Quality Characteristics and Antioxidant Activity of Syrup Added with Maca (Lepidium meyenii) Extract (마카 추출액 첨가에 따른 시럽의 품질특성과 항산화성)

  • This study was conducted to develop a functional syrup by using maca extract. Five different levels (0%, 7%, 14%, 21%, 28%) of maca extract was added and the physicochemical properties were investigated. The pH of control syrup was lower than that of syrup added with maca extract. Viscosity increased with increasing amount of maca extract. Hunter L value decreased while b value increased as maca extract level increased. Results of consumer acceptance test revealed that no significant differences in flavor, sweet taste and overall acceptability were observed among samples. Therefore, it is suggested that maca extract can be incorporated into syrup up to 28% without depressing the quality. Total polyphenol content and DPPH radical, superoxide anion radical and hydroxyl radical scavenging tests were conducted in view of estimating the functionality of maca syrup. Results showed that incorporation of maca extract into syrup exhibited higher polyphenol content and DPPH radical and superoxide anion radical scavenging activity than control syrup (p<0.05).

Study on the Antioxidant Activity of Extracts from the Fruit of Elaeagnus multiflora Thunb (뜰보리수(Elaeagnus multiflora Thunb) 추출물의 항산화 효과에 관한 연구)

  • This study was investigated to analyze the antioxidant activity or extracts form Elaeagnus multiflora Thunb for development to the functional materials. The antioxidative activities of water ethanol and methanol extracts from the Elaeagnus multiflora Thunb were analyzed by electron donating ability (EDA), anti-oxidization activity, superoxide dismutase (SOD)-like activity, The superoxide anion radical-scavenging activity, and nitrite scavenging ability. The Etectron donating ability of methanol extract was higher in 1.0 mg/mL of extraction solution than those of ethers. The anti-oxidization activity of ethanol and methanol extracts by thiocyanate method using linolenic acid system was higher than those of the water extract. The SOD-like activity was increased with increase of the extract concentration in each extracts. The SOD-like activity was highest in 2.0 mg/mL of methanol extract. The superoxide anion radical-scavenging activity was increased with increase of the concentration in the ethanol extract and methanol extract. The nitrite scavenging ability of water extracts in 1.0 mg/mL of extraction soiution in pH 1.2 was higher than ethanol extrats and methanol extracts. The nitrite scavenging ability of all extracts was decreased according to increase of pH.

Hepatopotective Effects of Black Rice on Superoxide Anion Radicals in HepG2 Cells

  • Cyanidin 3-glucoside (C3G) isolated from black rice was investigated for hepatoprotective effects in HepG2 cells under oxidative stress. When an increase in the production of reactive oxygen species (ROS) was induced by gramoxone, cell viability was drastically decreased by 42%. However, in the presence of C3G, no hepatocytic damage was observed in HepG2 cells treated with gramoxone. C3G was found to manifest a stronger scavenging effect (91%) on superoxide anion radical ($O_2\;^{.-}$) than any of the other natural and synthetic antioxidants. Results suggest that C3G from black rice possesses hepatoprotective effects in vitro, which may be, at least in part, due to $O_2\;^{.-}$ scavenging.

Reactive Oxygen Species Scavenging Activity of Jeju Native Citrus Peel during Maturation (수확시기별 제주재래종 감귤과피의 활성산소종 소거활성)

  • This study was conducted to investigate total polyphenolic contents and reactive oxygen species (ROS) scavenging effects of extracts from peels of ten Jeju native citrus fruits according to the harvest from August 2006 to February 2007. Total polyphenolic contents from methanol extracts of citrus peel were the highest in Jigak (Citrus aurantium) and Hongkyool (C. tachibana) by over 200 mg% in the unmatured period, from the late August to the late September, and all the citrus peels mostly decreased while ripening. Scavenging effect of superoxide anion radical showed good correlation with total polyphenolic contents. The unmatured periods of Hongkyool and Jigak were the highest with more than 60%. Hydrogen peroxide scavenging activity was the highest in Sadoogam (C. pseudogulgul) at 73.8% in late August and the second highest activity was observed in Jigak at near 70%, and all the citrus peels decreased during ripening. Hydroxy radical scavenging activity were the highest among all the ROS scavenging activities, especially in the Jigak and Dangyooja (C. grandis) at 75.1% and 74.6%, respectively, and not much affected by increased maturity of the fruits. Nitric oxide radical scavenging activity was the highest in Bungkyool (C. platymama) at 58.4% in late February, and increased with fruit ripening. In this study, Jigak was generally the highest in the polyphenolic contents and ROS scavenging activities, so the further studies are needed for industrial applications.

  • Production of free radicals of superoxide anion in tissues by cadmium, activities of superoxide dismutase and catalase to protect tissue damages caused by the free radicals and ATPase that plays an important role in energy metabolism at cellular level were investigated. Experiments in vivo were conducted with liver, kidney and testicle tissue homogenates of rats adding $0.05{\sim}0.50mM$ cadmium chloride, and in vivo experiments administering single dose of 5 mg of cadmium/kg of body weight in 0.1% cadmium chloride solution intraperitoneally 48 hours prior to evisceration. Production of superoxide radicals in liver and testicle increased with addition of cadmium in vitro, but not in kidney. In vivo experiments, however, superoxide radicals slightly increased in liver and kidney but not in testicle. Superoxide dismutase (Cu, Zn-SOD and Mn-SOD), catalase and ATPase (total, $Mg^{++}-\;&\;Na^+,\;K^+-$) activity decreased in the presence of cadimium in dose dependent manner. Reduction of these enzyme activities varied not only with dosage of cadmium but also with type of tissue and between in vitro and in vivo experiment.

