Changes in Cell Ca2+ Distribution in Loquat Leaves and Its Effects on Cold Tolerance

  • Calcium has been associated with improved cold tolerance in many crops. The aim of this study was to investigate the changes in leaf cell $Ca^{2+}$ distribution and cell organelle ultrastructure of loquat (Eriobotrya japonica Lindl.) plants in response to cold stress at $-3^{\circ}C$, using transmission electron microscopy (TEM). Two loquat accessions, Zaozhong 6 (a commercial cultivar) and oakleaf loquat (a wild relative) were used. Cold tolerance, as measured by leaf browning rate, was higher in oakleaf plants, and calcium treatment improved cold tolerance in both species. Cold stress first induced inward transport of $Ca^{2+}$ from the intracellular space. Then, the imported $Ca^{2+}$ was aggregated around the chloroplast membrane, finally entering the chloroplast. This pattern of $Ca^{2+}$ distribution in leaf cells occurred earlier in Zaozhong 6 than in the wild loquat. With increasing time of cold exposure, the chloroplast membranes of Zaozhong 6 leaves were damaged, blurred and even disappeared, while those of wild oakleaf loquat leaves maintained their structure longer. In Zaozhong 6, cold stress induced a clear cavity between poorly structured granal thylakoids and vesicles appearing inside the chloroplast, while in oakleaf leaves cold stress had little effect on the ultrastructure of chloroplasts (although chloroplast membranes looked blurred). Loquat leaves accumulated free calcium ions around chloroplasts in response to cold stress, with earlier calcium accumulation occurring in the cold-sensitive cultivar Zaozhong 6 than in wild oakleaf loquat. These results demonstrate that these two loquat species have differences in both cold tolerance and calcium accumulation dynamics.

  • Objective: The present study was carried out to evaluate the effect of high dietary cation and anion difference (DCAD) rations on diurnal variations in eating and meal patterns, water intake and urination patterns in dairy goats fed under high ambient temperature (HTa). Methods: Ten crossbred dairy goats during peri-parturition period were selected and divided into two groups of five animals each. Experimental diets were control DCAD (control, 22.8 mEq/100 g dry matter [DM]) and high DCAD (DCAD, 39.1 mEq/100 g DM). The composition of two diets consisted of 44% corn silage and 56% concentrate. From the 2nd week to 8th week postpartum, goats were fed ad libitum twice daily either with the control or DCAD total mix ration with free access to water. The spontaneous eating and drinking patterns were determined. Results: The environmental conditions in the present experiment indicated that goats were fed under HTa conditions (average peak THI = 85.2) and were in heat stress. In addition to the typical HTa induced tachypnoea in both groups, the respiratory rate in the DCAD group was significantly higher than the control group (p<0.05). Although the goats from both groups showed comparable level of eating, drinking and urination during experiment, the meal pattern and water intake were different. High DCAD apparently increased eating and meal patterns compared with the control. At week 8 postpartum, goats from high DCAD group had significant (p<0.05) bigger meal size and longer meal duration. Moreover, high DCAD appeared to increase night-time water intake (p<0.05). Conclusion: Both meal pattern and night-time drinking effects of DCAD suggested that feeding with high DCAD ration may alleviate the effect of heat stress in dairy goat fed under HTa conditions.

Effects of Taurine Supplementation on Heat Shock Protein 70 and In Vitro Protein Syntheses in Liver of Broiler Chicks under Chronic Heat Stress (고온 스트레스 하에 타우린 첨가가 육계 간의 Heat Shock Protein 70 및 In Vitro의 단백질 합성에 미치는 영향)

