The antioxidant activities of methanol and dichloromethane/acetone extracts of twenty three seaweeds were tested by using 1,1-Diphenyl-2-picryl-hydrazyl(DPPH) at a $100{\mu}g/ml$ concentrations. The dichloromethane/acetone extracts of three seaweeds(Symphyocladia latiuscula, Gloiopoltis furcata, Sagassum thunbergii), were found to be most effective in DPPH radical scavenging activity. The DPPH radical scavenging effect of these seaweeds was Symphyocladia latiuscula(85.82%), Gloiopoltis furcata(82.83%), Sagassum thunbergii(74.05%) in order. These seaweeds were evaluated using the pyrogallol UV-VIS spectrophotometeric method to generate superoxide anion. Among them, the methanol extracts of six seaweeds were showed weak superoxide dismutase-like activities. The dichloromethane/acetone extracts obtained from Symphyocladia latiuscula was fractionated with $CH_2Cl_2$, n-hexane, 15% aq.MeOH, n-BuOH, $H_2O$. The 15% aq.MeOH soluble fraction exhibiting the strongest antioxidative activities was further purified by C18 column flash chromatography and reversed HPLC. The two active principles of Symphyocladia latiuscula were isolated and characterized as 2,3,6-tribromo-4,5-dihydroxybenzyl alcohol(1), 2,3,6-tribromo-4,5-dihydroxybenzyl methyl ether(2).
The objective of this study was to evaluate the antioxidant and anti-aging effects of marigold methanol extract (MGME) in human dermal fibroblasts. Total polyphenolic and flavonoid contents in MGME were 74.8 mg TAE (tannic acid equivalent)/g and 85.6 mg RE (rutin equivalent)/g, respectively. MGME ($500{\mu}g/mL$) increased 1,1-diphenyl-2-picryl hydrazyl (DPPH) and 2,2'-azino-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical-scavenging, and superoxide dismutase (SOD)-like antioxidant activities by 36.5, 54.7, and 14.8%, respectively, compared with the control. At $1,000{\mu}g/mL$, these activities increased by 63.7, 70.6, and 20.6%, respectively. MGME ($100{\mu}g/mL$) significantly increased the synthesis of type 1 procollagen by 83.7% compared with control treatment. It also significantly decreased Matrix Metalloproteinase-2 (MMP-2) activity and MMP-1 mRNA expression by 36.5% and 69.5%, respectively; however, it significantly increased laminin-5 mRNA expression by 181.2%. These findings suggest that MGME could protect human skin against photo-aging by attenuating oxidative damage, suppressing MMP expression and/or activity as well as by stimulating collagen synthesis.

To produce bioactive compound enriched yeast using medicinal Gugiga (Lycium chinensis Mill), several edible Saccharomyces species were cultured in Gugija extracts added yeast extract, peptone and dextrose medium (GE - YEPD medium) at $30^{\circ}C$ for 24 hr, and their growth were determined. Growth of Saccharomyces cerevisiae K-7 and Sacchromyces cerevisiae ACTC 7904 were better than those of the other yeasts. Two yeasts were selected and then determined their some physiological functionalities after cultivated the yeasts in the GE - YEPD medium and compared those grown on YEPD medium. Antihypertensive angiotensin I-converting enzyme (ACE) inhibitory activity of S. cerevisiae K-7 grown on GE - YEPD medium was about 20% higher than that grown on YEPD medium. Superoxide dismutase-like activity of S. cerevisiae ACTC 7904 was also about 12% more high. However, the other physiological functionalities were almost same or lower. Optimal addition concentration of Gugija extract was 10%, and maximally growth and ACE inhibitory activity of S. cerevisiae K-7 were shown when the strain was cultured in 10% Gugija extracts containing YEPD medium at $30^{\circ}C$ for 12 hr.
Native soy protein isolate (SPI) was hydrolyzed with 4 different proteolytic enzymes, including bromelain, papain, Neutrase, and Flavourzyme. SPI hydrolysates with the degree of hydrolysis (DH) in range of 6 to 15% were prepared by each enzyme. The angiotensin 1 converting enzyme (ACE) inhibitory and the antioxidant activities of the SPI hydrolysates, such as superoxide dismutase-like activity and inhibition of the linoleic acid autoxidation, were evaluated. Overall, as the DH increased, all evaluated bioactivities of the SPI hydrolysates significantly increased. The significantly highest ACE inhibitory and antioxidant activities were found in hydrolysates made with papain and bromelain, respectively. SPI hydrolysates by Flavourzyme showed the significantly lowest activity in all tested bioactivities. The results suggested that ACE inhibitory and antioxidant activities of SPI hydrolysates were determined by the DH and by the enzyme used.
