이플루오린화 암모늄 시료분해 및 ICP/MS에 의한 암석 및 퇴적물 중 게르마늄 분석 (Analysis of germanium in rock and sediment by ICP/MS after ammonium bifluoride(NH4HF2) digestion)

  • 본 연구에서는 암석과 퇴적물 시료를 대상으로 이플루오린화 암모늄을 이용한 시료 분해법을 적용하여 open vessel digestion 후 유도결합 플라즈마 질량분석법(Inductively coupled plasma mass spectrometry, ICP/MS)으로 게르마늄을 분석하였다. 미국 지질조사소 표준물질 중 QLO-1, SDO-1을 사용하였고, 실제 시료로 GeoPT용 퇴적물, 현무암, 점토를 사용하였다. 휘발하여 손실되기 쉬운 것으로 알려진 게르마늄을 이플루오린화 암모늄($NH_4HF_2$) 시료 분해법을 사용함으로서 게르마늄 휘발 억제와 동시에 암석 및 퇴적물을 open vessel digestion하에서 간단하고 빠르게 분해 가능하였으며, 또한 이플루오린화 암모늄 분해법에 과산화수소를 함께 사용해도 게르마늄의 회수율에 영향을 끼치지 않았다. ICP/MS에 의한 게르마늄 분석 결과, MDL (method detection limit)은 0.015 ${\mu}g/g$, 게르마늄 회수율은 106~128% 이었다.

Preparation, Structural Investigation and Thermal Decomposition Behavior of Two High-Nitrogen Energetic Materials: ZTO·2H2O and ZTO(phen)·H2O

  • Two new high-nitrogen energetic compounds $ZTO{\cdot}2H_2O$ and $ZTO(phen){\cdot}H_2O$ have been synthesized (where ZTO = 4,4-azo-1,2,4-triazol-5-one and phen = 1,10-phenanthroline). The crystal structure, elemental analysis and IR spectroscopy are presented. Compound 1 $ZTO{\cdot}2H_2O$ crystallizes in the orthorhombic crystal system with space group Pnna and compound 2 $ZTO(phen){\cdot}H_2O$ in the triclinic crystal system with space group P-1. In $ZTO(phen){\cdot}H_2O$, there is intermolecular hydrogen bonds between the -NH group of ZTO molecule (as donor) and N atom of phen molecule (as acceptor). Thermal decomposition process is studied by applying the differential scanning calorimetry (DSC) and thermo thermogravimetric differential analysis (TG-DTG). The DSC curve shows that there is one exothermic peak in $ZTO{\cdot}2H_2O$ and $ZTO(phen){\cdot}H_2O$, respectively. The critical temperature of thermal explosion ($T_b$) for $ZTO{\cdot}2H_2O$ and $ZTO(phen){\cdot}H_2O$ is $282.21^{\circ}C$ and $195.94^{\circ}C$, respectively.

침전법으로 제조된 전구체 성질에 의한 LiCoO2의 특성에 관한 연구 (Characterization of LiCoO2 Synthesized via Structural and Compositional Variations of Precursors Prepared by Precipitation)

  • 본 연구에서는 수산화물 전구체로부터 HT-$LiCoO_2$ 분말을 제조하였다. 사용된 $Co(OH)_2$${\alpha}$상 및, 또는 ${\beta}$ 상의 수산화물이며 여러 조건에서 침전법으로 제조되었다. 제조시에 $[OH^-]/[CO^{2+}]$ 농도 비와 숙성시간을 조절함으로써 원하는 특성을 지닌 $Co(OH)_2$ 제조가 가능하였다. 침전법으로 합성한 $Co(OH)_2$와 LiOH를 메탄올 수용액을 이용하여 혼합한 후 이어서 증발, 건조 그리고 숙성 조작에 의하여 HT-$LiCoO_2$의 전구체를 합성하였다. 숙성기간에 tailor-made ${\beta}$-phase $Co(OH)_2$는 CoOOH로 변환되면서 고용체를 형성한다. 이 같은 결과로 HT-$LiCoO_2$가 낮은 온도($600^{\circ}C$), 짧은 시간(10분)에 합성이 가능하게 된다.

