UVB 광손상에 대한 백작약의 보호 효과 (Protective Effect of Paeoniae Radix Alba against UVB Photodamage)

  • Objective : UVB damages skin health by causing skin redness and intense inflammation, sunburn, and skin cancer. Paeoniae Radix Alba has been used to relieve gynecological symptoms, muscle spasms, and skin ailments. This study was conducted to confirm whether it has a protective effect against UVB photodamage. Methods : Ethanol extract of Paeoniae Radix Alba (PRA) was prepared by extracting 100 g Paeoniae Radix Alba in 1 L of ethanol for 48 h. Apoptosis was monitored by the terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay and expression levels of apoptosis indicator proteins, and tyrosinase activity was measured with a colorimetric commercial kit. Results : In human keratinocyte HaCaT cells, PRA reduced UVB-induced cell death through apoptosis by inhibiting PARP cleavage and caspase-3 and -9. UVB-induced increase in cellular reactive oxygen species (ROS) was suppressed by PRA pretreatment. PRA also showed dose-dependent ABTS and DPPH radical scavenging activities. Furthermore, the inhibitory effect of tyrosinase activity by PRA was confirmed. Conclusion : These results demonstrated the protective role of PRA in UVB photodamage of human keratinocytes, mainly due to its antioxidant and antiapoptotic properties. We also suggest that PRA can be considered as an effective natural agent to prevent skin photodamage.

흰쥐에 조사한 자외선B가 Nitric Oxide의 활성에 미치는 효과 (I) (Effects of ultraviolet light B irradiation on nitric oxide activity in the sprague-dawley rat in vivo (I))

  • Recent studies have revealed that Nitric oxide(NO) was one of the demonstration for the physiological regulator, endothelial derived relaxing factor(EDRF) and that NO was produced by ultraviolet irradiation in human. Thus, the present author have carried out a experimental study on the change of hematological, histological value of ultraviolet irradiation in sprague-dawley rats. The subjects were divided into four groups of ten rats each selected at random. There were 4 groups: 1. no irradiation control; 2. ultraviolet $75mJ/cm^2$ irradiation group; 3. ultraviolet $150mJ/cm^2$ irradiation group; 4. ultraviolet $225mJ/cm^2$ group. After a irradiation, hematological and histological tests were performed to observe erythrocyte, hemoglobin, hematocrit, MCH, MCHC, MCV, $O_2$ saturation, pH, $PO_2,\;PCO_2$ value and to observe histological changes. In hematological tests, erythrocyte, hemoglobin, hematocrit significantly increased in $75mJ/cm^2$ than control group and more $150mJ/cm^2$ ultraviolet irradiation group respectively. Also In blood gas tests, $PO_2$ significantly increased in $75mJ/cm^2$ and more $150mJ/cm^2$ group than control group. Whereas $PCO_2$ significantly decreased in $75mJ/cm^2$ and more $150mJ/cm^2$ group than control group (Duncan-Tukey test, P<0.05). In histological tests, control and $75mJ/cm^2$ group unchanged, but more $150mJ/cm^2$ group changed that it was cytolysis, cytotoxic effect, acanthosis, proliferation of keratinocyte, appearance of amorphous cell and pyknotic nucleus, production of sunburn cell. In conclusion, the present author results support the importance of the relation between NO effect and hematological, histological value by ultraviolet B irradiation.

대학생들의 화장습관에 따른 소변 중 벤조페논 농도 비교 (Urinary Concentrations of Benzophenones in University Students and Association with Cosmetics Habits)

