Antibiosis against Super Bacteria from Natural Dyeing with Elm Bark Extract (느릅나무껍질 추출액을 이용한 천연염색의 슈퍼박테리아에 대한 항균성)

  • In this study, a cotton knit was dyed with elm bark extract; subsequently, the dyed fabric was measured according to the types of mordants and the preprocessing cationizers used. Additionally, antibiosis against super bacteria was examined. The results follow. First, the color of the dyed cotton knit appeared reddish and yellowish for fabrics treated with non-mordants and mordants. When preprocessing with a cationizer was conducted, the dyeing properties were the best. Second. even when mordants were not used for dyeing, color fastness after washing, sweating, and rubbing was generally good Grade 4 and 5. Color fastness after exposure to sunlight was the best Grade 4 for fabric prepared with ferrous sulfate as the mordant. Third. as for antimicrobial properties, or resistance to super bacteria, the growth of bacteria was suppressed in a meaningful way for fabrics treated with non-mordants and mordants, compared to the control group fabric. The dyeing methods with the most powerful antimicrobial effects were dyeing after preprocessing with a cationizer and preparing fabric with copper sulfate as the mordant. The results stated above show that in case of dyeing with elm bark extract, preprocessing of the cotton knit with a cationizer and dying with copper mordant displayed high levels of antimicrobial properties that were useful for resisting super bacteria. Of these the dyeing properties were the best when preprocessing with a cationizer.

Effect of Ulmus Davidiana var. Japonica Nakai Extract on Antibiotic Resistant Bacteria in Dyed Cotton (유백피 추출액을 이용한 염색 면포의 항생제 내성균주 증식 억제효능)

  • This research verifies stainability, super bacteria antibacterial features and antibiotic resistance of Ulmus davidiana var. japonica Nakai (UD) extract in dye cotton cloth. UD was extracted with hot water, the test cloth dyed, and then processed by mordanting treatment using $FeSO_4$ $7H_2O$, $CuSO_4$ $5H_2O$ and $Al_2(SO_4)_3$. The surface color and color fastness of the three kinds (original cloth, the cloth without mordancy and mordant cloth) were measured and the influence of dying of cotton cloth and the mordant method on the genesistasis of Methicillin-resistant Staphylococcus aureus (MRSA), antibiotic resistant bacteria. 1. The surface color of the cotton cloth for dyeing (without and with mordancy) displayed a reddish and yellowish color. Stainability was greatest when the mordant of $FeSO_4$ $7H_2O$ was used. 2. When a mordant was not used for dyeing, the color fastness to washing, perspiration and friction of the contaminated cloth was satisfactory at 4 to 5 grade in general. 3. As for the antibiotic effect to super bacteria, the growth of germ was meaningfully suppressed both on the cloth without and with mordancy compared to the comparison cloth; in addition, the dyeing method with the biggest antibacterial impact was found to be the mordancy with the mordant of $CuSO_4$ $5H_2O$ after dyeing. The results of the experiments that involved dyeing with UD extract showed that cotton cloth processed through mordancy with the mordant of $CuSO_4$ $5H_2O$ had the biggest antibiosis to super bacteria and that processed with the mordant of $FeSO_4$ $7H_2O$ had the greatest stainability.

Biosynthesis of Polymyxins B, E, and P Using Genetically Engineered Polymyxin Synthetases in the Surrogate Host Bacillus subtilis

  • The development of diverse polymyxin derivatives is needed to solve the toxicity and resistance problems of polymyxins. However, no platform has generated polymyxin derivatives by genetically engineering a polymyxin synthetase, which is a nonribosomal peptide synthetase. In this study, we present a two-step approach for the construction of engineered polymyxin synthetases by substituting the adenylation (A) domains of polymyxin A synthetase, which is encoded by the pmxABCDE gene cluster of Paenibacillus polymyxa E681. First, the seventh L-threonine-specific A-domain region in pmxA was substituted with the L-leucine-specific A-domain region obtained from P. polymyxa ATCC21830 to make polymyxin E synthetase, and then the sixth D-leucine-specific A-domain region (A6-D-Leu-domain) was substituted with the D-phenylalanine-specific A-domain region (A6-D-Phe-domain) obtained from P. polymyxa F4 to make polymyxin B synthetase. This step was performed in Escherichia coli on a pmxA-containing fosmid, using the lambda Red recombination system and the sacB gene as a counter-selectable marker. Next, the modified pmxA gene was fused to pmxBCDE on the chromosome of Bacillus subtilis BSK4dA, and the resulting recombinant strains BSK4-PB and BSK4-PE were confirmed to produce polymyxins B and E, respectively. We also succeeded in constructing the B. subtilis BSK4-PP strain, which produces polymyxin P, by singly substituting the A6-D-Leu-domain with the A6-D-Phe-domain. This is the first report in which polymyxin derivatives were generated by genetically engineering polymyxin synthetases. The two recombinant B. subtilis strains will be useful for improving the commercial production of polymyxins B and E, and they will facilitate the generation of novel polymyxin derivatives.

