Extraction of Resveratrol Containing Grade Seed Oil with Supercritical Carbon Dioxide (초임계 이산와탄소를 이용한 Resveratrol 함유 포도씨유 추출)

  • Grape seed oil made by press or organic solvent extraction does not contain resveratrol, a bioactive compound. Supercritical carbon dioxide could extract oil containing resveratrol from grape seed. The extraction efficiency was mainly dependent on the water content in grape seed. More resveratrol was contained in the oil extracted with un-dried grape seed. No resveratrol was extracted with dried grape seed. Time course changes of grape seed oil extraction also resulted that resveratrol could be extracted by supercritical carbon dioxide with the positive influence of water.

  • The effects of modifiers such as methanol, water diethylamine in methanol (10 v/v %), and diethylamine in water (10 v/v %) were investigated at three different concentrations (1, 5, and 10 v/v %) of the modifiers in supercritical $CO_2$ (SC-$CO_2$) in order to enhance the supercritical fluid extraction (SFE) efficiency of cephalotaxine from Cephalotaxus wilsoniana Leaves. Among the modifiers employed, methanol basified with diethylamine was found to greatly enhance the extraction efficiency relative to any other modifiers employed. The results suggest that cephalotaxine in plant matrices may be readily changed from SC-$CO_2$-insoluble salt to SC-$CO_2$-soluble free base by basified modifiers. In addition, SC-$CO_2$modified with basified methanol could enhance the extraction efficiency of cephalotaxine more than 30% when compared to the conventional organic solvent extraction.

  • Acer tegmentosum is a tree used to treat various liver diseases in Korea. There have been some concern regarding the safety of Acer tegmentosum due to some toxic chemical compounds in its stems. Supercritical fluid extraction (SFE) was employed to develop a removing method of toxic compounds from Acer tegmentosum. The toxic compounds were effectively extracted with ethanol modified supercritical fluid $CO_2$. The optimum condition of SFE was 100 bar of pressure, $40^{\circ}C$ of extraction temperature, 3 mL/min of $CO_2$ flow rate, 0.2 mL/min of modifier (ethanol) flow rate.

  • Pressure and flowrate control systems for a supercritical extraction process are designed and analyzed. To do this, the dynamic model was first set up using the performance equations of control valves, CO2 compressor and the state equation of the supercritical fluid. Using this model, optimum pairs of manipulated and controlled variable which give least steady stat interaction are determined though the relative gain analysis.

Quality and characteristics of fermented ginseng seed oil based on bacterial strain and extraction method

  • Background: In this study, the fermentation of ginseng seeds was hypothesized to produce useful physiologically-active substances, similar to that observed for fermented ginseng root. Ginseng seed was fermented using Bacillus, Pediococcus, and Lactobacillus strains to extract ginseng seed oil, and the extraction yield, color, and quantity of phenolic compounds, fatty acids, and phytosterol were then analyzed. Methods: The ginseng seed was fermented inoculating 1% of each strain on sterilized ginseng seeds and incubating the seeds at $30^{\circ}C$ for 24 h. Oil was extracted from the fermented ginseng seeds using compression extraction, solvent extraction, and supercritical fluid extraction. Results and Conclusion: The color of the fermented ginseng seed oil did not differ greatly according to the fermentation or extraction method. The highest phenolic compound content recovered with the use of supercritical fluid extraction combined with fermentation using the Bacillus subtilis Korea Food Research Institute (KFRI) 1127 strain. The fatty acid composition did not differ greatly according to fermentation strain and extraction method. The phytosterol content of ginseng seed oil fermented with Bacillus subtilis KFRI 1127 and extracted using the supercritical fluid method was highest at 983.58 mg/100 g. Therefore, our results suggested that the ginseng seed oil fermented with Bacillus subtilis KFRI 1127 and extracted using the supercritical fluid method can yield a higher content of bioactive ingredients, such as phenolics, and phytosterols, without impacting the color or fatty acid composition of the product.