  • Objectives: Ultraviolet (UV) radiation may cause skin cancer, photo-ageing, erythema, and sunburn. Benzophenone (BP) is commonly used to protect skin from UV radiation. In Korea, sunscreen, sunblock, socalled 'blemish balm' (BB) and 'color correcting' (CC) creams, and foundation may contain from 0.5 to 5% benzophenone in order to protect skin from UV radiation. The purpose of this study is to understand the levels of benzophenone derivatives in urine among a group of university students and identify the contribution of cosmetics use. Methods: Forty volunteers (20 women and 20 men) were asked about skin type, frequency of use of cosmetics, and recognition of related health effects, etc. in a survey. Subjects were divided into several subgroups and were compared for concentration of benzophenone-1 (BP-1) and benzophenone-3 (BP-3). Their urine was pretreated with enzyme hydrolysis and solid phase extraction. Determinations of BP-1 and BP-3 in the urine were made with LC-MS/MS. Results: Among the study subjects, 82.5% used basic cosmetics at least once per day, and 77.5% used sunscreens at least once per day. The concentrations of BP-1 and BP-3 of the males were 4.36 ng/mL and 9.16 ng/mL, respectively. Those of the females were 3.98 ng/mL and 5.07 ng/mL, respectively. The use of cosmetics was positively related to urinary benzophenone levels. Conclusions: BP-1 and BP-3 were widely detected among the university students. Cosmetic use was identified as a potential source. Implications of such exposure deserve further investigation.

Sun Exposure and the Risk of Prostate Cancer in the Singapore Prostate Cancer Study: a Case-control Study

  • Background: Most of the epidemiology studies on the effects of sun exposure and prostate cancer were conducted among the temperate countries of North America and Europe. Little is known about the influence on Asian populations. The purpose of current study was to evaluate any association of sun exposure with risk of prostate cancer in Chinese, Malays and Indians who reside in the tropics. Methods: The Singapore Prostate Cancer Study is a hospital-based case-control study of 240 prostate cancer incident cases and 268 controls conducted in Singapore between April 2007 and May 2009. Detailed information on outdoor activities in the sun, skin colour, sun sensitivity and other possible risk factors were collected in personal interviews. Cases were further classified by Gleason scores and TNM staging. Odds ratios (OR) and 95% confidence intervals (CI) were calculated using unconditional logistic regression analysis, adjusted for age, ethnicity, education, family history of any cancers, BMI and skin colour. Results: We found that prostate cancer risk was increased in subjects with black/dark-brown eyes (OR 5.88, 95%CI 3.17-10.9), darker skin colour e.g. tan/dark brown/black (OR 7.62, 95%CI 3.41-17.0), frequent sunburn in lifetime (OR 4.30, 95%CI 1.7-11.2) and increased general sun exposure in adulthood per week (OR 2.03, 95%CI 1.09-3.81). The increased risk was consistent for high grade tumours and advanced stage prostate cancers. Conclusion: The findings from this study suggest that excessive sun exposure is a risk factor for prostate cancer in Asians.

Sun Burn Incidence and Knowledge of Greek Elementary and High School Children about Sun Protection

  • Introduction: Overexposure to sun radiation and particularly its accumulation during childhood and adolescence is a significant risk factor for skin cancer development. The sun burn is particularly important. Aim: To estimate sun burn incidence in young pupils in a coastal area of Greece. Materials and Methods: Two surveys were conducted in a school population in the same district in Greece, over different periods of time, in young people 9 to 18 years old (n=2 977). Anonymous questionnaires were completed. Levels of significance were two-tailed and statistical significance was set at p=0.05. SPSS 17.0 software was used for statistical analysis. Results: From the individual characteristics of the participants it was shown that the majority of them had dark hair and fair skin, whereas a significant percentage reported the existence of moles on face and their body (83.4% vs 68.1%). The sun burn incidence was high in adolescents and the younger pupils (41.9% vs 55.6%). The younger aged children who were living in an urban area had significantly higher rates of sun burn than those living in semi-urban areas (33.8% vs 24.8%, p=0.020). As far as the knowledge of pupils about the risks of sun radiation it was shown that the elementary school pupils had better knowledge than those at high school. Finally, those with better knowledge had the fewer sun burns (Mean 2.83 SD 0.87, p<0.001). Conclusions: The contribution of knowledge to the decrease of sun burn incidence is important as long as this is continuous. Therefore, the education should concern not only children but also teachers and parents in the context of continuous and systematic programs of health education.

