Radical scavenging activity of ethanol extract and solvent partitioned fractions of lotus seeds

  • This study focused on the evaluation of the antioxidative effects of lotus seeds from golden colored flowers. The lotus seeds were extracted with ethanol and then fractionated into 4 fractions, ethyl acetate (EtOAc), n-butanol, methylene chloride, and n-hexane. The comparison of antioxidative activities of the extract and fractions from the lotus seeds was carried out using an in vitro radical scavenging model and the total phenol content was analyzed. Of the tested extracts and fractions, the EtOAc fraction of the lotus seeds showed the strongest 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity with 96.24% at a concentration of $100{\mu}g/mL$. In addition, the hydroxyl radical scavenging activity of the lotus seed EtOAc fraction was also increased in a concentration dependent manner with the concentrations tested ranging from 5 to $100{\mu}g/mL$. Moreover, the EtOAc fraction showed the highest scavenging activity for nitric oxide and superoxide anion radicals. In particular, of all the extracts and fractions, the EtOAc fraction showed highest contents of total phenols. These results indicate that lotus seeds have potential as an antioxidative agent against oxidative stress involving reactive oxygen species. Furthermore, the EtOAc fraction of lotus seeds includes promising oxidative stress-protective compounds.

Antioxidative Effect of Rhus javanica Linne Extract Against Hydrogen Peroxide or Menadione Induced Oxidative Stress and DNA Damage in HepG2 Cells

  • The free radical scavenging activities and the protective effects of Rhus javanica extracts against oxidative damage induced by reactive oxygen species (ROS) were investigated. n-Hexane, ethyl acetate and water fractions were prepared from a methanol extract. DPPH radical, superoxide anion and hydroxyl radical scavenging activities were estimated. Intracellular ROS formation was quantified using fluorescent probes, 2', 7'-dichlorofluorescin diacetate (DCFH-DA) for hydroxyl radical and dihydroethidium (DHE) for superoxide anion. The oxidative DNA damage was investigated by the comet assay in HepG$_2$ cells exposed either to $H_2O$$_2$ or to menadione. The highest $IC_{50}$/ values for DPPH radical scavenging activity was found in the ethyl acetate fraction with a value of 5.38 $\mu\textrm{g}$/mL. Cells pretreated with $\geq$ 1 $\mu\textrm{g}$/mL of the ethyl acetate extract had significantly increased cell viability compared to control cells, which were not pretreated with the extract. Intracellular ROS formation and DNA damage in HepG$_2$ cells, which were pretreated with the various concentrations of Rhus javanica ethyl acetate extract and then incubated either with $H_2O$$_2$ or with menadione, reduced in a dose-dependent manner. These findings suggest that Rhus javanica might have biologically active components which have strong protective effects against ROS induced oxidative damages to the biomolecules, such as cell membranes and DNA.

하엽(荷葉) 추출물이 항산화 효능 및 피부노화에 미치는 영향 (Antioxidant and Anti-wrinkling Effects of Extracts from Nelumbo nucifera leaves)

  • Objective : The purpose of this study was to investigate anti-aging and antioxidant effects of extracts of Nelumbo nucifera leaves (NN-L) using ethanol on skin .Methods : Each part of leaves(NN-L), flowers(NN-F) and stem(NN-S) was extracted with 70% ethanol. We performed radical scavenging assay(DPPH, ABTS+, Superoxide anion radical), elastase inhibition assay, collagenase inhibition assay. NN-L extracts were tested for cell viability(MTT assay), MMP-1 inhibition and MMP-1 protein expression on CCD-986sk cells (human fibroblast line).Results : Recently, many studies have reported that elastin is also involved in inhibiting or repairing wrinkle formation, although collagen is a major factor in the skin wrinkle formation. We measured its free radical scavenging activity, elastase inhibitory activity and expression of MMP-1 (matrix metalloprotease-1) in human fibroblast cells. Among the parts of Nelumbo nucifera, NN-L showed the highest antioxidant activities and in radical scavenging. DPPH, ABTS+ and Superoxide anion radical scavenging activity of NN-L at concentration of 1,000 μg/mL were 91.43%, 99.31% and 73.7% respectively. In vitro elastase and collagenase inhibition effects of NN-L at concentration of 1,000 μg/mL was 42.8% and 55.3% respectively. The ethanol extract of NN-L showed cell viability of 95.4% in 50 μg/mL concentration. In addition, The results from Western blot assay showed that NN-L decreased the expression of MMP-1 protein in a dose-dependent manner (by up to 35.0% at 50 μM).Conclusion : The findings suggest that the NN-L great potential as a cosmeceutical ingredient with antioxidant and anti-wrinkling effects.

