Synthesis and in vitro Activity of Novel 1β-Methylcarbapenems Having Spiro[2,4]heptane Moieties. Part II

  • The synthesis of a new series of 1$\beta$-methylcarbapenems having spiro[2,4]heptane moieties is described. Their in vitro antibacterial activities against both Gram-positive and Gram-negative bacteria were tested and the effect of substituents at the pyrrolidine ring was investigated. Most of the compounds were found to be more active compared to imipenem against Gram-negative bacteria. A particular compound (IIIc) having 7-oxo-5- azaspiro[2,4]heptane moiety showed the most potent antibacterial activity.

  • To extend the knowledge of triple bonding between group 6 transition metal and heavier group 14 elements, the structural and bonding aspects of ($η^5-C_5H_5$)$(CO)_2$M≡ER (M = Cr, Mo, W; E = Si, Ge, Sn, Pb) are investigated by hybrid density functional calculations at the B3PW91 level. Substituent effects are also investigated with R = H, Me, $SiH_3$, Ph, $C_6H_3-2,6-Ph_2$, $C_6H_3-2,6-(C_6H_2-2,4,6-Me_3)_2$, and $C_6H_3-2,6-(C_6H_2-2,4,6- iPr_3)_2$.

  • The rate constants for the addition reaction of thiourea to nitrone derivatives were determind at various pH and reaction rate equation which could be applied over a wide pH were obtained. The substituent effects and general base catalysis for the addition of thiourea to nitrone derivatives were observed. On the basis of these findings, a plausible reaction mechanism for the nucleophilic addition of thiourea to nitrone was proposed.

  • Novel diphenylphenanthrenes (DPPs) were prepared by a photocyclization method, and the substituent effects of the phenyl groups on the photophysical properties of the phenanthrene chromophore were investigated based on measurements of fluorescence yields, lifetimes, and transient absorption. Fluorescence activities in DPPs are increased by introducing phenyl rings that can enhance the transition moment along the short axis of the phenanthrene skeleton. Intersystem crossing from the fluorescent states to the triplet manifolds is shown to be operative through the triplet-triplet absorption spectra obtained by laser photolysis techniques.

  • The catalytic activities of several metalloporphyrin, wherein the porphyrins are TPP(5,lO,l5,20-Tetraphenyl-21H,23H-porphyrin) and (p-X)TPP (X =$CH_{3}O$, $CH_{3}$, F, Cl), are reported for the oxidation of styrene and it's derivatives. The electronic effects of substrates and porphyrins on the catalytic activity of metalloporphyrin containing the transition metal ion such as Mn(III) was discussed. Investigating the correlation between the Michaelis-Menten's rate parameters and the substituent constants, we are going to analyze the influences on the changes of catalytic activity or rate determining step during the processes of the formation and the dissociation of the M-oxo-olefin.

  • The rate constants of hydrolysis of N-tert-butyl-${\alpha}$-phenylnitrone and its derivatives have been determined by UV spectrophotometry at $25^{\circ}C$ and a rate equation which can be applied over a wide pH range was obtained. On the basis of rate equations derived and judging from the hydrolysis products obtained and general base and substituent effects, plausible mechanism of hydrolysis in various pH range have been proposed. Below pH 4.5, the hydrolysis was initiated by the protonation and followed by the addition of water to ${\alpha}-carbon$. Above pH 10.0, the hydrolysis was proceeded by the addition of hydroxides ion to ${\alpha}-carbon$. In the range of 4.5${\sim}$10.0 the addition of water to nitrone was rate controlling step.

  • In order to evaluate the potential of partial hydrocarbon substitution to improve the safety of hydrogen use in general and the performance of internal combustion engines in particular, the outward propagation and development of surface cellular instability of spark-ignited spherical premixed flames of mixtures of hydrogen, hydrocarbon, and air were experimentally studied at NTP (normal temperature and pressure) condition in a constant-pressure combustion chamber. With propane being the substituent, the laminar burning velocities, the Markstein lengths, and the propensity of cell formation were experimentally determined, while the laminar burning velocities and the associated flame thicknesses were computed using a recent kinetic mechanism. Results show substantial reduction of laminar burning velocities with propane substitution, and support the potential of propane as a suppressant of both diffusional-thermal and hydrodynamic cellular instabilities in hydrogen-air flames.

Precursor and Substituent Effects on the Alternate Adsorption of Polycation and Anionic Dyes as Studied by Second Harmonic Generation and Absorption Spectroscopies

  • An alternately assembled polycation-anionic dye film was prepared on a precursor assembly of poly(allylamine)hydrochloride (PAH) and poly(sodium styrenesulfonate) (PSS) or of poly (djallyldimethyl)ammonium chloride (PDDA) and PSS. The (PAH/PSS) precursor assembly gave better adsorptivity to the anionic dye than the (PDDA/PSS) one. Four kinds of anionic dyes (EB-T, AR-17 AR-18, AR-27) with different numbers of sulfonate substituents were compared. The extent of dye assembling was higher for the dye with a smaller number of sufonate substituents. Second harmonic generation (SHG) was clearly observed from the films. The SHG signal and the absorption intensity correlated well up to four polycation/dye bilayers, while further assembly did not increase the SHG signal appreciably. Second order nonlinear coefficients for ten bilayer assemblies were the order of pm/V.

Synthesis of Novel Fluorescent Dye Based on Fluorescein

  • The functional materials have been developed as a promising research topic toward the end uses for organic materials and applications. In this study, fluorescein based dye was synthesized by three step reaction. We have designed and synthesized the colorimetric dye through the reactions of fluorescein and methoxy group and ethylene diamine and squaric acid. The structure of the non-fluorescent spirolactam was elucidated by $^1H$-NMR, LC-Mass and FT-IR analyzes. Further studies are in progress to understand the effects of various substituent during the recognition process and to develop fluorescein based sensors for cations or anions.

  • Electronic spectra and quenching constants for ten N,N-dimethylanilines (DMA) have been determined. The $250m{\mu}$ absorption band (C band) of DMA was shown to be mainly responsible for the quenching. This band was confirmed as an $n{\to}{\pi}^{\ast}$ band through substituent and medium effects on the spectra shifts. The energy absorbed by this band then transfers to triplet and down to the lowest triplet state, $^{3}La$, where the energy is lost by non-radiative collisions.
