Relaxation Effects of Epimedium Koreanum Nakai in Isolated Rabbit Corpus Cavernosum Smooth Muscle (음양곽(淫羊藿)의 토끼 음경해면체 평활근 이완효과)

  • This study aimed to investigate the relaxation effects and its underlying mechanisms of Epimedium koreanum Nakai(EK) in phenylephrine(PE) treated isolated rabbit corpus cavernosum smooth muscle. The dose-dependent relaxation responses of phenylephrine(PE, $1{\times}10^{-6}M$)-precontracted strips to EK at $0.01-3.0mg/m{\ell}$ were measured and also observed after endothelial denudation using organ bath. To analyze the underlying mechanisms of EK-induced relaxation, $N{\omega}$-nitro-L-arginine(L-NNA), methylene blue(MB), tetraethylammonium chloride(TEA), indomethacin(IM) were pretreated before EK extract infused into precontracted strips induced by PE. To investigate cytotoxic activity and nitric oxide(NO) concentration of EK extract on EA.hy926 cells, mitochondrial dehydrogenase activity(MTT) assay and nitric oxide detection kit were used. The cavernous strips were significantly relaxed by EK extract at $0.3mg/m{\ell}$, $1.0mg/m{\ell}$, $3.0mg/m{\ell}$ and the relaxation responses of PE-precontracted strips denuded endothelium also inhibited in comparison with intact endothelium. The pretreatment of L-NNA, MB, TEA reduced EK extract-induced endothelium-dependent relaxation, but the pretreatment of IM didn't affect EK extract-induced endothelium-dependent relaxation. When EK extract was applicated on EA.hy926 cells, the NO concentration was increased. Our findings have shown that EK extract exerts a relaxing effect on corpus cavernosum in part by suppressing influx of extracellular $Ca^{2+}$ through activating the NO-cGMP system.

  • Red seabream strips were decontaminated by dipping with solutions of 0.25~1.0% acetic, lactic, or citric acids for 5min. Control strips were dipped with tap water only for 5min. All strips were individually placed in plastic bags and stored at 4$^{\circ}C$. Acetic acid(AA) treatments were completely inhibited aerobic spoilage bacteria(areobic plate count : APC) compared to the initial controls for 6 days. Treatments of either lactic acid(LA) or citric acid(CA) completely inhibited APC compared to the initial controls for 3 days. Red seabream strips treated with AA extended microbiological shelf-life for 12 days.

Effects of frequency - amplitude electrical stimulation on sympathetic neurotransmitter and vasoactive intestinal peptide (SSP 주파수 진폭변조가 Vasoactive Intestinal Peptide와 $\beta$-endorphin, cGMP에 미치는 영향)

  • Vasoactive intestinal peptide (VIP) is a very potent dilatator and a nonadrenergic, noncholinergic (NANC) neurotransmitter or neuromodulator in the peripheral and the central nervous systems. The mechanisms of action of VIP were examined in aortic circular and in uterine longitudinal smooth muscle strips of the rat. The effects of sympathetic neurotransmitter were investigated in gastric and aortic circular muscle strips of the mouse and the rat. The effects of silver spike point, SSP, low frequency electrical stimulations of VIP, sympathetic neurotransmitter and $\beta$-endorphin were examined in plasma, serum and 24h urine from the healthy volunteer. In gastric smooth muscle strips from the mouse, adrenergic neurotransmitter norepinephrine was inhibitory effected, followed by caused phasic and tonic contraction to the, muscrine receptor agonist carbachol and acetylcholine, respectively. In urine from the healthy volunteer, both norepinephrine and epinephrine were significantly decreased in continue type and low frequency (3 Hz) of SSP electrical stimulations. The contractile responses to S-HT in uterine longitudinal smooth muscle strips of the rats were completely decreased by a VIP 1 $\mu$M. The contractile responses to PGF2$\alpha$ were not decreased by a VIP. In plasma and serum from the healthy volunteer, both VIP and $\beta$-endorphin were significantly increased in continue type and low frequency (3 Hz) of SSP electrical stimulations. Therefore, this study demonstrate that VIP has the capacity to relax vascular or gastric smooth muscles in part by stimulating the generation of NO, and silver spike point low frequency electrical stimulation has the capacity both to decrease sympathetic neurotransmitters and to increase VIP, $\beta$-endorphin.