평위산(平胃散) 약침액(藥鍼液)의 활성산소 및 활성질소 소거능 (Scavenging Property of Pyungwi-san Herbal-acupuncture Solution on ROS and RNS)

  • Pyungwi-san(PWS) have been using as a basic prescription of digestive disorder in Korean traditional medicine. This study was performed to examine the in vitro antioxidant activity of the extract using different antioxidant tests including by 1,1-diphenyl-2-picryl-hydrazil (DPPH) radical scavenging, superoxide anion radical scavenging, metal chelating hydrogen peroxide scavenging, lipid peroxydation protective effect and scavenging effect of nitric oxide and peroxynitrite. Herbal-acupuncture solution of PWS(PWS-HS) exhibited a concentration-dependent inhibition of DPPH radical adduct formation and it showed dose-dependent free radical scavenging activity onto superoxide anions. In addition, the result of metal chelating hydrogen peroxide scavenging and ammonium thiocyanate experiments showed that PWS-HS was an active scavenger of hydroxyl radicals. Furthermore, it was also found to be effective in scavenging nitric oxide and peroxynitrite, well-known cytotoxic species that can oxidize several cellular components such as proteins, lipids and DNA.

TOSC 방법을 이용한 참당귀 뿌리 용매분획(Angelica gigas)의 항산화 활성 평가 (Evaluation of Antioxidant Activity of Solvent Fractions of Angelica gigas Root Using TOSC Assay)

  • Objectives: The aim of this study was to elucidate the antioxidant effect of solvent fractions of Angelica gigas root. Methods: The ethanol extract of Angelica gigas root was suspended in water and then partitioned with dichloromethane (MC Fr.), ethyl acetate (EA Fr.) and butanol (BuOH Fr.), sequentially. The antioxidant activities of solvent fractions of Angelica gigas root were evaluated for radical scavenging activity against stable free radicals (2,2-diphenyl-1-picrylhydrazyl) DPPH, hydrogen peroxide and superoxide anion radicals. In addition the antioxidant activities of solvent fractions of Angelica gigas root against peroxyl radicals, hydroxyl radicals and peroxynitrites were determined by the total oxy-radical scavenging capacity (TOSC) assay. Results: Among the solvent fractions of MC Fr., EA Fr., and BuOH Fr., BuOH Fr. was found to have stronger antioxidant activity with $IC_{50}$ values of 59.72, 14.36, 30.96 and $44.75\;{\mu}g/m{\ell}$ on the DPPH radical, nitrite, superoxide anion and hydrogen peroxide, respectively, than BHA used as a positive control. Moreover, specific TOSC values(564.8, 276.4 and 405.5 TOSC/mM) of BuOH fr. against peroxyl radicals, hydroxyl radical and peroxynitrite were 4 times higher than GSH (136.5, 67.4 102.6 TOSC/mM) used as a positive control. Conclusions: These results suggest that the BuOH fr. of Angelica gigas root has a high antioxidant activity and can be useful to develop functional food against oxidative stress conditions.

