Physiological Activities of Ethanol Extracts from Different Parts of Ailanthus altissima (가죽나무(A. altissima) 부위별 에탄을 추출물의 생리활성)

  • The study was carried out to optimize the electron donating ability (EDA), superoxide dismutase (SOD)-like activity, nitrite scavenging ability, and the inhibitory activities of xanthine oxidase and tyrosinase of Ailanthus altissima ethanol extracts from roots, stems and leaves. The EDA of the roots and stem extracts were 64.04% and 63.27% at $1,000{\mu}g/mL$, respectively. The SOD-like activity of the leaves extract was the highest (50.00%) at $1,000{\mu}g/mL$. The nitrite scavenging ability of the leaves extracts was over 98% at pH 1.2 and 3.0. The xanthine oxidase inhibitory rates of the extracts were $93.62%{\sim}95.40%$ and the tyrosinase inhibitory rate of the roots was the highest (62.01%) at the concentration of $1,000{\mu}g/mL$. These results indicated that the roots extract showed the highest EDA and tyrosinase inhibition, while the leaves extract had tile highest SOD-like activity and nitrate scavenging ability.

Antioxidant Effect of Citri Reticulatae Pericarpium Extract on Oxidative Stress-Mediated Cytotoxicity In Cultures

  • In order to examine oxidative stress of reactive oxygen species and the antioxidant effect of Citri Reticulatae Pericarpium (CRP) extract, human skin melanoma cells were treated with various concentrations of hydrogen peroxide ($H_2O_2$). Antioxidant effect of CRP extract on $H_2O_2$-induced cytotoxicity, cell viability, DPPH-radical scavenging activity and superoxide dismutase (SOD)-like activity. In this study, $H_2O_2$ decreased cell viability of cultured human skin melanoma cells in dose- and time-dependent manners, and then, midcytotoxicity value (MCV) was determined at $60\;{\mu}M$ after human skin melanoma cells were cultured for 5 hours in the media containing $20{\sim}60\;{\mu}M$ of $H_2O_2$, respectively. The $H_2O_2$ was on cultured human skin melanoma cells because MCV of $H_2O_2$ was lower than $100\;{\mu}M$. In the antioxidant effect of CRP extract, CRP extract increased cell viability DPPH-radical scavenging activity and SOD-like activity. From these results, it is suggested that $H_2O_2$ was very toxic on cultured human skin melanoma cells. And also, CRP extract has the antioxidant effect on $H_2O_2$-induced cytotoxicity.

Antioxidant Activity and Ginsenoside Pattern of Fermented White Ginseng

  • Ethanol and water extracts of white and fermented ginseng were prepared and their ginsenoside composition and antioxidant effects were assessed. The main ginsenosides in white ginseng were $Rb_1$ > Re > $Rg_1$, and those in fermented ginseng were $Rb_2+Rb_3$ > Rd > $Rg_1$. Ginsenosides Rd and $Rg_3$ in fermented ginseng were enriched 11 and 58 times, respectively, over that in white ginseng through fermentation with five Bacillus spp. The greatest levels of 2-deoxyribose and superoxide anion dismutase-like activities were found in 50% ethanol extracts of fermented ginseng. Thus, these data suggest that white ginseng has the greatest free radical scavenging activity and that fermented ginseng has the highest antioxidant activity.

Antioxidative Activities and Tyrosinase Inhibition of Water Extracts from Ailanthus altissima (가죽나무(Ailanthus altissima) 물 추출물의 항산화 활성과 Tyrosinase 저해)

  • Water extracts from root, stem and leaf of Ailanthus altissima were utilized to determine antioxidant properties such as electron donating ability (EDA), nitrite scavenging ability, superoxide dismutase (SOD)-like activity, and the inhibitory activities of xanthine oxidase (XO) and tyrosinase. The EDA of root extract was the highest as 77.33% at 0.5 mg/mL concentration and that of stem extract was 70.01% at 1.0 mg/mL. The nitrite scavenging ability of leaf extract revealed the highest effect as 95.18% at pH 1.2, 1.0 mg/mL while those of stem and root extracts were 55.17% and 33.33%, respectively. The leaf extract showed the highest SOD like activity as 26.77% at 1.0 mg/mL, the measurement of root extract was 3.82% and that of stem extract was not effective. All kinds of extracts had strong inhibitory activities on XO of over 92% at 1.0 mg/mL. The highest activity on tyrosinase inhibition was obtained from leaf extract of 16.33% at 2.0 mg/mL. The results indicated that among the three extracts, the leaf extract has a strong and extensive antioxidant activity.