Effects of Oja-Shingiwhan in Contracted Corpus Cavernosum Smooth Muscle (五子腎氣丸이 음경해면체 평활근의 수축에 미치는 영향)

  • The purpose of this study is to investigate the effects of Oja-Shingiwhan(OS) in contracted corpus cavernosum smooth muscle and its mechanism. To evaluate the relaxation of OS in contracted corpus cavernosum, OS was treated in strips which were precontracted with phenylephrine(PE). To examine its mechanism, OS was treated into corporal strips contracted by PE after pretreatment of Nω-nitro-L-arginine(L-NNA) and compared with non-pretreatment of L-NNA. In calcium chloride(Ca2+)-free krebs solution, Ca2+ 1 mM was treated into corporal strips contracted by PE after pretreatment of OS and compared with non-pretreatment of OS. action were measured by histochemical, immunohistochemical methods. OS significantly affected on the relaxation of corporal strips, and the relaxation effects were inhibited by pretreatment of L-NNA. Contractions induced by Ca2+ influx were inhibited by pretreatment of OS in Ca2+-free krebs solution. OS increased eNOS positive reaction in corpus cavernosum, but decreased PDE-5 positive reaction. These result suggest that the effect of OS in contracted corpus cavernosum smooth muscle are shown by suppressing extracellular Ca2+ influx and increase of eNOS, NO production and decrease of PDE-5.

Studies on Sympathetic Innervation of the Jejunum in the Chick (병아리 공장(空腸)의 교감신경지배(交感神經支配)에 관한 연구(硏究))

  • It has been generally understood that the intestinal tracts are under the control of the autonomic nerves; the parasympathetics are excitatory and the sympathetics inhibitory. However, it is recently reported that the actions of these autonomic nerves in the newborn animals are shown to be different from those in the adult animals in some species. In order to elucidate the role of sympathetic innervation to the intestinal tracts, the effects of periarterial nerve stimulation were studied in the periarterial sympathetics-jejunum preparations of the chick and the effects of some autonomic drugs on the isolated muscle strips were also studied. The results obtained were as follows: 1. The periarterial stimulation in the periarterial sympathetics-jejunum preparation elicited the responses of three patterns; 1) contrcation followed by relaxation 2) contraction only 3) relaxation only. The excitatory response was most effective in the stimulus frequencies of 40 cps, whereas the inhibitory response was maximal in the stimulus frequencies of 30 cycle per second. 2. The excitatory response to the periarterial stimulation was not affected by the pretreatment with phenoxybenzamine, dibenamine, propranolol and atropine, whereas the inhibitory response was completely blocked by the pretreatment with phenoxybenzamine and propranolol. 3. In the periarterial syrnpathetics-jejunum preparation treated with reserpine, the periarterial stimulation evoked only contraction, and the contraction was not affected by the pretreatment with phenoxybenzamine, propranolol and atropine. 4. The administration of norepinephrine evoked a relaxation in the isolated jejunum muscle strips and the effect was completely blocked by the pretreatment with phenoxybenzamine. 5. The administration of isoproterenol produced a relaxation in the isolated jejunum muscle strips and the effect was not affected by pretreatment with phenoxybenzamine, whereas the effect was completely blocked by the pretratment with propranolol. 6) The administration of acetylcholine produced a marked contraction in the isolated jejunum muscle strips and the effect was completely abolished by the pretreatment of atropine. These experimental evidences indicate that the inhibitory response to the periarterial stimulation is due to adrenergic fibers and the excitatory response is due to neither adrenergic nor cholinergic component.

