Optimization of Enzymatic Synthesis Condition of Structured Lipids by Response Surface Methodology (반응표면분석에 의한 기능성 유지의 효소적 합성 조건 최적화)

  • Synthesis conditions were optimized using response surface methodology for producing structured lipids (SL) by interesterification of DHA-enriched algae oil derived from microalgae, Schizochytrium sp. and corn oil. Reaction was performed fer 24 hr at $55^{\circ}C$ catalyzed by immobilized lipase from Rhizomucor miehei (RM IM) in shaking water bath. Major fatty acids of SL were palmitic (21.70 mol%), oleic (20.20 mol%), and linoleic (27.34 mol%) acids, and DHA (15.06 mol%). To separate newly synthesized SL-triglycerides (TG) species, HPLC with evaporative light scatting detector (ELSD) was used. Production conditions were optimized using central composite design with reaction temperature $(35-75^{\circ}C,\;X_1)$, reaction time $(2-42\;hr,\;X_2)$, and enzyme concentration $(2-14%,\;X_3)$ as variables. When variables were $70.28^{\circ}C\;(X_1),\;28.74\;hr\;(X_2),\;and\;11.30%\;(X_3)$, maximum content of selected three peaks of synthesized SL-TG species was predicted as 6.97 area%.

Optimization of Lipase-Catalyzed Production of Structured Lipids from Canola Oil Containing Similar Composition of Triacylglycerols to Cocoa Butter (Canola Oil로부터 코코아버터와 유사한 Triacylglycerol 조성을 가진 재구성지질의 효소적 합성 최적화 연구)

  • Synthesis conditions of cocoa butter equivalents were optimized using the response surface method (RSM) by interesterification of canola oil (Ca), palmitic ethyl ester (PEE), and stearic ethyl ester (StEE). The reaction was catalyzed by immobilized lipase (Lipozyme TLIM) from Thermomyces lanuginosa to produce structured lipids containing a composition of triacylglycerols similar to cocoa butter. Reaction conditions were optimized using D-optimal design with the three reaction factors of the substrate molar ratio of canola oil to palmitic ethyl ester and stearic ethyl ester (Ca : PEE : StEE=1:1:3, 1:1.66:5, 1:2:6, 1:2.33:7, 1:3:9, $X_1$), enzyme ratio (2~6%, $X_2$), and reaction time (30~270 min, $X_3$). The optimal conditions that minimized acyl-migration while maximizing 1-palmitoyl-2-oleoyl-3-stearoyl glycerol (POS), 1,3-distearoyl-2-oleoyl glycerol (SOS), and 1,3-dipalmitoyl-2-oleoyl glycerol (POP) were predicted, resulting in Ca : PEE : StEE=1:3:9, 6% of enzyme ratio, and 40 min of reaction time. The reaction product of structured lipids was synthesized again under the same conditions, showing 10.43 area% of acyl-migration, 25.31 area% of POS/PSO, 19.79 area% of SOS, and 11.22 area% of POP.

Synthesis and Characterization of Structured Lipids from Evening Primrose Seeds Oil and Rice Bran Oil (달맞이꽃 종자유와 미강유로부터 효소적 합성한 재구성 지질의 이화학적 특성 분석)

  • Structured lipids (SLs) were synthesized by enzymatic interesterification with evening primrose oil (EPO) and rice bran oil (RBO) in a batch-type reactor. The interesterification was performed using a water shaker for 24 hr at $55^{\circ}C$. Mixing speed was set at 200 rpm and Lipozyme RM IM (immobilized lipase from Rhizomucor miehei, 10% by weight of total substrates) was used as a biocatalyst. Rice bran oil and evening primrose oil were interesterified with various molar ratios (RBO : EPO, 1:3, 1:4, and 1:5 mol/mol). Reversed-phase high performance liquid chromatography connected with evaporative light-scattering detector was performed to separate the triacylglycerol (TAG) species of SLs. In the fatty acid analysis, $\gamma$-linolenic acid (7.9 mol%), linoleic acid (67.3 mol%) and oleic acid (13.2 mol%) were the most abundant fatty acids in the SLs. During 24 hr reaction, most of the reaction occurred within 3 hr. TAG compositions, tocopherols and phytosterols were also analyzed. In the TAG species analysis, LLL (ECN=42, L=linoleic acid) dramatically decreased when the reaction time increased.

