해상 교량기초용 대형 원형강관 가설공법의 무선 원격 안정성 모니터링을 위한 현장실험 (Field Study on Wireless Remote Sensing for Stability Monitoring of Large Circular Steel Pipe for Marine Bridge Foundation)

  • 해상 교량기초용 대형 원형강관 가설공법은 가물막이와 작업대의 역할을 함께 수행할 수 있는 공법으로 개발되었다. 본 연구의 목적은 현장실험을 통해 시공과 운용 중 대형 원형강관의 안정성을 모니터링하기 위한 무선 원격계측시스템을 검증하는 것이다. 현장실험을 위해 수심 4m의 인공 해상 지반을 조성하였으며, 직경 5m, 높이 9.5m인 대형 원형강관을 석션 관입하였다. 대형 원형강관의 근입 깊이는 5m이다. 경사계와 변형률계는 상부 모듈의 서로 다른 네 면에 설치되었으며, 강관의 운반, 시공, 해체 전 과정에 걸쳐 경사각과 응력을 모니터링하였다. 실험결과, 경사계로 측정된 경사각은 석션 관입 중 일정하게 유지되었으나 x축 방향으로 더 크게 기울어진 것으로 나타났다. 또한, 서로 다른 면에 설치되어 있어도 같은 축 방향의 경사각은 거의 같게 측정되었다. 변형률계로 측정된 응력은 석션 관입 시 증가하였고, 인발 시 감소하였다. 또한, 측정된 응력으로부터 강관에 편심이 작용하고 있음을 알 수 있었다. 본 연구의 결과는 경사계와 변형률계로 구축된 무선 원격계측 시스템이 대형 원형강관의 안정성 모니터링에 유용하게 활용될 수 있음을 보여준다.

금형을 이용한 정밀전단가공에서 펀치의 변형거동 (Behavior of Punch Deformation in Precision Shearing Process Using Press Die)

  • Uneven clearances in the left and right sides of a press die cause deformation of the punch in precision shearing process. This deformation results from the compression stress and bending moment from shearing force in vertical direction and from the side force in horizontal direction acting to the punch, In this study the behavior of punch deformation is investigated in order to clarify the deformation state of the punch by using strain gauge deformation to shearing force side force bending moment radius of curvature and shear plane of the punch. Also we presented the calculation method of deformation size for the punch.

  • Stress waves monitored on the surface of structures under various loading conditions can provide useful information on the structural health status. In this paper, stress waves are measured by several sensors when a steel beam is impacted by a ball drop. The sensors used include the piezo-electric film Sensor, the electrical strain gage, and the ultrasonic transducer, and special attention is given to the pieza film sensor. The wavelet transform is used for the time-frequency analysis of dispersive waves propagating in the beam. The velocities of the wave produced in the team due to the lateral impact is found to be frequency-dependent and identified as the flexural wave velocity based on the comparisons with the Timoshenko beam theory. A linear impact site identification method is developed using the flexural wave, and the impact sites of the beam can be accurately estimated by the piezo film sensors. It is found that the piezo film sensor is appropriate for sensing stress waves due to impact and for locating impact sites in the beam.

Design, calibration and application of wireless sensors for structural global and local monitoring of civil infrastructures

  • Structural Health Monitoring (SHM) gradually becomes a technique for ensuring the health and safety of civil infrastructures and is also an important approach for the research of the damage accumulation and disaster evolving characteristics of civil infrastructures. It is attracting prodigious research interests and the active development interests of scientists and engineers because a great number of civil infrastructures are planned and built every year in mainland China. In a SHM system the sheer number of accompanying wires, fiber optic cables, and other physical transmission medium is usually prohibitive, particularly for such structures as offshore platforms and long-span structures. Fortunately, with recent advances in technologies in sensing, wireless communication, and micro electro mechanical systems (MEMS), wireless sensor technique has been developing rapidly and is being used gradually in the SHM of civil engineering structures. In this paper, some recent advances in the research, development, and implementation of wireless sensors for the SHM of civil infrastructures in mainland China, especially in Dalian University of Technology (DUT) and Harbin Institute of Technology (HIT), are introduced. Firstly, a kind of wireless digital acceleration sensors for structural global monitoring is designed and validated in an offshore structure model. Secondly, wireless inclination sensor systems based on Frequency-hopping techniques are developed and applied successfully to swing monitoring of large-scale hook structures. Thirdly, wireless acquisition systems integrating with different sensing materials, such as Polyvinylidene Fluoride(PVDF), strain gauge, piezoresistive stress/strain sensors fabricated by using the nickel powder-filled cement-based composite, are proposed for structural local monitoring, and validating the characteristics of the above materials. Finally, solutions to the key problem of finite energy for wireless sensors networks are discussed, with future works also being introduced, for example, the wireless sensor networks powered by corrosion signal for corrosion monitoring and rapid diagnosis for large structures.