  • Metal ions such as Cu and Cr were extracted from the Cu, Cr and Ag impregnated active carbon by contacting the solid surfaces with supercritical carbon dioxide(Sc-$CO_2$) containing chelating agents. About 10g of the active carbon sample were loaded in a vertical tube extractor contacting with $CO_2$ flowing from the bottom of the tube for 6hrs. The ligands used were acetyl acetone(AA) and Cyanex-302(C-302). Water and methanol were used as entrainers to study the effect of co-solvent to $CO_2$. Experimental results showed that C-302 was more effective than AA in removing Cu with the maximum extraction of 42.0wt%, while 57.6wt% of Cr was extracted with AA from the sample.

Comparison of Antimicrobial and Antioxidant Activities by Different Extraction Methods in Korean Bamboos (한국산 대나무의 추출방법에 따른 항균 및 항산화 특성)

  • To develop potential cosmetic ingredients with antimicrobial and antioxidant activities of 4 Korean bamboo species (P. bambusoides, P. nigra var. henonis, P. pubescens and Sasa coreana) using three different extraction methods-water, ethanol and supercritical fluid extraction. Antimicrobial activities and DPPH assay have been examined. Among the antimicrobial activities against two test strains, Escherichia coli and Staphylococcus aureus, ethanol extracts of 3 bamboo trees, P. bambusoides, P. nigra var. henonis, and P. pubescens, showed stronger than those of supercritical extracts. However, 4 bamboo supercritical extracts showed dose-dependent increase in antioxidant activity by DPPH assay. These results suggest that water fraction of bamboo extracts may be useful for the cosmetic ingredient with low cytotoxicity.

  • In order to maximize the utility of vacuum residue, supercritical solvent extraction technique where n-pentane was used as a supercritical solvent was applied to obtain deasphalted oil from vacuum residue. Oil-extraction yield at various temperatures and pressures and the contents of metal complex and sulfur of extracted oil were investigated. In supercritical state, extraction yield of deasphalted oil was found to be strongly dependent on the n-pentane density, and the metal complex content of extracted oil was effectively lowered when compared with that of vacuum residue. However, the sulfur content of extracted oil showed little difference when compared with that of vacuum residue.

Coffee Deodorization with Supercritical Carbon Dioxide (초임계 이산화탄소를 이용한 커피의 탈취)

  • Supercritical carbon dioxide was used to remove coffee odors. The odor removal efficiency was tested with coffee drink prepared by the espresso extraction method. Five typical odors in coffee were analyzed with GC and these odors in deodorized coffee were compared to those in control. Supercritical carbon dioxide extraction conditions were optimized as 350 bar and 70$^{\circ}C$ because the solvating power of supercritical fluid is depend on the density which is determined by temperature and pressure. A modified head space method was applied to collect coffee odors in coffee drink prepared by the espresso extraction method. Odors generated in coffee drink made with deodorized coffee powder were reduced by 73% in total mass of typical five coffee odors.

  • This study is about a technique for obtaining collagen by extracting fat by treating collagen-containing liposuction effluent in the presence of supercritical fluid. Using a supercritical solvent, a collagen extract could be obtained from animal-derived fat in a short time (about 6 hours), and about 2-3% of collagen by mass compared to the raw material could be obtained. The presence of collagen in the extract obtained by supercritical extraction was confirmed by SDS-PAGE, and it was confirmed that it was type 1 collagen having a relatively large molecular weight. In addition, the growth factors of IGF-1, bFGF, VEGF and NGF were analyzed to find out which growth factors were present in the collagen obtained by supercritical extraction, and it was found that these growth factors were contained in the extract. There was no significant difference in DNA content per mg of sample before and after supercritical treatment. Further in-depth studies are likely to be needed on decellularization technology using the supercritical process. In conclusion, the extracellular matrix obtained through the solvent extraction process using a supercritical fluid contains growth factors above a certain amount even after decellularization and removal of fat, so that it was found that not only biocompatibility is greatly increased, but also tissue regeneration can be rapidly induced.
