암세포는 종양의 성장을 지지하는 다양한 성분으로 구성되어 있는 환경에서 자란다. 암미세환경에 존재하는 주요 세포등은 섬유아세포, 내피세포, 면역세포들이며 이들세포들은 암세포들과 서로 소통을 하고 있다 종양조직에 유입된 면역세포중에서 대식세포가 종양미세환경의 주요성분으로서 다양한 면역현상들을 조절한다. 면역세포유입에 의한 암촉진과 항암효과 간의 복잡한 균형은 종양의 성장과 진행에 필요한 만성염증 환경을 생성시킬 수 있다. 대식세포는 M1과 M2 극성화로 규정된 미세환경 신호에 반응하여 기능적으로 다른 프로그램을 작동시킬 수 있다. 종양관련대식세포는 다양한 사이토카인, 케모카인, 단백질분해효소들을 분비함으로써 암 신생혈관형성, 증식, 전이 및 면역억제를 촉진시킨다. 최근에, 종양관련대식세포는 암줄기세포와 상호작용하여 종양의 진행, 전이 및 항암제 내성을 유도하는 것으로 알려져 있다. 종양관련대식세포는 암미세환경을 유지하기위해 면역억제 기능을 획득하며, 종양의 이질성과 가소성의 특성을 갖고 있어 암관련인자 및 감염등의 노출에 의해 서로 다른 극성형질로 리프로그래밍된다. 종양관련대식세포는 기질인자의 자극에 의해 암특이적인 케모카인들을 생성하기 때문에 케모카인은 질병의 활성을 반영하는 바이오마커로 작용할 수 있다. 종양조직에 종양관련대식세포가 많이 유입될수록 환자의 예후가 좋지 않으며 항암치료에 대한 저항성이 생긴다. 따라서 종양에서 대식세포를 표적화하는 항암치료는 유망한 치료전략이 될 수 있다.
림프절은 체내로 침입한 병원체에 반응하여 성숙한 림프구들이 활성화 되는 곳이다. 림프구들은 스트로마의 구조적 뼈대를 따라 동계항원을 제시하고 있는 항원제시세포의 표면을 탐색한다. Fibroblastic reticular cells(FRC)는 림프절 T zone에서 3차원구조 네트워크를 형성하는데 관여하는 스트로마 세포로 유입되는 T 림프구들에 대한 안내길을 제공한다. 이런 상호 협력적인 환경에서 FRC와 T세포의 양방향적 관계는 림프절의 정상적 기능을 수행하는데 필수적이다. FRC는 물리적으로 림프절 조형물을 형성 할 뿐만 아니라 T세포 생물학적 기능조절에도 필수적이다. FRC는 T 림프구와 상호 반응하며 T세포에 발판을 제공하고 T세포 면역반응에 영향을 미치는 용해성 인자들을 방출한다. 최근에는 FRC는 말초에서 자기 관용 T세포 생성에도 관여하며 림프절에서 활성화된 T세포 분열을 조절하는데도 관여하고 있다. 따라서, FRC와 T세포 상호간 협력은 림프절에서 T세포기능을 조절하는데 중요한 결과를 야기한다. 더욱이, FRC는 염증 상황에서 항생펩타이드, 보체 등의 분비를 통한 선천성 면역에도 중요한 역할도 한다. 결론적으로 FRC와 T세포 상호간에 T세포 생물학적 효능을 증대를 위해 양방향성 접촉을 하며 이러한 상호 협력적 피드백은 면역반응 동안 조직기능 유지를 돕게 된다.
줄기세포와 전구세포 사이의 기능 분석은 여러 조직 특히 혈액에서 잘 확립되어 있다. 특히 조혈줄기세포는 골수 니쉬에서 자가재생능 및 재구성능을 가지고 있으며, 골수 내 기질세포는 조직 기능 조절에 큰 영향을 미친다. 최근 연구에서는 포유동물 줄기세포의 기능은 니쉬 세포 내에서 실험적으로 처음 증명되었고, 특히 미세환경에 의해 종양발생이 가능하다는 증거를 나타내고 있다. 고대에서부터 뼈와 피의 관계는 생체 내 필수불가결인 관계로 진화 과정을 거쳐 포유류의 줄기세포에 대해 최초로 제안되었고, 실험적으로 증명된 니쉬세포를 포함한 미세환경과의 복잡한 상호 관계를 규명하였다. 여러 골수 기질세포는 조혈줄기세포의 기능 조절을 하며, 일부의 기능장애는 골수 이형성 및 백혈병을 유발할 수 있다. 현재까지 여러 기질세포에 대한 맵핑이 되지 않아 현재 많은 연구자들이 단일 분자 수준에서 개개의 기질세포 유형을 파악하는 데이터가 필요하다고 주장하고 있으며 이를 바탕으로 골수 내 조혈줄기세포의 특정 기능을 파악할 수 있다고 볼 수 있다. 따라서 본 총설을 통해 조혈줄기세포 및 미세환경에 대한 이전 연구들의 흥미로운 문제를 논의하고, 조혈줄기세포와 골수 니쉬에 대한 현재 및 새로운 개념을 요약하고자 한다.
