The applicability of a commercial enzymatic time-temperature integrator (TTI) for monitoring spoilage of ground beef was investigated under isothermal storage condition at different temperatures. The volatile basic nitrogen (VBN) value was used as a spoilage index for ground beef. The time taken to reach the spoilage of ground beef stored at 4, 10, 15, 20, and ${25^{\circ}C}$ were 168, 114, 60, 48, and 24 hrs, respectively. However, these quality losses of beef were not coincided with the endpoints of the three different C-type TTIs (C-1, C-4, and C-7). In order to match the TTI response to the quality loss of beef, some ingredients such as enzyme and substrate solutions were extracted from C-1TTI and remixed with different amount of them in the tubes to constitute the modified TTIs. The responses of the modified CM-1 TTI were very close to the quality loss of beef stored at 20 and ${25^{\circ}C}$, but not at other temperatures tested. The response of the other modified CM-2 TTI was only matched to the quality loss of beef stored at ${15^{\circ}C}$. Therefore, systematic kinetic studies of food spoilage and the TTI response are required to apply the TTI as a quality indicator for a specific food.

In this study, reduced graphene oxide/polyaniline composite was fabricated tomaximize their advantages with electrochemical performances and use as a electrodematerial for supercapcaitor. Polyaniline as an electrode material was synthesized bychemical polymerization of aniline monomer and reduced graphene oxide wasintroduced to prepare composite with polyaniline without any pre-treatment. Thereduced graphene oxide, polyaniline and their composite electrodes were fabricatedon gold coated PET(polyethylene terephthalate) substrate through spray coatingmethod which can also apply to industrial scale. we have also prepared reducedgraphene oxide and polyaniline single material electrode to compare theirelectrochemical properties with reduced graphene oxide/polyaniline composite electrode. We have analyzed and compared electrochemical properties of eachelectrodes by using cyclic voltammetry(CV), galvanostaticcharge-discharge(GCD) and electrochemical impedancespectroscopy(EIS) at same condition. As a result, reduced graphene oxide /polyaniline composite electrode showed higher capacitance value more thanpolyaniline and reduced graphene oxide electrode, respectively. Internal resistanceof reduce graphene oxide/polyaniline composite electrode was 24% and 58% lessthan polaniline and reduced graphene oxide electrode respectively. These resultsconsidered that reduced graphene oxide/polyaniline composite electrode has potential ability and enable to apply flexible energy storage and wearable devices.
This study was conducted to investigate the optimal culture method for gametophyte and sporophyte propagation in Coniogramme japonica (Thunb.) Diels, which can be used in various fields. The propagation of prothallus were cultured in 1/4 - 1 Murashige and Skoog medium and Knop medium for 10 weeks. The results indicated that the fresh weight of prothallus was the highest (14.5 g) in 1MS medium. Subsequently, various concentrations of sucrose, activated charcoal and nitrogen source were also added to 1MS medium and cultured for 8 weeks. The results provided that the sucrose concentration was 3% and the fresh weight of prothallus was the highest 10.8 g. According to the concentration in the range of 8.8 ~ 10.8 g, in the case of activated charcoal, the four treatments showed no significant difference. The nitrogen source was added at a concentration of 30, 60 and 120 mM with the ratio of ${NH_4}^+:{NO_3}^-$ being 1 : 2. As a result, the fresh weight of all treatments increased to similar level but there was no significant difference. We investigated sporophyte formation according to soil type and the highest number of sporophytes at 228.0 was formed in soils mixed with horticultural substrate and decomposed granite at 2 : 1 (v : v). On the other hand, sporophyte was not formed in soils containing peatmoss except for the one with peatmoss and decomposed granite at 2 : 1 (v : v).

To cope with automobile exhaust gas regulations, ISG (Idling Stop & Go) and charging control systems are applied to HEVs (Hybrid Electric Vehicle) for the purpose of improving fuel economy. These systems require quick charge/discharge performance at high current. To satisfy this characteristic, improvement of the positive electrode plate is studied to improve the charge/discharge process and performance of AGM(Absorbent Glass Mat) lead-acid batteries applied to ISG automotive systems. The bonding between grid and A.M (Active Material) can be improved by applying the Sand-Blasting method to provide roughness to the surface of the positive grid. When the Sand-Blasting method is applied with conditions of ball speed 1,000 rpm and conveyor speed 5 M/min, ideal bonding is achieved between grid and A.M. The positive plate of each condition is applied to the AGM LAB (Absorbent Glass Mat Lead Acid Battery); then, the performance and ISG life characteristics are tested by the vehicle battery test method. In CCA, which evaluates the starting performance at -18 ℃ and 30 ℃ with high current, the advanced AGM LAB improves about 25 %. At 0 ℃ CA (Charge Acceptance), the initial charging current of the advanced AGM LAB increases about 25 %. Improving the bonding between the grid and A.M. by roughening the grid surface improves the flow of current and lowers the resistance, which is considered to have a significant effect on the high current charging/discharging area. In a Standard of Battery Association of Japan (SBA) S0101 test, after 300 A discharge, the voltage of the advanced AGM LAB with the Sand-Blasting method grid was 0.059 V higher than that of untreated grid. As the cycle progresses, the gap widens to 0.13 V at the point of 10,800 cycles. As the bonding between grid and A.M. increases through the Sand Blasting method, the slope of the discharge voltage declines gradually as the cycle progresses, showing excellent battery life characteristics. It is believed that system will exhibit excellent characteristics in the vehicle environment of the ISG system, in which charge/discharge occurs over a short time.

