Antioxidant and Protective Effects of Leonurus sibiricus L. Extract on Ultraviolet B (UVB)-induced Damage in Human Keratinocytes (익모초 추출물의 항산화 효과와 자외선으로 유도된 각질형성세포 손상에 대한 보호효과)

  • In this study, we investigated the anti oxidative potential and protective effects of water extract of Leonurus sibiricus L. leaf (LSLW) against ultraviolet B (UVB)-induced oxidative damage in human keratinocytes (HaCaT cells). To evaluate the anti oxidative activity of LSLW, we measured DPPH radical, hydroxyl radical, hydrogen peroxide, superoxide anion scavenging activities, lipid peroxidation inhibition, and reducing power of LSLW. For induction of oxidative stress in HaCaT cells, the cells were irradiated with UVB at 40 mJ/㎠. To investigate the protective effects of LSLW against UVB, we measured cell viability and apoptotic bodies using annexin V staining. LSLW showed anti oxidative activities by scavenging DPPH radical, hydroxyl radical, hydrogen peroxide, superoxide anion and by reducing lipid peroxidation. In addition, LSLW showed high reducing values. The UVB-induced oxidative conditions led to cell apoptosis. However, treatment with LSLW ameliorated oxidative stress conditions, including inhibition of cell death, apoptotic body. Taken together, LSLW exhibited anti oxidative and protective effects against UVB-induced damage in HaCaT cells. Thus, LSLW could be useful for the development of cosmetics for UVB-induced skin aging.

Protective Effects of Opuntia Ficus-Indica and Saururus Chinensis on Free Radical-Induced Neuronal Injury in Mouse Cortical Cell Cultures (생쥐 피질세포배양에서 Free Radical 유발 신경손상에 대한 손바닥선인장 및 삼백초의 보호효과)

  • The author examined whether the methanol extracts of Opuntia ficus-indica fruit and Saururus chinensis have the inhibitory action on xanthine/xanthine oxidase (X/XO)-, $FeCl_2/ascorbic$ acid- and arachidonic acid-induced neurotoxicity in mouse cortical cell cultures. The methanol extracts ($10\;{\mu}g/ml{\sim}1\;mg/ml$) of Opuntia ficus-indica and Saururus chinensis were exhibited 53-89% and $48{\sim}100%$ inhibitory action on X/XO-induced neurotoxicity, respectively. At the range of same concentration, both extracts also attenuated the $FeCl_2/ascorbic$ acid-induced neurotoxicity by $35{\sim}100%$ and $15{\sim}98%$, respectively. In arachidonic acid neurotoxicity, the methanol extract (1 mg/ml) of Opuntia ficus-indica and Saururus chinensis reduced neuronal injury by 22% and 38%, respectively. These results suggest that Opuntia ficus-indica fruit and Saururus chinensis may contribute the neuroprotection in certain free radical-mediated neuronal injury.

In vitro anti oxidant activity of methanol extract of Clerodendrum infortunatum Linn

  • The antioxidant potency of methanolic extract Clerodendrum infortunatum Linn. (MECI), which are widely used in the Indian indigenous system of medicine for different purposes, was studied. The antioxidant potential was evaluated using different established in vitro antioxidant tests viz. determination of total amount of polyphenolics compounds, DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging, nitric oxide scavenging, superoxide anion radical scavenging, hydroxyl radical scavenging and reductive power assay. It was found that MECI contain a high amount of polyphenolics and possesses significant free radical scavenging activity in all the assay. The higher activity was may be due to presence of richest amount of polyphenolics and flavonoids in it.

Antioxidant Activities of Traditional Wine and Liquor Produced in Chungcheong-do (충청지역 민속주의 항산화활성)

  • Antioxidant activities of traditional liquors produced in Chungcheong-do were studied. The contents of organic acids, carbohydrate, and protein of the traditional liquors were 0.30~0.95%, 1.4~11.1% and 1.5~37.3 mg%, respectively. Especially, the contents of phenolic compounds of L-3 was the highest (205.8 $\mu\textrm{g}$/mL). L-5, L-4 and L-3 showed higher antioxidant activities than the others on hemoglobin-induced linoleic acid system. L-5 also showed high reducing power and scavenging effect on DPPH radical. Scavenging effects on hydroxyl radicals in L-6, L-5 and L-4 showed higher activities (42~53%) than those of the others. The highest scavenging effect on superoxide anion radical was observed in L-5 ($IC_{50}$/: 1.1 $\mu\textrm{g}$).

