Effect of Korean Red Ginseng Crude Drug-Combined Preparations (RGCDPs) on Memory Enhancement in Mice

  • Anti-amnestic activities of Korean red ginseng (Ginseng Radix Rubra) and Crude drug-combined preparations (RGCDP-1, RGCDP-2, and RGCDP-3) were evaluated by the animal experiment. RGCDP-1 and RGCDP-2 were prepared based on Korean folk prescriptions, 'Chongmyongtang' and 'Guibitang', respectively, while RGCDP-3, by a combination of both. Among the three preparations, RGCDP-3 was found to show the most potent anti-amnestic activity as evaluated by the passive avoidance test with mice, indicating synergistic action by combined effect of RGCDP-1 and RGCDP-2.

In vitro efficacy of formalin, hydrogen peroxide and copper sulfate on the scuticocilliate Uronema marinum at low salinity

  • The scuticocilliate, Uronema marinum is a histophagous ciliate and the causative agent of 'scuticociliatosis'in cultured olive flounder Paralichthys olivaceus. In the present study, in vitro efficacy of hydrogen peroxide, formalin and copper sulfate on the scuticocilliate at low salinity was investigated. Each chemical showed synergistic parasiticidal effects with low salinity (salinity in 5 ppt) compared to each chemical alone (salinity in 33 ppt). At low salinity (5‰), ciliates were killed completely within 1.5h by exposure to 50ppm formalin (37% formaldehyde), at 100ppm hydrogen peroxide (30% solution) and at 100ppm copper sulfate (20% solution). The formalin was the most effective chemical against the parasites at low salinity.

  • Dially disulfide (DADS), a component of garlic (Allium sativum), has been known to exert potent chemopreventive activity against various cancers. In this study, the synergistic effect of DADS and daunorubicin on the cytotoxicity of HL-60 cells, a human leukemia cell line, was investigated. DADS at 25 M greatly potentiated daunorubicin-induced cell death, decreasing cell viabilityto50%ofthe control. Daunorubicin-induced apoptosis was accompanied by the activation of caspase-3, the degradation of poly-(ADP-ribose) polymerase (PARP) and D4-GDI, and DNA fragmentation, which were blocked by pre-treatment with acetyl-Asp-Glu-Val-Asp- dialdehyde (Ac-DEVD-CHO). Treatment that combined 25 M DADS and 100 nM daunorubicin caused a similar degree of caspase-3 activation, PARP and D4-GDI degradation, and DNA fragmentation to that caused by treatment with 250 nM daunorubicin alone. These results indicate that combined therapy using daunorubicin with DADS, a component of food, and garlic can effectively decrease the therapeutic dose of daunorubicin, preventing the severe side effects of daunorubicin.

  • The soot formation characteristics have been studied experimentally in concentric co-flow ethylene/propane diffusion flames. Comparing to the homogeneously mixed propane/ethylene case, the increase of soot formation is observed when propane is supplied through the outer nozzle, while the decrease is observed when propane is supplied through the inner nozzle. The reaction path of PAHs formed from the pyrolysis process of propane is likely to be responsible to the observed difference. When propane is supplied through the outer nozzle, PAHs formed during the combustion process are easy to be exposed to the oxidization environment; however, when propane is supplied through the inner nozzle, PAHs are not likely to be oxidized and thus get involved in soot formation process. The synergistic effect in ethylene/propane diffusion flames is affected not only by the composition of mixture but also by the way of mixing.

  • Isoguanosine and berberine 1:1 mixture[l:[1(mole:mole)] has been prepared and evaluated by measuring antitumor effects against various tumor cell lines in culture and in mice. We reported that the synergistic effect of isoguanosine and berberine mixture has been revealed compared with each of isoguanosine and berberine increased by $3{\sim}8$ times than that of each components in various tumor cell lines in vitro. The most effective dose of isoguanosine and berberine mixture was 60 mg/kg/day in mice bearing S-180 solid tumor, the %(1-T/C) values were 70%. Against the P-388 leukemia, isoguanosine-berberine mixture was the most effective at the dose of 60 mg/kg/day, the %T/C values were 163%.

