The Generation Mechanism of Radical in the Silver-ion-fluted Aqueous Solution from A-type Zeolite Supporting Silver tons and Its Applications (은 이온교환 A 형 제오라이트에 의한 라디칼 발생메카니츰과 그 응용에 관한 연구)

  • We confirm that when A-type zeolite supporting silver lone is placed in ion-exchanged distilled water, silver ions is eluted and eluted silver ions generates hydroxyl-radical (.OH) and hydro-radical (.H) continuously, the amount of those is proportion to the silver-ion concentration. Hydroxyl-radical is not generated by super-oxide anion-radical (.O2) but by directly dissolved water. To know such a above discussed mechanism, we prepare A-type zeolite supporting silver ions, and measure the amount of the eluted silver tons by atomic absorption spectroscopy and the generated radical by ESR The radical generated by A-type zeolite supporting silver ions is discussed in the application of elecrical and electronic materials.

An Antioxidant Hispidin from the Mycelial Cultures of Phellinus linteus

  • In the course of screening for reactive oxygen species scavengers from natural products, an antioxidant was isolated from the mycelial culture broth of Phellinus linteus and identified as hispidin. The hispidin content was reached its maximum level at 12 days after onset of inoculation. About 2.5 mg/mL of hispidin was produced by P linteus in a yeast-malt medium (pH 5.8, $25^{\circ}C$). Hispidin inhibited 22.6 and 56.8% of the super oxide anion radical, 79.4 and 95.3% of the hydroxyl radical, and 28.1 and 85.5% of the DPPH radical at 0.1 and 1.0 mM, respectively. The positive control ${\alpha}-tocopherol$ scavenged 25.6 and 60.3%, 74.6 and 96.3%, and 32.7 and 77.5% of each radical, respectively, at the same concentrations. However, hispidin showed no significant activity on the hydrogen peroxide radical.

Antioxidant and Oxidative DNA Damage Protection Potential of Methanol Extract of Red Tea Stem

  • This study was undertaken to determine free radical scavenging capacity and oxidative DNA damage protecting activity of methanol extract of red tea stem. The extract was subjected to assess their antioxidant potential using various in vitro systems such as $DPPH^{\bullet}$, $ABTS^{{\bullet}+}$, super oxide and nitric oxide free radicals and it exhibited $IC_{50}$ values of $68.88{\pm}1.1$, $12.08{\pm}0.65$, $404.38{\pm}1.6$, $93.6{\pm}2.7{\mu}g/mL$ respectively. Red tea extract also showed ferric reducing ability (FRAP) with 2606.85 mmol Fe (II)/g of extract. Furthermore, Methanol extract of red tea stem showed significant DNA damage protecting activity in concentration dependent manner against $H_2O_2+UV$ induced photolysis on pUC19 plasmid DNA. Results of this study showed that the methanol extract of Red Tea stem has strong antioxidant potential along oxidative DNA damage protecting capacity that would be the significant sources of natural antioxidants, which might be helpful in preventing the progress of various oxidative stress generated diseases. Further study is necessary for isolation and characterization of the active antioxidants, which may serve as a potential source of natural antioxidant.

Flavonol Glycosides from the Aerial Parts of Aceriphyllum rossii and Their Antioxidant Activities

  • The methanol extract obtained from the aerial parts of Aceriphyllum rossii (Saxifragaceae) was fractionated into ethyl acetate (EtOAc), n-BuOH and $H_2O$ layers through solvent fractionation. Repeated silica gel column chromatography of EtOAc and n-BuOH layers afforded six flavonol glycosides. They were identified as kaempferol 3-O-$\beta$-D-glucopyranoside (astragalin, 1), quercetin 3-O-$\beta$-D-glucopyranoside (isoquercitrin, 2), kaempferol 3-O-$\alpha$-L-rhamnopyranosyl $(1{\to}6)-\beta$-D-glucopyranoside (3), quercetin 3-O$\alpha$-L-rharnnopyranosyl $(1{\to}6)-\beta$-D-qlucopyrano-side (rutin, 4), kaempferol 3-O-[$\alpha$-L-rharnnopyranosyl $(1{\to}4)-\alpha$-L-rhamnopyranosyl $(1{\to}6)-\beta$-D-glucopyranoside] (5) and quercetin 3-O-[$\alpha$-L-rhamnopyranosyl $(1{\to}4)\alpha$-L-rhamnopyranosyl $(1{\to}6)\beta$-D-glucopyranoside] (6) on the basis of several spectral data. The antioxidant activity of the six compounds was investigated using two free radicals such as the ABTS free radical and superoxide anion radical. Compound 1 exhibited the highest antioxidant activity in the ABTS $\{2,2-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid)\}$ radical scavenging method. 100 mg/L of compound 1 was equivalent to $72.1\pm1.4\;mg/L$ of vitamin C, and those of compounds 3 and 5 were equivalent to $62.7\pm0.5\;mg/L$ and $54.3\pm1.3\;mg/L$ of vitamin C, respectively. And in the superoxide anion radical scavenging method, compound 5 exhibited the highest activity with an $IC_{50}$ value of $17.6{\pm}0.3{\mu}M$. In addition, some physical and spectral data of the flavonoids were confirmed.