An Advanced Kinetic Method for HO2·/O2-· Determination by Using Terephthalate in the Aqueous Solution

  • Hydroperoxyl radical/superoxide anion radical ($HO_2{\cdot}/O_2^-{\cdot}$, $pK_a$=4.8) as an intermediate is of considerable importance in oxidation processes. Hence, the method of detecting $HO_2{\cdot}/O_2^-{\cdot}$ with high sensitivity is necessary to be developed. To achieve this objective, this study newly employed terephthalate (TA) as a probe for the measurement of $HO_2{\cdot}/O_2^-{\cdot}$ in the kinetic method presented in our previous study. This method was based on the hydroxylation of TA to produce mainly hydroxyterephthalic acid or hydroxyterephthalate (OHTA), which was analyzed by fluorescence detection (${\lambda}_{ex}$=315nm, ${\lambda}_{ex}$=425nm). The life-time of $HO_2{\cdot}/O_2^-{\cdot}$ and its concentration formed from the photolysis technique of $H_2O_2$ were reported in this study. At range of pH 2-10, the life-time of $HO_2{\cdot}/O_2^-{\cdot}$ was 51-422 sec. In particular, an increase in the life-time with pH was observed. The sensitivities of the kinetic method by using TA were always higher with 1.7-2.5 times at pH 8.0 than those by using benzoic acid. From these results, this study can contribute to understanding the basic functions of $HO_2{\cdot}/O_2^-{\cdot}$ in oxidation processes.

Determination of Hydroperoxyl/superoxide Anion Radical (HO2·/O2·-) Concentration in the Decomposition of Ozone Using a Kinetic Method

  • A novel kinetic method for determination of $HO_2{^{\cdot}}/O_2{^{{\cdot}-}}$ in ozone decomposition in water is described. In this study, potential interferences of $O_3$ and the hydroxyl radicals, $^{\cdot}OH_{(O3)}$, are suppressed by $HSO_3{^-}/SO_3{^{2-}}$. $HO_2{^{\cdot}}/O_2{^{{\cdot}-}}$ formed in ozone decomposition reduces $Fe^{3+}$-EDTA into $Fe^{2+}$-EDTA and subsequently the well-known Fenton-like (FL) reaction of $H_2O_2$ and $Fe^{2+}$-EDTA produces the hydroxyl radicals, $^{\cdot}OH_{(FL)}$. Benzoic acid (BA) scavenges $^{\cdot}OH_{(FL)}$ to produce OHBA, which are analyzed by fluorescence detection (${\lambda}_{ex}=320nm$ and ${\lambda}_{ex}=400nm$). The concentration of $HO_2{^{\cdot}}/O_2{^{{\cdot}-}}$ in ozone decomposition has been determined by the novel kinetic method using the experimentally determined half-life ($t_{1/2}$). The steady-state concentration of $HO_2{^{\cdot}}/O_2{^{{\cdot}-}}$ is proportional to the $O_3$ concentration at a given pH. However, the steady-state concentration of $HO_2{^{\cdot}}/O_2{^{{\cdot}-}}$ in ozone decomposition is inversely proportional to pH values. This pH dependence is due to significant loss of $O_2{^{{\cdot}-}}$ by $O_3$ at higher pH conditions. The steady-state concentrations of $HO_2{^{\cdot}}/O_2{^{{\cdot}-}}$ are in the range of $2.49({\pm}0.10){\times}10^{-9}M(pH=4.17){\sim}3.01({\pm}0.07){\times}10^{-10}M(pH=7.59)$ at $[O_3]_o=60{\mu}M$.

