Vegetative Anatomy and Tetrasporogenesis in Stoechospermum marginatum (C. Agardh) Kűtzing (Dictyotales, Phaeophyceae)

  • Anatomical organization of Stoechospermum marginatum reveals small cortical cells with moderately dense cytoplasm, overlying a multilayered medulla comparatively poor in cytoplasmic contents. The anticlinal walls of cortical cells show local thickenings rich in alginic acids. Sori form on both thallus surfaces and show tetrasporangia, paraphyses and sterile-cells. The unicellular paraphyses are rich in sulphated polysaccharides whereas multicellular ones have abundance of not only polysaccharides, but also of vacuoles and phenols. The sterile-cells are modified cortical cells present on either side of the tetrasporangium and bear cytoplasmic strands towards soral cavity. Various stages of tetrasporogenesis are seen in a single sorus. The developing tetrasporangium shows a two layered wall, where the outer one is rich in alginic acid and inner has sulphated polysaccharides. An apical pad aids tetraspore release. Also involved in the release process are sterile-cells, paraphyses and polysaccharides.

  • The integumental secretory structure is exocrine unicellular gland located in the epidermis of goldfish, Carassius auratus, and divided into two groups, mucous and granular cells. By the histochemical studies of integumental secretions the mucos cells reacted for acidic polysaccharides, and the granular cells for neutral glycoprotein. According to concentration of the secretion the integumental mucous are gradually sulphated. The mucous cells are typical form of goblet cell located in the upper region of the epidermis, and membrane bounded vesicles of the mucous are observed several size and electron densities by the cellular differentiation. The granular cells in middle and lower epidermis are present syncitial forms occasionally, and contain electron dense granules sized $1.0{\mu}m$ which are accumulated in cytoplasmic process held the cells to the basal lamina. The precursors of the integumental secretory materials are originated from the rough endoplasmic reticulum and next transported through the Golgi apparatus as a form of membrane bounded vesicles. After accomplish this process mature secretions are extruded to integumental surface by the mechanism of merocrine secretion in response to nerve stimulations respectively.

Preparation of Agarose from Gelidium amansii for Gel Electrophoresis using Various Purification Methods and Its Resolution Characteristics for DNA (다양한 정제방법에 의한 전기영동용 한천유래 아가로즈의 제조 및 DNA분리 특성)

  • The present study was conducted to investigate the preparative methods of agarose for gel electrophoresis from agar. Naturally occuring agar consists of two main polysaccharides, the neutral polysaccharide agarose and the acid sulphated polysaccharide agaropectin. The sulphate and carboxyl functions of the agar are accumulated in the agaropectin. The hydrophilic, non-ionogenic, rigid and transparent gel matrix of the agarose was found to be suitable for gel electrophoresis gel filtration and affinity chromatography. Agar was purified by chitosan treatment, cetylpyridinium chloride (CPC) treatment, and polyethylene glycol (PEG) treatment. Yields of agarose purified from agar with chitosan, CPC and PEG were 56.7%, 55.6% and 62.3%. It was proper to treat with chitosan in preparative methods of agarose for gel electrophoresis from agar.
