Alzheimer's disease (AD) is a progressive neurodegenerative disease characterized by cognitive decline. Several recent studies demonstrated that impaired adult neurogenesis could contribute to AD-related cognitive impairment. Adult subventricular zone (SVZ) neurogenesis, which occurs in the lateral ventricles, plays a crucial role in structural plasticity and neural circuit maintenance. Alterations in adult SVZ neurogenesis are early events in AD, and impaired adult neurogenesis is influenced by the accumulation of intracellular Aβ. Although Aβ-overexpressing transgenic 5XFAD mice are an AD animal model well representative of Aβ-related pathologies in the brain, the characterization of altered adult SVZ neurogenesis following AD progression in 5XFAD mice has not been thoroughly examined. Therefore, we validated the characterization of adult SVZ neurogenesis changes with AD progression in 2-, 4-, 8-, and 11-monthold male 5XFAD mice. We first investigated the Aβ accumulation in the SVZ using the 4G8 antibody. We observed intracellular Aβ accumulation in the SVZ of 2-month-old 5XFAD mice. In addition, 5XFAD mice exhibited significantly increased Aβ deposition in the SVZ with age. Next, we performed a histological analysis to investigate changes in various phases of adult neurogenesis, such as quiescence, proliferation, and differentiation, in SVZ. Compared to age-matched wild-type (WT) mice, quiescent neural stem cells were reduced in 5XFAD mice from 2-11 months of age. Moreover, proliferative neural stem cells were decreased in 5XFAD mice from 2 to 8 months of age. Furthermore, differentiations of neuroblasts were diminished in 5XFAD mice from 2-11 months of age. Intriguingly, we found that adult SVZ neurogenesis was reduced with aging in healthy mice. Taken together, our results revealed that impairment of adult SVZ neurogenesis appears with aging or AD progression.
미토콘드리아는 세포안에서 에너지 공급, 칼슘 이온 저장, 활성산소 생성, 세포 자살과 같은 다양한 기능을 수행한다. 이러한 기능을 통해, 미토콘드리아는 줄기세포의 유지, 증식, 그리고 분화에 관여한다. 뇌에서 뇌실 하 영역(subventricular zone, SVZ)에는 일평생 새로운 신경세포를 생성하는 신경줄기세포(neural stem cell, NSC)가 존재한다. 하지만, SVZ NSCs에서 미토콘드리아의 역할에 대한 연구는 많이 알려져 있지 않다. 이번 연구에서 우리는 미토콘드리아의 complex I 저해제인 rotenone이 SVZ NSCs의 증식과 분화를 다른 방식으로 방해한다는 것을 보여주었다. 증식 중인 신경줄기세포에서, rotenone은 세포분열을 감소시켰는데, 이때 세포분열은 히스톤 H3에 인산기가 붙어있는 지를 측정하여 확인하였다. Rotenone을 50 nM 농도로 증식 중인 신경줄기세포에 처리했을 때, 세포사멸은 발생하지 않았다. 한편, 분화 중인 신경줄기세포에 rotenone을 처리한 경우, 신경세포와 희소 돌기아교 세포(oligodendrocyte)으로의 분화가 억제되었고, glial fibrillary acidic protein (GFAP)를 발현하는 성상세포(astrocyte)에는 영향이 없었다. 흥미롭게도, 4-6일 동안의 분화 과정 동안 rotenone이 처리된 신경줄기세포에서 대조군 보다 더 많은 세포 수가 관찰 되었는데, 이는 증식 과정 중의 rotenone의 효과와 다른 것이다. 이에, 우리는 rotenone이 세포 자살은 감소시켰으나, 세포 분열에는 영향을 끼치지 않았음을 관찰하였다. 세포 자살의 경우는 cleaved caspase-3를 측정함으로써 확인하였다. 이러한 결과들은 SVZ 신경줄기세포의 증식과 분화 모두에 제대로 작동하는 미토콘드리아가 있어야 함을 제안하고 있다. 게다가, 이러한 과정에서 미토콘드리아는 세포 분열과 세포자살에 관여할 수도 있을 것이다.
