Tunicamycin-Induced ER Stress Upregulates the Expression of Mitochondrial HtrA2 and Promotes Apoptosis Through the Cytosolic Release of HtrA2

  • Recent studies provide some evidence that the HtrA2 protein is intimately associated with the pathogenesis of neurodegenerative disorders and that endoplasmic reticulum (ER) quality control and ER stress-associated cell death play critical roles in neuronal cell death. However, little is known about the intimate relationship between HtrA2 and ER stress-associated cellular responses. In the present study, we have demonstrated that the HtrA2 protein level was gradually and significantly increased by up to to-fold in the mitochondria under tunicamycin (Tm)-induced ER stress, which eventually promoted cell death through the release of HtrA2 into the cytoplasm. Using an ecdysone-inducible mammalian expression system, we demonstrate that the extent of cell death in 293-HtrA2 cells was approximately 20 times higher under Tm-induced ER stress, indicating that the increase in the HtrA2 protein level in the mitochondria itself is necessary but not sufficient for the promotion of cell death. Taken together, these results suggest that HtrA2 may serve as a mediator of ER stress-induced apoptosis and ER-mitochondrial cross-talk in some cellular processes.

Protective role of Populus tomentiglandulosa against hydrogen peroxide-induced oxidative stress in SH-SY5Y neuronal cells

  • Oxidative stress caused by the overproduction of reactive oxygen species (ROS) is known as an etiology of neurodegenerative diseases. Populus tomentiglandulosa (PT), a member of the Salicaceae family, is widely grown in Korea and has been reported to exert protective effects on cerebral ischemia by attenuating of oxidative stress and neuronal damage. In the present study, we investigated the antioxidant activity and neuroprotective effects of an ethanol extract and four fractions [n-butanol, ethyl acetate (EtOAc), chloroform, and n-hexane] of PT under in vitro and cellular systems. The extract and four fractions of PT showed 1,1-diphenyl-2-picrylhydrazyl (DPPH), •OH, and O2- radical scavenging activities in a dose-dependent manner. In particular, the EtOAc fraction of PT had the strongest DPPH, •OH, and O2- radical scavenging activities among the extract and other fractions. Therefore, we further investigated the neuroprotective effect of the EtOAc fraction of PT against oxidative stress in H2O2-induced SH-SY5Y cells. Treatment with H2O2 significantly decreased cell viability and lactate dehydrogenase (LDH) release, and it also increased the ROS levels compared to the normal group. However, treatment with the EtOAc fraction of PT significantly increased cell viability. Moreover, the EtOAc fraction of PT-treated group significantly suppressed ROS production and LDH release compared to the H2O2-induced control group. In conclusion, our findings indicated that PT had in vitro antioxidant activity and neuroprotective effects against oxidative stress. Therefore, PT could be used as a natural agent for protection against oxidative stress.

  • In this paper, we examine the singular stresses and electric fields in a functionally graded piezoelectric ceramic strip containing an eccentric crack off the center line under anti-plane shear loading with the theory of linear piezoelectricity. It is assumed that the properties of the functionally graded piezoelectric ceramic strip vary continuously along the thickness. Fourier transforms are used to reduce the problem to the solution of two pairs of dual integral equations, which are then expressed to a Fredholm integral equation of the second kind. Numerical values on the stress intensity factor and the energy release rate are obtained.

  • When the exposed surface of rock slopes investigated, various factors have not been considered. The factors required to be considered include the vlume swelling of rock caused by stress release at the fault zone, unexposed discontinuities in the earth and so on. Also, rock slope failure occurs sometimes due to these factors. In this paper, we intend to help engineers to judge about the stability of similar new cutting-slopes or the maintenance of already cut-slope, so that they can consider these factors.

Characteristics of Fracture Energy on Steel Fiber-Reinforced Lightweight Polymer Concrete

  • In this study, unsaturated polyester resin, artificial lightweight coarse aggregate, artificial lightweight fine aggregate, heavy calcium carbonate and steel fiber were used to produce a steel fiber-reinforced lightweight polymer concrete with which mechanical properties were examined. Results of this experimental study showed that the flexural strength of unnotched steel fiber-reinforced lightweight polymer concrete increased from 8.61 to 13.96 MPa when mixing ratio of fiber content increased from 0 to 1.5%. Stress intensity factors($K_{IC}$) increased with increasing fiber content ratio while it did not increase with increasing notch ratio. Energy release rate ($G_{IC}$) turned out to depend upon the notch size, and it increased with increasing steel fiber content.

  • This study was analyzed dynamically by finite element method about the results of experiments which materials were applied by dynamic load. And they were compared with each other as the simulation data applied onto dynamic impact velocities of 6.4, 16.7 and 18.47m/s. The crack energy release rate, von-Mises stress and the displacement according to the load applied by block were calculated numerically by computer. As the numerical simulation data of specimen analyzed in this study approached the experimental data, the inspection of this specimen model suggested in this paper could be reasonable for the numerical simulation.

  • Polysilicon surface mdoification tecnique is developed to reduce the sticking of microstructures fabricated by micromachining. Modified anti-sticking grain holes are simply formed by two-step dry eth without additional photolithography nor deposition of thin films. Both process-induced sticking and in-use sticking are successfully reduced more than two times by adopting grain holed polysilicon substrate. A sticking model for cantilever beam is derived. This model includes bending moment stems from stress gradient along the thickness directionof structural polysilicon. Because the surface tension of rinse liquid and the surface energy of the solids to be stuk tend to decrease in recently developed anti-sticking techniques, the effect of stress gradient will play an important role to analyze the sticking phenomena. Effect of the temperature during post-release rinse and dry is modelled and verified experimentally. Based on developed anti-sticking polysilicon structure and the sticking model, sticking of microstructure, fabricated by simple wet process including sacrificial layer etch and rinse with deionized water without special equimpment for post-release rinse and dry was alleviated more than 3.5 times.

An analysis method for rubber toughened PVC is suggested to evaluate critical strain energy release rates(Gc) from the Charpy impact energy measurements. An Instrumented Charpy impact tester was used to extract ancillary information concerning fracture properties in addition to total fracture properties and maximum critical loads. The stress intensity factor Kd was computed for varying amounts of rubber contents from the obtained maximum critical loads and also toughening effects were investigated as well.

An instrumented Charpy impact tester was used to extract ancillary information concerning fracture parameters in addition to total fracture energies and maximum critical loads. The dynamic stress intensity factor $K_{Id}$ was computed for varying amounts of rubber contents from the obtained maximum critical loads and also toughening effects were investigated as well. The fracture surfaces produced under low velocity impact fur PVC/MBS composites were investigated by SEM. The results show that MBS rubber is very effective reinforcement material for toughening PVC.C.