The present study deals with the amelioration by Lagerstroemia speciosa Linn. leaf extract against hepatotoxicity induced by carbon tetrachloride ($CCl_4$), which was evaluated in terms of serum marker enzymes like serum glutamate pyruvate transaminase, serum glutamate oxaloacetate transaminase, alkaline phosphatase, serum total bilirubin, total protein levels along with concomitant hepatic and antioxidants like superoxide dismutase, catalase, glutathione, glutathione peroxidase and lipid peroxidation enzymes were monitored. These biochemical parameters altered by the single dose level of $CCl_4$ (0.75 ml/kg body weight, i.p). Pre treatment with L. speciosa prior to the administration of $CCl_4$, at the doses of 50 and 250 mg/kg. body weight/day, p.o. for 7 days, significantly restored all the serum and liver tissue parameters near to the normal levels, respectively. Silymarin was used as a reference standard, prior to the administration of $CCl_4$ to rats. These findings indicate the protective potential of L. speciosa against hepato toxicity which possibly involve mechanism related to its ability of selective inhibitors of (reactive oxygen species like antioxidants brought about significant inhibition of TBARS suggesting possible involvement of $O_2{\cdot}-$, $HO_2{\cdot}$, and ${\cdot}OH$. In conclusion, the amelioration may be attributed to the synergistic effects of its constituents rather than to any single factor as the leaves are rich in tannins, sterols, flavonoids, saponins etc.
본 연구에서는 머핀의 품질향상을 위한 기능성 천연소재의 개발을 위해 현미분말을 0, 20, 40, 60% 농도로 첨가하여 제조한 머핀의 품질특성을 살펴보았다. 머핀의 중량은 대조군이 높았으나 현미분말 첨가군 간에 유의적인 차이는 없었고, 높이와 pH 또한 대조군이 높았던 반면 현미분말 첨가군은 첨가량이 증가할수록 감소하였다(p<0.05). 현미분말 첨가 머핀 수분함량의 측정결과는 제조 직후 대조군이 현미분말 첨가 머핀보다 높았지만, $30^{\circ}C$에서 저장기간이 길어짐에 따라 수분함량은 대조군과 현미분말 첨가군 간에 유의적인 차이는 없었다(p<0.05). 경도와 복원성은 대조군이 높았고 현미분말 첨가량이 증가할수록 높아졌으며, 응집성, 탄력성은 대조군이 높았으며 현미분말 첨가량이 증가할수록 낮아졌다(p<0.05). 이에 반해 검성과 씹힘성은 대조군이 낮았고 현미분말 첨가량이 증가할수록 높았으며, 부착성은 시료 간에 유의적인 차이가 없었다(p<0.05). 관능검사 결과 외부색은 20% 첨가군이, 내부색은 대조군이 높게 나타났고(p<0.05), 맛, 향, 조직감, 전반적인 기호도는 60% 첨가군이 가장 높게 나타났다. 현미분말 첨가 머핀의 항산화활성을 측정한 결과는 현미분말 첨가 농도가 증가할수록 DPPH radical 소거능과 SOD 유사활성이 증가하는 것으로 나타났다. 이상의 결과를 종합해 볼 때 현미를 첨가할 경우 관능특성 및 기능성을 개선하여 품질향상에 도움을 주는 것으로 나타났는데 이는 현미를 이용한 기능성식품 개발의 기초자료로 제공될 수 있을 것으로 사료된다.
Doxorubicin induces oxidative stress leading to cardiotoxicity causing electrocardiogram abnormalities and increases in biomarkers associated with toxicity. Green tea extract (GTE) is reported to possess antioxidant activity mainly via its polyphenolic constituent, catechins. This study was intended to determine the effect of various doses of GTE (25, 50 and 100 mg/kg/day p.o. for 30 days) on doxorubicin-induced electrocardiographic and biochemical changes in rat heart. The latter included lactate dehydrogenase, creatine kinase, and glutamic oxaloacetate transaminase in serum and superoxide dismutase, catalase, and reduced glutathione, as well as membrane bound enzymes like $Na^+K^+ATPase,\;Ca^{2+}ATPase,\;Mg^{2+}ATPase$ and decreased lipid peroxidation in heart tissue Results demonstrated that rats which received GTE were less susceptible to such changes indicating protection afforded by GTE.