The aspiration cytologic finding of myositis ossificans is not well documented but similar to that seen in nodular fasciitis except less cellularity. Myositis ossificans is a reactive condition that is sometimes mistaken microscopically for extraosseous osteosarcoma. Cytologically, myositis ossificans may be distinguished from extraosseous osteosarcoma by the presence of uniform benign stromal cells composed of mature fibroblasts and osteoclastic giant cells. However, the differential diagnosis may be difficult in the early stage of this reactive and proliferative process. We recently experienced a case of myositis ossificans. The patient was a 67-year -old woman with painful swelling of the left index finger for one month. Simple x-ray finding showed a soft tissue mass with calcific center at middle phalanx of the left hand. Fine needle aspiration cytology revealed a few individual or clusters of spindle cells in dense eosinophilic stroma with osteoclastic giant cells. The spindle cells were uniform smooth-bordered, and oval nuclei with single small inconspicuous nucleoli, and elongated cytoplasm. The scattered individual cells had eccentric nuclei with one or two nucleoli and abundant, basophilic cytoplasm.
The present study was underve the histochemical nature of various follicles and interstitial tissues in the ovaries of Korean native goats and cattle as well as the histochemical changes of those in the ovaries of Korean native goats treated with dexamethasone. Much more lipid granules appeared in the granulosa and theca cells of atretic follicles compared with normal follicles in these ruminant ovaries. In the ovaries of Korean native goats the interstitial tissue derived from the theca interna of atretic follicles appeared the form of patches of cells and the interstitial tissue derived from stromal cells appeared the form of diffuse, obscure bounds. In the ovaries of Korean native cattle the interstitial tissue derived from theca interna of atretic follicles showed sparsely scattered form of pathes of cells. The histochemical components were described on the basis of lipids in the granulosa and theca cells of normal follicles, atretic follicles and interstitial tissue. In the ovaries of Korean natve goats treated with dexamethasone, the granulosa and theca cells of atretic foillicle contained plenty lipid granules that were increased in size and number, however, lipid granules were markedly decreased in the interstitial tissue.

Taurine is present in a variety of tissue and exhibits many important physiological functions in many tissues. Although it is known that many tissues mediate taurine transport, its functions of taurine transport in bone have not been identified yet. In the present study, we investigated the expression of taurine transporter (TauT) and taurine uptake using mouse stromal ST2 cells and osteoblast-like MC3T3-El cells, which is bone related cells. Detection of TauT mRNA expression in these cells were performed by reverse transcription polymerase chain reaction (RT-PCR). The activity of TauT was assessed by measuring the uptake of [$^3$H]taurine in the presence or absence of inhibitors. TauT mRNA was detected in these cells. [$^3$H]Taurine uptake was dependent upon the presence of extracellular sodium, chloride and calcium ions, and inhibited by cold-taurine and ${\beta}$-alanine. These results suggest that taurine has biological functions in bone and some effect on the bone cells.
Purpose: Nodal metastasis is the main prognostic factor in the patients with oral squamous cell carcinoma (OSCC). We investigated the association between tumor-associated lymphatics and OSCC characteristics. Methods: Thirty-four specimens were used for the immunohistochemical staining with the antibody for vascular endothelial growth factor (VEGF)-C, VEGF-D, VEGF receptor (VEGFR)-3, phosphorylated VEGFR-3, D2-40, and matrix metallproteinases (MMPs). We observed the distribution of the lymphangiogenic factors and quantified the degree of expression. We determined lymphatic vessel density (LVD) and lymphatic vessel dilatation with D2-40 immunostaining. We assessed the association of LVD or lymphatic vessel dilatation with tumor progression or tumor differentiation. Results: OSCC cells expressed lymphangiogenic ligands. Lymphangiogenic receptor, VEGFR-3, was expressed and activated in some tumor cells as well as in tumor-associated endothelial cells. LVD was not associated with tumor size or nodal status, but lymphatic vessel dilatation was higher in tumors with nodal metastasis, and also higher in poorly differentiated tumors. In stromal area of OSCC, MMP-1 and MMP-10 were up-regulated and the basement membrane of tumor-associated endothelial cells was destroyed by these collagenases. Conclusion: In the primary tumors with nodal metastasis, especially in poorly differentiated OSCC, tumor cells invaded the dilated lymphatic vessels via ruptured sites. MMP-1 and MMP-10 are important in the lysis of the glycocalyx inside the tumor-associated lymphatic endothelial cells.
Current immunosuppressants have nonspecific immuosuppressive effects, and are not helpful for tolerance induction. Consequently, transplant patients cannot discontinue using them, and their nonspecific immunosuppressive effects result in many side effects, including infection and malignancy. However, most of cellular immunotherapy can have donor antigen-specific immunsuppressive effects. Therefore, cell therapy could be an alternative or adjunctive to nonspecific immunosuppressants. Polyclonal or antigen-specific Foxp3+ regulatory T cells have been actively tried for prevention of acute rejection, treatment of chronic rejection, or tolerance induction in clinical trials. Regulatory macrophages are also under clinical trials for kidney transplant patients. IL-10-secreting type 1 regulatory T cells and donor- or recipient-derived tolerogenic dendritic cells will also be used for immunoregulation in clinical trials of kidney transplantation. These cells have antigen-specific immunoregulatory effects. Mesenchymal stromal cells (MSCs) have good proliferative capacity and immunosuppressive actions independently of major histocompatibility complex; therefore, even third-party MSCs can be stored and used for many patients. Cell therapy using various immunoregulatory cells is now promising for not only reducing side effects of nonspecific immunosuppressants but also induction of immune tolerance, and is expected to contribute to better outcomes in transplant patients.