Synthesis of Structured Lipids from Corn Oil and Conjugated Linoleic Acid with Immobilized Lipase-Catalyzed Reaction (옥수수유와 conjugated linoleic acid로부터 고정화 효소를 이용한 재구성지질의 합성)

  • Structured lipids (SL) were synthesized by esterification of corn oil and conjugated linoleic acid (CLA) in a continuous packed-bed column reactor. The effects of flow rate, reaction temperature, and substrate molar ratios were studied. The reaction was catalyzed by TL IM (immobilized lipase from Thermomyces lanuginosa). Results of triacylglycerol (TAG) analysis by GC showed that the incorporated CLA isomers were mainly cis9, trans11- and trans 10, cis12-CLA. Slower flow rates yielded higher incorporation, and maximum incorporation of CLA was obtained with a molar ratio of 1:3 (corn oil: CLA) at a temperature of $55^{\circ}C$. The obtained SLs had iodine values ranging from 120 to 128. The SLs were composed of TAG $(98{\sim}99%)$, 1,2- and 1,3-diacylglycerol ($0.7{\sim}1.3%$), and a small amount of monoacylglycerol.

Optimization for the Phytosterol Extraction and Production of Structured Lipids from Safflower seed (홍화씨로부터 Phytosterol 추출의 최적화 및 재구성지질의 합성)

  • Response surface methodology was used for monitoring and optimizing the extraction conditions of campesterol, stigmasterol, ${\beta}$ -sitosterol, and total sterols from the safflower seed. The conditions of phytosterol extraction were optimized by using central composite design with the temperature(35∼75$^{\circ}C$, X$_1$), the time (1∼11hr, X$_2$), and the preheating temperature(60∼100$^{\circ}C$, X$_3$) as three variables. The extraction conditions for maximum campesterol content were 59.01$^{\circ}C$(X$_1$), 2.88hr(X$_2$), and 75.04$^{\circ}C$(X$_3$). But stigmasterol, ${\beta}$ -sitosterol and total sterols were not significantly different under designed extraction condition in this study. Besides, oil was extracted from safflower seed at various conditions and yields were 23.44% at 35$^{\circ}C$ and 20.05% at 80$^{\circ}C$, respectively. Total tocopherol content increased from 0.172% to 0.207% as the extraction temperature increased from 35$^{\circ}C$ to 80$^{\circ}C$. A structured lipids(SL) was synthesized enzymatically by extracted safflower oil and conjugated linoleic acid(CLA). After 24hr reaction, 31.79 mol% CLA was incorporated into the extracted safflower oil.

Analysis of Phytosterols and Tocopherols, and Production of Structured Lipids from the Extracted Plant Oils (선택된 식물자원에서 추출조건에 따른 Phytosterol과 Tocopherol의 함량분석 및 추출유로부터 재구성지질의 합성)

  • To obtain the oil sesame, walnut, whole wheat, and rice bran were extracted for 1, 3, or 6 hr by a shanking water bath(35$^{\circ}C$ and 100 rpm), and by soxhlet extractor(80$^{\circ}C$) for 1, 3 or 6 hr, respectively. The highest yield of extracted oil was obtained from the walnut(63.07% weight) and the whole wheat showed the lowest extraction yield of oil(1.13% weight). Major fatty acids from the extracted oils were linoleic, oleic, and palmitic acid. The maximum contents of total phytosterol in sesame, walnut, whole wheat, and rice bran were 0.44, 1.57, 2.25, and 2.03(% weight), respectively. Besides, total tocopherol contents in sesame, walnut, whole wheat, and rice bran were maxima 3.42, 0.16, 2.92, and 0.07(% weight), respectively. From the extracted oils, structured lipids(SL) were synthesized by the interesterification reaction with conjugated linoleic acid(CLA) in a shanking water bath at 55$^{\circ}C$. When the reactions(1:3 substrate molar ratio, extracted oil:CLA) were conducted for 24 hr, maxima 23.75 mol% of CLA incorporation was obtained from walnut oil and, in other cases, 16.28 - 19.15 mol% of CLA was found in the produced SL triacylglycerol molecules.

Enzymatic Synthesis of Structured Lipids Containing Conjugated Linoleic Acid from Extracted Corn and Peanut Oil (추출된 옥수수유와 땅콩유로부터 Conjugated Linoleic Acid를 함유한 재구성지질의 효소적 합성 및 분석)

  • Structured lipids (SLs) were synthesized by acidolysis of crude oils (corn and peanut oil) and conjugated linoleic acid (CLA) with a molar ratio of 1:3 (extracted oil:CLA) in a shaking water bath. The reaction was performed for various reaction time (1, 2, 3, 6 and 24 hr) at 55$^{\circ}C$ with 175 rpm catalyzed by sn -1,3 specific IM 60 lipase from Rhizomucor miehei. The content of the incorporated CLA increased with the prolonged reaction time, showing 7.5∼9.3 ㏖% in the synthesized SL triacylglycerol molecules from the extracted corn and peanut oil. However, total tocopherol content in SLs decreased up to about 20% compared to the content in extracted oils. Among the CLA isomers, 6.3∼7.5 ㏖% of cis 9,trans 11- and trans 10,cis 12-CLA known as physiologically active compounds are contained in corn and peanut SLs.

