Effect of the Lycii Fructus on multiple low-dose streptozotocin-induced diabetic rats (Streptozotocin 저용량 반복투여로 유도한 당뇨 흰쥐에서 구기자 추출물의 효능연구)

  • Objectives : This study was performed to investigate the anti-diabetic effect of the fruit extract of Lycium chinense Mill(Lycii Fructus, LF) on multiple low-dose streptozotocin-induced diabetic rats.Methods : Male Sprague-Dawley rats were divided into three groups; normal, STZ-control, Lycii Fructus extractorally administrated 300 ㎎/㎏ group (STZ-LF). Diabetes was induced in rats by consecutive injection of streptozotocin (STZ) at doses of 30 ㎎/㎏ for 5 days. After 4 weeks, all rats were sacrificed, Fasting blood glucose, total cholesterol (CHO), triglyceride (TG) and HDL-Cholesterol were measured in sera of rats. Histopathological changes of pancreas, kidney, liver and lung tissues were observed by microscope after H&E, Periodic acid-Schiff (PAS) and Masson's trichrome staining. The changes of body weight, blood glucose, and food and water intake were also measured.Results : There were no differences in body, food intake and water intake in LF-administrated groups compared with STZ control group. However, LF extract significantly decreased the levels of serum glucose, CHO, TG and HDL-Cholesterol in diabetic rats. In histopathological analysis of kidney, liver and lung, LF-administrated groups showed the inhibition of morphological damage.Conclusions : These results suggest that LF have a biological action on multi low-dose STZ-induced diabetes in rats via decreasing the serum glucose, TG and TG levels and may protect the morphological changes of kidney, liver and lung.

  • The increased level of glucose, ketone bodies, non-esterified fatty acids and lactate in blood, decrease of glycogen content, phosphofructokinase activities and glucokinase activity and the increased level of glucose-6-phosphatase activity in the liver of streptozotocin injected rats were significantly moderated by ginseng saponin administration. It is not likely, however, that the hypoglycomic action of ginseng saponin might be due to their direct action on enzyme activities, since the saponin effect of servers enzymes in vitro was not enough to explain such an appreciable hypoglycemic activity of the saponin in streptozotocin induced diabetic rats, for which much work have to be done.

Effects of KIOM-79 on streptozotocin-induced insulin-dependent diabetes in Sprague-Dawley rats (Streptozotocin을 이용한 제1형 당뇨 유발 흰쥐에서 KIOM-79의 효과)

  • We investigated the effect of KIOM-79, 80% ethanolic extract of herbal prescription isolated from Magnolia officinalis, Pueraria lobate, Glycyrrhiza uralensis, and Euphorbia pekinensis, on streptozotocin-induced diabetes in Sprague-Dawley rats. The rats were treated orally with KIOM-79 (500 mg/kg/day) 1) for 3 days prior to streptozotocin (60 mg/kg, intraperitoneal) injection or 2) for 9 weeks after establishing diabetes model to examine acute and chronic effects on hyperglycemia and biochemical variables, respectively. As a result, KIOM-79 had little effects on hyperglycemic changes in acute model. Sexual comparison, however, showed reduced hyperglycemia in female rats, especially 24 hours after streptozotocin injection with or without KIOM-79 pretreatment. In chronic model, streptozotocin-induced hyperglycemia was well established, but KIOM-79 treatment showed no statistically significant effects on all variables. Thus, based on our findings KIOM-79 might have little effects on streptozotocin-induced insulin-dependent diabetes although it has been known to have hypoglycemic and antidiabetic effects on non-insulin-dependent diabetes models.

Effects of Indongdeung Jigolpi-tang applied to Soyangin on Hyperglycemic Rats induced by Streptozotocin (소양인(少陽人)에 응용(應用)되는 인동등지골피탕(認冬藤地骨皮湯)이 Streptozotocin 투여(投與) 고혈당(高血糖) 백서(白鼠)에 미치는 영향(影響))

