The purple acid phosphatases comprise a family of binuclear metal-containing enzymes. The metal centre contains one ferric ion and one divalent metal ion. Spectroscopic studies of the monomeric, ${\sim}$36 kDa mammalian purple acid phosphatases reveal the presence of an Fe(III)Fe(II) centre in which the metals are weakly antiferromagnetically coupled, whereas the dimeric, ${\sim}$110 000 kDa plant enzymes contain either Fe(III)Zn(II) or Fe(III)Mn(II). The three dimensional structures of the red kidney bean and pig enzymes show very similar arrangements of the metal ligands but some significant differences beyond the immediate vicinity of the metals. In addition to the catalytic domain, the plant enzyme contains a second domain of unknown function. A search of sequence databases was undertaken using a sequence pattern which includes the conserved metal-binding residues in the plant and animal enzymes. The search revealed the presence in plants of a 'mammalian-type' low molecular weight purple acid phosphatase, a high molecular weight form in some fungi, and a homologue in some bacteria. The catalytic mechanism of the enzyme has been investigated with a view to understanding the marked difference in specificity between the Fe-Mn sweet potato enzyme, which exhibits highly efficient catalysis towards both activated and unactivated phosphate esters, and other PAPs, which hydrolyse only activated esters. Comparison of the active site structures of the enzymes reveal some interesting differences between them which may account for the difference. The implications fur understanding the physiological functions of the enzymes will be discussed.
Antagonists of the d -opioid receptor are effective in overcoming resistance against analgesic drugs such as morphine. To identify novel antagonists of the d -opioid receptor that display high potency and low resistance, we performed 3D-QSAR analysis using chemical feature-based pharmacophore models. Chemical features for d -opioid receptor antagonists were generated using quantitative (Catalyst/HypoGen) and qualitative (Catalyst/HipHop) approaches. For HypoGen analysis, we collected 16 peptide and 16 non-peptide antagonists as the training set. The best-fit pharmacophore hypotheses of the two antagonist models comprised identical features, including a hydrophobic aromatic (HAR), a hydrophobic (HY), and a positive ionizable (PI) function. The training set of the HipHop model was constructed with three launched opioid drugs. The best hypothesis from HipHop included four features: an HAR, an HY, a hydrogen bond donor (HBD), and a PI function. Based on these results, we confirm that HY, HAR and PI features are essential for effective antagonism of the d -opioid receptor, and determine the appropriate pharmacophore to design such antagonists.
The development of expression systems for heterologous proteins has been greatly demanded not only for the study of the structure/function relationships of these proteins but also for their biotechnological and pharmaceutical applications. During the past decades, the methylotrophic yeast Hansenula polymorpha and Pichia pastoris have drawn attention as one of promising hosts for the production of a variety of heterologous proteins. The increasing popularity of H. polymorpha and P. pastoris as the host systems can be attributed to the several advantages over the traditional yeast Saccharomyces cerevisiae, such as the availability of very strong and tightly regulated promoters from the enzymes involved in the metabolism of methanol, a very high-cell density even on simple mineral media, and a high stability of expression plasmids. Furthermore, it has been observed that glycoproteins from these two yeasts are less hyperglycoylated compared to those from S. cerevisiae. Despite substantial similarities as methylotrophic yeasts, however, these two expression systems have some unique features distinguished from each other. In this paper we present a brief overview on the present status of the expression systems developed in methylotrophic yeast, mainly focusing on the similarities and differences between the H. polymorpha and P. pastoris systems.
The paper deals with the structural vibration and noise characteristics of pipe structures. In general, A structure bone noise has a great effect on the drain noise of toilets, and depends on the natural frequency of the related structures. In order to measure and to find the relationships between structural vibration and noise of the pipe structures, some experiments have been performed. Through the experiments, impact acceleration signal in time domain and magnitude of transfer function in the frequency domain have been investigated for three kinds of pipes. Transmission loss of pipes depending on the frequency ranges was also found by using small speakers as a sound source.

