PC12 손상 세포 및 전뇌허혈 유발 Gerbil에 대한 백지의 세포보호효과 (Protective Effect of Angelicae Dahuri Radix on Hypoxia Reperfusion Induced by PC12 Cell Damage and Global Ischemia in Gerbil)

  • Objective : This research was performed to investigate the protective effect of Angelicae Dahuri Radix against ischemic damage using PC12 cells and global ischemia in gerbils. Methods : To observe the protective effect of Angelicae Dahuri Radix on ischemia damage, viability and changes in activities of superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase and production of malondialdehyde (MDA) were observed after treating PC12 cells with Angelicae Dahuri Radix during ischemic insult. Gerbils were divided into three groups : a normal group, a 5-min two-vessel occlusion (2VO) group, and an Angelicae Dahuri Radix administered after 2VO group. The CCAs were occluded by microclip for 5 minutes. Angelicae Dahuri Radix was administered orally for 7 days after 2VO. The histological analysis was performed at 7 days after surgery. For histological analysis, the brain tissue was stained with 1% cresyl violet solution. Results : 1. Angelicae Dahuri Radix has a protective effect against ischemia in the CA1 area of the gerbil hippocampus 7 days after 5-minute occlusion, 2. In the hypoxia/reperfusion model using PC12 cells, Angelicae Dahuri Radix has a protective effect against ischemia in the dose of $0.2\mu\textrm{g}/ml$, $2\mu\textrm{g}/ml$ and $20\mu\textrm{g}/ml$, 3. Angelicae Dahuri Radix increased the activities of glutathione peroxidase and catalase. 4. The activity of superoxide dismutase (SOD) was increased by ischemic damage, which might represent self protection. This study suggests that Angelicae Dahuri Radix has some neuroprotective effect against neuronal damage following cerebral ischemia in vivo with a widely used experimental model of cerebral ischemia in Mongolian gerbils, and it also has protective effects on a hypoxia/reperfusion cell culture model using PC12 cells. Conclusions : Angelicae Dahuri Radix has protective effects against ischemic brain damage at the early stage of ischemia.

Sequence Variation in Superoxide Dismutase Gene of Toxoplasma gondii among Various Isolates from Different Hosts and Geographical Regions

  • Toxoplasma gondii, an obligate intracellular protozoan parasite of the phylum Apicomplexa, can infect all warm-blooded vertebrates, including humans, livestock, and marine mammals. The aim of this study was to investigate whether superoxide dismutase (SOD) of T. gondii can be used as a new marker for genetic study or a potential vaccine candidate. The partial genome region of the SOD gene was amplified and sequenced from 10 different T. gondii isolates from different parts of the world, and all the sequences were examined by PCR-RFLP, sequence analysis, and phylogenetic reconstruction. The results showed that partial SOD gene sequences ranged from 1,702 bp to 1,712 bp and A + T contents varied from 50.1% to 51.1% among all examined isolates. Sequence alignment analysis identified total 43 variable nucleotide positions, and these results showed that 97.5% sequence similarity of SOD gene among all examined isolates. Phylogenetic analysis revealed that these SOD sequences were not an effective molecular marker for differential identification of T. gondii strains. The research demonstrated existence of low sequence variation in the SOD gene among T. gondii strains of different genotypes from different hosts and geographical regions.

