Toxicological Relevance of Transporters

  • Transporters are membrane proteins that mediate the transfer of substrate across the cellular membrane. In this overview, the characteristics and the toxicological relevance were discussed for various types of transporters. For drug transporters, the overview focused on ATP-binding cassette transporters and solute carrier family 21A/22A member transporters. Except for OCTN transporters and OATP transporters, drug transporters tend to have broad substrate specificity, suggesting drug-drug interaction at the level of transport processes (e.g., interaction between methotrexate and non-steroidal anti-inflammatory agents) is likely. For metal transporters, transporters for zinc, copper and multiple metals were discussed in this overview. These metal transporters have comparatively narrow substrate specificity, except for multiple metal transporters, suggesting that inter-substrate interaction at the level of transport is less likely. In contrast, the expressions of the transporters are often regulated by their substrates, suggesting cellular adaptation mechanism exists for these transporters. The drug-drug interactions in drug transporters and the cellular adaptation mechanisms for metal transporters are likely to lead to alterations in pharmacokinetics and cellular metal homeostasis, which may be linked to the development of toxicity. Therefore, the transporter-mediated alterations may have toxicological relevance.

  • Growth mechanism of carbon nanotubes(CNTs) synthesized by chemical vapor deposition is abided by two growth modes. These growth modes are classified by the position of activated catalytic metal particle in the CNTs. Growth mode can be also affected by interaction between substrate and catalytic metal and induced energy such as thermal and plasma. We studied the reaction of catalytic metal to the substrate and growth mode of CNTs. Various substrates such as Si(100), graphite plate, coming glass, sapphire and AAO membrane are used to study the relation between catalytic metal and substrate in the synthesis of CNTs. For catalytic metal, thin film was deposited on various substrate via sputtering technique with a thickness of ∼20nm and magnetic fluids with none-sized particles were dispersed on AAO membrane. After laying process on AAO membrane, it was dried at 80$^{\circ}C$ for 8 hour. Synthesizing of CNTs was carried out at 900$^{\circ}C$ in NH3/C2H2 mixture gases flow for 10minutes.

패럴린 박막을 이용한 기계화학적 폭발물 센서 (Parylene membrane based chemomechanical explosive sensor)

  • This paper reports a chemomechanical explosive sensor based on a thin polymer membrane. The sensor consists of thin parylene membrane and electrodes. Parylene membrane is functionalized with 4-mercaptophenol which interacts strongly with nitrotoluene based explosives. The membrane deflection caused by molecular interaction between the surface and explosives is monitored by capacitance between the membrane and the substrate. To measure the capacitance, electrodes are formed on the membrane and the substrate. While the previous cantilever system requires a bulky optical measuring system, this purely electric monitoring method offers a compact and effective system. Thus, this explosive sensor can be readily miniaturized and used in the field. The developed sensor can reliably detect dinitrotoluene and its limit of detection is evaluated as approximately 110 ppb.

Mechanics of lipid membranes subjected to boundary excitations and an elliptic substrate interactions

  • We present relatively simple derivations of the Helfrich energy potential that has been widely adopted in the analysis of lipid membranes without detailed explanations. Through the energy variation methods (within the limit of Helfrich energy potential), we obtained series of analytical solutions in the case when the lipid membranes are excited through their edges. These affordable solutions can be readily applied in the related membrane experiments. In particular, it is shown that, in case of an elliptic cross section of a rigid substrate differing slightly from a circle and subjected to the incremental deformations, exact analytical expressions describing deformed configurations of lipid membranes can be obtained without the extensive use of Mathieu's function.