  • This study was conducted to investigate the effect of taurine supplementation on heat shock protein 70 and in vitro protein turnover in broiler chicks under chronic heat stress. Chicks were allocated into 3 groups of 10 birds per group; the control group was maintained at a temperature of $24^{\circ}C$ without taurine (CO group), the heat-stressed group maintained at a temperature of $34^{\circ}C$ without taurine (HO group), and heat-stressed group maintained at a temperature of $34^{\circ}C$ with taurine (HT group). The final body and liver weights of broilers in the HO and HT groups were significantly lower than those of broilers in the CO group (P<0.05). However, these parameters of the broilers in the HT group were significantly higher than those of broilers in the HO group (P<0.05). The heat shock protein 70 (hsp70) concentration in the liver of broilers in the HO group was significantly higher than that of broilers in the CO and HT groups, but the hsp70 concentration in the liver of broilers in the HT group was not different from that of broilers in the CO group. Liver homogenates of 21 day-old broilers were incubated at temperatures of $37^{\circ}C$ and $45^{\circ}C$ to prove the effect of high temperature and taurine on total protein syntheses. Neither high temperature nor taurine supplementation affected protein syntheses in liver homogenates of the broilers. However, the more the temperature increased, the more the degradation rates of cytoplasmic protein in liver homogenates increased; however, taurine supplementation had no effects on the protein syntheses in the liver of the broiler. It is possible that taurine indirectly affected protein turnover via various physiological mechanisms.

Heat Transfer and Flow Measurements on the Turbine Blade Surface (터빈 블레이드 표면과 선형익렬에서의 열전달 및 유동측정 연구)

  • An experimental study has been conducted to investigate the effects of the free stream turbulence intensity and Reynolds number on the heat transfer and flow characteristics In the linear turbine cascade. Profiles of the time-averaged velocity, turbulence intensity, and Reynolds stress were measured in the turbine cascade passage. The static pressure and heat transfer distributions on the blade suction and pressure surfaces were also measured. The experiments were made for the Reynolds number based on the chord length, Rec = $2.2{\times}10^4$ to $1.1{\times}10^5$ and the free stream turbulence intensity, $FSTI_1$ = 0.6% to 9.1 %. The uniform heat flux boundary condition on the blade surface was created using the gold film Intrex and the surface temperature was measured by liquid crystal, while hot wire probes were used for the flow measurements. The results show that the free stream turbulence promotes the boundary layer development and delays the flow separation point on the suction surface. It was found that the boundary layer flows on the suction surface for all Reynolds numbers tested with $FSTI_1$ = 0.6% are laminar. It was also found that the heat transfer coefficient on the blade surface increases as the free stream turbulence intensity increases and the flow separation point moves downstream with an increasing Reynolds number. The results of skin friction coefficients are in good agreement with the heat transfer results in that for $FSTI_1{\geq}2.6%$, the turbulent boundary layer separation occurs.

Effect of TiO2 on the Properties of ZnO-V2O5-P2O5 Low Temperature Sealing Glasses (저온실링용 ZnO-V2O5-P2O5계 봉착재의 물성에 미치는 TiO2 의 영향)

  • We designed new compositions for lead free and low temperature sealing glass frit of $ZnO-V_2O_5-P_2O_5$ system, which can be used for PDP (Plasma Display Panel) or other electronic devices. The $ZnO-V_2O_5-P_2O_5$ system can be used as a sealing material at temperatures even lower than 430$^{\circ}C$. This system, however, showed lower bonding strength with glass substrate compared to commercialized Pb based sealing materials. So, we added $TiO_2$ as a promoter for bonding strength. We examined the effect of $TiO_2$ addition on sealing behaviors of $ZnO-V_2O_5-P_2O_5$ glasses with the data for flow button, wetting angle, temporary & permanent residual stress of glass substrate, EPMA analysis of interface between sealing materials and glass substrate, and bonding strength. As a result, sealing characteristics of $ZnO-V_2O_5-P_2O_5$ system glasses were improved with $TiO_2$ addition, but showed a maximum value at 5 mol% $TiO_2$ addition. The reason for improved bonding characteristics was considered to be the chemical interaction between glass substrate and sealing glass, and structural densification of sealing glass itself.

Effect of Salicylic Acid on Growth and Chilling Tolerance of Cucumber Seedlings

  • The present study was undertaken to investigate the effect of low temperature and salicylic acid(SA) on the chilling tolerance of acclimated and nonacclimated cucumber(Cucurmis sativus L.) seedlings. The acclimation phenomenon was characterized in chilling-sensitive cucumber seedlings and found to have a significant effect on the survival and shoot dry weights. The injuries experienced by the acclimated seedlings in the third leaf stage were on average smaller by half than those experienced by the nonacclimated seedlings. Chilling also caused a large increase in the free proline levels, regardless of the acclimation status. Exogenous treatment with SA(0.5mM) resulted in improved growth and survival of the nonacclimated chilled seedlings, indicating that SA induced chilling tolerance and SA and acclimation had common effects. The application of cycloheximide in the presence of SA restored the acclimation-induced chilling tolerance. The elevated proline level observed in the cold-treated and SA-treated plants was more pronounced in the light than in the dark at a chilled temperature, indicating that endogenous proline may play a role in chilling tolerance by stabilizing the water status in response to chilling. From these results it is suggested that SA provided protection against low-temperature stress by increasing the proline accumulation, and pre-treatment with SA may induce antioxidant enzymes leading to increased chilling tolerance.