Lipase-catalyzed Transeseterification of Corn Oil, Conjugated Linoleic Acid, and Capric Acid in Batch Type Reactor

  • Structured lipids (SLs) are defined as triacylglycerols to change the fatty acid composition in the glycerol backbone and lipases are known as a powerful tool for the syntheses of SLs. Structured lipid from corn oil, capric acid, and conjugated linoleic acid (CLA) by transesterification reaction and using several amounts of immobilized lipase RM IM (from Rhizomucor miehei) was studied, and 4% of lipase amount was selected for further study as the optimal amount. Comparison the chemical properties (free fatty acid value, iodine value, saponification value, tocopherols, and color analysis), solidification behavior, and volatile fractions (from headspace SPME GC-MS) between com oil and SL com oil was obtained.

Serum Lipids by Gender, Age and Lifestyle in Korean Adults (일부 한국성인의 성, 연령 및 생활습관에 따른 혈청지질 농도)

  • The purpose of this study was to measure serum lipids concentrations by gender and age, and to investigated the relation of serum lipids levels to cigarette smoking alcohol intake exercise and menopausal status in Korean adults. The subjects consisted of 463 healthy adult(29- men and 173 women) volunteers who were visiting the health Promotion Center in the Asan Medical Center from July 1, 1994 to July 22, 1994 Subjects were ambulatory free-living people and aged 20- 80 years, Anthropometric measurement was performed and serum triglyceride(TG), HDL-cholesterol and total cholesterol(TChol) concentrations were measure by automatic system(Hitachi 736-40). Data on life styles were obtained using structured questionnaires issued by a trained interviewer, All data were statistically analyzed by PC-SAS(Version 6.04) Serum concentrations of triglyceride and total cholesterol was higher(p<0.05) in men than in women, and serum HDL-cholesterol was lower(p<0.05) in men than in women. The prevalence of hyperlipidemia(TG>170 mg/dL and TChol>220 mg /dL) was 12.5% in men and 4.1% in women. The levels of serum triglyceride, total cholesterol and BMI increased with age in women. The distribution curves of serum triglyceride, total cholesterol and LDL-cholesterol of women were located in lower increased more rapidly with aging than in men, The distribution curves of BMI of women increased with aging after the thirties, The BMI of women was located in higher levels than of men in their sixties. The serum concentration of triglyceride and total cholesterol of male current-smokers and non- smokers were significantly(p<0.05) higher than those of ex-smokers. The concentrations triglyceride the male current0drinkers were higher(p<0.05) than those of male non-drinkers. In non-smokers and non- drinkers, serum TG concentration were lower (p<0.05) than those of current-smokers and current-drinkers. In exercisers, the serum triglyceride concentration was significantly(p<0.05) lower than in non- exercisers. In postmenopausal woman BMI and serum concentrations of TChol, TG and LDL-Chol were significantly by age and gender and smoking alcohol drinking, exercise and menopausal staus might be a better indicator to predict serum lipids in both genders.

Relationships among Lifestyle, BMI, BP, and Serum Lipid Profiles in Working Men (성인 직장 남성의 생활습관과 체질량지수, 혈압 및 혈중지질농도의 상관성)

  • Purpose: The purpose of this study was to examine lifestyle, BMI, BP, and lipids profiles in male subjects and to explore the relationships among variables. Methods: A total of 148 male subjects were recruited from one life insurance company from December 1, 2005 to February 28, 2006. Data collection methods were structured questionnaire, anthropometry and serum analysis. The relationships among lifestyle, BMI, BP, and serum lipid profiles were assessed by descriptive analysis, t-test, ANOVA, and partial Pearson's correlation coefficient of variables after controlling for age, educational level, and economic status. Results: The mean BMI of the participants was relatively high as 25.38(range: 18.38 - 32.83). The differences of serum lipid profiles according to age, educational level, and economic status were significant. 'Use of caffeine and drugs'(r = -.187, p < .05) and 'consciousness of safety'( r= -.200, p < .05) was negatively related to BMI. Higher score of 'type of personality' domain was correlated with lower systolic BP(r = -.221, p < .01) and lower diastolic BP(r = -.195, p < .05) and was positively correlated with HDL(r = .191, p < .05). Conclusion: 'Use of caffeine and drugs', 'consciousness of safety' and 'type of personality' of lifestyle as well as 'dietary habit' and 'exercise' played a key role in circulatory disease.