This study was conducted to create a technology to remove acne bacteria with human-friendly materials. First, the Cutibacterium acnes (C. acnes) were adsorbed to the mica disc to grow, and then the biofilm was checked through an atomic microscope to see if the biofilm had grown. Based on the topographic image, the shape changed round, the size was 17% longer on average, and the phase value of the resonance frequency separating materials was observed as a single value, the biofilm grown by covering the extracellular polymeric substrate (EPS). As a result of processing 50 mM of amino acids in the matured biofilm, the concentration of C. acnes decreased when valine, serine, arginine and leucine were treated. Scanning with nanoindentation and AFM contact modes confirmed that the hardness of biofilms treated with Valine (Val) increased. This indicates that an AFM tip measured cell which may have more solidity than that of EPS. The experiment of fluorescent tagged to EPS displays an existence of EPS at the condition of 10 mM Val, but an inhibition of growth of EPS at the 50 mM Val. Number of C. acnes was also reduced above 10 mM of Val. Weak adhesion of biofilm generated from an inhibition of EPS formation seems to induce decrease of C. acnes. Accordingly, we elucidated that Val has an efficiency which eliminates C. acnes by approach of an inhibition of EPS.
MMBR system has been suggested as a promising system to resolve harvesting problems induced from low settling efficiency of microalgae. And recently, a lot of research on reducing fouling at the MMBR system has investigated focused on EPS in many cases. EPS of microalgae mainly consists of polysaccharides and protein components, and is produced through photosynthesis and nitrogen-carbon metabolic pathways. Especially, P-EPS is one of major compounds which occur membrane fouling phenomenon, as its hydrophobic protein components cause floc formation and cake layer accumulation. And it is already known that almost every microalgae can metabolize P-EPS or Chl-a when nitrogen sources as a substrate is insufficient or exhausted situation. With the above backgrounds, uptake rates of P-EPS or Chl-a by Scenedesmus quadricauda according to the type of carbon source and nitrogen concentration were evaluated in order to verify correlation between carbon source vs P-EPS production, and indeed Scenedesmus quadricauda uses P-EPS or Chl-a when the amounts of nitrogen sourc es in the feed is not satisfied. As a result, it was shown that P-EPS and Chl-a production were increased proportional to nitrogen concentration under organic carbon condition. And especially, the amo unts of P-EPS and Chl-a in the cell were diminished with the nitrogen source becomes insufficient or exhausted. Because P-EPS accelerates fouling at the MMBR system, P-EPS degradation by Scenedesmus quadricauda in order to get nitrogen source may contribute to reducing fouling. About a affects of N-consumed Chl-a to the MMBR fouling, more survey is needed. On the contrary, considering the purpose of MMBR system of this study, i.e. harvesting useful high value microalgae efficiently feeding adequate industrial process wastewater, it seems like difficult to maintain satisfied metabolic activity and to harvest with high yield rate using nitrogen-poor MMBR feed.
The effects of a high-fat diet and fasting on resting energy expenditure and energy substrate utilization were examined using the method of measuring whole body energy metabolism and oxygen uptake. Eight 4-week old male Sprague-Dawley rats were used for the high-fat diet experiment. Energy metabolism was measured using acrylic metabolic chambers over 24 hours. After 1-week of preliminary feeding, 4 rats were fed a chow diet, whereas the remaining 4 rats were fed a high-fat diet (HF) ad libitum, which contained 40% (w/w, calorie base 60%) more fat than that in the chow diet. The flow rate to measure energy metabolism inside the chamber was controlled at a mean of 3.5 L/min, and five chambers were subjected to measurement. One of the five chambers was used to correct errors by measuring the atmosphere. As a result of 5 weeks of control diet and high-fat diet feeding, body weight of the high-fat diet group tended to increase more than that in the control diet fed group, but the difference was not significant. Oxygen uptake and carbon dioxide production changed as time went on over the 24 hr. The respiratory exchange ratio also changed during the 24 hr, and the difference between the groups was significant. The control group showed significantly more carbohydrate oxidation than that of the high-fat diet fed group. A fasting experiment was conducted using six 7-week old Sprague-Dawley male rats. Energy metabolism measurements were performed using the same method as that used in the high-fat diet experiment; resting metabolism was measured prior to fasting, and a fasting condition began from 9:00 am the next day for 3 days to calculate energy metabolism. Both body weight and 24-hour oxygen uptake decreased significantly as a result of 3-day fasting. Total oxygen uptake in the first day decreased, and declined significantly on day 3 of fasting. Total 24-hour carbon dioxide production decreased significantly over the 3 days. The mean 24-hour respiratory exchange ratio decreased significantly. Additionally, energy expenditure during the dark period (20:00-08:00), which is the active period for rats, decreased significantly with fasting, whereas energy expenditure during the light period (08:00-20:00) did not increase by fasting.