Evidence for Regulation of Interaction of Endogenous Protein Kinase C(Pkc) Substrates with Plasma Membrane by PKC Down-Regulation in K562 Cells

  • In the particulate fraction obtained from PKC-down regulated K562 cells by treatment for 24 h with 200nM TPA, phosphorylation of two proteins with molecular weight, 100 kDa and 23 kDa (designated p100 and p23, respectvely) was depleted and addition of exogenous purified PKC to this fraction failed to testore their phosphorylation. However, in the soluble fraction, all of phosphoproteins abolished by long-term treatment with TPA were restored by exogenously added PKC. Phosphorylation of two proteins was increased by short-term tretment (20 min), and diminished with the persistant exposure to TPA as well as at a concentration as low as 1nM. When K562 cells were treated with 1 nM and 200 nM TPA for 24 h, phosphorylation of p100 was restored with or without exogenous PKC on 2-3day and 6day after removal of treated TPA, respectively. Two-dimensional electrophoresis of phosphoproteins revealed that phosphorylated p100 (pl=5.9) and p66 species were completely absent from the particulate fraction of K562 cells treated with 200nM TPA for 24 h. These results suggest that the interaction of sensitive endogenous substrates, p100 and p23 with the plasma membrane might be regulated by PKC-down regulation without displacement to the cytosol and the interaction of p100 with the membrane might be reveersible.

한외여과막 반응기를 이용한 어피젤라틴의 연속적 가수분해 (Continuous Hydrolysis of Cod Skin Gelatin in an Ultrafiltration Reactor)

  • 효소를 이용하여 대구피 젤라틴 가수분해물을 연속적으로 생산하기 위해 한외여과막 반응기에서의 젤라틴 가수분해 최적 공정조건 및 한외여과막 반응기를 장시간 작동하였을 때의 효소활성 저하원인을 규명하였다. 효소농도가 0.4mg/ml까지 증가함에 따라 기절의 전환율은 증가하였으나 기질의 경우는 농도가 증가함에 따라 전환율이 감소하였다. 유량(Flux)이 클수록 전환율은 감소되었으며 잔류시간이 길수록 전환율을 증가하였다. 한외여과막 반응기 장치의 작동시 가질의 전환율은 반응시간 1시간 부근에서 최대 전환율을 나타내었으며 시간당(8시간 작동) 평균 전환율 감소는 2.1%였다. 한외여과막을 통한 효소 누출은 반응시간 10~60분 범위에서 최대였으며 6시간 이후에는 거의 누출되지 않았다. 전체 효소량 중 누출량은 $50^{\circ}C$에서 16%인데 비해 $35^{\circ}C$에서는 12%였다. 그러나 효소 누출과 기질 전환율 사이에 명백한 상호관계는 나타나지 않았다. 막에 의한 효소의 활성저하는 작동시간 120분까지 나타났으며, 이때 초기속도의 36%가 저하하였다. 연속식(CSTMR)에서 $K_m$ 값은 회분식의 그것보다 20배 큰 반면 $K_2$ 10.5배 낮았다. 최적 가수분해조건은(기질농도 10%, $50^{\circ}C$, pH 8.0, 유량 7.31ml/min, 잔류시간 82분, S / E = 50w / w) 하에서의 생산량은 효소 mg당 가수분해물이 285mg으로 회분식 44.8mg에 비해 6배 이상이었다.

고온 구동형 고분자 전해질 막 연료전지용 폴리벤즈이미다졸계 고분자 전해질 막의 개발 동향 (Research Trends of Polybenzimidazole-based Polymer Electrolyte Membranes for High-temperature Polymer Electrolyte Membrane Fuel Cells)

  • 고온 구동형 고분자 전해질 막 연료전지(high temperature polymer electrolyte membrane fuel cell, HT-PEMFC)는 전극의 빠른 활성과 피독 현상에 대한 높은 저항성으로 인해 저온 구동형 PEMFC의 대안으로 많은 연구가 진행되고 있다. 폴리벤즈이미다졸(polybenzimidazole, PBI)을 기반으로 한 PEM의 경우 고온 구동 조건에서 이온 전도성 물질과의 높은 상호 작용과 우수한 열적ㆍ기계적 안정성 특징으로 인해 HT-PEMFC용 PBI 기반 전해질 막 개발과 관련된 다양한 연구들이 진행되고 있다. 본 총설에서는 고성능/고내구성의 PBI 기반 PEM을 개발하기 위해 1) 인산 및 다양한 이온전도성 물질이 도핑된 PBI 막의 특성 분석과 막 제조법에 따른 PBI 막의 물성 비교에 관한 연구를 우선적으로 살펴본 후 2) 다공성 폴리테트라플루 오르에틸렌 지지체 및 무기 입자 혼입을 통한 PBI 복합 막의 성능 개선 연구 및 3) 고분자 블렌딩을 통해 가교 구조가 도입된 PBI 기반 가교 막의 내구성 향상에 관한 연구 동향에 대하여 소개하고자 한다.

