석웅황의 시험관내 위암, 신경교종 및 전립선암 세포에 대한 항암 연구 (Anti-tumor effects of Realgar on Stomach Cancer Cells (AGS), Glioma Cells (T98G, A172, SNU-489) and Prostate Cancer Cells (LNCaP))

  • Objectives : The purpose of this study was to identify the anti-tumor effects of realgar on various cancer cells through molecular biologic and cellular biologic methods. Materials & Methods : We used 5 kinds of cancer cell lines:stomach cancer cell (AGS), glioma cells (T98G, A172, SNU-489) and prostate cancer cells (LNCaP). We injected the boiled extract of realgar. $50{\mu}$g/ml and $100{\mu}$g/ml to culture media (ml) for 24 hours. We examined the morphological changes under an inverted microscope and a fluorescence microscope. We measured the suppressive effect on viability of 5 kinds of cancer cells via XTT assay. We examined the effect on the revelation of PARP cleavage, Bcl-2 protein and Bax protein by western blot analysis. Results : The extract of realgar caused markedly morphological changes on AGS, T98G, SNU-489, and LNCaP. All of them showed withdrawn and floating appearance. The suppressive effect on viability of AGS, T98G, A172, SNU-489, and LNCaP showed that each test group had more suppressive effect on viability of AGS, T98G, A172, SNU-489, and LNCaP than the control group, which was statistically significantly (p<0.01). The extract of realgar did not induce PARP cleavage in AGS, T98G, A 172, SNU-489, or LNCaP. In the revelation of protein related to apoptosis, the protein levels of Bcl-2 decreased and the protein levels of Bax increased in AGS, T98G, SNU-489, and LNCaP treated with realgar. The protein levels of Bcl-2 decreased and the protein levels of Bax did not change in A172 treated with realgar. Conclusions : This experiment showed that realgar has anti-tumor effect on stomach cancer cells (AGS), glioma cells (T98G, SNU-489L and prostate cancer cells (LNCaP)

DDT의 Aromatase 증가에 의한 Testosterone 감소효과 (DDT Reduced Testosterone and Aromatase Activity Via ER Receptor in Leydig Cell)

  • 본 연구는 환경호르몬(endocrine disruptors)으로 분류되었으며, 제초제로 널리 사용되었던 Dichlorodiphenyltrichloroethane(DDT)가 설치류의 생식세포 중 Leydig 세포의 testosterone(T)생성억제 및 그 관련 작용메카니즘을 규명코자 수행되었다. 먼저 흰쥐의 웅성 생식세포주인 R2C세포에 T의 양 및 aromatase 활성도를 radio immunoassay (RIA)방법을 이용하여 측정하였다. 그 결과 R2C 세포에 황체형성호르몬(LH)를 처리하여 testosterone생성을 증가시킨 후, DDT를 처리한 군들은 대조군에 비하여 농도 의존적으로 T의 양은 감소하였으며, aromatase 활성도는 증가하였다. 또한 DDT자체만 처리한 군에서도 대조군에 비하여 testosterone의 생성이 감소하였다. 이러한 aromatase활성 증가가 estradiol receptor(ER)와의 상호 관련성을 확인하기 위해 ER antagonist인 ICI 182.780를 처리한 후 T의 양 및 aromatase 활성도를 측정한 결과 DDT에 의해 증가된 aromatase활성도가 ICI 182.780에 의해 다시 감소됨을 확인하였다. 또한 DDT에 의해 감소된 T의 양도 ICI 182.780에 의해 다시 회복되었다. In vivo실험으로 흰쥐에 DDT를 직접 투여한 후 정소 내 성 호르몬들을 측정해 본 결과 T의 양은 유의성 있게 감소하였으며, estradiol(E$_2$)의 양은 증가하였으며, aromatase 활성도도 감소하였다. 이러한 결과를 종합해 볼 때 DDT는 aromatase를 감소시키고, 이렇게 감소된 aromatase에 의해 testosterone 생성량을 억제하고, 이러한 DDT의 aromatase의 감소는 ER을 경유하는 것으로 추정할 수 있다.

  • Analgesic, anticonvulsive, sedative and antipyretic actions of Yangkyuksanwhatang which is composed of nine crude drugs including Rehmanniae Radix, and its effects on isolated ileum and blood vessel were investigated. The results of the studies were summerized as follows; 1. Analgesic and antipyretic effects were observed. 2. Suppressive actions were not shown on the convulsion induced by strychnine, but significant effects were noted on the convulsion induced by picrotoxin. 3. A prolongation of anesthetic time induced by pentobarbital sodium was significantly observed. 4. Relaxing actions were noted on the ileum of mice, and also, same effects were recognized on the contraction of the ileum due to acetylcholine chloride and barium chloride. 5. The expansion of blood vessels by relaxation of smooth muscle and hypotensive effects were noted.

  • Some of organophosphate pesticides which are the most heavily used in Korea, were examined for their effects on the murine immune system. Immunotoxicological assay parameters adaopted in this study were Arthus reaction for humoral immunity, delayed type hypersensitivity reaction for cell mediate immunity, carbon clearance for macrophage function and susceptiility to tumor challenge. Subchronic exposure of rodents to the pesticides resulted in the marked suppression of immune functions and enhancement of susceptibility to tumor challenge. Among the pesticides tested (fenitrothion, fenthion, diazinon and EPN), fenitrothion was the most suppressive in Arthus and delayed type hypersensitivity reaction.