Recently, electronic devices with bendable electronic devices based on flexible substrates are being sold, and therefore, the purpose of this study is to evaluate the possibility of flexible substrates of conductive transparent tapes. As a transparent electrode, carbon nanotube (CNT) was formed by the coating method developed by the research team, and samples coated up to 5 times were fabricated. The surface resistance and transmittance of the substrate were measured, and both resistance and transmittance decreased as the number of CNT coatings increased. After the tape was detached from the glass, the surface resistance slightly increased in all samples, and the transmittance increased by about 10% in all measured wavelength ranges because the glass was removed. Next, the tape coated with CNT twice was used to a bending test 20,000 times under the condition of a radius of curvature of 2 mm. The electrical and optical properties before and after bending did not change, which means that there was no change in CNT properties due to bending.
Protein modification by N-acetyl-${\beta}$-D-glucosamine (O-G1cNAc) on the hydroxyl groups of Ser or Thr ubiq-uitously occurs in eukaryotic cells and is involved in many cellular phenomena. The level of O-G1cNAc-mod-ified protein is regulated by OGT and O-GlcNAcase enzymes. We have tried to produce recombinant O-GlcNAcase in E. coli as an effort to establish in vitro screening system for modulators of O-GlcNAcase. The culture conditions for improvement of O-GlcNAcase productivity, were as follows: induction temperature, $30^{\circ}C$; the concentration of L-arabinose, 0.02% and induction time, 5 hr. Under these culture conditions, E. coli cells containing O-GlcNAcase gene had no enzyme activity until up to 3 hr culture. However, O-GlcNAcase activity dramatically increased from 3 to 5 hr culture. It almost maintained the same level after 5 hr culture. Western blot analysis verified the amount of expressed O-GlcNAcase increased with culture time, being con-sistent with activity data. The optimal reaction condition determined in this study was as follows: protein quan-tity, $5{\mu}g$; reaction time, 30 min; reaction temperature, $45^{\circ}C$; substrate concentration, 2 mM; reaction pH, 6.5. Methanol had little effect on O-GlcNAcase activity and 90% of activity were retained at 10%. Only 15% resid-ual activity were detected at 5% of chloroform.
Purpose: Type 2 diabetes mellitus is a metabolic condition marked by persistent elevated blood sugar levels resulting from insulin resistance. The effective management of diabetes mellitus involves strict regulation of the blood glucose levels. This study examined the effects of Autumn olive (Elaeagnus umbellata Thunb.) berry (AOB) on insulin resistance and hyperglycemia using a type 2 diabetes mellitus animal model. Methods: Eight-week-old C57BL/6J mice were divided into four groups. The control group received a basal diet, while the high-fat, high-sucrose (HFHS) group was fed a HFHS diet containing 27% sucrose and 33% lard for 12 weeks. The low AOB (LAOB) and high AOB (HAOB) groups were offered a HFHS diet with a 0.5% and 1.0% AOB extract, respectively. Results: The HAOB group showed significantly lower epididymal fat pad weight than the HFHS group. The LAOB and HAOB groups showed lower serum glucose levels and homeostasis model assessment for insulin resistance values than the HFHS group, and the HAOB group has lower serum insulin levels than the HFHS group. Supplementation with HAOB decreased serum cholesterol levels significantly compared with the HFHS group. The consumption of LAOB and HAOB reduced the serum triglyceride and hepatic total lipids and triglyceride levels compared to the HFHS group. In addition, LAOB and HAOB consumption in mice fed a HFHS diet increased adenosine monophosphate-activated protein kinase protein expression. Insulin receptor substrate-2 protein expression in the HAOB group was significantly higher than the HFHS group. Conclusion: AOB can alleviate hyperglycemia in type 2 diabetes mellitus partly by mitigating insulin resistance.