Studies on Antioxidant Effect of Mushroom Complex (버섯복합물의 항산화 효과)

  • In this study, we evaluated the antioxidant effects of Cordyceps, Ganoderma lucidum (Curtis) P. Karst and Phellinus linteus complexes. Three mushrooms were extracted using 70% ethanol, respectively, in ratios of 80:10:10(?). The electron-donating ability was measured using the DPPH radical mushroom complexes and showed effects of 66% in 500 ${\mu}g/ml$. ABTS radical cation decolorization measured 73% in 50 ${\mu}g/ml$ and had an effect similar to BHA in 500 ${\mu}g/ml$. In their superoxide anion radical scavenging ability, the mushroom complexes showed an effect of 41% in 10 ${\mu}g/ml$. The effect on the control of BHA was more than three times as high as BHA in 50 ${\mu}g/ml$ and 100 ${\mu}g/ml$ concentration. The results showed that lipid oxidation is an excellent inhibitor of lipid oxidation $Cu^{2+}$, compared to the $Fe^{2+}$ group. The results of superoxide dismutase (SOD) activity in the mushroom complexes showed a low effect of 13% in 500 ${\mu}g/ml$. Thus, the results of this study showed that the antioxidant effect of the mushroom complexes was excellent, indicating their potential as an antioxidant material.

Cytotoxic Effect of Adriamycin in Cultured Skin Cells of Fetal Rat (백서 태자의 배양 피부세포에서 Adriamycin의 세포독성에 관한 연구)

  • Local extravasation during intravenous administration of adriamycin (doxorubicin HCl) can cause severe skin ulceration and necrosis. To investigate the mechanism of adriamycin-induced skin toxicity, effects of adriamycin on reactive oxygen radical metabolism using cultured skin cells of fetal rat. Adriamycin produced significant release of lactic dehydrogenase from cultured skin cell preparations dose- and time-dependently. The production of superoxide anion in sonicated suspensions of cultured skin cells was significantly increased by adriamycin under the presence of NADPH and NADH. The drug also stimulated malondialdehyde (MDA) production, an index of lipid peroxidation, in NADPH- and NADH-supported cell preparations. The increased production of MDA was significantly inhibited by oxygen radical scavengers (superoxide dismutase, catalase, thiourea) and antioxidants (butylated hydroxytoluene, ${\alpha}-tocopherol$). Treatment of cultured skin cells with 1, 3,-bis (2-chloroethyl)-1-nitrosourea (BCNU), an inhibitor of glutathione reductase, enhanced the lipid peroxidation induced by adriamycin. The present study suggests that lipid peroxidation which is resulted from the stimulated production of reactive oxygen radical causes cellular damage in adriamycin-treated skin cells of rat.

Flavonoids and chlorogenic acid from Eriobotrya japonica scavenge peroxynitrite

  • Peroxynitrite is a cytotoxic intermediate produced by the reaction between the superoxide anion radical and nitric oxide. Flavonoids (afzelin, quercitrin and quercetin 3-O-sambubioside), and chlorogenic acid and its methyl ester obtained from leaves of loquat (Eriobotrya japonica) have recently been shown to scavenge 1,1-diphenyl-2-picrylhydrazyl radical and to inhibit lipid peroxidation in mouse liver homogenate. The aim of this study is to investigate the inhibitory effects of the above components on peroxynitrite produced stimulated by 3-morpholinosydnonimine (SIN-1) to produce superoxide anion radical and nitric oxide at the same time. In addition, the present study tests whether or not the components directly scavenge peroxynitrite itself. The results showed that the components with the aromatic ortho-dihydroxyl groups (catechol) were more potent inhibitors of peroxynitrite formation by SIN-1. In particular, the methyl ester form of chlorogenic acid showed the most potent inhibition. At $5\;{\mu}M$ concentration, the order of minimizing peroxynitrite formation were : methyl chlorogenic acid > quercitrin > quercetin 3-O-sambubioside > chlorogenic acid > afzelin. Authentic peroxynitrite was directly scavenged by the components in a manner similar to peroxynitrite formation by SIN-1. In particular, when compared with penicillamine as a positive control, methyl chlorogenate was as effective in inhibiting peroxynitrite formation and approximately 2 times more effective in scavenging an authentic peroxynitrite. These results demonstrate therefore, that components extracted from the leaves of Eriobotrya japonica effectively scavenged peroxynitrite.