The most extensively used synthetic antioxidants are butylated hydroxyanisole (BHA), butylated hydroxy toluene (BHT) and tert-butylhydroquinone (TBHQ). However BHT and BRA have been suspented of being responsible for liver damage andcar cinogenesis. Therefore, the importance of the search and exploitation of natural antioxidant, especially of plant origin, has greatly increased in recent years. Plant contain a wide variety of chemicals that have potent biological effects. As a result, there has been a growing interest in the use of herbs as a source of therapeutic drugs. The aim of this study was to assess the antioxidant and cosmeceutical of natural materials. The antioxidant and cosmeceutical activity of natural materials were investigated by hydroxyl radical scavenging, superoxide dismutase (SOD) -like, xanthine oxidase inhibition, tyrosinase inhibition, anti-microbial and astringent.
조경수목(造景樹木)들의 대기오염물질(大氣汚染物質)들에 대한 감수성(感受性) 및 저항성(抵抗性)을 규명하기 위해서 야외조사(野外調査)와 실내실험(室內實驗)을 통하여 엽내(棄內) 유황함량(硫黃含量)과 superoxide dismutase(SOD) 함량(含量)을 측정(測定) 분석(分析)하였다. 전(全) 수종(樹種)에서 엽내(棄內) 유황함량(硫黃含量)과 SOD 활성간(活性間)에 정(正)의 상관(相關)을 나타내었다. 식물체(植物體)가 오염물질(汚染物質)에 대하여 자체방어(自體防禦)를 위하여 초기(初期)에는 효소활성(酵素活性)을 증대(增大)시키지만, 식물체(植物體)의 수용한계(收容限界)를 넘을 때는 오히려 효소(酵素)의 실활(失活)과 함께 식물체(植物體)가 피해(被害)를 받았다. 활엽수(闊葉樹)는 대기오염물질(大氣汚染物質)의 정화능력(淨化能力)이 큰 반면 침엽수(針葉樹)는 오염물질(汚染物質)에 대한 방어기능(防禦機能)이 크게 나타났다. 엽내(葉內) SOD 활성(活性)은 비오염지역(非汚染地域)과 오염지역간(汚染地域間)에 차이(差異)가 뚜렷하였는데, SOD 활성(活性)은 활엽수(闊葉樹)에 비하여 침엽수(針葉樹)에서 높게 나타났으며 묘목(苗木)보다는 성목(盛木)에서 높았고, 소나무보다는 잣나무에서 활성(活性)이 높았으며, 튜립나무는 은행나무와 양버듬나무보다 낮은 것으로 나타나 SOD 활성(活性)이 높은 것이 대기오염물질(大氣汚染物質)에 대한 내성(耐性)이 강(强)함을 알 수 있었다. 고유(固有)의 SOD 활성(活性)이 높은 소나무 2년생(年生)잎과 잣나무 1, 2년생(年生)잎에서는 $SO_2$ 처리농도(處理濃度)가 강(强)해질수록 SOD 활성(活性)이 증가(增加)하였으나, 고유(固有)의 SOD 활성(活性)이 나은 수종(樹種)은 저농도(低濃度(0.5ppm)처리(處理)에서는 증가(增加)하고 고농도(高濃度) 처리(處理)에서는 감소(減少)하는 경향(傾向)이었다.
Objectives: This study was carried out to analyze the cytotoxicity of cadmium sulfate ($CdSO_4$) and the antioxidative effect of Typha orientalis L. (TO) extract on the oxidative stress induced by cytotoxicity of $CdSO_4$ in the cultured NIH3T3 fibroblasts. Methods: For this study, the cell viability and the antioxidative effects such as the inhibitory activity of lipid peroxidation (LP) and superoxide dismutase (SOD)-like activity and xanthine oxidase (XO)-inhibitory activity were assessed. Results: The cadmium sulfate significantly decreased cell viability in dose-dependently, and $XTT_{50}$ value was measured at $47.4{\mu}M$ of $CdSO_4$. The cytotoxicity of $CdSO_4$ was determined as highly toxic by Borenfreund and Puerner's toxic criteria. The butylated hydroxytoluene (BHT) as antioxidant significantly increased cell viability injured by $CdSO_4$-induced cytotoxicity in these cultures. In the protective effect of TO extract on $CdSO_4$-induced cytotoxicity, TO extract remarkably increased the inhibitory ability of LP and XO as well as SOD-like ability. Conclusions: From the above results, it is suggested that the oxidative stress is involved in the cytotoxicity of $CdSO_4$, and TO extract effectively protected $CdSO_4$-induced cytotoxicity by antioxidative effects. The natural component like TO extract may be a putative therapeutic agent for treatment of the toxicity induced by heavy metallic compound like $CdSO_4$ correlated with the oxidative stress.