LC/MS/MS를 이용한 산화성 스트레스 지표로써 소변 중 8-iso-PGF 분석 (Determination of 8-iso-PGF as Oxidative Stress Marker in Human Urine by High Performance Liquid Chromatography with Tandem Mass Spectrometry)

  • This study aimed to develop analytical method for 8-isoprostanes as biomarkers for oxidative stress with LC/MS/MS technique and to apply the method for human urine samples. Analyzed compounds for urinary oxidative stress markers were 7 stereo-isomers of prostaglandins and the internal standard (iso-$PGF_{2{\alpha}}-d_4$) was used to adjust the recovery rate. The method for determining urinary iso-$PGF_{2{\alpha}}$ consisted of solid phase extraction and LC/MS/MS detection. Separation of isomers of prostaglandins completed by porous graphitic carbon column and buffer solution. Detection limits for urinary markers of oxidative stress, iso-$PGF_{2{\alpha}}$ with LC/MS/MS were 0.01 ng/ml by S/N ratio 3 and 0.028 ng/ml by calculated as to FDA method. The recovery (92.8~101.9%) and precision (8.8~20.7%) of analysis were feasible for detecting iso-$PGF_{2{\alpha}}$ in real human urine samples. We detected 4 isomers of prostaglandins in human urine samples. Mean (standard deviation) of urinary iso-$PGF_{2{\alpha}}$ concentration were 0.231 (0.117) ng/mg creatinine for smoking group and 0.154 (0.082) ng/mg creatinine for non-smoking group.

다양한 아미노실란을 이용한 이산화탄소 흡착제 합성 및 흡착 특성 (Synthesis of CO2 Adsorbent with Various Aminosilanes and its CO2 Adsorption Behavior)

  • 넓은 비표면적과 큰 기공 부피를 갖는 실리카에 다양한 아미노실란 화합물을 in-situ 중합법을 통해 기능화 후 이산화탄소 흡착 특성을 확인하였다. 이산화탄소 흡착 기능기로 아민기가 포함된 아미노실란 화합물이 사용되었다. 흡착제의 흡착 특성 분석을 위해 질소 흡 탈착 실험과 원소분석, in situ FT-IR, TGA를 이용하였다. 흡착제 합성 전후를 비교하였을 때 폴리아미노실란이 기능화되면 표면적과 기공부피 및 크기가 감소하였으며 실리카 기공 부피의 70%에 해당하는 폴리아미노실란 화합물을 기능화 시켰을 경우 기공 부피의 100% 기능화 보다 이산화탄소 흡착능이 향상되었다. 흡착 온도를 비교하며 $30^{\circ}C$보다 $75^{\circ}C$에서 폴리아미노실란 화합물의 열팽창과 자유부피 증가로 흡착능이 증가하였고, 2NS/NPS-2의 경우 기공 부피 70% 기능화와 흡착 온도 $75^{\circ}C$에서 9.2 wt%의 높은 $CO_2$ 흡착능을 보였다.

The Neuro-Protective Effect of the Methanolic Extract of Perilla frutescens var. japonica and Rosmarinic Acid against H2O2-Induced Oxidative Stress in C6 Glial Cells

  • Neurodegenerative diseases are often associated with oxidative damage in neuronal cells. This study was conducted to investigate the neuro-protective effect of methanolic (MeOH) extract of Perilla frutescens var. japonica and its one of the major compounds, rosmarinic acid, under oxidative stress induced by hydrogen peroxide ($H_2O_2$) in C6 glial cells. Exposure of C6 glial cells to $H_2O_2$ enhanced oxidative damage as measured by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide and thiobarbituric acid-reactive substance assays. The MeOH extract and rosmarinic acid prevented oxidative stress by increasing cell viability and inhibiting cellular lipid peroxidation. In addition, the MeOH extract and rosmarinic acid reduced $H_2O_2-indcued$ expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) at the transcriptional level. Moreover, iNOS and COX-2 protein expression was down-regulated in $H_2O_2-indcued$ C6 glial cells treated with the MeOH extract and rosmarinic acid. These findings suggest that P. frutescens var. japonica and rosmarinic acid could prevent the progression of neurodegenerative diseases through attenuation of neuronal oxidative stress.

Polyphenolic Biflavonoids Inhibit Amyloid-Beta Fibrillation and Disaggregate Preformed Amyloid-Beta Fibrils

  • Alzheimer's disease (AD) is a devastating neurodegenerative disease and a major cause of dementia in elderly individuals worldwide. Increased deposition of insoluble amyloid β (Aβ) fibrils in the brain is thought be a key neuropathological hallmark of AD. Many recent studies show that natural products such as polyphenolic flavonoids inhibit the formation of insoluble Aβ fibrils and/or destabilize β-sheet-rich Aβ fibrils to form non-cytotoxic aggregates. In the present study, we explored the structure-activity relationship of naturally-occurring biflavonoids on Aβ amyloidogenesis utilizing an in vitro thioflavin T assay with Aβ1-42 peptide which is prone to aggregate more rapidly to fibrils than Aβ1-40 peptide. Among the biflavonoids we tested, we found amentoflavone revealed the most potent effects on inhibiting Aβ1-42 fibrillization (IC50: 0.26 µM), as well as on disassembling preformed Aβ1-42 fibrils (EC50: 0.59 µM). Our structure-activity relationship study suggests that the hydroxyl groups of biflavonoid compounds play an essential role in their molecular interaction with the dynamic process of Aβ1-42 fibrillization. Our atomic force microscopic imaging analysis demonstrates that amentoflavone directly disrupts the fibrillar structure of preformed Aβ1-42 fibrils, resulting in conversion of those fibrils to amorphous Aβ1-42 aggregates. These results indicate that amentoflavone affords the most potent anti-amyloidogenic effects on both inhibition of Aβ1-42 fibrillization and disaggregation of preformed mature Aβ1-42 fibrils.

