Morphological Study of the Digestive Tract of the Mud Crab (Hemigrapsus Penicillatus De Haan) and the Symbiotic Crab (Pinnotheres cyclinus Shen)

  • The influence of eating habits and food type on the ultrastructural characteristics of the digestive tracts was studied under the scanning and transmission electron microscopes in two crustacean decapods (Hemigrapsus penicillatus De Haan; mud crab, Pinnotheres cyclinus Shen; symbiotic crab). The relative ratio of the length of midgut versus hindgut was 1:1 in the mud crab, but 4:1 in the symbiotic crab. Observation through the scanning electron microscope revealed that the midguts of both species have densely-arranged longitudinal mucosal folds with a smooth surface. In the hindgut of the mud crab, mucosal folds were longitudinally oriented, clusters of two to five spines were observed on the cuticular surface, and the length of the spine in the distal hindgut was longer than that in the proximal portion. In the symbiotic crab, the mucosal folds were irregulary arranged, and numerous rudimentary spinal structures were noted on the cuticular surface. Through observation of a transmission electron microscope, the epithelial cells of the midgut in both species had numerous microvilli, but the length of the microvilli was slightly longer in the mud crab than in the symbiotic crab. The central layer of the basement membrane and the muscular layer of the midgut were more developed in the mud crab than in the symbiotic crab. The thickness of the cuticular layer over the hindgut surface in the mud crab was about 4 times than that of the symbiotic crab.

  • Sponges are host organisms for various symbiotic microorganisms such as archaea, bacteria, cyano-bacteria and microalgae. Sponges are also sources of a wide variety of useful natural products like cyto-toxins. antifouling agents, antibiotics, and anti-inflammatory and antiviral compounds, Symbiotic microorganisms is sponges can be sources of various natural products, because metabolites previously ascribed to sponges have recently been demonstrated to be biosynthesized by symbionts. If a symbiotic microorganisms from which some natural products are derived can be cultured, the microorganism could be used in a mass production of the bioactive comopounds. We summarize recent research on iso-lation and cultivation of sponge-symbiotic microorganisms and the symbiotic relationship.

A Comparative Study on the Nitrogen Metabolism of Symbiotic Chlorella from Paramecium bursaria with Chlorella ellipsoidea (Paramecium bursaria와 공생하는 Chlorella와 Chlorella ellipsoidea의 질소대사에 관한 비교연구)

  • The excretion of ammonia and glutamine synthetase activities were measured in aposymbiotic Paramecium and symbiotic Paramecium. The uptake of nitrate and ammonia, and specific enzyme activities of nitrate reductase, glutamate dehydrogenase and glutamine synthetase were investigated in symbiotic Chlorella from Paramecium bursaria and Chlorella ellipsoidea. The ammonia concentration in the culture media of aposymbiotic Paramecium was increased according to the growth of the Paramecium but it was not changed in symbiotic Paramecium. Nitrate, the major nitrogen source, was taken up at a rate of 0.635 nmol/ 106 Chlorella/hr in Chlorella ellipsoidea. Most of ammonia was assimilated to glutamine by glutamine synthetase, of which acitivty was 1,467 $\mu$mol/mg protein/min in Chlorella elliposidea. Contrary to Chlorella ellipsoidea, ammonia and glutamine transported from the Paramecium were the nitrogen source of symbiotic Chlorella and ammonia was taken up at a rate of 3.854 nmo./106 Chlorella/hr into synmbiotic Chlorella. Most of ammonia were assimilated to glutamate by glutamate dehydrogenase in symbiotic Chlorella. The glutamate dehydrogenase (GDH/NADH) activity was 0.851 $\mu$mol/mg protein/min.

  • Symbiotic evolutionary algorithms are stochastic search algorithms that imitate the biological coevolution process through symbiotic interactions. In the algorithms, the fitness evaluation of an individual requires first selecting symbiotic partners of the individual. The symbiotic partner effects the change of individual's fitness and search direction. In this study we are to analyze how much partnering strategies can influence the performance of the algorithms. For this goal extensive experiments are carried out to compare the performance of partnering strategies. The NKC model and the binary string covering problem are used as the test-bed problems. The experimental results indicate that there does not exist statistically significant difference in their performance.