감마선 조사전 홍삼 추출물의 투여가 생쥐 간에서의 Superoxide dismutase의 활성과 지질 과산화에 미치는 영향

  • 생쥐에 5.5mg의 홍삼추출물을 투여한 후 6.5Gy의 감마선을 전신 조사하여 생쥐 간에서의 항산화 효소들(Superoxide dismutase, Catalase, Peroxidase)의 활성도와 지질 과산화 수준의 변화를 측정 비교하여 이들의 상호 관련성과 인삼이 방사선 장해에 미치는 영향을 검토하였다. 방사선 조사로 인하여 SOD, Catalase 그리고 Peroxidase의 활성은 계속적으로 감소하였으나 홍삼추출물 투여군은 방사선 조사 후 21일에 생리적 식염수 투여군보다 Cu, Zn-SOD($40.7\%$), Mn SOD($26.9\%$), Catalase($20.0\%$) 그리고 Peroxidase($20.1\%$)의 활성이 증가되었으며, 또한 방사선 조사로 인하여 상승되는 지질 과산화의 함량이 억제되었다($70.5\%$). 이러한 결과는 인삼 성분이 방사선 조사로 인하여 생성이 촉진된 free radicals를 제거시킴으로서 지질 과산화물의 생성을 억제시켰거나 방사선에 의하여 손상된 기관들의 회복능력을 강화시킨 결과로 보이며 이는 인삼이 방사선 장해에 대하여 회복 또는 방호효과도 있음을 보여 주는 것이라 생각된다.

Superoxide Dismutase Activity in Small Mesenteric Arteries Is Downregulated by Angiotensin II but Not by Hypertension

  • Many studies reported reduced antioxidant capacity in the vasculature under hypertensive conditions. However, little is known about the effects of antihypertensive treatments on the regulation of vascular antioxidant enzymes. Thus, we hypothesized that antihypertensive treatments prevent the reduction of antioxidant enzyme activity and expression in the small vessels of angiotensin II-induced hypertensive rats (ANG). We observed the small mesenteric arteries and small renal vessels of normotensive rats (NORM), ANG, and ANG treated with a triple antihypertensive therapy of reserpine, hydrochlorothiazide, and hydralazine (ANG + TTx). Systolic blood pressure was increased in ANG, which was attenuated by 2 weeks of triple therapy (127, 191, and 143 mmHg for NORM, ANG, and ANG + TTx, respectively; p < 0.05). Total superoxide dismutase (SOD) activity in the small mesenteric arteries of ANG was lower than that of NORM. The protein expression of SOD1 was lower in ANG than in NORM, whereas SOD2 and SOD3 expression was not different between the groups. Reduced SOD activity and SOD1 expression in ANG was not restored in ANG + TTx. Both SOD activity and SOD isoform expression in the small renal vessels of ANG were not different from those of NORM. Interestingly, SOD activity in the small renal vessels was reduced by TTx. Between groups, there was no difference in catalase activity or expression in both the small mesenteric arteries and small renal vessels. In conclusion, SOD activity in the small mesenteric arteries decreased by angiotensin II administration, but not by hypertension, which is caused by decreased SOD1 expression.

급성 간손상의 실험동물 피부조직에 있어서 Oxygen Free Radical의 대사효소 활성 변동 (Change of Dermal Oxygen Free Radical Metabolizing Enzyme Activities in Acute Liver Damage Induced with $CCl_4$ in Rats)

  • 실험동물에 있어서 $CCl_4$에 의한 급성 간손상시 피부조직의 oxygen free radical 대사효소 활성 변동을 검토하기 위해 흰쥐에 $CCl_4$와 olive oil의 동량 혼합액을 체중 100g당 0.1 ml씩 복강으로 투여하여 처치하였다. $CCl_4$ 투여로 인한 혈청 alanine aminotransferase 및 xanthine oxidase (XO) 활성은 현저히 증가되었으며 체중당 간무게 (%)및 간조직의 malondialdehyde함량 역시 유의하게 증가되었다. 그리고 병리조직 검사에서도 $CCl_4$투여군에서 간조직의 괴사성 병변이 관찰되었다. 따라서 $CCl_4$를 투여한 실험동물이 급성 간손상 모델로 확인되었다. 이와 같은 간손상 실험동물의 피부조직중 oxygen free radical의 생성 효소인 XO 활성은 대조군과 별다른 차이를 볼 수 없었으나 oxygen free radical의 scavenging 효소인 superoxide dismutase, glutathione peroxidase 및 catalase 활성은 대조군에 비하여 유의하게 감소되었다. 또한 세포화학적 검사에서 cerrous perhydroxide과립이 간손상 실험동물의 피부조직에서 많이 나타났다. 이상 실험결과는 $CCl_4$에 의한 급성 간손상 유도 실험동물의 피부조직에 $H_2O$$_2$의 축적이 나타나는 현상을 시사해주고 있다.