Antioxidant Properties and Total Phenolic Contents of Cherry Elaeagnus (Elaeagnus multiflora Thunb.) Leaf Extracts

  • In Korea and China, cherry elaeagnus (Elaeagnus multiflora Thunb.) has been used traditionally to treat cough, diarrhea, itching, and foul sores. Therefore, in this study, the ethanol and water extracts of cherry elaeagnus leaves were examined for their antioxidant activities. The ethanol extract of the cherry elaeagnus leaves contained more phenolics than the water extract. All the cherry elaeagnus leaf extracts had higher 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging ability than ascorbic acid at concentrations of $250-1,000\;{\mu}g/mL$. The ethanol extract also showed higher superoxide dismutase (SOD)-like activity compared to the water extract. Furthermore, the SOD-like activity of the ethanol extract amounted to 89% of that of ascorbic acid at a concentration of $500\;{\mu}g/mL$. The nitrite scavenging ability and xanthine oxidase inhibitory (XOI) activity of the ethanol extract were higher than those of the water extract. In particular, the ethanol extract had higher XOI activity than ascorbic acid at a concentration of $1,000\;{\mu}g/mL$.

Antioxidant and Angiotensin I Converting Enzyme Inhibitory Activities of Red Snow Crab Chionoecetes japonicas Shell Hydrolysate by Enzymatic Hydrolysis

  • We investigated the antioxidant and angiotensin I converting enzyme (ACE) inhibitory activities of red snow crab Chionoecetes japonicas shell (RSCS) hydrolysate by enzymatic hydrolysis and its molecular weight cut-off fractions. The RSCS hydrolysate was fractionated through two ultrafiltration membranes of 3 and 10 kDa cut-offs. Three fractions (<3 kDa, 3-10 kDa, and >10 kDa) were evaluated for total amino acid composition, antioxidant activities using 2'-azino-bis[3-ethylbenzthiazoline-6-sulfonic acid] ($ABTS^+$) radical scavenging and superoxide dismutase (SOD)-like activities and reducing power assays, and ACE inhibitory activity using Hou's method. Although all fractions showed activity, the <3 kDa fraction of RSCS hydrolysate exhibited the greatest $ABTS^+$ radical scavenging, SOD-like and ACE inhibitory activities. However, these fractions exhibited low reducing power. These results suggest that the low-molecular-weight enzymatic hydrolysate of RSCS could be used as a functional ingredient to control oxidative stress and ACE activity.

Antioxidant and Neuronal Cell Protective Effects of Eugenia caryophyllata Thunb. by Extraction Solvent (용매별 정향 추출물의 항산화 및 신경세포 보호 효과)

  • This study examined the antioxidant and neuronal cell protective effects of the water and methanol extracts of Eugenia caryophyllata Thunb. The total polyphenol content was significantly higher in the methanol extract than in the water extract. The DPPH radical scavenging activity in the water extract was similar to Vit. C at a concentration of $100{\sim}200{\mu}g/mL$. The ABTS radical scavenging activity in the water and methanol extract was similar to Vit. C at a concentration of $800{\sim}1,000{\mu}g/mL$. The superoxide dismutase (SOD)-like activity in the methanol extract was similar to Vit. C at a concentration of $800{\sim}1,000{\mu}g/mL$. The DPPH, ABTS radical scavenging and (SOD)-like activity increased with increasing extract concentration. In a cell viability using MTT, the water extract (50 and 100 ppm) and methanol extract (100 ppm) had a protective effect against $H_2O_2$-induced neurotoxicity.The result ssuggest that the extract of E. caryophyllata Thunb. has antioxidant activities and may be useful for treating neurodegenerative disorders.