EFFECTS OF COLLISIONAL DE-EXCITATION ON THE RESONANCE DOUBLET FLUX RATIOS IN SYMBIOTIC STARS AND PLANETARY NEBULAE

  • Resonance doublets including O VI 1032, 1038, NV 1239, 1243 and C IV 1548, 1551 constitute prominent emission lines in symbiotic stars and planetary nebulae. Spectroscopic studies of symbiotic stars and planetary nebulae from UV space telescopes show various line ratios of these doublets deviating from the theoretical ratio of 2:1. Using a Monte Carlo technique, we investigate the collisional de-excitation effect in these emission nebulae. We consider an emission nebula around the hot component of a symbiotic star characterized by the collisional de-excitation probability $p_{coll}\;{\sim}\;10^{-3}\;-\;10^{-4}$ per each resonance scattering, and the line center optical depths for major resonance doublets in the range ${\tau}_0\;{\sim}\;10^2\;-\;10^5$. We find that various line ratios are obtained when the product $p_{coll}{\tau}_0$ is of order unity. Our Monte Carlo calculations show that the flux ratio can be approximately fitted by a linear function of ${\log}{\tau}_0$ when ${\tau}_0p_{coll}\;{\sim}\;1$. It is briefly discussed that this corresponds to the range relevant to the emission nebulae of symbiotic stars.

Population, Symbiotic Effectiveness, and Protein Profile Patterns of Indigenous Rhizobium leguminosarum biovar viciae to Korean Soils

  • Some symbiotic characteristics of native Korean Rhizobium leguminosarum biovar viciae were analysed to get some informations desirable for cultivation of hairy vetch (Vicia villosa Roth) using its symbiont in Korea. The size of indigenous populations of R. leguminosarum biovar viciae was higher in seven upland soils showing $1.7{\times}10^2{\sim}5.8{\times}10^4cells\;g{\cdot}soil^{-1}$, which appeared to be 10% and 37% higher for cultivated and uncultivated soils of hairy vetch, respectively, than seven paddy soils with $1.7{\times}10^2{\sim}1.7{\times}10^4cells\;g{\cdot}soil^{-1}$. In symbiotic potentials, however, the yields of hairy vetch treated with 10-fold-diluted ($10^{-1}$) inoculum and 1000-fold-diluted ($10^{-3}$) one was 11.2% and 8.8% more, respectively, in paddy than upland. Hairy vetch inoculated with either strain KHR 106 from Sacheon or strain KHR 120 from Yesan among native Korean R. leguminosarum biovar viciae isolates was of similar yield increment of 16% (p < 0.05) in upland soils with native R. leguminosarum biovar viciae of $5.8{\times}10cells\;g{\cdot}soil^{-1}$. In case of coinoculation of the two strains, however, the yields was not significantly increased. In especial, isolate KHR 106, KHR 120, and KHR 122 from Suwon, which has also good symbiotic effectiveness, showed different protein profile patterns each other. As a result, hairy vetch is possibly able to use atmospheric nitrogen through symbiotic relationship with diverse native R. leguminosarum biovar viciae in Korean arable lands. For safe and good production of hairy, however, the use of superior strains with high symbiotic effectiveness and competitiveness will be desirable.

Enzymatic Characteristics for Xylanase Activity of Tremella fuciformis and its Symbiotic Fungi (흰목이버섯 및 공생균이 분비하는 Xylanase 효소적 특성)

  • Effects of cultural conditions on the production of xylanase by Tremella fuciformis, symbiotic fungi and mixed fungi were investigated. The optimum carbon source for high production of xylanase by T. fuciformis, symbiotic fungi and mixed fungi was xylose. The optimum nitrogen source for both T. fuciformis and symbiotic fungi was $KNO_3$, whereas mixed fungi was $(NH_4)_2SO_4$. The optimum culture period for high production of xylanase was 5 days for both T. fuciformis and mixed fungi, and 6 days for symbiotic fungi, respectively. The optimum temperature for T. fuciformis and symbiotic fungi was $40^{\circ}C$, and the corresponding value for mixed fungi was $45^{\circ}C$. Xylanase activity was high at pH 6 for T. fuciformis and symbiotic fungi, and pH 7 for mixed fungi. Except $Hg^{2+}$ and $Pb^{2+}$, metal ions in T. fuciformis inhibited the activity of xylanase, and, thermal stability of xylanase in T. fuciformis, symbiotic fungi and mixed fungi maintained 80% of activity until $50^{\circ}C$. The Michaelis constant (Km) of xylan was $6.25{\times}10^{-5}\;M$ in T. fuciformis, $5.6{\times}10^{-2}\;M$ in symbiotic fungi, $5.2{\times}10^{-2}\;M$ in mixed fungi.

