A comparison of Multilayer Perceptron with Logistic Regression for the Risk Factor Analysis of Type 2 Diabetes Mellitus (제2형 당뇨병의 위험인자 분석을 위한 다층 퍼셉트론과 로지스틱 회귀 모델의 비교)

  • The statistical regression model is one of the most frequently used clinical analysis methods. It has basic assumption of linearity, additivity and normal distribution of data. However, most of biological data in medical field are nonlinear and unevenly distributed. To overcome the discrepancy between the basic assumption of statistical model and actual biological data, we propose a new analytical method based on artificial neural network. The newly developed multilayer perceptron(MLP) is trained with 120 data set (60 normal, 60 patient). On applying test data, it shows the discrimination power of 0.76. The diabetic risk factors were also identified from the MLP neural network model and the logistic regression model. The signigicant risk factors identified by MLP model were post prandial glucose level(PP2), sex(male), fasting blood sugar(FBS) level, age, SBP, AC and WHR. Those from the regression model are sex(male), PP2, age and FBS. The combined risk factors can be identified using the MLP model. Those are total cholesterol and body weight, which is consistent with the result of other clinical studies. From this experiment we have learned that MLP can be applied to the combined risk factor analysis of biological data which can not be provided by the conventional statistical method.

Structural Analysis of the Antitumor Active Exo-polysaccharide Produced by Submerged Cultivation of Ganoderma lucidum Mycelium (영지(Ganoderma lucidum) 균사체의 액체배양에 의한 세포외 항암활성 다당류의 구조분석)

  • Exo-polysaccharide obtained from the submerged cultivation of Ganoderma lucidum mycelium was fractionated. The structural analysis of the acidic exo-polysaccharide fraction (BWS-DA-GI), showing high antitumor activity, was carried out and compared to the mycelial acidic fraction (MWS-DA-GI). The major sugar constituents of the fraction of BWS-DA-GI were glucose, galactose and mannose in the molar ratio of 2.5 : 2.1 : 2.5. The minor components in this fraction were xylose and fucose. While the major sugar constituents of the mycelial acidic fraction of MWS-DA-GI were galactose, fucose, mannose and glucose. The trace components in this fraction was xylose. From the results of periodate oxidation, Smith degradation, affinity chromatography and methylation analysis, the chemical structures of the two fractions, BWS-DA-GI and MWS-DA-GI were both determined as ${\beta}-1,3$ glucans. It was also estimated that BWS-DA-GI had a $1{\rightarrow}6$ glucosidic linkage and MWS DA-GI had $1{\rightarrow}4$ and $1{\rightarrow}6$ glucosidic linkages. The molecular weights of these fractions, MWS-DA-GI and MWSDA-GI were estimated as $1.2{\times}10^6\;and\;1.0{\times}10^6$ dalton, respectively.

Effect of Squid and Octopus on the Quality Characteristics of Kimchi during Fermentation (문어와 오징어의 첨가량에 따른 배추김치의 숙성 중 품질특성에 미치는 영향)

  • This study was carried out to investigate the chemical and microbiological properties of Kimchi containing different amounts of squid and octopus during fermentation at $4^{\circ}C$ for 56 days. Kimchi were assigned to one the of the following 7 treatment groups: CK (control Kimchi; no addition), SK-5 (added 5% of squid), SK-10 (added 10% of squid), SK-20 (added 20% of squid), OK-5 (added 5% of octopus), OK-10 (added 10% of octopus), and OK-20 (added 20% of octopus). The quality characteristics of CK and other treatments were determined by measuring the pH, titratable acidity, reducing sugar, volatile basic nitrogen (VBN), and microbial analysis. During fermentation, Kimchi containing squid and octopus had higher pH and lower acidity values than the CK group (P<0.05). The titratable acidity was increased in all treatment groups, whereas the reducing sugar content decreased gradually during fermentation. The reducing sugar contents were highest in the CK group. The VBN value increased in all treatment groups with increasing storage period, and the CK group had lower VBN value than the other treatment groups containing squid and octopus. The numbers of total bacteria and lactic acid bacteria rapidly increased up to the second week, and this slowly increased thereafter.

Nutritional Constituent Analysis of Korean Chestnuts (국내산 밤들의 영양성분 분석)

  • Eighteen Korean chestnut cultivars were collected from various places and the proximate compositions, dietary fiber, amino acid and free sugar contents in three parts (whole kernel, white kernel, yellow kernel) of eighteen Korean chestnut cultivars were analyzed. The white kernel of Chukfa and the yellow kernel of Ipyung contained the highest amount of moisture and crude protein, respectively. Carbohydrate content of whole kernel showed a range of $30.8{\sim}52.0%$ and crude ash content of whole kernel showed a range of $0.9{\sim}1.8%$. The amount of crude lipid was the highest in Byunggo. The amount of dietary fiber in Kwangeun, Daebo, Parkmi 1 ho, Yooma and Pyeonggi were higher than that of other Korean chestnut cultivars. Seventeen amino acids were detected. Major amino acids of the various chestnuts were aspartic acid and leucine. The amount of amino acids was higher in Ichui, Ipyung and Pyeonggi but was lower in Dantaek and Sandae than that of other samples. The major free sugar in the chestnuts was glucose. The free sugar amount of yellow kernels was higher than the white kernels.