Anti-oxidant, Anti-aging, and Whitening Effects of Viscum album var. coloratum In Vitro (한국산 겨우살이의 항산화, 항노화 및 미백 효과)

  • In this study, we investigated the anti-oxidant, anti-aging, and skin whitening effects of Korean mistletoe (Viscum album var. coloratum). The mistletoe fraction was composed of four types: hexane (HX), ethyl acetate (EA), butanol (BU), and methylene chloride (MC). In total phenol content assay, HX showed the highest phenol content among four fractions. In addition, EA significantly increased 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and catalase-like activities, and MC significantly increased superoxide dismutase (SOD)-like activity. When we compared IC50 value in the hyaluronidase and elastase inhibition assay, MC had the lowest IC50. In addition, we also performed tyrosinase inhibition assay to demonstrate the possibility of Korean mistletoe as a cosmetic component. HX showed the highest tyrosinase inhibition rate among the fractions.

Changes of Chlorophyll and SOD-like Activities of Chinese Chives Dehydrated at Different Heat Treatments (부추의 건조 온도 조건별 클로로필, Superoxide Dismutase 유사활성의 변화)

  • Allium tuberosum Rotter (Liliaceae, Chinese chives) is a perennial herb of which leaves are used for food. This study investigated the effect of pretreatment on quality of dehydrated Chinese chives. Chinese chives was blanched at $80^{\circ}C$ for 20 sec, followed by drying at $70^{\circ}C$, $80^{\circ}C$, $100^{\circ}C$, or drying at $100^{\circ}C$ for 30 min and subsequent drying at $70^{\circ}C$, or $100^{\circ}C$ for 60 min and subsequent drying at $70^{\circ}C$. Optimum drying temperature and time was $100^{\circ}C$ for 30 min and subsequent drying at $70^{\circ}C$, or $100^{\circ}C$ for 60 min and subsequent drying at $70^{\circ}C$. These conditions were shortened time for dehydration and showed smaller decrease than others in Hunter color L, a, b. Dehydrated Chinese chives showed a constant decrease in greenness with storage, probably due to destruction of chlorophyll by light. In the measurement of Hunter color L, a, b, these conditions showed smaller decrease than others in Hunter color for 15 week storage. Chlorophyll content and SOD (superoxide dismutase)-like activity in that condition was higher than others. It was assumed that a phenolic compound that forms its thermostable activity. The fitness of drying models was conducted in order to explain reducing chlorophyll loss and SOD (superoxide dismutase)-like activity loss. Based upon the chlorophyll content, SOD-like activity, and retention of green color of the vegetable, optimum drying conditions was $100^{\circ}C$ for 30 min followed by $100^{\circ}C$ for 30 min and subsequent drying at $70^{\circ}C$, or $100^{\circ}C$ for 60 min and subsequent drying at $70^{\circ}C$.

Quality Characteristics and Antioxidant Activities of Soybean Curd Products Containing Small Black Soybean (쥐눈이콩을 첨가한 두부제조에 따른 두부, 비지 및 순물의 항산화성)

  • Soybean curds were prepared by adding different levels (20, 40, and 60%) of small black soybean, and the quality characteristics of the soybean curds were investigated. The yield of soybean curd increased slightly with the increment of the levels of small black soybean, whereas L value (lightness) and b value (yellowness) decreased. Analysis of textural properties demonstrated that the addition of small black soybean lowered the hardness and chewiness of soybean curd, while it slightly increased the cohesiveness and adhesiveness. Antioxidant activities of soybean curd, soybean curd residue, and soybean curd whey were estimated by determining electron donating ability (EDA) to DPPH radical and superoxide dismutase (SOD)-like activity. These activities in all of the samples increased with the addition of small black soybean, especially in soybean curd residue and soybean curd whey; also, soybean curd whey containing small black soybean showed the highest activities.
