Hypoglycemic Effect of Yacon Tuber Extract and Its Constituent, Chlorogenic Acid, in Streptozotocin-Induced Diabetic Rats

  • Smallanthus sonchifolius (Yacon, Asteraceae) was originally cultivated in South America and used in food and traditional medicine by Andean inhabitants. Yacon is potentially beneficial for the management of diabetes and is composed of fructooligosaccharides, proteins, minerals and phenolic compounds. The aim of this study was to investigate the hypoglycemic effect of Yacon tuber extract (YTE) and its constituent, chlorogenic acid (CGA), in streptozotocin (STZ)-induced diabetic rats. In this study, a HPLC method was developed for simultaneous determination of major active phenolic components, CGA and caffeic acid in YTE. We investigated the hypoglycemic effect of YTE and CGA in STZ-induced diabetic rats and studied glucose tolerance test (GTT). The effect of orally administered multiple doses of YTE and CGA on plasma biochemical parameters was examined using diabetic rats. We also measured free radical scavenging activity by the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. Oral administration of YTE (200 mg/kg) and CGA (10 mg/kg) for 6 weeks produced a significant hypoglycemic effect in STZ-induced diabetic rats. YTE and CGA-treated groups exhibited significantly decreased plasma glucose surge during the GTT. Total cholesterol (TC) and triglyceride (TG) concentrations were significantly decreased by 33% and 49%, respectively, in YTE-treated rats. TC and TG concentrations were also significantly decreased by 26 % and 41%, respectively, in CGA-treated rats. In the DPPH assay, free radical scavenging activity of CGA was similar to that of vitamin E, a positive control. This study suggests that YTE and its constituent, CGA, may be a useful option for management of hyperglycemia and diabetic nephropathy.

The Effects of Korean Wild Vegetables on Blood Glucose Levels and Liver-muscle Metabolism of Streptozotocin-induced Diabetic Rate (한국산 야생식용식물이 당뇨유발 흰쥐의 혈당 및 간과 근육내 에너지원 조성에 미치는 영향)

  • Attempts were made to determine the effects of 5-Korean wild vegetabel consumptions on blood glucose leveles and orgen-energy metabolisms in streptozotocin-induced diabetic rats. The 5-Korean wild vegetables were : Cassia tora L.(C.t), Lycium chinese Mill(L.c), Trichosanthes kirilowii Max(T.k), Polygonatum odoratum var. Pluriflorum Ohwi(P.o) and Arctium lappa L(A.l). Sixty male Spargue-Dawley rats(160-220g) were induced diabetes mellitus by streptozotocin injection into the tail vein and were devided into 6 groups : a diabetic control and 5-experimental groups. All groups of the rats were fed on a AIN-76 diet, and the 5-experimental groups were fed with each wild vegetable (10%) for four weeks. An increased tendency in body weight of all the groups was observed and the tendency was more significant in L.c, T.k. and P.o. groups. The organ weight of liver and kidney were higher in L.c. and A.l. groups and lower in T.k. group which has shown the improvement from diabetes. Plasma glucose levels were markedly decreased from the 1st week in C.t, T.k. and P.o. groups and the tendency has lasted throughout the four weeks experimental period. The consumption of P.o. has decreased plasma cholesterol level while any significant difference was not seen in plasma protein levels from all the experimental groups. The level of plasma triglyceride was decreased in P.o. group and the levels of plasma free fatty acids were also significantly lower in P.o. and T.k. groups. The liver protein levels were significantly higher in P.o. and T.k. groups and these two groups also showed the negative or relatively small amount of urinary glucose excretion. The experimental group of T.k. has revealed the decreased level of muscles protein and the increased level of muscle glycogen. The 5-Korean wild vegetables contained dietary fiber and 9-analyzed minerals comparable to the ordinary use vegetables.

The Effect of BuOH Fraction of Polygonatum odoratum with Selenium on Blood Glucose Level and Lipid Peroxidation in Streptozotocin Induced Diabetic Rats (둥굴레 분획물과 Selenium이 Streptozotocin 유발 당뇨 흰쥐의 혈당수준과 지질과산화에 미치는 영향)

