초고온 Archaeon인 Thermococcus profundus로부터 Thermosome유사 단백질의 분리 및 구조 분석 (Thermosome-like Protein from Hyperthermophilic Archaeon Thermococcus Profundus; Purification and Structural Analysis)

  • 초고온 archaeon인 Thermococcus profundus에서 실린드형태의 단백질 복합체를 분리, 생화학적 특성과 구조를 규명하였다. 전자현미경과 영상처리 (image processing)를 이용한 구조 분석으로 8개의 subunit으로 이루어진 링 (ring) 형태가 두 개 겹쳐져 가운데 통로를 가지는 실린드형태로 이루어진 복합체임을 알 수 있었다. 또한 60kDa단백질(P60 complex)로 구성된 homomultimer임을 보여 주고 있다. 이 복합체는 $80^{\circ}C$ 이상에서 도 강한 ATPase의 활성을 보여주는 강한 내열성 단백질임을 알 수 있다. P60 complex는 초고온 archaeon이 높은 온도에서 적응하며 생존하는데 매우 중요한 역할을 할 것으로 추정할 수 있다.

후기 청소년의 건강위험행동 구조모형: 2010 한국 청소년 건강실태조사 기반 (A Structural Model for Health Risk Behavior of Late Adolescents: Based on 2010 Korea Adolescent Health Survey)

  • Purpose: This study was done to construct and test a structural model to explain health risk behavior of late adolescents. Methods: Data for this study were secondary data from the 2010 Korea Adolescent Health Survey based and 3,675 high school students who participated. Data were analyzed using SPSS 18.0 and AMOS 19.0 programs. Results: After 7 lines were removed, fitness statistics for the hypothetical model were appropriate (${\chi}^2$=559.13, p<.001, GFI=.98, SRMR=.03, RMSEA=.04, NFI=.88, IFI=.90, CFI=.90, TLI=.86, AIC=671.13). The result showed that drinking-smoking is directly affected by 5 variables (32.5%), obesity is directly affected by 2 variables (0.7%), lack of physical activity is directly affected by 5 variables (22.2%), skipping of breakfast is directly affected by 3 variables (11.9%), improper sleep is directly affected by 3 variables (7.5%), and psychological adaptation is directly affected by 4 variables (26.8%). Conclusion: The results of this study, indicate that late adolescents' health risk behavior is affected by many factors with complicate correlations suggesting further study compare youth health risk behaviors in a variety of environments.

면진격리 고무베어링 설계법을 이용한 구조물의 성능점 예측 (Determination of Structural Performance Point Utilizing The Seismic Isolation Rubber Bearing Design Method)

  • 면진격리 고무베어링의 설계법을 수정하여 구조물의 성능점 예측을 위한 간편한 해석방법을 제안하였다. 이러한 적용이 가능한 것은 구조물이 지진력의 작용으로 인하여 손상을 입게 되면 구조물의 항복 후 강성은 연화되고, 이로 말미암아 구조물의 동적 특성이 장주기화 하기 때문이다. 제안된 해석법이 기존의 방법에 비하여 우월한 것은 능력스펙트럼법이 요구하는대로 보유능력곡선과 요구량스펙트럼을 가속도-변위 좌표계로 치환하지 않고서도 비교적 정확한 성능점을 예측할 수 있다는 것이다. 제안된 방법의 타당성은 문헌에서 보이는 정확한 값과의 비교에 의하여 입증하였다.

가압경수로(PWR)용 고준위폐기물 처분용기의 구조적 안전성 평가 보완 해석 (A Complementary Analysis for the Structural Safety Evaluation of the Spent Nuclear Fuel Disposal Canister for the Pressurized Water Reactor)

  • 가압경수로(PWR)에서 배출되는 고준위폐기물을 지하 500m의 화강암 암반의 처분장에 장기간(약 10,000년 동안) 처분하기 위하여 여러 구조적 안전성 평가 수행을 통하여 처분용기모델이 개발되었다. 기존에 설계된 가압경수로용 처분용기 모델은 구조적 안전성은 문제가 없으나 너무 무거운 단점이 지적되었다. 따라서 구조적 안전성을 유지하면서 좀 더 경량화 된 처분용기모델을 개발하는 것이 요구된다. 기존의 처분용기모델이 무거워진 한가지 이유는 처분용기 개발 시 적용된 외력조건 및 안전계수 등에 대한 조건들을 너무 엄격하게 적용했기 때문이라고 사료되기 때문에 이런 조건들을 완화하여 처분용기의 재원들을 조정하여 구조해석을 다시 수행하는 것이 요구된다. 따라서 본 논문에서는 설계 완성된 기존의 처분용기에 대하여 외력 조건 및 용기의 재원(두께 등) 들을 변화시키면서 구조해석을 재 수행하여 구조적 안전성 평가를 보완하였다. 이를 바탕으로 외력 조건에 따른 처분용기의 재원 등을 재 산출한다. 보완 해석 결과 기존의 122cm의 처분용기의 직경을 102cm까지 줄여 경량화 시킬 수 있음이 확인되었다.