Toward Complete Bacterial Genome Sequencing Through the Combined Use of Multiple Next-Generation Sequencing Platforms

  • PacBio's long-read sequencing technologies can be successfully used for a complete bacterial genome assembly using recently developed non-hybrid assemblers in the absence of second-generation, high-quality short reads. However, standardized procedures that take into account multiple pre-existing second-generation sequencing platforms are scarce. In addition to Illumina HiSeq and Ion Torrent PGM-based genome sequencing results derived from previous studies, we generated further sequencing data, including from the PacBio RS II platform, and applied various bioinformatics tools to obtain complete genome assemblies for five bacterial strains. Our approach revealed that the hierarchical genome assembly process (HGAP) non-hybrid assembler resulted in nearly complete assemblies at a moderate coverage of ~75x, but that different versions produced non-compatible results requiring post processing. The other two platforms further improved the PacBio assembly through scaffolding and a final error correction.

  • In the field of medical and marine industries, antibacterial surfaces have been emerged as one of the most important issues. Recently, many researchers have been studying antibacterial surfaces to kill bacteria or prevent the adhesion of bacteria. In their researches, various materials and structures are suggested to inhibit the adhesion of bacteria or kill the attached bacteria. However, chemical materials such as antibiotics or metal could be toxic. Moreover, frequent use of antibiotics causes super bacteria having resistance to antibiotics. In this study, nano-pillar structured surface was fabricated using polyurethane acrylate (PUA) and the mechanically induced antibacterial function was confirmed based on the fabricated nanostructures. Nanostructures can damage the bacterial membrane of Gram-negative bacteria through stretching of bacterial membrane via interaction with the nanostructures and the bacterial membrane. Consequently, the proposed transparent, flexible and nanostructured PUA films can be one of promising candidates for antifouling and antibacterial surfaces which can be applied in various industries.

In vitro antibacterial activities of Pozzolan as a dietary silicate minerals supplementation to animals (가축에 대한 보조사료 규산염제로서 포졸란의 in vitro 항균 효과)

  • The purpose of this study was to investigate in vitro antibacterial activities of pozzolan against super bacteria and intestinal bacteria. There were four treatment groups: 1) CON, pozzolan free control group; 2) DP0.3, microbial culture medium prepared by mixing distilled water and 0.3% of pozzolan powder; 3) DP0.5, microbial culture medium prepared by mixing distilled water and 0.5% of pozzolan powder; and 4) PE, microbial culture medium prepared with pozzolan powder extracts without adding distilled water. The count of Lacctobacillus casei was significantly higher in the DP0.3 group compared to CON (P<0.05). However, it showed no significant difference compared to other treatment groups. Numbers of Clostridium butyricum, Escherichia coli, and Salmonella typhimurium were significantly lower in pozzolanic treatment groups compared to CON (P<0.05). Clostridium butyricum and Salmonella typhimurium counts were significantly different among DP0.3, DP0.5, and PE groups (P<0.05). Counts of E. coli were also significantly between DP0.5 and PE groups (P<0.05). Counts of MRSA and VRE were significantly lower in pozzolanic treatment groups compared to CON (P<0.05). MRSA counts were significantly different among DP0.5, DP0.3 and PE groups. VRE counts were significantly higher in the order of PE > DP0.3> DP0.5> CON (P<0.05). These results suggest that pozzolan could be used as a dietary silicate supplement and a natural antibacterial agent for livestock if its antimicrobial activity against super bacteria and harmful bacteria in the intestine is confirmed.

Dietary Value of Live Food for the Seedling Production of the Sweetfish, Plecoglossus altivelis (은어, Plecoglossus altivelis 종묘생산을 위한 먹이생물학적 연구)

  • For the effective seedling production of the sweetfish, Plecoglossus altivelis, dietary value of live food in the growth and survival rate of the larvae was examined. To investigate dietary value of three types of rotifers (L-type, S-type, Us-type rotifer) for the sweetfish larvae, the larvae were fed with alone or mixed with each type and with mixtures of two or three types. Although the survival rates of the larvae fed different types of rotifers were not significantly different among them, the larvae fed a mixture of three types of rotifers showed the highest growth significantly. A feeding trial was conducted to determine the effect of the dietary values of rotifer and Artemia nauplius enriched with Chlorella ellipsoidea, $\omega-yeast$, spray-dried Spirulina platensis, and Super Selco for the sweetfish larvae. The dietary values of rotifer and Artemia nauplius enriched with Super Selco mixed with spray-dried S. platens is were distinctly improved for the survival rate and growth of the larvae. With regard to effect of the green water with Chlorella ellipsoidea, spray-dried Spirulina platensis, and the addition of photosynthetic bacteria (Rhodopseudomonas capsulatus), the green water with Spirulina platens is mixed with photosynthetic bacteria also enhanced the growth and survival rate of the larvae.