Antioxidative Effects of Polyozellin and Thelephoric Acid Isolated from Polyozellus multiflex (까치버섯(Polyozellus multiplex)으로부터 분리한 polyozellin과 thelephoric acid의 항산화 활성)

  • In the course of screening for reactive oxygen species scavengers from natural products, two antioxidants were isolated from the edible mushroom Polyozellus multiflex and identified as polyozellin and thelephoric acid. Thelephoric acid inhibited 45.7% of the super oxide anion radical, 74.6% of the hydroxyl radical, and 44.0% of the DPPH radical at 0.1 mM, while the positive control ${\alpha}-tocopherol$ did 22.1%, 75.6%, and 26.5% of each radical, respectively. Polyozellin, the reductive acetylated form of thelephoric acid, showed almost same scavenging activity against above mentioned radicals. The isolated compounds showed scavenging activity on the superoxide anion radical in the ESR method ($IC_{50}$ of polyozellin and thelephoric acid were $218.0\;{\mu}M$ and $21.1\;{\mu}M$, respenctively). However, they showed no significant activity on the hydrogen peroxide radical.

Growth Characteristic, Mono-strain Mass Culture and Antioxidant Effects of Two Benthic Diatoms Amphora coffeaeformis and Achnanthes longipes from Korea

  • Amphora coffeaeformis and Achnanthes longipes are commonly found as dominant benthic microalgae in Jeju coastal water throughout the year. In order to investigate pharmaceutical uses of these diatoms, each single species was isolated with micropipette under phase contrast microscope and subcultured with synthetic seawater media which was enriched with F/2 media, trace metal solution and $Na_2SiO_3$). Growth characteristics of these species were also determined with different combination of salinity, nutrients concentration and temperature. Thereafter, mass culture of each species was done based on the maximum growth condition. Biomass was collected after two weeks of mass culture and freeze dried for antioxidant study. The antioxidant properties of different fractions (n-hexane, chloroform and ethylacetate) obtained by solvent fractionation of 80% methanolic extract of two microalgae were investigated for free radical, reactive oxygen species scavenging (Super oxide, Hydrogen peroxide, Hydroxyl radical and Nitric oxide), metal chelating and lipid peroxidation inhibition activities. All fractions of A. longipes showed higher $DPPH^{\cdot}$ (free radical) scavenging activities (n-hexane: 89.0%, Chloroform: 76.0%, Ethylacetate: 66.0%, Methanol: 90.6% and aqueous residue: 63.0%). N-hexane fraction of A. longipes showed significantly higher activity (49.0%) on nitric-oxide. Ethylacetate fraction of A. longipes and aqueous residue of A. coffeaeformis exhibited 64.0% and 75.6% metal chelating activity which was higher than commercial antioxidants (${\alpha}$-tocopherol: 18.0% and BHT: 16.0%). The n-hexane fraction of A. coffeaeformis had 67.5% activity on $DPPH^{\cdot}$. Chloroform and n-hexane fractions of A. coffeaeformis exhibited 46.2% and 47.6% $H_2O_2$ scavenging effects which were closely similar to commercial antioxidants (${\alpha}$-tocopherol: 49.2% and BHT: 58.6%). Chloroform and ethylacetate fractions of A. longipes and fraction of n-hexane and chloroform of A. coffeaeformis showed better lipid peroxidation activities than ${\alpha}$-tocopherol. These data suggest that both organic and aqueous fractions have good antioxidative compounds with different antioxidant properties.

  • This experiment was designed to investigate the effects of So-Oochim-tang-Gagam-bang (SOCT-G) on oxidative stress and serotonin metabolism in P815 Mast Cells The effects of SOCT-G on activity of 2,2-Diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging and Super Oxide Dismutase (SOD) in P815 mast cells were investigated. The effect of SOCT-G on content of serotonin in P815 mast cells was investigated. The effects of SOCT-G on expression of 5-hydroxytryptamine transporter (5-HTT), Tryptophan hydroxylase 1 (TPH-1) mRNA in P815 mast cells were investigated. The SOCT-G increased DPPH radical-scavenging activity in P815 mast cells. The SOCT-G increased SOD activity in P815 mast cells. The SOCT-G decreased the intracellular content of serotonin in P815 mast cells. The SOCT-G decreased 5-HTT and TPH-1 mRNA expression in P815 mast cells. This experiment shows that So-Ochm-Tang-Gagam-bang has a significant effect of oxidative stress that help prevent free radical damage. And So-Ochim-Tang-Gagam-bang decreased the intracellular content of serotonin and mRNA expression of 5-HTT and TPH-1. Therefore, further researches are suggested to reveal the anti-depressive effectiveness of So-Ochim-Tang-Gagam-bang.