Structures and Formation Energies of LixC6 (x=1-3) and its Homologues for Lithium Rechargeable Batteries

  • Using first principles density functional theory the formation energies of various binary compounds of lithium graphite and its homologues were calculated. Lithium and graphite react to form $Li_1C_6$ (+141 mV) but not form $LiC_4$ (-143 mV), $LiC_3$ (-247 mV) and $LiC_2$ (-529 mV) because they are less stable than lithium metal itself. Properties of structure and reaction potentials of $C_5B$, $C_5N$ and $B_3N_3$ materials as iso-structural graphite were studied. Boron and nitrogen substituted graphite and boron-nitrogen material as a iso-electronic structured graphitic material have longer graphene layer spacing than that of graphite. The layer spacing of $Li_xC_6$, $Li_xC_5B$, $Li_xC_5N$ materials increased until to x=1, and then decreased until to x=2 and 3. Nevertheless $Li_xB_3N_3$ has opposite tendency of layer spacing variation. Among various lithium compositions of $Li_xC_5B$, $Li_xC_5N$ and $Li_xB_3N_3$, reaction potentials of $Li_xC_5B$ (x=1-3) and $Li_xC_5$ (x=1) from total energy analyses have positive values against lithium deposition.

Metabolic Activities of Ginseng and Its Constituents, Ginsenoside Rb1 and Rg1, by Human Intestinal Microflora

  • To evaluate the difference in expressing pharmacological effects of ginseng by intestinal microflora between Koreans, metabolic activities of ginseng, ginsenoside $Rb_1$ and $Rg_1$ by 100 fecal specimens were measured. The ${\beta}$-glucosidase activity for p-nitrophenyl-${\beta}$-D-glucopyranoside was 0 to 0.42 mmol/min/mg and its average activity (mean${\pm}$SD) was $0.10{\pm}0.07$ mmol/min/mg. The metabolic activities of ginsenosides Rb1 and Rg1 were 0.01 to 0.42 and 0.01 to 0.38 pmol/min/mg, respectively. Their average activities were $0.25{\pm}0.08$ and $0.15{\pm}0.09$ pmol/min/mg, respectively. The compound K-forming activities from ginsenoside Rb1 and ginseng extract were 0 to 0.11 and 0 to 0.02 pmol/min/mg, respectively. Their average compound K-forming activities were $0.24{\pm}0.09$ pmol/min/ mg and $2.14{\pm}3.66$ fmol/min/mg, respectively. These activities all were not different between males and females, or between ages. Although compound K-forming activity from the aqueous extract of ginseng was low compared to that from ginenoside $Rb_1$, their profiles were similar to those of isolated compounds. Based on these findings, we believe that the intestinal bacterial metabolic activities of ginseng components are variable in individuals and may be used as selection markers for responders to ginseng.

카드뮴으로 유발된 산화 스트레스에 대한 진피의 간세포 보호 및 항산화 효과 (Hepatocyte protection and antioxidant effect of Citri Unshius Pericarpium against cadmium-induced oxidative stress)

  • Objective : Citri Unshius Pericarpium is the dried peel of mature fruit of Citrus unshiu Markovich and has been used to treat indigestion, vomiting, and removal of phlegm. This study investigated the hepatoprotective and antioxidant effect of CEE (Ethanol extract of Citri Unshius Pericarpium) in cadmium (CdCl2)-treated HepG2 cells. Methods : Component analysis of Citri Unshius Pericarpium was analyzed by UPLC with C18 column. Cell viability was determined by MTT assay. The enzyme activity of superoxide dismutase (SOD) and the level of reactive oxygen species (ROS) and reduced glutathione (GSH) were analyzed using commercially available kits. Results : Cadmium caused severe HepG2 cell death. Cadmium also increased ROS production, consistent with depletion of GSH and inhibition of the SOD enzyme. However, CEE treatment reduced cell death and relieved oxidative stress caused by cadmium toxicity. CEE lowered ROS levels and improved depletion of GSH levels. CEE also enhanced the enzymatic activity of SOD. In component analysis, hesperidin was the most abundant of the five marker compounds (Narigenin, Narigin, Narirutin, Hesperidin and Hesperidin), which assumes that hesperidin partially contributed to the antioxidant activity of CEE. Conclusion : These results suggested that CEE could be a potential substance to solve heavy metal-related health problems. In particular, inhibition of oxidative stress by CEE can be a way to treat liver damage caused by cadmium.