Detection of edge delamination in surface adhered active fiber composites

  • A simple method has been developed to detect the bonding condition of active fiber composites (AFC) adhered to the surface of a host structure. Large deformation actuating capability is one of important features of AFC. Edge delamination in adhesive layer due to large interfacial shear stress at the free edge is typically resulted from axial strain mismatch between bonded materials. AFC patch possesses very good flexibility and toughness. When an AFC patch is partially delaminated from host structure, there remains sensing capability in the debonded part. The debonding size can be determined through axial resonance measured by the interdigitated electrodes symmetrically aligned on opposite surfaces of the patch. The electrical impedance and modal response of the AFC patch in part adhered to an aluminum plate were investigated in a broad frequency range. Debonding ratio of the AFC patch is in inverse proportion to the resonant frequency of the fundamental mode. Feasibility of in-situ detecting the progressive delamination between AFC patch and host plate is demonstrated.

Complete Genome Sequence of Levilactobacillus brevis KL251 Isolate from Kimchi

  • In this study, we performed whole-genome sequencing of Levilactobacillus brevis KL251 (KL251) isolated from kimchi. The KL251 genome, characterized by a circular chromosome spanning 2,345,062 base pairs with a GC content of 45.78%, was analyzed. KL251 contains 2,275 coding DNA sequences (CDSs), 56 transfer RNAs (tRNAs), and 4 ribosomal RNAs (rRNAs). Genes associated with gamma-aminobutyric acid (GABA) production and CRISPR-Cas systems were identified and could potentially be used for GABA synthesis and defense against foreign DNA. Additionally, the presence of functional genes involved in isoprenoid biosynthesis, glutathione generation, and redox sensing showed that cellular metabolism and stress responses were important characteristics of this genome. These genomic findings suggest that the KL251 strain could potentially have several applications, including food fermentation, probiotics, dairy product starters, and the development of health-enhancing products.

Cold-Stress Response of Probiotic Lactobacillus plantarum K25 by iTRAQ Proteomic Analysis

  • To understand the molecular mechanism involved in the survivability of cold-tolerant lactic acid bacteria was of great significance in food processing, since these bacteria play a key role in a variety of low-temperature fermented foods. In this study, the cold-stress response of probiotic Lactobacillus plantarum K25 isolated from Tibetan kefir grains was analyzed by iTRAQ proteomic method. By comparing differentially expressed (DE) protein profiles of the strain incubated at 10℃ and 37℃, 506 DE proteins were identified. The DE proteins involved in carbohydrate, amino acid and fatty acid biosynthesis and metabolism were significantly down-regulated, leading to a specific energy conservation survival mode. The DE proteins related to DNA repair, transcription and translation were up-regulated, implicating change of gene expression and more protein biosynthesis needed in response to cold stress. In addition, two-component system, quorum sensing and ABC (ATP-binding cassette) transporters also participated in cell cold-adaptation process. These findings provide novel insight into the cold-resistance mechanism in L. plantarum with potential application in low temperature fermented or preserved foods.

Composite Fracture Detection Capabilities of FBG Sensor and AE Sensor

  • Non-destructive testing methods of composite materials are very important for improving material reliability and safety. AE measurement is based on the detection of microscopic surface movements from stress waves in a material during the fracture process. The examination of AE is a useful tool for the sensitive detection and location of active damage in polymer and composite materials. FBG (Fiber Bragg Grating) sensors have attracted much interest owing to the important advantages of optical fiber sensing. Compared to conventional electronic sensors, fiber-optical sensors are known for their high resolution and high accuracy. Furthermore, they offer important advantages such as immunity to electromagnetic interference, and electrically passive operation. In this paper, the crack detection capability of AE (Acoustic Emission) measurement was compared with that of an FBG sensor under tensile testing and buckling test of composite materials. The AE signals of the PVDF sensor were measured and an AE signal analyzer, which had a low pass filter and a resonance filter, was designed and fabricated. Also, the wavelength variation of the FBG sensor was measured and its strain was calculated. Calculated strains were compared with those determined by finite element analysis.