A study of domestic furniture arrangements may reveal the living style relevant to the room as conceived and coded by occupants and the effects of the physical environment on the structure of behavior settings. The purpose of this study was to investigate, through analizing the furniture usage and behavior as a non-reactive and activity oriented behavioral measures, the occupants` domestic habits as a living style using MDS. MDS(multidimensional scaling technique) is a statistical technique for creating a spatial representation of data. It Is a particularly appropriate technique for analizing qualitative data such as the furniture usage and behavior because it takes into account all of the relationships between items. For the MDS analysis, the furniture usage and behavior examined by housing types based on 114 households in Seoul. The result of spatial configuration by MDS has three dimensions : recogn;lion of room function, pattern of room organization, understanding of room meaning. The effect of housing types for dimensions is identical but configuration of furniture items is different.
The cytoplasm in mammalian cells is a battlefield between the host and invading microbes. Both the living organisms have evolved unique strategies for their survival. The host utilizes a specialized autophagy system, xenophagy, for the clearance of invading pathogens, whereas bacteria secrete proteins to defend and escape from the host xenophagy. Several molecules have been identified and their structural investigation has enabled the comprehension of these mechanisms at the molecular level. In this review, we focus on one example of host autophagy and the other of bacterial defense: the autophagy receptor, NDP52, in conjunction with the sugar receptor, galectin-8, plays a critical role in targeting the autophagy machinery against Salmonella; and the cysteine protease, RavZ secreted by Legionella pneumophila cleaves the LC3-PE on the phagophore membrane. The structure-function relationships of these two examples and the directions of future research will be discussed.
Knowledge classification and expression of constructed knowledge have been main research issues in the field of knowledge representation. Constructed design knowledge of the former product loses its utility when new products with different structures are introduced to the market. In order to construct the design knowledge for a new product. designers need to reconstruct the design knowledge with new relationships. The design knowledge has been constructed with level trees, but it is difficult to rearrange the relations. Design DNA is proposed in this work in order to facilitate the rearrangement of design knowledge and give flexibility to knowledge structure. Design DNA is based on Layout-oriented domain knowledge and Function-oriented domain knowledge, which enables to generate new design knowledge that will result in new part geometries for given constraints on the part functions. Design DNA is applied to the design knowledge of lever system of the automatic transmission of passenger cars as an example.
Organisms living in the Arctic and Antarctic regions are exposed to strong constraints, of which temperature is a driving factor. Evolution has led to special adaptations, some with important implications at the biochemical, physiological, and molecular levels. The northern and southern polar oceans have very different characteristics. Tectonic and oceanographic events have played a key role in delimiting the two polar ecosystems and influencing evolution. Antarctica has been isolated and cold longer than the Arctic; its ice sheet developed at least 10 million years earlier. As an intermediate system, the Arctic is a connection between the more extreme, simpler Antarctic system and the very complex temperate and tropical systems. By studying the molecular bases of cold adaptation in polar fish, and taking advantage of the information available on hemoglobin structure and function, we analysed the evolutionary history of the ${\alpha}\;and\;{\beta}globins$ of Antarctic and Arctic hemoglobin using the molecular clock hypothesis as a basis for reconstructing the phylogenetic relationships among species.
An optimization method has been developed to find the minimum weight design of steel building structures which consist of the commercially available discrete sections. In this study, an emphasis was particularly placed on the practical applicability of optimization algorithm in engineering practice. The structure Is optimized through element optimization under the element level constraints first and then, if there is any violation of structural level constraints, it is adequately compensated by the constraint error correction vector obtained through the sensitivity analysis. A scaling procedure is introduced for the problems of large violated displacement constraint. The oscillation control in the objective function is also discussed. By dividing the available H-sections into two groups based on their section characteristics, much improved relationships between section variables were obtained and used efficiently in searching the optimum section in the section table.