적절한 농도의 활성산소종(ROS)은 병원체에 대한 세포의 방어, 신호 전달, 세포 성장 및 유전자 발현을 포함한 다양한 정상 세포 기능을 매개한다. 최근의 연구는 ROS와 ROS를 생성하는 NADPH 산화 효소(Nox)가 성인 쥐 뇌의 뇌실 하 영역(SVZ)에 있는 신경 줄기세포의 자가 복제와 신경 세포 분화에 중요하다는 것을 보여 주었다. 본 연구에서 세포 내 ROS가 갓 태어난 쥐의 뇌에서 적출되어 배양된 SVZ 신경 줄기세포에서 검출된 것으로 나타났다. Nox 유사 유전자들 중 Nox4가 배양된 세포에서 주로 발현되었고, Nox1과 Nox2는 거의 발현되지 않았다. 또한, Nox4 유전자는 신경 세포 분화 동안 최대 10배까지 발현이 크게 증가하였다. Immunocytochemistry결과 Nox4 단백질은 신경 세포 특이적인 tubulin인 Tuj1-양성 신경 세포에서 주로 발견되었다. 이와 맥을 같이 하여, 내인성 ROS는 분화 후 축삭돌기를 가지고 있으며 신경 세포로 보이는 세포에서만 검출되었다. 또한, ROS를 제거하는N-acetyl cysteine에 의해 세포 산화 환원 상태가 교란되었을 때, 신경 세포로의 분화가 크게 감소하였다. 마지막으로, shRNA를 이용하 여 Nox4를 knockdown한 세포에서 신경 세포로의 분화가 감소하였다. 이러한 연구 결과는 Nox4가 갓 태어난 쥐의 SVZ 신경 줄기 세포의 주요한 ROS 생성 효소이고, Nox4에 의한 ROS생성이 신경 세포 분화에 중요하다는 것을 암시한다.
Glioblastoma (GBM) is the most common and aggressive form of human adult brain malignancy. The identification of the cell of origin harboring cancer-driver mutations is the fundamental issue for understanding the nature of GBM and developing the effective therapeutic target. It has been a long-term hypothesis that neural stem cells in the subventricular zone (SVZ) might be the origin-of-cells in human glioblastoma since they are known to have life-long proliferative activity and acquire somatic mutations. However, the cell of origin for GBM remains controversial due to lack of direct evidence thereof in human GBM. Our recent study using various sequencing techniques in triple matched samples such as tumor-free SVZ, tumor, and normal tissues from human patients identified the clonal relationship of driver mutations between GBM and tumor-free SVZ harboring neural stem cells (NSCs). Tumor-free SVZ tissue away from the tumor contained low-level GBM driver mutations (as low as 1% allelic frequency) that were found in the dominant clones in its matching tumors. Moreover, via single-cell sequencing and microdissection, it was discovered that astrocyte-like NSCs accumulating driver mutations evolved into GBM with clonal expansion. Furthermore, mutagenesis of cancer-driving genes of NSCs in mice leads to migration of mutant cells from SVZ to distant brain and development of high-grade glioma through the aberrant growth of oligodendrocyte precursor lineage. Altogether, the present study provides the first direct evidence that NSCs in human SVZ is the cell of origin that develops the driver mutations of GBM.
뇌실하 영역과 subgranular zone은 뇌에서 평생 새로운 신경 세포를 만들어 내는 곳이다. 이 부위에 있는 신경줄기세포는 세포 분열을 통해서 줄기 세포군을 계속 유지할 뿐만 아니라, 신경 세포와 신경 교세포로 분화한다. 세포 분열을 측정하기 위해 thymidine 유사체인 5-ethynyl-2'-deoxyuridine (EdU)가 사용되어 왔다. 몇몇의 경우에서는 새롭게 만들어지는 신경 세포를 표지하려는 목적으로 사용되었다. 이번 연구에서는, EdU가 쥐의 뇌실 하영역에서 분리해낸 신경줄기세포의 분열과 분화에 어떠한 영향을 미치는 지를 보여주었다. 첫째, 신경줄기세포가 EdU를 포함하는 세포 증식 배양액에서 24시간 동안 배양되었을 때, 추후에 분화를 유도하여도 신경세포로 분화가 전혀 일어나지 않았다. EdU를 1시간 동안 처리했을 때도 신경세포로의 분화가 상당부분 저해되었다. 둘째, EdU는 농도가 높을수록, 처리시간이 많을수록 신경줄기세포의 증식을 더욱 많이 저해하였다. 끝으로, EdU는 신경 교세포 중에서 oligodendrocyte으로의 분화는 억제하였지만, astrocyte로의 분화는 오히려 증가시켰다. 본 연구결과는 뇌실하 영역 신경줄기세포의 분화에 EdU가 어떠한 영향을 미치는 지를 처음으로 보여주었고, 이러한 결과들은 신경 세포와 oligodendrocyte로의 분화에 세포 분열이 반드시 필요하다는 것을 제안하고 있다.

Glioblastoma (GBM) is a disease without any definite cure. Numerous approaches have been tested in efforts to conquer this brain disease, but patients invariably experience recurrence or develop resistance to treatment. New surgical tools, carefully chosen samples, and experimental methods are enabling discoveries at single-cell resolution. The present article reviews the cell-of-origin of isocitrate dehydrogenase (IDH)-wildtype GBM, beginning with the historical background for focusing on cellular origin and introducing the cancer genesis patterned on firework. The authors also review mutations associated with the senescence process in cells of the subventricular zone (SVZ), and biological validation of somatic mutations in a mouse SVZ model. Understanding GBM would facilitate research on the origin of other cancers and may catalyze the development of new management approaches or treatments against IDH-wildtype GBM.