Fabrication of the thermopile using SOI structure (SOI 구조를 이용한 열전쌍열(Thermopile) 제작)

  • In this paper, a thermopile which is applied to wide uses of temperature measuring was fabricated and its characteristic was improved by appling SOI structure to the fabrication. We improved characteristic of the thermopile by using single crystal silicon strips that has high seebeck coefficient and dielectric isolating the silicon strips from substrate with silicon dioxide film which dramatically decrease thermal conductivity between hot and cold junction compared to a silicon strip which was fabricated by ion implantation. The thermopile consists of 17 p-type single crystal silicon strips and 17 n-types by serial connection. The result of electromotive force measuring showed very good characteristic as 130mV/K when temperature difference between the two ends of the thermopile occurs by applying light on the thermopile fabricated with silicon strips of $1600{\mu}m$ length, $40{\mu}m$ width, $1{\mu}m$ thickness.

Effect of Prostaglandin $E_2$ and Indomethacin on Electrical Activity of Isolated Cat Stomach (적출한 고양이 위(胃)의 전기활동에 미치는 prostaglandin $E_2$ 및 indomethacin의 영향)

  • This study was conducted to investigate the effect of prostaglandin $E_2(PGE_2)$ and indomethacin upon the electrical activity of the isolated cat stomach muscle strips$(1.5{\times}7.0\;cm)$. Fifty-seven muscle strips, obtained from 57 cat stomachs(including corpus and antrum) were studied in a muscle chamber filled with Krebs solution(pH 7.4, temperature $36{\pm}0.5^{\circ}C$) aerated with 5% $CO_2$ in $O_2$. The electrical activity was recorded by five capillary electrodes (Ag-AgCl), of which two were placed on the corpus and three on the antrum. After recording of the electrical activity in normal Krebs solution, $PGE_2$ in concentrations of 0.25(N=7), 0.5(=7), 1(N=7) and $2{\times}10^{-7}\;M(N=6)$ were administered to 27 muscle strips, while indomethacin was applied in concentretions of 0.25(N=9), 0.5(N=10), 1(N=6) and $2{\times}10^{-3}\;M(N=5)$ to the remaining 30 strips. The mean frequency were minutely measured from each electrogastrogram. 1) By adding $PGE_2$ in all doses, gastric slow wave frequency increased significantly compared with that in resting state. 2) Following $PGE_2$ administration, peak slow wave frequency increased dose-dependently. 3) After indomethacin addition in all doses, the slow wave frequency decreased significantly compared with that in resting state. 4) Following indomethacin administration, incidence of complete abolition of slow wave increased dose-dependently, and its latent period decreased also in a dose-dependent manner. It is inferred from the above results that prostaglandin $E_2$ has a facilitatory role in the development of gastric slow wave in cat.

The Endothelium-Dependent Vasorelaxation Effect of Cynomorii Herba (쇄양(鎖陽)의 내피세포 의존성 혈관이완효과)

  • Objectives : The purpose of this study was to investigate the endothelium-dependent vasorelaxation effect of Cynomorii Herba(CH) extract on contracted rabbit carotid artery.Methods : To clarify the vasorelaxation effect of CH extract, arterial strips with intact was used, to endothelium -dependent vasorelaxation effect of CH extract, arterial strips damaged endothelium was used for experiment using organ bath. Arterial strips was contracted with phenylephrine(PE) before treated with CH extract(0.01, 0.03 and 0.1 ㎎/㎖). To study mechanisms of CH-induced vasorelaxation effect, CH extract infused into arterial rings after treatment by indomethacin(IM), tetraethylammonium chloride(TEA), Nω-nitro-L-arginine (L-NNA), methylene blue(MB) for comparing with non-treated. And calcium chloride(Ca2+) 1 mM was treated into precontracted arterial ring induced by PE after treatment of CH extract in Ca2+-free krebs solution. Cytotoxic activity of CH extract on human umbilical vein endothelial cell(HUVEC) was measured by MTT assay, and nitric oxide(NO) concentration was measured by Griess reagent.Results : PE-induced arterial strips was significantly relaxed, but the damaged endothelium arterial ring wasn't relaxed by CH extract. Pretreatment of IM, TEA didn't inhibit the vasorelaxation of CH extract, but pretreatment of L-NNA, MB inhibited the vasorelaxation of CH extract. Pretreatment of CH extract reduced the increase of contraction by influx of extracellular Ca2+ in contracted arterial ring induced by PE, CH extract increased nitric oxide concentration on HUVEC significantly.Conclusions : This study shows that CH extract have the vasorelaxation effect by blocking the influx of extracellular Ca2+ through the activating NO-cGMP system.