  • Exposure to ultraviolet (UV) radiation is associated with the development of adverse effects in skin. Among the three types of UV rays, UV-B causes the most damaging effects, inducing sunburn and penetrating the outer skin, resulting in DNA mutations and skin cancer. The objective of this study was to formulate a UV-protective film by incorporating Ecklonia cava extracts. Cells covered with the film were exposed to UV-B (50, 80, and $100mJ/cm^2$). To determine the protective effects of the film, we evaluated cell viability, intracellular ROS generation, and apoptosis. We found that all E. cava extracts absorbed UV light and exhibited protective effects against UV-B-induced photodamage. Among the protective films examined in this study, that incorporating an E. cava 70% ethanol extract (70EX) formed the most effective protection against UV-B in HaCaT cells. These findings suggest that the application of film containing E. cava extract could prevent UV-B-induced photodamage, and offer protection against the detrimental effects of UV radiation, thus maintaining physiological condition.

  • The skin exposure to solar irradiation and photoreactive xenobiotics may produce abnormal skin reaction, phototoxicity. Phototoxicity is an acute light-induced response, which occurs when photoreacive chemicals are activated by solar lights and transformed into products cytotoxic against the skin cells. Multifarious symptoms of phototoxicity are identified, skin irritation, erythema, pruritis, and edema that are similar to those of the exaggerated sunburn. Diverse organic chemicals, especially drugs, are known to induce phototoxicity, which is probably from the common possession of UV-absorbing benzene or heterocyclic rings in their molecular structures. Both UVB (290~320 nm) and UVA (320~400 nm) are responsible for the manifestation of phototoxicity. Absorption of photons and absorbed energy (hv) by photoactive chemicals results in molecular changes or generates reactive oxygen species and depending on the way how endogenous molecules are affected by phototoxicants, mechanisms of phototoxcity is categorized into two modes of action: Direct when unstable species from excited state directly react with the endogenous molecules, and indirect when endogeneous molecules react with secondary photoproducts. In order to identify phototoxic potential of a chemical, various test methods have been introduced. Focus is given to animal alternative test methods, i.e., in vitro, and in chemico assays as well as in vivo. 3T3 neutral red uptake assay, erythrocyte photohemolysis test, and phototoxicity test using human 3-dimensional (3D) epidermis model are examples of in vitro assays. In chemico methods evaluate the generation of reactive oxygen species or DNA strand break activity employing plasmid for chemicals, or drugs with phototoxic potential.

Phloroglucinol Attenuates Ultraviolet B-Induced 8-Oxoguanine Formation in Human HaCaT Keratinocytes through Akt and Erk-Mediated Nrf2/Ogg1 Signaling Pathways

  • Ultraviolet B (UVB) radiation causes DNA base modifications. One of these changes leads to the generation of 8-oxoguanine (8-oxoG) due to oxidative stress. In human skin, this modification may induce sunburn, inflammation, and aging and may ultimately result in cancer. We investigated whether phloroglucinol (1,3,5-trihydroxybenzene), by enhancing the expression and activity of 8-oxoG DNA glycosylase 1 (Ogg1), had an effect on the capacity of UVB-exposed human HaCaT keratinocytes to repair oxidative DNA damage. Here, the effects of phloroglucinol were investigated using a luciferase activity assay, reverse transcription-polymerase chain reactions, western blot analysis, and a chromatin immunoprecipitation assay. Phloroglucinol restored Ogg1 activity and decreased the formation of 8-oxoG in UVB-exposed cells. Moreover, phloroglucinol increased Ogg1 transcription and protein expression, counteracting the UVB-induced reduction in Ogg1 levels. Phloroglucinol also enhanced the nuclear translocation of nuclear factor erythroid 2-related factor 2 (Nrf2) as well as Nrf2 binding to an antioxidant response element located in the Ogg1 gene promoter. UVB exposure inhibited the phosphorylation of protein kinase B (PKB or Akt) and extracellular signal-regulated kinase (Erk), two major enzymes involved in cell protection against oxidative stress, regulating the activity of Nrf2. Akt and Erk phosphorylation was restored by phloroglucinol in the UVB-exposed keratinocytes. These results indicated that phloroglucinol attenuated UVB-induced 8-oxoG formation in keratinocytes via an Akt/Erk-dependent, Nrf2/Ogg1-mediated signaling pathway.