Thermographic analysis of failure for different rock types under uniaxial loading

  • Mining activities focus on the production of mineral resources for energy generation and raw material requirements worldwide and it is a known fact that shallow reserves become scarce. For this reason, exploration of new resources proceeds consistently to meet the increasing energy and raw material demand of industrial activities. Rock mechanics has a vital role in underground mining and surface mining. Devices and instruments used in laboratory testing to determine rock mechanics related parameters might have limited sensing capability of the failure behavior. However, methodologies such as, thermal cameras, digital speckle correlation method and acoustic emission might enable to investigate the initial crack formation in detail. Regarding this, in this study, thermographic analysis was performed to analyze the failure behaviors of different types of rock specimens during uniaxial compressive strength experiments. The energy dissipation profiles of different types of rocks were characterized by the temperature difference recorded with an infrared thermal camera during experiments. The temperature increase at the failure moment was detected as 4.45℃ and 9.58℃ for andesite and gneiss-schist specimens, respectively. Higher temperature increase was observed with respect to higher UCS value. Besides, a temperature decreases of about 0.5-0.6℃ was recorded during the experiments of the marble specimens. The temperature change on the specimen is related to release of radiation energy. As a result of the porosity tests, it was observed that increase in the porosity rate from 5.65% to 20.97% can be associated to higher radiation energy released, from 12.68 kJ to 297.18 kJ.

Sensing the Stress: the Role of the Stress-activated p38/Hog1 MAPK Signalling Pathway in Human Pathogenic Fungus Cryptococcus neoformans

  • All living organisms use numerous signal-transduction pathways to sense and respond to their environments and thereby survive and proliferate in a range of biological niches. Molecular dissection of these signalling networks has increased our understanding of these communication processes and provides a platform for therapeutic intervention when these pathways malfunction in disease states, including infection. Owing to the expanding availability of sequenced genomes, a wealth of genetic and molecular tools and the conservation of signalling networks, members of the fungal kingdom serve as excellent model systems for more complex, multicellular organisms. Here, we employed Cryptococcus neoformans as a model system to understand how fungal-signalling circuits operate at the molecular level to sense and respond to a plethora of environmental stresses, including osmoticshock, UV, high temperature, oxidative stress and toxic drugs/metabolites. The stress-activated p38/Hog1 MAPK pathway is structurally conserved in many organisms as diverse as yeast and mammals, but its regulation is uniquely specialized in a majority of clinical Cryptococcus neoformans serotype A and D strains to control differentiation and virulence factor regulation. C. neoformans Hog1 MAPK is controlled by Pbs2 MAPK kinase (MAPKK). The Pbs2-Hog1 MAPK cascade is controlled by the fungal "two-component" system that is composed of a response regulator, Ssk1, and multiple sensor kinases, including two-component.like (Tco) 1 and Tco2. Tco1 and Tco2 play shared and distinct roles in stress responses and drug sensitivity through the Hog1 MAPK system. Furthermore, each sensor kinase mediates unique cellular functions for virulence and morphological differentiation. We also identified and characterized the Ssk2 MAPKKK upstream of the MAPKK Pbs2 and the MAPK Hog1 in C. neoformans. The SSK2 gene was identified as a potential component responsible for differential Hog1 regulation between the serotype D sibling f1 strains B3501 and B3502 through comparative analysis of their meiotic map with the meiotic segregation of Hog1-dependent sensitivity to the fungicide fludioxonil. Ssk2 is the only polymorphic component in the Hog1 MAPK module, including two coding sequence changes between the SSK2 alleles in B3501 and B3502 strains. To further support this finding, the SSK2 allele exchange completely swapped Hog1-related phenotypes between B3501 and B3502 strains. In the serotype A strain H99, disruption of the SSK2 gene dramatically enhanced capsule biosynthesis and mating efficiency, similar to pbs2 and hog1 mutations. Furthermore, ssk2, pbs2, and hog1 mutants are all hypersensitive to a variety of stresses and completely resistant to fludioxonil. Taken together, these findings indicate that Ssk2 is the critical interface protein connecting the two-component system and the Pbs2-Hog1 pathway in C. neoformans.