Inhibitory Effects of Polyphenol-Rich Fraction Extracted from Rubus coreanum M on Thoracic Aortic Contractility of Spontaneously Hypertensive Rats

  • The purpose of the present study was to investigate whether polyphenol-rich fraction extracted from fruit wine of Rubus coreanum M (PCRC) can affect the contractility of the thoacic aortic strips isolated from spontaneously hypertensive rats (SHRs), and to clarify its mechanism of action. PCRC (200-800 ${\mu}g/mL$) concentration-depenedently blocked phenylephrine (10 ${\mu}M$)-induced contractile responses of the isolated aortic strips of SHRs. PCRC (400 ${\mu}g/mL$), added in to bath medium, also depressed the contractile active tension evoked by both phenylephrine (3 and 10 ${\mu}M$) and high potassium (25 and 56 mM). In the simultaneous presence of PCRC (400 ${\mu}g/mL$) and L-NAME (a selective inhibitor of NO synthase, 300 ${\mu}M$), the contractile responses evoked by phenylephrine and high $K^+$ were recovered to considerable level of the corresponding control contractility compared with those effects of PCRC-treatment alone. However, in the simultaneous presence of indomethacin (10 ${\mu}M$, a selective cyclooxygenase inhibitor) and PCRC (400 ${\mu}g/mL$), they were not affected. In the endothelium-denuded aortic strips by CHAPS-treatment, PCRC did not affect the contractile responses induced by phenylephrine or high potassium. Interestingly, PCRC (1.0, 3.0 and 10.0 mg/kg/30 min, i.v., respectively) dose-dependently suppressed norepiphrine-induced vasopressor responses in anesthetized SHRs. Collectively, we concluded that PCRC causes vasorelaxation in the thoracic aortic strips with intact endothelium of SHRs at least partly by the increased NO production through the activation of NO synthase of vascular endothelium, but not through the activation of cyclooxygenase. These results suggest that PCRC might be helpful to prevent or alleviate cardiovascular diseases, including hypertension.

Compact UWB Log Periodic Right Triangle-Shaped Dipole Array Antenna Appended With Strips (스트립이 추가된 소형 UWB 대수 주기 직각 삼각형-모양 다이폴 배열 안테나)

  • A compact LPDA antenna consisting of right triangle-shaped dipole elements appended with strips is proposed for UWB applications. First, right triangle-shaped dipole elements are used instead of conventional strip dipole elements to reduce the width of the LPDA antenna. Second, the spacing between the LPDA elements is decreased to reduce the length of the LPDA antenna. Finally, strips are appended at the ends of the right triangle-shaped dipole elements in order to further reduce the width of the antenna. A prototype of the proposed antenna with 16 elements and gain > 4 dBi is fabricated on an FR4 substrate with dimensions of 44 mm×30 mm. Measured frequency band of the fabricated antenna is 2.99-14.76 GHz for a VSWR < 2, which ensures UWB operation, and measured gain range is 4.0-5.5 dBi with a front-to-back ratio larger than 10 dB. The length and width of the proposed compact LPDA antenna are reduced by 40.9% and 20.6%, respectively, compared to the conventional LPDA.