  • Micro gas sensors including thin film catal) tic type require stress-free memhrancs for etch stop of Si anisotropic etching and sublayer of sensing elements hecause stress is one of the main factors affecting breakdown of thin membranes. This paper reports the effects of deposition conditions on stress and refractive index of $SiN_{x}/SiO_{x}/(NON)$ films deposited by low pressure c11ernic;rl vapor deposition(L, t'CVI)) 2nd reactve sputtering. In the case of I.PCVI1, the stresses of $SiN_{x}$ and NON films arc $7.6{\times}10^{8}dyne/cm^2$ and $3.3{\times}10^{8}dyne/cm^2$, respectibely, and the refractive indices are 3.05 and 152, respectively. In the cxse oi the sputtered SiN, , compressi\e stress decreased in magnitude and then turned to tensility as increasing proc, ess pressure by lmtorr to 30mtorr and cicreasmg applied power density by $2.74W/cm^2$ to $1.10W/cm^2$. The hest value of film stress obt;~ined under condition of lOmtorr and $1.37W/cm^2$ in this' experiment was $1.2{\times}10^{9}dyne/cm^2$ cnnipressive. The refr~ict~ve index decreased from 2 05 to 1 89 as decreasing applied power density by lnitorr to 3Orntorr and increasing process pressure hy $2.74W/cm^2$ to $1.10W/cm^2$. Stresses of films deposited by both LPCVL) and sputtering decreased as incre;lsing temperature and showed plastic behavior as decreasing temperature.

Hydroformability and mechanical properties of A16061 tubes on different extrusion type (알루미늄 6061 압출재의 제조공정에 따른 온간액압성형성과 기계적 특성 연구)

For the investigation, as-extruded, full annealed and T6-treated Al 6061 tubes at different extrusion type were prepared. To evaluate the hydroformability, uni-axial tensile test and free bulge test were performed at room temperature and $200^{\circ}C$. Also mechanical properties of hydroformed part at various pre- and post-heat treatments were estimated by tensile test. And the tensile test specimens were obtained from hexagonal prototype hydroformed tube at $200^{\circ}C$. As for the heat treatment, hydroformability of full annealed tube is 25% higher than that of extruded tube. The tensile strength and elongation were more than 330MPa and 12%, respectively, when hydroformed part was post-T6 treated after hydroforming of pre- full annealed tube. However, hydroformed part using T6 pre treated tube represents high strength and low elongation, 8%. Therefore, the T6 treatment after hydroforming for as-extruded tube is cost-effective. Hydroformability of Al6061 tube showed similar value for both extrusion types. But flow stress of seam tube showed $20{\sim}50MPa$ lower value.

  • In this study, the characteristics of aluminum tube hydroformed products at different extrusion type and heat treatment conditions were investigated. For the investigation, as-extruded, full annealed and T6-treated Al 6061 tubes at different extrusion type were prepared. To evaluate the hydroformability, free bulge test was performed at room temperature to $300^{\circ}C$. Also mechanical properties of hydroformed products at various pre- and post-heat treatments were estimated by hexagonal prototype hydroforming test at $250^{\circ}C$. And the tensile test specimens were obtained from hexagonal prototype hydroformed tube. As a results, hydroformability of full annealed tube is $5{\sim}8%$ higher than that of extruded and T6-treated tube. The tensile strength and elongation of T6-post heat treated indirect extrusion tube were more than 330MPa and 12%, respectively. However, T6 pre treated hydroformed product represents high strength, 330MPa and low elongation, 8%. Therefore, Hydroformability of Al6061 tube showed similar value for both extrusion types. However flow stress of direct tube showed $20{\sim}50MPa$ lower value than indirect tube.