  • Observations were made on Crinum and Catharanthus pollen growth in artificial media by an ultraviolet transmission fluorescence microscope showing synergistic effect on pollen growth with calcium (Ca) and aureomycin. Bright yellow fluorescence of aureomycin enabling to trace out at tissue or cellular level did reveal that the greater accumulation of fluorescence occurred in the pollen tube wall if Ca was supplemented to the media than when aureomycin alone was present. The promotive pollen growth the media of Ca alone was further enhanced by the addtion of aureomycin. It was assumed that the promoted pollen growth with aureomycin in the Ca media was probably brought about by a supporting role of aureomycin in the Ca action.

Synergistic Antimicrobial Action of Thymol and Sodium Bisulfate against Burkholderia cepacia and Xanthomonas maltophilia Isolated from the Space Shuttle Water System

  • A combination of thymol and sodium bisulfate was found to be an effective biocidal agent against strains of Burkholderia cepacia and of Xanthomonas maltophilia that were found in the space shuttle water system. Potassium iodide (KI), the biocidal agent used in the past, had a minimum inhibitory concentration (MIC) of 50,000 ppm against the two B. cepacia (541 STS-81 and 1119 STS-91) strains, whereas that of thymol and sodium bisulfate was 2,400 and 950 ppm, which was 21 and 53 times lower than that of KI for B. cepacia, respectively. The MIC value for the combination of thymol and sodium bisulfate was 4 times lower than that for thymol or sodium bisulfate alone against B. cepacia (541 STS-81, 1119 STS-91) or Pseudomonas cepacia (ATCC 31941). The fractional inhibitory concentration (FIC) of the combination of thymol and sodium bisulfate for all organisms tested was less than 0.5, indicating a strong synergistic effect.

  • Synergistic effect was observed in the extraction of trivalent lanthanides by the mixed solvent of TBPO (tri-n-butyl phosphine oxide) and TOPO (tri-n-octyl phosphine oxide) in toluene diluent. The reason of the enhancement was verified as mainly due to the formation of new extractable species $M(NO_3)_3(TBPO)_2\;(TOPO)$ besides of the formation of $M(NO_3)_3(TBPO)_3$and $M(NO_3)_3(TOPO)_3$.

  • The effect of garlic extract and vitamin C mixture on the various cancer cell lines in vitro and in vivo have been examined. Proliferation of human colon cancer (HT-29), human rectal cancer (HRT-18) and human hepatoma (HepG2) cells was inhibited by garlic extract and vitamin C, respectively. Based on the cytotoxic activity, mixture of garlic extract and vitamin C was demonstrated to possess a synergistic growth inhibition on HT-29, HRT-18 and HepG2 cancer cells. Mixture of garlic extract and vitamin C significantly arrested G2/M phase cells in the HepG2 cell cycle. Oral administration of mixture of garlic extract and vitamin C to sarcoma-180 tumor-bearing mice prolonged survival time compared to that of control group. These results suggested that addition of vitamin C enhances anticancer activity of garlic extract in vitro, and mixture of garlic extract and vitamin C has antitumor effect in vivo.

  • 1-Acetyl-$\beta$-carboline was isolated as an anti-MRSA agent from the fermentation broth of a marine actinomycete isolated from marine sediment. The producing strain was identified to be Streptomyces sp. by phylogenetic analysis of the 16S rRNA gene sequence. The anti-MRSA agent was isolated by bioactivity-guided fractionation of the culture extract by solvent partitioning, ODS open flash chromatography, and purification with a reversed-phase HPLC. Its structure was elucidated by extensive 2D NMR and mass spectral analyses. Combination of 1-acetyl-$\beta$-carboline with ampicillin exhibited synergistic antibacterial activity against MRSA.