  • In order to investigate experimented effects of Indongdeung Jigolpi-Tang, which is applied to Soyangin, to diabetes, the measurement, comparison and observation made on the contents of volumes of blood glucose, total cholesterol, triglyceride, phospholipid, total protein and electrolyte($K^+$, $Na^+$) in blood serum by giving a high blood glucose, induced by streptozotocin, to hyperglycemic rats, dosing the cooked liquid of I.J.T., and following conclusion was attained. 1. The volume of glucose urine, induced by streptozotocin to hyperglycemic rats, was shown an increase in comparision to a normal flock when $12.50m{\ell}/kg$ of I.J.T. dosed to the rats and checked after two and four weeks and thereby, a sinificant reduction in the volume of blood glucose was made. 2. The volume of total cholesterol of blood serum in the diabetic rats, induced by streptozotocin, was increased in comparision to the normal flock, and in the fouth week of dosing $1.25m{\ell}/kg$ of I.J.T., a significant reduction of total cholesterol volum was shown in the blood serum. 3. The volume of triglyceride in the blood serum of the diabetic rats, induced by the streptozotocin, was increased in comparision to the normal flock, and with dosing $1.25m{\ell}/kg$ and $12.50m{\ell}/kg$ of I.J.T., a significant reduction of triglyceride volium was shwon in the blood serum after second and fourth week. 4. The volume of phospholipid in the blood serum of the diabetic rats, induced by the streptozotocin, was somewhat reduced in comparision to the normal flock, and with dosing $1.25m{\ell}/kg$ of I.J.T., a significant reduction of phospholipid volium in the blood serum was shown after first and second week. 5. The volume of total protein in the blood serum of the diabetic rats, induced by the streptozotocin, was somewhat reduced in comparison to the normal flock however, with dosing the I.J.T., no significant change on the volium of total protein was made in the blood serum. 6. The volume of potassium in the blood serum of the diabetic rats, induced by the streptozotocin, was increased in comparision to the normal flock however, with dosing the I.J.T., no significant change of potassium content was made in the blood serum. 7. The volume of sodium in the blood serum of the diabetic rats, induced by the streptozotocin, was somewhat reduced in comparision to the normal flock, and with dosing $12.50m{\ell}/kg$ of the I.J.T., a significant increase was shown in the blood serum after first week. Soyangin, accompanying with a middle and upper heat, the I.J.T. - that has a merits of cleaning up fever, solving fever, solving poison, solving thirst and restoring exhaustion - can be proved that it has a blood sugar reduction effect and it is considered that it can be meritorious for strengthening vitality, further, many other exhausting diseases, and can actively used for the cases such as thirst causing disease and a disease that causes an abnormal food hunger, thirst causing and frequent going to pass urine, in particular.

Effect of Natural Functional Mixture on the Descent of BloodGlucose Level in Streptozotocin-Induced Diabetic(type I) Rats(I) (천연 기능성 소재 혼합물이 Streptozotocin 유발 제1형 당뇨 쥐의 혈당 강하 효과(I))

  • Hypoglycemic efficacy of natural functional mixture(FM) and level of the diabete related hormones in streptozotocin(STZ)-induced diabetic rats were investigated. Male Sprague-Dawley rats were divided into three groups (normal, diabetic fed diets with/without FM). Supplement of FM did not affect the body weight and feed intake of STZ-induced diabetic rats. The increase in the weight of liver of STZ-induced diabetic rats was weakened by supplement of FM, whereas the weight of kidney and heart was not affected. Blood glucose level was slightly, and glucose tolerance of post-feeding was significantly improved by functional mixture. The mixture significantly reduced the elevated HbA1C level of diabetic rats by 15%, and it increased the level of insulin and C-peptide in blood and decreased glucagon level. Therefore, we conclude that FM in this study has a potency of prevention and treatment of diabetes mellitus.

  • This study evaluated effects of Yanggyuksanhwa-tang(YST) on diabetic rats. Diabetic condition in rats was induced by streptozotocin injection. Then control effect of YST was observed with changes of serum glucose, insulin, and triglyceride levels and hepatic glucokinse activity. YST treatment was resulted significant decrease of high serum glucose level of diabetic rats induced by streptozotocin at 5th day of YST treatment. YST treatment was resulted increase of low serum insulin level of diabetic rats induced by streptozotocin, but it was not significant statistically. YST treatment was resulted significant decrease of high serum triglyceride level of diabetic rats induced by streptozotocin at 3rd and 5th of YST treatment. YST treatment was resulted significant increase of low hepatic glucokinase activity of diabetic rats induced by streptozotocin at 5th day of YST treatment. These results suggest that YST has effects on serum glucose control against cerebral inchemic damage under diabetic condition.

The Effect of Vitamin B6 Deficiency on Energy Metabolite in Streptozotocin-induced Diabetic Rats (Vitamin B6 결핍이 Streptozotocin 유발 당뇨 흰쥐의 에너지 대사물 농도에 미치는 영향)

  • The purpose of this study was to investigate the effect of vitamin B6 deficiency on the concentration of energy metabolite in streptozotocin-induced diabetic rats. Thirty rats were fed a vitamin B6 deficient diet(-B6) or a control diet(+B6) for 5 weeks and then subdivided into 3 groups respectively ; base group, one day diabetic group and three day diabetic group. Diabetes of rats were induced by streptozotocin injection into the tail vein. Glucose, glycogen, protein, alanine, triglyceride and free fatty acids were compared in plasma, liver skeletal muscle of rats. Also, the total urinary nitrogen and glucose excretion were compared. Compared with +B6 rats, the increase of plasma glucose in -B6 rats due to the diabetes was smaller. After diabetes was induced, the level of plasma alamine was not changed in -B6 rats while increased significantly(p<0.05) in +B6 rats. The increase of urinary nitrogen excretion was smaller and the increase of muscle protein was larger in -B6 rats at the first day diabetes was induced. The levels of plasma free fatty acid and liver triglyceride were significantly (p<0.05) higher in -B6 rats after diabetes was induced. These results suggest that vitamin B6 deficiency may impair the adaptation of animals to the energy metabolism related due to a decrease of the body protein catabolism of fatty acid oxidation in diabetes and aggravate fatty liver which is one of the chronic complications of diabetes.