Functional Layer-by-Layer Assembled Multilayers Based on Nucleophilic Substitution reaction

  • Ultrathin polyelectrolyte (PE) multilayer films prepared by the versatile layer-by layer (LbL) assembly method have been utilized for the preparation of light-emitting diodes, electrochromic, membrane, and drug delivery system, as well as for selective area patterning and particle surface modification because the various materials with specific properties can be inserted into the film with nano-level thickness irrespective of the size or the shape of substrate. Since the introduction of the LbL technique in 1991 by Decher and Hong, various hydrophilic materials can be inserted within LbL films through complementary interactions (i.e., electrostatic, hydrogen-bonding or covalent interaction). In this study, it is demonstrated that LbL SA multilayer films based on nucleophilic substitution reaction can allow the preparation of the highly efficient magnetic and/or optical films and nonvolatile memory devices.

Effect of Nitric Oxide on the Sinusoidal Uptake of Organic Cations and Anions by Isolated Hepatocytes

  • The issue of whether or not the presence NOx (NO and oxidized metabolites) in the hepatocytes at pathological levels affects the functional activity of transport systems within the sinusoidal membrane was investigated. For this purpose, the effect of the pretreatment of isolated hepatocytes with sodium nitroprusside (SNP), a spontaneous NO donor, on the sinusoidal uptake of tributylmethylammonium (TBuMA) and triethylmethyl ammonium (TEMA), representative substrates of the organic cation transporter (OCT), and taurocholate, a representative substrate of the $Na^+$/taurocholate cotransporting polypeptide (NTCP), was measured. The uptake of TBuMA and TEMA was not affected by the pretreatment, as demonstrated by the nearly identical kinetic parameters for the uptake ($i.e., V_{max}, K_{m} and CL_{linear}$). The uptake of mannitol into hepatocytes was not affected, demonstrating that the membrane integrity remained constant, irregardless of the SNP prutreatment. On the contrary, the uptake of taurocholate was significantly inhibited by the pretreatment, resulting in a significant decrease in V_{max}$, thus providing a clear demonstration that NOx preferentially affects the function of NTCP rather than OCT on the sinusoidal membrane. A direct interaction between NOx and NTCP or a decrease in $Na^+/K^+$ ATPase activity as the result of SNP pretreatment might be responsible for this selective effect of NOx.

Novel Composite Membranes Comprising Silver Salts Physically Dispersed in Poly(ethylene-co-propylene) for the Separation of Propylene/Propane

  • Novel composite membranes, which delivered high separation performance for propylene/propane mixtures, were developed by coating inert poly(ethylene-co-propylene) rubber (EPR) onto a porous polyester substrate, followed by the physical distribution of $AgBF_4$. Scanning electron microscopy-wavelength dispersive spectrometer (SEM-WDS) revealed that silver salts were uniformly distributed in the EPR layer. The physical dispersion of the silver salts in the inert polymer matrix, without specific interaction, was characterized by FT-IR and FT-Raman spectroscopy. The high separation performance was presumed to stem from the in-situ dissolution of crystalline silver ionic aggregates into free silver ions, which acted as an active propylene carrier within a propylene environment, leading to facilitated propylene transport through the membranes. The membranes were functional at all silver loading levels, exhibiting an unusually low threshold carrier concentration (less than 0.06 of silver weight fraction). The separation properties of these membranes, i.e. the mixed gas selectivity of propylene/propane ${\sim}55$ and mixed gas permeance ${\sim}7$ GPU, were stable for several days.