Determination of Shelf-life of a Packaged Paralichthys Olivaceus for Super-chilled Distribution (슈퍼칠링 유통을 위한 포장광어 (Paralichthys Olivaceus)의 유통기한 설정 연구)

  • Maintaining steady and low temperature during distribution process is a key technique to inhibit microbial growth, one of the most important factors in determining the shelf life. It is possible to provide high-quality fresh products to consumers only when precise and adequate temperature control is guaranteed throughout the entire distribution process for fresh seafood. This study investigated the shelf-life of packaged fresh flounder (Paralichthys Olivaceus) in order to learn the feasibility of super-chilled distribution of fresh seafood. To estimate the shelf life, weight, number of bacteria such as E. coli, pH, sensory test and volatile basic nitrogen were investigated. As a result of the study, the difference in shelf life of 6 days at the super-chilling temperature (0±1℃) and 1 day at the general refrigeration temperature (8±2℃) (based on volatile base nitrogen) showed the market possibility of super-chilling distribution. Through additional empirical studies such as packaging methods and economic feasibility, it is expected to promote commercialization of super-chilling containers and packaging system developed in the future and secure customer reliability.

The Distribution of Indicator Microorganisms and Identification of Antibiotic Resistant Strains in Domestic Animal Feces (가축 분변 유래 지표미생물 분포 및 항생제 내성 균주의 동정)

  • Objectives: To estimate the microbial contaminant load discharged from livestock farms, we randomly selected livestock farmers of cattle, swine, and fowl and collected bacterial strains from domestic animals' feces and compost samples. Recently, as multi-antibiotic-resistant bacteria and super bacteria showing resistance to a variety of antibiotics have been reported one after another, the ecological and health hazard of antibiotic-resistant bacteria is emerging as an important issue. Methods: Monitored indicator microorganism constituents were totak coliform (TC), fecal coliform (FC), and aerobic bacteria. The multi-antibiotic-resistant bacteria were identified from investigated indicator microorganisms by 16S rRNA sequencing. Results: By microbiological analysis, the largest population of aerobic bacteria ($1.5{\times}10^5$ CFU/g) was found in cattle fecal compost, and total coliforms ($1.1{\times}10^7$ CFU/g) and fecal coliforms ($1.0{\times}10^5$ CFU/g) were found primarily in swine fecal compost, while the lowest population was found in fowl fecal compost. Among the 67 strains separated from aerobic bacteria, five strains expressing high antibiotic resistance were selected in each sample. We found the multi-antibiotic resistant strains to be Shigella boydii, Staphylococcus lentus, Acinetobacter sp. and Brevibacterium luteolum. Conclusions: These results suggest that increasing numbers of multi-antibiotic-resistant bacteria in the environment have a close relation to the reckless use of antibiotics with livestock.

Quality Characteristics of Ginseng Treated by Hot Air Drying after Being Dried using Super-heated Steam (초급속 과열증기를 이용한 인삼의 품질 특성)

  • This research aims to develop a new drying technology using super-heated steam to reduce the time required for drying ginseng and to enhance its quality. The drying rate of ginseng powder treated with hot-air drying after drying with super-heated steam was about 40% faster than the drying rate of ginseng powder treated with hot-air drying. Minimal difference was found between hot-air drying and hot-air drying after drying with super-heated steam in the general composition of the ginseng powder, such as its crude lipid, crude protein, crude fiber, and ash. Moreover, no difference was found between hot-air drying and hot-air drying after drying with super-heated steam in terms of the ginseng powder's mineral and free sugar contents. The crude saponin contents was 5.43% in the ginseng treated with hot-air drying and 5.4% in the ginseng treated with hot-air drying after drying with super-heated steam. The ginseng powder that was treated with hot-air drying after drying with super-heated steam had a slightly higher redness compared to the ginseng powder that was treated with hot-air drying alone. The number of microorganisms of the ginseng powder treated with hotair drying after drying with super-heated steam showed a reduction of one log cycle in the total bacteria and two log cycles in the yeast molds compared to the ginseng powder treated with hot-air drying alone.