Free Radical Scavenging Activities and Inhibitory Effects on Xanthine Oxidase by Ethanol Extract from Capsella bursa-pastoris (냉이(Capsella bursa-pastoris)에탄을 추출물의 유리라디칼 소거 및 Xathine Oxidase 저해활성)

  • To examine the characteristics of antioxidative compounds from Capsella bursa-pastoris, ethanol extracts were separated into five organic solvent fractions; hexane(Fr.H), diethyl ether (Fr.E), ethyl acetate(Fr.EA), butanol (Fr.B), and water(Fr.D) fractions. Fr.B showed the greatest electron donating ability and inhibitory effect on lipid peroxidation. Whereas Fr.E had the most excellent activity in the superoxide radical scavenging activity by xanthine/xanthine oxidase-cytochrome c reduction system. The inhibitory effect of each fraction on xanthine oxidase was also measured. Fr.E had the strongest inhibitory effect on xanthine oxidase and $IC_{50}$ was $5.65\;{\mu}g$. The results indicate that the superoxide radical scavenging activity of Fr.E is caused by the inhibitory effect on radical generating system of xanthine oxidase. Also the order of inhibitory effect on xanthine oxidase was Fr.B

Effect of p-Phenylenediamine to Rat Skin (p-Phenylenediamine이 흰쥐의 피부 조직에 미치는 영향)

  • Red brownish p-pheylenediamine (PPD) has been widely used hair dye for women. The dye was known to cause systemic anaphylaxis, dermatitis and bladder cancer. But the effect of PPD toxicity with oxygen free radical has not been studied. This study investigated the degree of skin injury by PPD. PPD ($2.5\%$ PPD in $2\%\;NH_{4}OH$) was applied to the rat skin ($25 mg/16.5\;cm^2$) 3 or 5 times every other day. Histopathological findings demonstrated the proliferation of epithelial cells and the increased keratinization by PPD. The activities of glucose 6-phosphatase (G6Pase) was decreased and acid phosphatase (ACP) was increased in PPD-applied rat skin. Groups in which PPD was applied 5 times were more damaged than groups applied 3 times. To examine the relationship between tissue damage and oxygen free radicals, effect of PPD on xanthine oxidase (XO) activity was measured and XO activity was more significantly increased in the group treated with PPD 5 times than 3 times. However, reduced glutathione (GSH) content, and the activities of catalase (CAT), super-oxide dismutase (SOD) and glutathione S -transferase (GST) were more decreased in PPD-applied groups than in controls. Even though the activities of XOD was not changed in the group treated with PPD 3 times, the decreased activities of oxygen free radical system and the damaged skin tissue were observed. This result might be caused by the production of toxic PPD metabolites in rat skin. In conclusion, topical PPD application led to skin injury in a dose-dependent manner, probably due to the generation rate of oxygen free radical.

Study of Antioxidant Effects of Platycladi Semen on Male Reproductive cells (백자인(柏子仁)이 수컷 생쥐의 생식세포에 미치는 항산화 효과 연구)

  • Objectives : The purpose of this study is to examine the antioxidant effects on male mouse reproductive cells of the extract of Platycladi Semen. Methods : The extract was studied for diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity, cell viability by a modified MTT assay, the effects on $H_2O_2$-induced cytotoxicity by MTT assay, lipid peroxidation by malondialdehyde (MDA) formation and super oxide dismutase (SOD), respectively. Results : The results showed that the extract scavenged DPPH radical in a dose-dependent manner by up to 74.87%. The cell viability of the extract was within 72~96% on Leydig cells and GC-2 cells at concentrations of 1, 5, 10, 50 and 100 ug/ml. The hydrogen peroxide-induced cytotoxicity of Leydig cells was protected to 72.09% by the extract at concentration of 100 ${\mu}g/ml$. The hydrogen peroxide-induced lipid peroxidation of MDA formation was decreased to 1.80 and 1.65 nmoles/mg protein by the extract at concentrations of 50 and 100 ${\mu}g/ml$. The extract at all concentrations, SOD activity was not significantly changed. Conclusions : In conclusion, the extract of Platycladi Semen has antioxidant effects on Leydig cells and protect male reproductive system against oxidative stress.