The Anti-diabetes and Vasoelasticity Effects of Mori Folium and Aurantii Fructus in Streptozotocin Induced Type II Diabetes Mellitus Model (Streptozotocin 유발 당뇨 백서에 상엽 지각(桑葉 枳殼) 혼합물의 항당뇨 및 혈관탄성개선 효과)

  • Objectives : This study was designed to evaluate the anti-diabetes and vasoelasticity effects of Mori Folium and Aurantii Fructus in streptozotocin-induced type II diabetes mellitus model. Methods : The anti-diabetic effect of Mori Folium and Aurantii Fructus on rats induced with diabetes by streptozotocin was investigated through analyses of changes in body weight, blood glucose, urine volume of rats, viability of human umbilical vein endothelial cells(HUVECs), and elasticity of descending thoracic aorta in rats. The subjects in this study were divided into four groups(n=15): a normal group without any treatment (Con), a normal group with Mori Folium and Aurantii Fructus treatment(Con+P), a diabetes group induced by streptozotocin(STZ), and a Mori Folium and Aurantii Fructus treatment group under diabetes induced by streptozotocin(STZ+P). Rats were administered streptozotocin to induce diabetes. Results : The study showed that Mori Folium and Aurantii Fructus significantly reduced highly increased blood glucose levels(p<0.01) and prevented the diabetic rats from weight loss(p<0.01) and polyurea(p<0.05), Mori Folium and Aurantii Fructus also recovered decreased viability of HUVECs(p<0.01) and damaged elasticity of aorta induced by the streptozotocin (p<0.01). Conclusions: It was concluded from the results that Mori Folium and Aurantii Fructus have a distinct anti-diabetes effect and they also prevent damage of blood vessel induced by diabetes. resulting in prevention of cardiovascular diseases ascribed to diabetes.

Effects of Polygonatum odoratum on In vivo Insulin Activity in Streptozotocin-Induced Diabetic Rats (둥글레 섭취가 Streptozotocin 유발 당뇨병 쥐의 In vivo 인슐린 작용에 미치는 영향)

  • This study investigated the in vivo insulin function of Polygonatum odoratum in normal and diabetic male Sprague-Dawley rats. Diabetes mellitus was induced by an i.p. injection of streptozotocin. Normal and diabetic rats were assigned to the diet groups of the control basal diet and Polygonatum odoratum diet. The animals were fed the diet and water ad libitum for 15 days. Initial and final body weights, total food intake and serum glucose and insulin levels were measured. An insulin suppression test was performed to elucidate the insulin function in the peripheral tissues. The results showed that the final serum glucose levels significantly decreased in the diabetic rats on the Polygonatum odoratum diet compared with the diabetic rats on the control diet. The final serum insulin levels were increased in the diabetic rats on the Polygonatum odoratum diet compared with the diabetic rats on the control diet. The in vivo function of the insulin increased in the diabetic rats on the Polygonatum odoratum compared with the diabetic rats on the control diet. These data indicate that Polygonatum odoratum may be beneficial in improving the in vivo insulin function in streptozotocin-induced diabetic rats.

The Effect of Vitamin $B_2$ Deficiency on Fuel Metabolism in Streptozotocin Induced Diabetic Rats (Vitamin $B_2$ 결핍이 Streptozotocin 유발 당뇨 흰쥐의 에너지대사에 미치는 영향)

Diabetes of the rats were induced by streptozotocin injection into the tail vein. Glucose, glycogen, protein, alanine, triglyceride and free fatty acid were compared in plasma, liver, skeletal muscle of rats. Also, the total urinary nitrogen and glucose excertion were compared. Compared with +B2 rats, the increase of plasm glucose in -B2 rats due to the diabetes tended to be smaller. After diabetes were induced, the levels of plasma protein and alanine was significantly decreased and the urinary nitrogen excretion was significantly increased in -B2 rats. The level of plasma free fatty acid was increased continuously in B2 rats while increased at the first day and decreased at the third day diabetes was induced in +B2 rats. These results suggest that vitamin B2 deficiency increase protein catabolism due to the decrease of fatty acid oxidation. Thus, vitamin B2 deficiency in diabetes impair the adaptation of animals to the fuel metabolism and aggravate the body protein wasting which is one of the chronic complications of diabetes.