The Chemical Reactions of Superoxide with Halopyrimidines

  • Halopyrimidines such as 2-chloro-, 5-bromo, and 4,6-dichloro-5-nitropyrimidine undergo substitution reactions with superoxide anion radical (superoxide) to give the corresponding hydroxypyrimidines under suitable conditions. Parallel experiments employing hydroxide instead of superoxide strongly indicate that the reactivity of superoxide is comparable to that of the hydroxide in the reaction with halopyrimidines. The results seem to provide a piece of information in favor of the nucleophilic substitution rather than electron-transfer mechanism in the title reaction.

Antioxidative Activity of Jeolpyun containing Suaeda glauca (Bunge) Bunge Powder and Extract (나문재 분말과 추출물 첨가 절편의 항산화 활성)

  • In this study, the antioxidative activity of Jeolpyun containing S. glauca (Bunge) Bunge powder (2%, 4%, 6%, and 8%) and extract (2%, 4%, 6%, and 8%) was evaluated based on the total polyphenol contents, electron-donating ability, scavenging of superoxide anion radical, and decomposition of hydrogen peroxide. Total polyphenol contents of Jeolpyun containing 8% S. glauca (Bunge) Bunge powder were the highest with a value of 2,280±0.00 ㎍ GAE/mL. It was observed that total polyphenol content exhibited a significant relation with concentrations at p<0.05. The electron-donating ability of Jeolpyun containing 8% S. glauca (Bunge) Bunge extract was the highest with a value of 83.93±0.69%. The electron-donating abilities revealed significant differences between concentrations (p<0.05). The superoxide radical scavenging ability of the blank was 0.01523±0.00068 and Jeolpyun containing 6% S. glauca (Bunge) Bunge powder was the highest with a value of 0.00977±0.00842. The hydrogen peroxide decomposing ability of Jeolpyun containing 0% S. glauca (Bunge) Bunge powder was -0.00377±0.0049; 8% S. glauca (Bunge) Bunge powder revealed the highest decomposing ability of -0.0476±0.0234. Hydrogen peroxide decomposing abilities revealed significant differences between concentrations (p<0.05). Jeolpyun containing S. glauca (Bunge) Bunge powder and extract demonstrated improved antioxidative activities.

Radical Scavenging Activities of Korean Traditional Rice Wine, Takju

  • The principal objective of this study was to assess the radical scavenging activities and total polyphenols, flavonoids, and flavonol contents of takju, a Korean traditional rice wine. The antioxidant properties of the wine and takju samples were evaluated using five distinct assays: specifically, 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2-azino-di-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS); hydroxyl; superoxide anion; and nitric oxide (NO) radical-scavenging activity assays. In this study, the takju evidenced strong scavenging activities against the hydroxyl, superoxide anion, and NO radicals. Furthermore, the total polyphenol contents of T-2 were similar to those previously observed in white wine (p<0.05). However, the flavonoids and flavonol contents of all takju samples were significantly (p<0.05) lower than that of white wine. The results of this study show that takju possesses a powerful radical scavenging activity against a variety of oxidative systems. The findings of this study also indicate that takju should be considered a useful antioxidant, and that their functional compound reduces oxidative stress.

An Antioxidant Homo-Flavoyadorinin-B from Korean Mistletoe (Viscum album var. coloratum) (한국산 겨우살이(Viscum album var. coloratum)로부터 분리한 homo-flavoyadorinin-B의 항산화 활성)

  • An antioxidant was isolated from Korean mistletoe (Viscum album var. coloratum) by consecutive purification using silica gel, Sephadex LH-20, and RP-HPLC. The active principle was identified as homo-flavoyadorinin-B (3',7-dimethoxyluteolin-4'-O-[apiosyl $(1{\rightarrow}2)$ glucoside]) by spectral analyses. It inhibited 74.6% of hydroxyl radical and 30.6% of superoxide anion radical at 0.01 mM; however, th~compound did not show any scavenging activity against hydrogen peroxide radical. At 0.1 mM, above compound scavenged superoxide anion radical about twice as effective as positive controls, BHT and ${\alpha}-tocopherol$. Radical scavenging activities of homo-flavoyadorinin-B on DPPH, hydroxyl, and hydrogen peroxide radicals were almost same with those of positive controls.

Research in the antioxidant of Phellinus linteus mycelia

  • Phellinus linteus mycelia have many pharmacological effects, although their pharmacological efficacy principles have not been known yet. In the course of screening for biological activity of the extracts of Phellinus linteus mycelia, we found strong antioxidative activity in some fraction of water-insoluble. Therefore, we tried to isolate the active principle(s) from the extract. The isolation of the active compound was guided by superoxide anion radical scavenging activity. As a result, caffeic acid was isolated as an active compound. The IC$\_$50/ of the compound was 3.05 $\mu$g/ml (16.9$\mu$M).