Prevention of UV-induced Skin Damage by Activation of Tumor Suppressor Genes p53 and $p14^{ARF}$

  • UV radiation is the most dangerous stress factor among permanent environmental impacts on human skin. Consequences of UV exposure are aberrant tissue architecture, alterations in skin cells including functional changes. Nowadays new kinds of outdoor leisure-time activities and changing environmental conditions make the question of sun protection more important than ever. It is necessary to recognize that self-confident consumers do not consider to change their way of life, they demand modern solutions on the basis of new scientific developments. In the past one fundamental principle of cosmetics was the use of physical and organic filter systems against damaging UV-rays. Today new research results demonstrate that natural protecting cell mechanisms can be activated. Suitable biological actives strongly support the protection function not from the surface but from the inside of the cell. A soy seed preparation (SSP) was proven to stimulate natural skin protective functions. The major functions are an increased energy level and the prevention of DNA damage. These functions can I be defined as biological UV protection. The tumor suppressor protein p53 plays a key role in the regulation of DNA repair. p53 must be transferred into the phosphorylated form to work as transcription factor for genes which are regulating the cell cycle or organizing DNA repair. A pretreatment with SSP increases the phosphorylation rate of p53 of chronically UV-irradiated human keratinocytes significantly. According to the same test procedure SSP induces a dramatic increase in the expression of the tumor suppressor protein p14$^{ARF}$ that is supporting the p53 activity by blocking the antagonist of p53, the oncoprotein Mdm2. Mdm2, a ubiquitin E3-ligase, downregulates p53 and at the same time it prevents phosphorylation of p53. The positive influence of the tumor suppressor proteins explains the stimulation of DNA repair and prevention of sunburn cell formation by SSP, which was proven in cell culture experiments. In vivo the increased skin tolerance against UV irradiation by SSP could be confirmed too. We have assumed, that an increased repair potential provides full cell functionality.y.

자외선 B 조사 마우스에서 피부손상에 대한 분죽 (Phyllostachys nigra var. henenis Strapf)잎 추출물의 효과 (The Effect of Bamboo (Phyllostachys nigra var. henenis Strapf) Leaf Extract on Ultraviolet B-induced Skin Damages in Mouse)

  • SKH1-hr 및 ICR 마우스에서 자외선 B(UVB) 조사에 의한 피부 일광화상세포(SBC) 및 ATPase 양성 표피 가지세포(DC)의 변화에 대한 대나무(분죽, Phyllostachys nigra var. henenis Strapf) 잎 추출물 (BLE)의 효과를 관찰하였다. 실험동물은 UVB ($200mJ/cm^2$) 조사 후 24시간에 희생시켰으며, BLE는 체중 kg당 50 mg의 용량으로 자외선 조사전 36시간, 12시간 및 조사 후 30분에 3회 복강내 주사하였다. 피부도포군은 0.2%의 용량으로 BLE 크림을 제조하고 자외선 조사전 24시간, 15분 및 조사후 즉시, 총 3회 도포하였다. 정상대조군 등쪽 피부에서는 길이 cm당 0.3개의 SBC가 관찰되었고, 자외선 조사에 따라 급격히 증가하였다. BLE 복강내 주사군(59.0%) 및 피부도포군(31.8%)에서 UVB에 의한 SBC의 발생은 유의성 있게 감소하였다. 정상대조군에서 귀등쪽 피부의 DC수는 $mm^2$$628.00{\pm}51.56$ 또는 $663.20{\pm}62.58$개 였으며, UVB 조사후 1일에 복강내 부영제 투여군에서는 39.0%, 부영제 피부 도포군에서는 27.1%감소하였다. 자외선 B조사에 의한 DC의 감소는 부영제를 처리한 UVB 조사군에 비하여, BLE 복강내 투여군에서는 25.7%, 피부도포군에서는 3.2%의 감소억제 효과를 나타냈다. 이상의 결과에서 BLE 투여가 UVB에의한 피부손상을 경감시킴을 알 수 있었다.