Effect of Red Pepper Seed on Kimchi Antioxidant Activity During Fermentation

  • In this study, the antioxidant activities of red pepper seed kimchi extracts were examined. The extracts were evaluated by various antioxidant assays that included determinations of total phenolic and flavonoid contents, DPPH radical scavenging, superoxide anion radical scavenging, nitric oxide scavenging, superoxide dismutase (SOD) activity, metal chelating activity, and reducing power. All the kimchi extracts showed strong antioxidant activities by the tested methods. The 7% red pepper seed kimchi that was fermented for 6 days possessed the highest activity according to the performed tests. Yet, the highest scavenging activity was exhibited against nitrite, by extracts from kimchi for 0 day of fermentation rather than 6 days. All the activities for the kimchi fermented for 0 day with the addition of 7% red pepper seed increased markedly with an increase in concentration. With the exception of metal chelating and SOD activities, for the antioxidant properties assayed, the red pepper seed kimchi extracts were found to be highly effective.

어유산화생성물의 DNA 손상작용 1. 총지질산화생성물의 DNA 손상작용 (The DNA Damage by Fish Oil Perokidation Products 1. DNA Damage by the Peroxidation Products of Total Lipid Fraction Extracted from Mackerel)

  • 고도불포화지방산함량이 많은 적색육어류지질의 산화에 의한 DNA 손상작용기구를 조사하기 위하여 고등어를 시료로 하여 추출한 지질을 plasmid pBR 322DNA와 $37^{\circ}C$에서 반응시켜 경시적인 DNA 손상정도를 조사하였으며 이와 함께 지질과 DNA의 반응계에 각종 활성산소소거제를 일정농도씩 첨가하고 지질산화로 일어나는 DNA 손상에 대한 활성산소종의 영향을 비교${\cdot}$ 검토하였다. 고등어 지질의 농도가 클수록 DNA 손상작용이 컸으며 이러한 작용은 고등어 지질의 POV가$100millieq{\cdot}/kg$이하에서 진행된 것으로 나타났다. 또한 활성산소종중 일중항산소$(^1O_2)$와 superoxide anion$({\cdot}O_2^-)$ DNA 손상작용이 가장 컸으며 수산 radical과 과산화 수소는 반응1일이후 거의 영향을 미치지 않아 반응초기의 DNA 손상작용에 관여할 것으로 생각된다. 또한, 고등어 지질의 산화에 대한 활성산소종의 영향을 살펴 본 결과, DNA 손상에 대한 결과와 마찬가지로 일중항산소와 superoxide anion의 영향이 큰 것으로 보아 고등어 지질의 산화반응이나 DNA 손상작용에 있어서 활성산소종이 밀접한 관계가 있는 것으로 나타났다.

호박분말 효소가수분해물의 항산화활성 (Antioxidant Activities in Enzymatic Hydrolysates of Pumpkin Powder (Cucurbita spp.))

  • 본 연구는 늙은호박, 단호박 및 애호박을 다양한 탄수화물 분해효소로 소화하여 조제한 효소가수분해물의 총 폴리페놀 함량과 항산화 활성을 측정하였다. 조회분, 조단백질 및 조섬유는 애호박이 높았으며, 탄수화물은 단호박이 높았고, 조지방은 서로 비슷하였다. 애호박 분말의 모든 효소처리군의 총 폴리페놀 함량은 늙은호박과 단호박의 가수분해물에 비하여 좀 더 높았다. 애호박 분말의 AMG와 Termamyl 소화로 조제된 효소가수분해물의 DPPH 유리기 소거활성 또한 늙은호박 및 단호박에 비하여 매우 높게 나타났다. 한편, Visozyme 처리 가수분해물을 제외한 늙은호박과 단호박의 대부분 가수분해물은 애호박보다 더 높은 superoxide anion 소거활성을 나타내었다. 애호박 가수분해물(Ultraflo 예외)의 hydrogen peroxide 소거활성은 다른 호박의 효소 분해물 보다 약간 높았으나, hydroxyl radical 소거활성은 모든 호박 품종에서 14% 미만으로 낮았다. Nitric oxide 소거활성은 단호박 및 애호박의 Viscozyme 분해물에서 매우 효과적이었으며, 다른 효소가수분해물의 경우에도 BHT보다는 낮았으나 $\alpha$-tocopherol보다 높은 활성을 나타내었다.
