuracy of each module was 96.39% for detection of car model, 99.94% for detection of license plates, and 79.06% for recognition of license plates. In addition, accuracy was measured using YOLO v3 tiny, a lightweight network of YOLO v3.

  • Penicillium sp I which produces a powerful hydrolysing enzyme was isolated from putrefid and dry Jerusalem artichoke medium. The strain was used to study on the optimum culture conditions for enzyme production. The results obtained are as follows: 1. Penicillium sp I was a vigorous strain to produce inulase. 2. The optimum culture conditions of the strain was examined in the Jerusalem artichoke extract medium and the synthetic medium. 3. Inulase productivity in the Jerusalem artichoke extract medium was higher than that of the synthetic medium. 4. The optimum culture period of the Jerusalem artichoke extract medium was four days, whereas that of the synthetic medium was five days. 5. The optimum temperature, pH and concentration in the Jerusalem artichoke extract medium were $30^{\circ}C$, 5.0 and 4.0% (W/V), respectively. Meanwhile, the optimum temperature, pH and concentration in the synthetic medium were $30{\sim}33^{\circ}C$, $5.0{\sim}6.0$, and $1.0{\sim}1.5%$ (W/V), respectively. 6. Corn steep liquor, peptone, $(NH_4)_2HPO_4,\;NH_4H_2PO_4,\;(NH_4)_2SO_4$, etc. were favorable as nitrogen sources. Of these, especially, Corn steep liquor and peptone as organic nitrogen sources caused an increase in inulase production in the synthetic medium. 7. All sugars except for inulin have no effect upon the inulase production. 8. KCl, $MgSO_4\;and\;FeSO_4$ were favourable mineral sources for inulase production.

Effect of Media Packing Ratio on the Sequencing Batch Biofilm Reator (연속회분식 생물막 반응기에서 여재 충진율의 영향)

  • This study was carried out to get more operational characteristics of the sequencing batch biofilm reactors with media volume/reactor volume ratio of 15 %, 25 % and 35 %. Experiments were conducted to find the effects of the media packing ratio on organic matters and nutrients removal. Three laboratory scale reactors were fed with synthetic wastewater. During studies, the operation mode was fixed. The organic removal efficiency didn't show large difference among three reactor of different packing media ratios. However, from the study results, the optimum packing media ratios for biological nutrient removal was shown as 25%. The denitrifying PAOs could take up and store phosphate using nitrate as electron acceptor.

Statistical Selection of Amino Acids Fortifying a Minimal Defined Medium for a High-level Production of the Kringle Fragments of Human Apolipoprotein(a)

  • A synthetic defined medium, fortified with amino acids, was developed for the stable production of the kringle fragments of human apolipoprotein(a) (apo(a)), rhLK68. Using a complex rich medium containing yeast extract and a high-cell-density fed-batch culture, the expression level of rhLK68 reached 17% of the total cellular protein, which corresponded to $5\;g\;l^{-1}$ of the culture. To replace the complex media with chemically defined media, several amino acids that positively affect cell growth and gene expression were chosen by a statistical method. The various combinations of the selected amino acids were tested for its fortifying effect on a minimal defined medium. When glutamine only was added, the overall expression level of rhLK68 reached 93% of the complex rich medium increasing the specific expression level by 22.4% and decreasing the cell growth by 24%. Moreover, the addition of glutamine resulted in a 2-fold increase in the concentration of rhLK68 in the culture broth, compared with the minimal defined medium. The synthetic defined media developed in this study could be generally applied to high-cell-density cultures of the recombinant Escherichia coli BL21(DE3), especially for the production of therapeutic proteins that require a strict quality control of the culture media and fermentation processes.

  • The application of microwave technology to synthesis of polymer in solvent media has been shown by the synthesis of copper phthalocyanine (CUPc). The increase of synthetic yield-was demonstrated successfully in this study. A power variable microwave synthetic system has been developed with modifying cavity of domestic microwave oven and attaching microwave power controller. The properties of the specimen synthesized at various condition under the conventional thermal processing and microwave processing has been characterized by the means of chemical analysis, X-ray diffractometry (XRD), scanning electron microscopy (SEM), and particle size analysis (PSA).