Ecological Study on Arbuscular Mycorrhizae(AM) at Coastal Reclaimed Lands (해안(海岸) 간척지(干拓地)에서 Arbuscular Mycorrhizae(AM)에 관한 생태학적(生態學的) 연구(硏究))

  • The symbiotic activities of arbuscular mycorrhizal fungi(AMF) such as spore density, symbiotic intensity and vesicle density, phytomasses of higher plants such as Calamagrostis epigeios, Imperata cylindria, Artemisia scoparia, Aster tripolium and Sonchus brachyotus and seasonal change of the AMF activities, electric conductivity and zinc contents in plant and soil were determined in the rhizospheres of higher plants at abandoned old coastal reclaimed lands, where constructed in 12 and 30 years ago. If plants of reclaimed land classified to salinity, symbiotic activities of AMF were high in order of obligate halophyte, facultative halophyte and glycophyte. Also, those plants classified to life form, symbiotic activities of AMF were high in order of annual, biennial and perennial plants. Seasonal variation of spore density, one of symbiotic activities showed that the plateau density maintained continuously from the end of growing season of the higher plants to next spring. For this reason, it regarded that reproduction of AMF spore would be formed in autumn, when the higher plants will be developed. Seasonal change of symbiosis intensity, other symbiotic activities, however, showed that the highest symbiosis intensity occurred in spring and summer but the lowest in autumn. In relationships among symbiotic activities, spore density was directry proportional increase of symbiosis intensity. Moreover, phytomass of higher plants also was directly proportional to increase the spore density as well as symbiosis intensity. Vesicle density, however, did not any correlation with the phytomass, spore density and symbiosis intensity. From these results, it can know that both spore density and symbiosis intensity are strongly possible to use as the measure of symbiotic activity owing to symbiosis of tho-AMF, the more absorption of zinc by the higher plants carried out the less concentration of zinc in the soil.

A Study on the Photosynthesis of Chlorella Symbiotic with Paramecium bursaria (Paramecium bursaria와 공생하는 Chlorella의광합성에 관한 연구)

  • The relationship of Paramecium bursaria with its symbiiotic Chlorella was studied in the view-point of photosynthesis and its product movement. The following results were obtained. In 1/10 dilution of bacterized medium green Paramecium grew and survived better than colorless Paramecium. P. bursaria was competitively related to its symbiotic algae as far as the change of total chlorophyll contents was concerned. Therefore, in continuous darkness total chlorophyll contents of green Paramecia decreased proportionally to the time course. Under the condition, on the other hand, that the bacterized medium was not replenished over 10 days, total chlorophyll contents in P. bursaria increased by over 2 times as much as those in normal condition. By means of Oxygen Diometer it was assured that green Paramecium was able to utilize thoitic the great part of oxygen evolved photosynthetically by its symbiotic algae. Also when was given many starch grains were found in symbiotic chlorella through electron micrography. These data suggested that P. bursaria was photosynthetically given lots of benifit by its symbiotic chlorella.

Seasonal Variation of Infection by Three Species of Symbiotic Copepods in Clam, Tresus keenae (왕우럭에 공생하는 요각류 3종에 대한 감염률의 계절 변동)

  • Seasonal variation of the infection by three species of symbiotic copepods in clam, Tresus keenae was investigated in south coast of Korea. The prevalence and abundance of three symbiotic copepods showed seasonal variation with a peak in winter. Reproduction of the copepods occurs from early autumn to spring as indicated by higher ratio of gravid copepods. The frequency distribution of Lichomolgus inflatus, Pseudomyicola spinosus and Anthessius progectus in their host population could be fitted with negative binominal distribution. Results from the present study suggest that seasonal fluctuation in prevalence, relative density and mean intensity is related to temperature and life history of the symbiotic copepods.