Cytoprotective and Antioxidative Effects of Crude Drug Preparation (E-kong-san) (이공산(異功散)의 세포보호 및 항산화 작용)

  • In the previous report, E-kong-san, which is usually used for recovering health in traditional medicine, has been shown to decrease cisplatin induced nephrotoxicity in vivo and in vitro. The significant reduction of E-kong-san on the cisplatin induced nephrotoxicity led us to investigate whether the effect of this water extract was a result of triggering antioxidation. In monkey kidney Vero cells, E-kong-san at $5{\sim}10\;mg/ml$ was able to attenuate 2mM cisplatin-stimulated cell death by 46.8% and 31.8%, respectively. E-kong-san showed strong free radical scavengering activities on 1,1-diphenyl-2-picrylhydrazil (DPPH) radical and xanthine/xanthine oxidase (XOD) generated superoxide anion radical $(O_2^{-.})$. We further studied the effects of E-kong-san on lipid peroxidation in rat liver microsomes induced by enzymatic and nonenzymatic methods. Moreover, E-kong-san exhibited significant inhibition on both ascorbic $acid/Fe^{2+}$ and $ADP/NADPH/Fe^{3+}$ induced lipid peroxidation in rat liver microsomes. Based on these results, we suggest that E-kong-san protects the cisplatin induced cytotoxicity by its antioxidative mechanism.

Antioxidant Effects of Solvent Fraction from Sanguisorbae officinalis L. with Acetone (오이풀 아세톤 추출물을 이용한 용매 분획물의 항산화 효과)

  • The solvent extracts of Sanguisorbae officinalis L. were investigated for the activities of antioxidants as a functional ingredient for cosmetic products. The electron donating effect of ethyl acetate layer and n-butyl alcohol layer was appeared similar activity with positive control butylated hydroxy anisole (BHA) at all concentrations. In addition, in 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid (ABTS) radical scavenging assay, ethyl acetate layer, n-butyl alcohol layer and water layer were over 99% effect at all concentrations and higher than that of BHA. Also in hydrogen peroxide scavenging assay, ethyl acetate layer and n-butyl alcohol layer were higher than that of positive control ascorbic acid. The measured superoxide dismutase (SOD)-like activity of n-butyl alcohol was more than 50% at concentration of 1,000 ${\mu}g/mL$ and superoxide anion radical scavenging ability showed more than 45% at 1,000 ${\mu}g/mL$ of n-butyl alcohol layer. All these findings suggested that ethyl acetate layer and n-butyl alcohol layer have a great potential as a cosmeceutical ingredient with an antioxidant effect.

  • The study was aimed at evaluating the antioxidant and free radical scavenging activities of ethanolic extract and its fractions of Cleome rutidosperma. The antioxidant activity, reducing power, total phenolic content, total flavonoid content, superoxide anion scavenging activity, nitric oxide anion scavenging activity, in vitro antilipid peroxidation activity and in vitro non-enzymatic hemoglobin glycosylation were studied. The results obtained in the study indicate that Cleome rutidosperma is a potential source of natural antioxidant. All the parameters were found to be concentration dependent and increased with increasing amounts of sample. Flavonoids, phenolic compound like tannins, terpenoids may be responsible for the antioxidant activity of the plant. Variation of solubility parameters in various models may be attributed to non-linearity of activity of ethanol extract fractions models. Further investigation on the isolation and identification of antioxidant component(s) in the plant may lead to chemical entities with potential for clinical use.