  • e-Government is an innovative phenomenon around the globe for refurbishing public administration. Research to date, however, has been deficient in empirical studies of the factors that emerge out of the interplay of structure and human interactions responsible for successful implementation of e-Government projects. From the perspective of structuration theory, this study examines and explains the impact of this interplay in the implementation of an e-Government system, eSeva for local administration at one of the Indian state, The results suggest that the success of implementation of e-Government projects can be conceptualized as the outcome of persisting constructive interrelationships among the human, programmatic and institutional elements of e-Government and the critical learning and adaptation as a result of efforts at the structuration level.

  • The purpose of this article is to understand of the muscle adaptation based on myosin heavy chain. Especially, skeletal muscle dadptation in related to aging, unloading, training will discussed. MHC expression is highly plastic in muscles of adult mammals in accordance with the environmental conditions. These changes is called muscle plasticity. The plasticity is the atility of muscle cell to alter either the quantity of protein or the type of protein. MHC is both an important structural and regulatory protein comprising the contractile apparatus.

Effects of Light on Disassembly of Chloroplast during Senescence of Detached Leaves in Phaseolus vulgaris

  • Effects of light on leaf senescence of Phseolus vulgaris were investigated by measuring the disassembly of chlorophyll-protein complexes in detached leaves which had been kept in the dark or under light. The loss of chlorophyll accompanied by degradation of chlorophyll- protein complexes. PSI (photosystem I) complex containing LHCI (light harvesting complex of PSI) apoproteins was rapidly decreased after the early stage of dark-induced senescence. RC(reaction center)-Cores was slightly increased until 4 d and slowly decreased thereafter. As disassembly of LHCII trimer progressed after the late stage of senescence, there was a steady increase in the relative amount of SC(small complex)-2 containing LHCII monomer. On the other hand, white and red light adaptation caused the structural stability of chlorophyll-protein complexes during dark-induced senescence. Particularly, red light was more effective in the retardation of LHCII breakdown than white light, whereas white light was slightly effect in protecting the disassembly of PSI complex compared to red light. These results suggest, therefore, that light may be a regulatory factor for stability of chlorophyll-protein complexes in the senescent leaves.

Calibration of model parameters for the cyclic response of end-plate beam-to-column steel-concrete composite joints

  • Composite joints, considering the composite action of steel and concrete, exhibit, in general, high strength and high ductility. As a consequence, the use of this type of joint has been increasing in many countries, especially in those that are located in earthquake-prone regions. In this paper, a hysteretic model with pinching is presented that is able to reproduce the cyclic response of steel and composite joints. Secondly, the computer implementation and adaptation of the model in a spring element within the computer code Seismosoft is described. The model is subsequently calibrated using a series of experimental test results for composite joints subjected to cyclic loading. Finally, typical parameters for the various joint configurations are proposed.

Fatigue experiments on steel cold-formed panels under a dynamic load protocol

  • A dynamic load protocol has been used to experimentally simulate fatigue behavior in cold-formed metal panels with screwed connections under wind loading. The specific protocol adopted is an adaptation of SIDGERS, originally developed for non-metallic membranes, which is composed of levels each under increasing load values. A total of 19 tests were performed on 3.35 m long by 0.91 m wide panels, identified as Type B-wide rib and Type E, both with screw connections at the edge and at the center, thus conforming two-span specimens. In some configurations the panels were fixed at the valleys, whereas crest-fixed connections were also investigated. Reinforcing the connections by means of washers was also investigated to evaluate their efficiency in improving fatigue capacity. The experimental results show maximum load capacities in improved connections with washers of approximately twice of those with classical connections.

Direct implementation of stochastic linearization for SDOF systems with general hysteresis

  • The first and second moments of response variables for SDOF systems with hysteretic nonlinearity are obtained by a direct linearization procedure. This adaptation in the implementation of well-known statistical linearization methods, provides concise, model-independent linearization coefficients that are well-suited for numerical solution. The method may be applied to systems which incorporate any hysteresis model governed by a differential constitutive equation, and may be used for zero or non-zero mean random vibration. The implementation eliminates the effort of analytically deriving specific linearization coefficients for new hysteresis models. In doing so, the procedure of stochastic analysis is made independent from the task of physical modeling of hysteretic systems. In this study, systems with three different hysteresis models are analyzed under various zero and non-zero mean Gaussian White noise inputs. Results are shown to be in agreement with previous linearization studies and Monte Carlo Simulation.