  • Objectives: This study was designed to find the effect of Phragmitis Rhizoma (PR) herbal-acupuncture solution on the inflammatory cytokine and chemokine secretion in human mast cell (HMC) and human alveolar epithelial cell 549 (A549) lines. Methods: Histamine levels in HMC after PR herbal-acupuncture solution treatment were measured with ELISA. Other cytokines and chemokines levels such as interleukin 8 (IL-8), monocyte chemoattractant protein-1 (MCP-1), and chemokine (C-C motif) ligand 5 (Ccl5, RANTES) in A549 were measured with flow cytometry CBA system. Results: In the PR herbal-acupuncture solution treatment group, the expression of histamine, IL-8, MPC-1, Ccl5, and RANTES decreased significantly. Conclusions: The results support that PR herbal-acupuncture solution had a suppressive effect on cytokine-induced inflammation.

  • Nitric oxide (NO) is a small diffusible molecule which plays an important role in various physiological activities. NO is a notable molecule, functioning as a cytotoxic agent and cellular messenger. There has been considerable interest in NO production by activated macrophages because this gaseous metabolite plays a fundamental role in the cytotoxic and cytostatic effects of macrophages towards invasive micro-organisms and tumour cells. No is a bioactive free radical that has been implicated in many physiological functions, plays a critical role during inflammation and therefore constitutes a potential target for developing therapeutics for inflammatory diseases. The use of medicinal plants by the population has been an important alternative the resource in the treatment of various diseases. Its growing acceptance in the medical community has been due to the fact that several plants with biological activities have been scientifically investigated and their efficacy and safety have been proven. In this review, discussed suppressive effects of No production by traditional medicines in RAW 264.7 and THP-1 macrophages.

  • The effects of various fractions from the aerial parts of Echinosophora koreensis Nakai on the antimutagenic and the immuno-regulating activities were evaluated by in vivo bone-marrow micronucleus test and HA titer reaction. No significant suppressive effects of these extracts and echinoisosophoranone were shown on cyclophosphamide-induced micronuclei, but HA titers were significantly enhanced in ether and BuOH extract treated-group. Tetracosanol and docosanol were isolated from the ether extract of this plant.

Suppressive Effects of Benincasae hispida on Allergic Inflammation

  • The effects of an extract of Benincasae hispida on allergic inflammation were examined in terms of histamine and $\beta$-hexosaminidase release, serum IgE level and inflammatory cytokine level. The B. hispida extract inhibited the release of histamine and $\beta$-hexosaminidase, a degranulation marker, from rat basophilic leukemia cells (RBL-2H3). When mice were first ovalbumin-challenged and then treated with B. hispida extract, there was a significant decrease in the IgE level in the mouse serum. The extract treatment reduced the serum IgE level prominently, compared with the ovalbumin-challenged mice. The extract also significantly reduced the TNF-$\alpha$ and IL-4 levels in the BAL fluid when challenged with antigen. Taken together, the Benincasae hispida extract may be efficacious against allergic inflammation.

  • We have studied some effects of the SOEUMIN-HYANGSAYANGYUI-TANG (S.H.Y.) on the C.N.S. and on the intestines. Several empirical remarks, depending on the different treatments, are investigated throughout this study as follows; 1) The treatment, using atropine bromide, represses the effect of the S.H.Y. on the contractive force of intestines. 2) The treatment, using pheniramine maleate, seems not be able to repress the effect of the S.H.Y. on the contractive force of intestines. 3) The treatment, using cyproheptadine, seems not be able to repress the effect of the S.H.Y. on the contractive force of intestines. 4) The analgesic effect as well as the antipyretic effect are remarked while the acetic acid is applied. 5) The suppressive action on the convulsion, induced by strychine, is not observed. However the significant effect on the convulsion induced by picrotoxin is remarked. 6) The sedactive effect of the S.H.Y., using the Rotor rod test, is not obviously observed.

Suppressive Effects of Divalent Cations on Self-splicing Inhibition by Spectinomycin of Group 1 Intron RNA

  • Effects of divalent cations on self-splicing inhibition by the antibiotic spectinomycin of the phage T4 thymidylate synthase intron (td) have been investigated. $Ca^{2+}$ ion at 1mM concentration suppressed splicing inhibition of spectinomycin by 10% and 50 ${\mu}M\;Co^{2+}$ ion also suppressed splicing inhibition of specinomycin by 10%. $Mg^{2+}$ ion at 6 mM concentration decreased splicing inhibition of spectinomycin by 42% while $Mn^{2+}$ ion decreased the splicing inhibition by 10%. $Zn^{2+}$ ion at 10 uM concentration lowered the splicing inhibition by spectinomycin of 15%. Of all divalent cations tested, $Mg^{2+}$ ion was the most effective in suppressing splicing inhibition by specinomycin whereas $Ca^{2+}$ ion was the least effective. The results suggest that spectinomycin may interact with specific and functional $Mg^{2+}$-binding sites within intron RNA that lead to a displacement of $Mg^{2+}$ essential for catalytic activity.