Experimental Study for the Irradiation Effect of High Energy X-ray on the Growth of Sprout Plants Seeds (고 에너지 X-선이 새싹식물 종자의 생장에 미치는 영향)

  • This study examined the irradiation effect of high energy x-rays on the growth of sprout plants seeds. particularly with regard to acidity, salinity and sugar content change. The experimental groups were one day old sprout plants seeds irradiated with 6MV x-rays at doses of 2, 5, 8, 10 Gy using a linear accelerator. The control group was not irradiated. After two weeks, sprout plants were grown and harvested for component analysis. The acidity of the x-ray irradiated radish and red radish groups were significantly higher than that of the control group and bok choi and buckwheat groups lower than that of the control group. The salinity and sugar content of the x-ray irradiated 5 groups were significantly lower than that of the control group and Soybean and Red radish groups higher than that of the control group. The cause of plants component change were estimated to have occurred by generate basic ions and radiation sensitivity by moisture content. Further genetic studies will be needed to confirm the correlation between high energy x-rays and the cause of plants component change.

An Analysis of Phenolic Compounds, Carbon Isotopes, and Sugar Components of Whiskys Based on the Period of Maturation (위스키의 종류와 숙성기간에 따른 페놀성 화합물, 탄소동위원소비 및 당 성분 분석)

  • This paper analyzes phenolic compounds, carbon isotopes, and sugar components of whiskys based on the maturation period. For this, the paper considers a total of 40 whiskys(mainly imports) distributed in Korea. It is important to analyze the presence phenolic compounds(e.g., furfural, syringaldehyde, vanillin, syringic acid, and vanillic acid) because these are found only in whiskys ripened in oak. The results indicate that the total content of phenolic compounds increased with the increase in the storage period regardless of the type of whisky. In terms of vanillin/syringaldehyde(V/S), Scotch whiskys had 0.4~0.5; American whiskies, 0.30~0.34; and Canadian whiskies, 0.31~0.33. In terms of Scotch whiskys, Macallan had 0.25~0.34, making it unique among Scotch whiskys. In terms of the ratio of carbon isotopes, there were clear differences between malt Scotch whiskys, blended Scotch grain whiskys, American whiskys, and Canadian whiskys: -23.4~ -24.3, -16.8~-21.0, -11.0~-11.5 and -9.5~13.9, respectively. In addition, malt Scotch whiskys contained 40~230 $mg/{\ell}$ of fructose; blended Scotch whiskys, 20~120 $mg/{\ell}$; American whiskys, 50~70 $mg/{\ell}$; and Canadian whiskys, 20~100 $mg/{\ell}$, demonstrating that the fructose content of single-malt whiskys was twice the average fructose content. On the other hand, malt Scotch whiskys contained 30~170 $mg/{\ell}$ of glucose; blended Scotch whiskys, 20~120 $mg/{\ell}$; American whiskys, 20~30 $mg/{\ell}$; and Canadian whiskys, 10~110 $mg/{\ell}$, demonstrating that the glucose content of single-malt whiskys exceeded the average glucose content. This study's results can be used as a database of classification for whiskys based on the fermentation of raw ingredients and the period of maturation for distinguishing between different types of whiskys. In addition, the results can facilitate the verification of genuine whiskys by allowing for the identification of different types of whiskys based on the period of maturation.

Detecting Defects from the Alcoholic Fermentation of Apple Concentrates (사과농축액의 알코올발효 불량화 자료 설정)

  • This study compared and analyzed the quality characteristics of five different apple juice concentrates (A~E) after alcoholic fermentation to establish test indicators for their defects. From our results, the titratable acidity was nearly similar in all diluted solutions. However, A and D showed a high pH of above 4.0 while B, C and E exhibited a low pH of below 3.0. In terms of free sugar content, maltose was undetected in A and D. In contrast, about 698 mg% maltose was found in C and more than 1,000 mg% maltose were detected in B and E. Malic acid, one of the main organic acids in apple, was measured at a high value of about 600 mg% in A and D and about 50 mg% in B, C and E. Potassium, one of the main minerals, was about 180 mg% in A and D, whereas a small amount of potassium, ranging between 6~9 mg% were present in B, C and E. Preservative (by sorbic acid) was not detected at all in all apple juice concentrates (A~E). When the above diluted apple concentrates were fermented, the alcohol contents were 11.2% and 10.8% in DAFB and AAFB, respectively. Alcoholic fermentation almost did not take place in BAFB, CAFB and EAFB. The use of maltose as the yeast may have influenced the fermentation. However, B, C and E were thought to be either defective or contaminated apple concentrates based on the analysis results of free sugar and organic acid.

