돼지, 닭 및 소유래 Staphylococcus hyicus에 대한 swine phage와 chicken phage의 용균능 비교 (A comparison of lytic activity of swine and chicken phages with Staphylococcus hyicus of swine, chicken and cattle origin)

  • The lytic activity of the set of swine and chicken phages which were derived from lysogenic Staphylococcus hyicus strains of swine and chicken origin was compared by means of S. hyicus isolated from swine, chicken and cattle. Of the 80 strains each from swine and chicken, 71 (88.8%) of strains from swine and all the strains of chicken origin were found to be lysogenic. Swine phages showed wider range of lytic activity to the examined strains than that of chicken phages. Using chicken phages at $100{\times}routine$ test dilution (RTD), 25.0%, 85.6% and 50.0% of swine, chicken and bovine strains were lysed, respectively. However, when the set of swine phages was used at $100{\times}RTD$, higher frequency of the typable strains was found in strains of swine and chicken origin (73.8% and 90.2%). Phage F12 and L16 from chicken set were found to be highly active with chicken and bovine strains. On the contrary, all the swine strains were completely resistant to lysis by the two phages at $100{\times}RTD$. Thirteen (12.5%) of 104 S. aureus strains, 1 (1.8%) of 55 S. simudance strains, 31 (58.5%) of 53 S. chromo genes strains, and none of 31 strains of other coagulase-negative Staphylococcus species isolated from bovine mastitis were typable with the set of swine phages.

한국의 동물과 사람에서 분리한 Salmonella enteritidis의 phage types (Phage types of Salmonella enteritidis isolated from animal and human in Korea)

  • The present study was conducted to investigate the phage types(PT) of Salmonella enteritidis(SE) isolated from animals and human patients in Korea during the period 1993-1999. SE were isolated from chickens including the layer and broiler, duck, rat, swine and human patients in nationwide scale. A total of 231 non-human and 14 human SE strains were phage typed and 80.4%(197/245) of SE isolates tested could discriminate as a ten SEPTs including the RDNC by the 10 standard phages, which were brought from the Central Public Health Laboratory in England. In analysing of phage types both animal and human SE isolates, SEPT 4(55.4%) was the most prevalent in domestic animals including the layer(39.3%) and broiler(64.5%), duck(55.5%), and swine(75.0%) since 1993. About 57% of the S enteritidis from human was SEPT 4. Along with SEPT 4, SEPT 1, SEPT 6a, SEPT 7 and SEPT 7a var, SEPT 9b, SEPT 15, SEPT 21b and SEPT 22 were also found from animal and human SE isolates. SEPT 4(40.0%) was isolated from frozen chicken meats imported from China along with SEPT 7 and 7a var strains in this study. In conclusion, SEPT 4 was the most predominant SEPT in our country regardless of animal or human hosts including the Che-Ju island and almost exclusively found in domestic poultry(layer and broiler). During the review period, SEPT 4 has already been spreading continually among the animal hosts since 1993.

이유자돈으로부터 분리한 Lactobacillus salivarius KLW001의 유전체 분석 (Draft genome sequence of Lactobacillus salivarius KLW001 isolated from a weaning piglet)

  • 이유자돈용 생균제 개발을 위해, 본 연구자들은 유산균의 일종인 Lactobacillus salivairus KLW001 균주를 대한민국 양돈가에서 사육 중인 이유자돈으로부터 분리하였다. 이 균주는 K88 antigen-positive Escherichia coli, Salmonella enterica serovar Typhimurium에 대한 항균 활성이 타 균주보다 우수하여, 우리는 이 균주의 유전체를 분석하였다. 유전체 초안 속의 166개 Contig (${\geq}500bp$)들에서, G+C content (%)가 33.0%였고, 2,326,706 bp 크기의 염기서열을 확보할 수 있었다. 유전체 초안으로부터 항생제 저항 유전자 4개, Phage 관련 유전자 36개, Bile 대사 유전자 3개, CRISPR Spacer 27개를 확인하였다.

Inhibition of Clostridium perfringens using Bacteriophages and Bacteriocin Producing Strains

  • In this study, we isolated and characterized a bacteriocin-producing strain and two bacteriophages (P4, A3), showing antimicrobial effects against Clostridium perfringens, from chicken and swine feces by the spot-on-the lawn antagonism method. The selected strain was identified as Streptococcus hyointestinalis by 16S rRNA gene sequencing. The bacteriocin from the isolated strain exhibited strong inhibitory activity against four strains of C. perfringens and all the tested strains of Listeria monocytogenes, and the bacteriocin were highly heat- and pH-stable even at pH 2, pH 10 and $121^{\circ}C$ for 15 min. We also evaluated the combined effects of the isolated bacteriocin and phages. Combining the phage treatments and bacteriocin resulted in a synergetic effect compared with the phage or the bacteriocin alone. In addition, during the probiotic test, the bacteriocin-producing S. hyointestinalis B19 strain reduced the population of C. perfringens significantly. Treatment with S. hyointestinalis B19 and a cocktail of lytic bacteriophages eradicated the C. perfringens KCTC $3269^T$, completely. Consequently, the isolated bacteriocin and bacteriophages represent candidates for effective biocontrol of C. perfringens, and bacteriocin-producing S. hyointestinalis B19 is a potential probiotic candidate for use in domestic animals.

용균성 박테리오파지에 의한 Cronobacter sakazakii와 Salmonella enterica Typhimurium의 생육저해 (Virulent Bacteriophage for Growth Inhibition of Cronobacter sakazakii and Salmonella enterica Typhimurium)

  • 즉석 편이식품에서 위해도가 가장 큰 C. sakazakii와 S. enterica Typhimurium을 박테리오파지로 제어하기 위하여 용균성 박테리오파지를 분리, 동정하였고 조제분유와 채소 주스에 이들 세균에 적용하여 그의 효과를 분석하였다. 박테리오 파지는 돼지 분변에서 C. sakazakii와 S. enterica Typhimurium균을 용해시키는 박테리오파지를 분리하였고 현미경과 그의 특성을 분석, 동정하였다. Cronobacter에 작용하는 ES2 파지와 Salmonella의 ST2 파지는 형태학적 특성이 각각 Myoviridae와 Siphoviridae로 각각 동정되었으며 제한효소지도와 SDS-PAGE 분석에 의하여 서로 다른 파지임을 확인하였다. ES2 파지의 경우 latent period는 약 40분 정도였으며, ST2 파지는 약 30분 정도를 나타냈으며, burst size는 ES2 파지는 약 $52{\pm}5PFU/cell$, ST2 파지는 약 $21{\pm}3PFU/cell$로 나타났다. 열안정성은 $60^{\circ}C$에서 ST2 파지의 경우 100분 동안 안정한 것으로 나타났으나, ES2 파지는 30분 이후부터는 확인되지 않았다. 따라서 ST2 파지가 ES2 파지에 비해 열안정성이 높은 것을 알 수 있었다. 이 분리 파지를 조제분유와 채소 주스에 직접 적용한 효과는 ES2에 의한 Cronobacter 제어는 접종 후 6시간까지는 균수가 일정하게 유지하였고 균의 증식을 일어나지 않는 것으로 나타났다. ST2 파지에 의한 Salmonella는 생육저해가 잘 일어나 접종시간이 지남에 따라 균수가 감소하는 것을 확인하였다. 그러므로 C. sakazakii와 S. enterica Typhimurium의 생육저해는 이들 용균성 박테리오파지를 활용하여 가능한 것으로 보인다.