급성 카드뮴중독시 랏트의 고환조직내 Catalase 및 Superoxide Dismutase 활성도와의 양-반응관계 (Dose-Response Relationship between Catalase and Superoxide Dismutase Activity in Testes of Acutely Intoxicated Rats by Cadmium)

  • Dose-response relationship among blood cadmium concentrations, catalase and superoxide dismutase activities were studied with acutely intoxicated rats by cadmium. The Sprague-Dawley male rats to which single dose of $1{\sim}32mg/kg\;CdCl_2$ were administered into peritoneal cavity were sacrificed by decapitation at $3{\sim}36$ hours after the administration. Cadmium concentrations in blood increased significantly with dose of $CdCl_2$ administered and reached peak level at 3 hours later. Catalase activities in rats' testes were not correlated with esposure time elapsed after the administration in rats to which $1{\sim}2mg/kg\;of\;CdCl_2$ were administered, but they showed linear relationship with time in groups to which $4{\sim}32mg/kg\;of\;CdCl_2$ were administered. Cu, Zn-SOD activities in testes of acutely intoxicated rats by cadmium were not altered either by dosage or by time elapsed after the administration. Mn-SOD activities in the testes were also not influenced by dosage of $1{\sim}2mg/kg\;CdCl_2$, but remarkably inactivated by dosage of $4{\sim}32mg/kg\;CdCl_2$ with time elapsed after the administration. Neither catalase, Cu, Zn-SOD nor Mn-SOD activities of testes were correlated with blood cadmium concentrations in acutely intoxicated rats by cadmium.

Dimethylformamide가 사람 혈청의 과산화지질 농도와 Superoxide dismutase 활성도 변화에 미치는 영향 (Effects of Dimethylformamide on Lipid Peroxide Level and Activity of Superoxide Dismutase in Human Serum)

  • The variation in the enzyme activities of human liver usually represents the particular physiological conditions of each individuals. Thus, we investigated the variation in the activities of SOD, HR and LPO of (1) non-exposed workers (56 subjects), and (2) exposed workers to DMF (43 subjects) in synthetic leather process. Serum levels of enzyme activities of exposed workers (AST:$30.26{\pm}20.041$U/L, ALT:$32.72{\pm}23.393$U/L, GGT:$28.47{\pm}18.635$ U/L, ALP:$81.77{\pm}34.879$ U/L)were slightly higher than those in nonexposed workers (AST:$24.00{\pm}9.441$ U/L, ALT:$23.89{\pm}18.305$U/L, GGT:$21.95{\pm}17.970$U/L, ALP:$70.84{\pm}24.678$U/L), but only the level of ALT was statistically significant (p< 0.05). Serum levels of LDH, TRF and CHOL in non-exposed workers were slightly higher than those of exposed workers. However, they were not statistically significant (p > 0.05). Serum levels of HR and LPO of the exposed workers appeared to be reduced, but not those of the non-exposed workers. The SOD activities of exposed workers were also slightly higher than those of non-exposed workers, but the results were not statistically significant (p > 0.05). The level of HR was increased with age, but the SOD level was not. These results suggest that the intermittent exposure to DMF at time-weighted average (TWA) level (10 ppm/$m^3$) has affected on the activities of enzymes such as AST, ALT, TRF, but not on the generation of HR, activity of SOD. However, if high dose of DMF was used, there would be severe effects for the generation of HR and LPO.

  • Superoxide dismutase (SOD), one of the essential element of the antioxidant defense system, mainly removes $O^{-10}$ $_2$ and also prevents $O^{-10}$ $_2$ mediated reduction of iron and subsequent OH$^{-10}$ generation, which is highly toxic to the organism. Of these SOD enzymes, Cu, Zn-containing SOD (SODI) is an important component of the antioxidant defense system in eucaryotic cells. The SODI enzyme binds one copper and one zinc ion and displays the Greek Key $\beta$-barrel fuld. (omitted)