Objective : Many recent reports have shown that the mature mammalian brain harbors multipotent stem cells, rendering the brain capable of generating new neurons and glia throughout life. Harvested stem cells from an adult rat are transplanted in order to evaluate the cell survival and differentiation. Methods : Using a percoll gradient with a high speed centrifugation method, we isolate neural stem/progenitor cells were isolated from the subventricular zone[SVZ] of a syngeneic adult Fisher 344 rats brain. For 14days expansion, the cultured cells comprised of a heterogeneous population with the majority of cells expressing nestin and/or GFAP. After expanding the SVZ cells in the presence of basic fibroblast growth factor-2, and transplanting then into the hippocampus of normal rats, the survival and differentiation of those cells were examined. For transplantation, the cultured cells were labeled with BrdU two days prior to use. In order to test their survival, the cells were transplanted into the dorsal hippocampus of normal adult Fisher 344 rats. Results : The preliminary data showed that at 7days after transplantation, BrdU+ transplanted cells were observed around the injection deposition sites. Immuno-fluorescent microscopy revealed that the cells co-expressed BrdU+ and neuronal marker ${\beta}$-tubulin III. Conclusion : The data demonstrate that the in vitro expanded SVZ cells can survive in a heterotypic environment and develop a neuronal phenotype in the neurogenic region. However more research will be needed to examine the longer survival time points and quantifying the differentiation in the transplanted cells in an injured brain environment.
뇌에서 뇌실하 영역은 자가 복제 및 신경세포와 교세포로 분화하는 신경줄기세포가 위치한 곳이다. 이러한 신경줄기세포는 태어난 직후 뿐만 아니라, 성인기까지 존재한다. 세포 증식과 분화에 대한 결정은 세포 안과 밖의 상황에 따라 조절될 필요가 있기에, 많은 세포 내부 또는 세포 외부의 인자들이 이러한 결정에 관여한다. 이러한 인자들 중에서 미토콘드리아는 신경줄기세포의 운명 결정에 관여함이 보고된 바 있다. 본 저자들의 이전 논문에서, 미토콘드리아 저해제인 rotenone을 장시간 처리했을 때, 신경세포로의 분화가 거의 일어나지 않았음을 보여주었다. 이번 연구에서, rotenone을 뇌실하 영역 신경줄기세포에 단기간 처리했을 때의 영향에 대해 조사하였다. 이를 통해 다음과 같은 결과를 관찰하였다. (4) sulfiredoxin 1 (Srxn1) 유전자 발현이 rotenone을 하루 처리한 후 증가하였는데, 이는 nuclear factor (erythroid-derived 2)-like 2 (Nrf2) 신호전달 경로가 활성화 되었음을 말해준다. 이러한 실험 결과는 기능을 갖춘 미토콘드리아가 신경세포 또는 희소 돌기 아교 세포로의 분화 뿐 아니라, 이미 분화가 끝난 신경세포의 유지에도 필요함을 확인해 주었다. 또한, 이러한 결과는 rotenone과 같은 미토콘드리아의 저해제에 짧은 시간 노출 되더라도 신경줄기세포의 신경세포로의 분화 가능성에 장기적인 영향을 미칠 수 있음을 시사한다.

Nifedipine은 신경세포로의 분화를 촉진하는 Dlx2 유전자의 전사를 증가시켰고, 초기 신경세포에서 보이는 Mash1의 양도 증가시켰다. Nifedipine 외 또다른 L-type 칼슘 채널의 저해제인 verapamil을 처리하자, 신경세포로의 분화가 소폭 증가하였으나, 통계적 유의미성은 매우 낮았다. T-type 칼슘 채널의 저해제 유전자인 Cav3.1, Cav3.2, Cav3.3가 발현함을 관찰하여, T-type 칼슘 채널의 저해제인 pimozide를 신경줄기세포에 처리하였으나, 신경세포로의 분화에는 변화가 없었다. 이러한 결과를 통해 nifedipine이 신경줄기세포의 초기 분화를 증진함을 알 수 있으며, L-type 칼슘 채널이 신경세포로의 분화에 관여함을 알 수 있다.
Neural stem cells are found in adult mammalian brain regions including the subgranular zone (SGZ) of the dentate gyrus (DG) and the subventricular zone (SVZ). In addition to these two regions, other neurogenic regions are often reported in many species. Recently, the subcallosal zone (SCZ) has been identified as a novel neurogenic region where new neuroblasts are spontaneously generated and then, by Bax-dependent apoptosis, eliminated. However, the development of SCZ in the postnatal brain is not yet fully explored. The present study investigated the precise location and amount of neuroblasts in the developing brain. To estimate the importance of programmed cell death (PCD) for SCZ histogenesis, SCZ development in the Bax-knockout (KO) mouse was examined. Interestingly, an accumulation of extra neurons with synaptic fibers in the SCZ of Bax-KO mice was observed. Indeed, Bax-KO mice exhibited enhanced startle response to loud acoustic stimuli and reduced anxiety level. Considering the prevention of PCD in the SCZ leads to sensory-motor gating dysfunction in the Bax-KO mice, active elimination of SCZ neuroblasts may promote optimal brain function.

제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