Effects of Isoamyl Acetate Production in Makgeolli According to Fermentation Conditions (막걸리 발효조건에 따른 isoamyl acetate 생성 비교)

  • Because of a continual reduction in its domestic market share, the quality of the Makgeolli, a Korean traditional liquor, needs to be upgraded. Among the several options for quality improvement, sufficient organoleptic expression of flavor is very important. We analyzed production changes of isoamyl acetate, which has a banana smell, based on fermentation temperature and sugar content through the cultivation of S. cerevisiae 98-5 KCCM 11396P using generally polished rice. The banana flavor of that fermentation mash was organoleptically high at 20℃, but a larger amount of isoamyl acetate was obtained with a higher sugar content at 10℃, based on analysis by GC-MS. Consequently, sufficient production of banana flavor from isoamyl acetate was based on the concentration of isoamyl alcohol as a substrate compound of isoamyl acetate, and the production depended highly on the maintenance of heat stability, since it is unstable in temperature and the minimized inhibition of alcohol acetyl transferase by unsaturated fatty acids. We also found that production of the flavor component required the addition of sugar and a slightly higher temperature of 20~25℃ at the beginning stage of fermentation, with additional mash fermentation and a gradual decrease in temperature to 10~15℃.

Optimization for Preparation of Perilla Jangachi according to Steaming Time and Onion Contents (찌는 시간과 양파 첨가량에 따른 깻잎장아찌의 최적화)

  • Korean native Perilla Jangachi (salted and fermented vegetable) is popular and well-known but little study on its composition and most palatable condition has been reported. This study was performed to observe the change in the contents of chemical components and sensory evaluation of Perilla Jangachi which was prepared through two processing. First, as a pretreatment, Perilla leaves were soaked in salt water. The optimal level of salt concentration and soaking time (salt 4%, soaking time 42hours) was determined. Second, soaked Perilla Jangachi was steamed and then fermented in various ingredients like soy sauce, sugar, onion and so on. In this study, Perilla Jangachi was made by 3 levels of steaming time(30 seconds, 60 seconds, 90 seconds) and onion contents (10%, 30%, 50%). The optimal level of onion contents and steaming time was determined with the results of sensory evaluation by response surface methodology and analysis of composition. Sodium contents were decreased as the levels of onion contents were increased. Flavonoids contents weren't increased although contents of onion were increased. But they were influenced on steaming time that as the levels of steaming time were shortened, flavonoids contents were increased. To lengthen the shelf-life of Perilla Jangachi, the activities of enzyme peroxidase - which changes Jangachi's quality bad - by various levels of steaming time were measured. After steaming 50 seconds, peroxidase became inactive. Reducing sugar contents were decreased relying on either increased steaming time and onion contents. While steaming 30 seconds and 10% onion contents, the content of reducing sugar was 0.858% but steaming 90 seconds and 50% onion contents, it became 0.372%. Among the sensory attributes, brownness was increased as onion contents increased and steaming more than 60 seconds. Saltiness, sweetness, perilla flavor were greater relying on decreased onion contents. Toughness was decreased as both the amounts of onion and steaming time were increased. With this results, the most optimal adding level of onion content and steaming time was determined. Steaming 72 seconds and adding onion 27% was established as a optimal condition of Perilla Jangachi.

Nucleotide Sequence and Cloning of sfs4, One of the Genes Involved in the CRP-Dependent Expression of E. coli mal Genes. (CRP 의존성 maltose 대사 촉진 유전자 sfs4의 클로닝 및 염기배열 결정)

  • In Escherichia coli, CRP forms a complex with cAMP and acts as a transcriptional regulator of many genes, including sugar metabolism operons. The E. coli MK2001, which is introduced the altered crp, is functional in the expression of lac, ara and man, in the absence of cAMP. However, the expression of mal gene is fully activated by the addition of cAMP or cGMP. The object of the study is cloning of the sfs (sugar fermentation stimulation) genes, which was involved in regulation of mal gene expression with the altered crp gene, and structural analysis and characterization of the genes at the molecular level. We have cloned 5 different E. coli genes which stimulate the maltose metabolism in a crp, cya::km (MK2001) background. Newly identified genes were designated as sfs. One of the sfs genes (pPC1), located at the 53.2 min map position on the E. coli chromosome, was further analyzed. Expression of the genes, which is involved in maltose metabolism, malQ (amylomaltase), was increased to 5.8-fold in the presence of a plasmid, pAP5, containing the subcloned sfs4 gene. The nucleotide seguence of a common 2,126 bp segment of the pPCM1 was determined and two open reading frames (ORF1 and ORF2) were detected. The ORF1 encodes the sfs4 gene and ORF2 encodes a truncated protein. Potential CRP binding site is located in the upstream of the putative promoter in the regulatory region. Expression of the cloned sfs4 gene was positively regulated by the cAMP-CRP complex.