  • The purpose of this study was to investigate the effect of butanol(BuOH) fraction of Polygonatum odoratum with selenium tr-eatment on blood glucose levels and lipid peroxidations in streptozotocin(STZ) induced diabetric rats. Male Sprague-Dawly rats weighing(180-200g) were divided into five groups : normal STZ-control and three expreimental groups(P, odoratum group P, odo-Se group and Se group) Diabetes mellitus was induced by injection STZ in the tail vein at the dose of 45mg/kg B.W The BuOH fraction of Polygonatum odoratum(500mg/kg. B,W) given orally administered for 14 days. The Se treated group were fed a AIN-76 recommendation diet mixed with Na2Seo3(2mg/kg diet). Diabetic rats showed the lower weight gain compared to the normal rats. the plasma glucose levels of the P. odo-Se group were significantly lower than the other experimental groups. The plasma cholesterol levels were higher in STZ-control and Se groups compared toP.odoratum and P. odo-Se groups and HDL-cholesterol levels were increased in the diabetic experimental groups fed on BuOH fraction of P. odoratum with Se supplementation. The liver and muscle glycogen levels were not significantly differ among all groups. The plasma free fatty acid levels were lower in diabetic experimental groups fed on BuOh fraction of P. odoratum or Se sup-plementation than STZ-control and Se groups. Diabetics rats showed the higher levels of triglyceride in plasma andlower levels in liver compared with the normal group. Supplementation with Se decreased significantly the liver triglyceride level. The MDA levels in liver and kidney were significantly reduced in all the experimental groups. In conclusion administration of BuOH fraction of Polygonatuum odoratum with selenium supplementation reduced blood glucose levels and peroxdative tissue damage in STZ induced diabetic rats showing the possibility of preventiave and therapeutic use of the wild edible plant to the diabetes mellitus.

Silymarin, a flavonoid antioxidant, protects streptozotocin-induced lipid peroxidation and β-Cell damage in rat pancreas

  • The present study is aimed at finding the influence of silymarin (a flavonoid) (25 mg/kg & 50 mg/kg) in streptozotocin (STZ)-induced diabetic rats. Type 2 diabetes was induced by single intraperitoneal injection of STZ (100 mg/kg) to 3 days old rat pups. Silymarin was administered for 15 days after the animals were confirmed diabetic (75 days after STZ injection). Blood glucose, glycosylated hemoglobin ($HbA_{1c}$), lipid peroxides (LPO) levels and reduced glutathione (GSH) contents in pancreas and liver were estimated following the established procedures. Biochemical observations were further substantiated with histological examination of pancreas. Blood glucose and $HbA_{1c}$ levels, which were elevated by STZ, were lowered to physiological levels by the administration of silymarin. The levels of LPO were significantly increased in STZ-induced diabetic rats. Silymarin reduced the LPO levels in both pancreas and liver. GSH contents which were reduced significantly in pancreas and liver of STZ-induced diabetic rats were brought back to near normal levels by silymarin treatment. Multifocal necrotic and degenerative changes of pancreas in STZ-diabetic rats were minimized to near normal morphology by administration of silymarin as evident by histopathological examination. Silymarin showed a dose dependent protective effect on STZ-induced $\beta$-cell damage. It could be attributed to the antioxidative and free radicals scavenging properties of the flavonoid. Thus, it may be considered as a natural antioxidant with potential therapeutic application in the treatment of type 2 diabetes.

Effects of Soyoligosaccharides on Blood glucose and Lipid Metabolism in Streptozotocin-Induced Diabetic Rats (대두올리고당이 당뇨 유발 흰쥐의 혈당 및 지질대상에 미치는 영향)

  • This study was conducted to investigate the effect of soyligosaccharides on improvement of diabetes mellitus. Sprague Dawley rats were divided into three groups. Control and one experimental group were fel AIN-93G diet(mixed diet) and the other experimental group was fed a similar diet containing 5% soyligosaccharides(oligosaccharide diet) for 6 weeks. Experimental groups were divided into two sub-groups after streptozotocin(STZ) injection and fed th mixed diet or the oligosaccharide diet for the next 2 weeks. Followings are final experimental and control groups: MM (mixed dietlongrightarrowSTZ injectionlongrightarrowmixed diet). MO(mixed dietlongrightarrowSTZ injectionlongrightarrowoligosaccharide diet), OM( oligosaccharide dietlongrightarrowSTZ injectionlongrightarrowmixed diet), OO(oligosaccharide dietlongrightarrowSTZ injectionlongrightarrowoligosaccharide diet), C(mixed dietlongrightarrowplacebo injectionlongrightarrowmixed diet). Blood glucose was analyzed at d1, d8 and d12 after streptozotocin injection. At the end of the experimental period, fecal bifidobacteria, plasma insulin, total lipid, triglyceride, cholesterol, and leptin concentrations was analyzed, After STZ injection, blood glucose level was significantly elevated in experimental group compared to a control group. Elevated blood glucose level declined faster in OO group compared to the other experimental groups. Meanwhile, insulin concentration decreased in all experimental groups compared to a control group. The number of bifidobacteria was significantly increased in oligosaccharide diet groups after STZ injection (MO and OO) than a mixed diet group(MM). Plasma cholesterol concentration significantly decreased in oligosaccharide diet groups(MO and OO) compared to mixed diet groups (MM and OM) after SZ injection. Plasma triglyceride and liver cholesterol concentrations significantly decreased in MO group compared to other experimental groups. Meanwhile, plasma leptin concentration was significantly lower in oligosaccharide diet groups after STZ injection(MO and OO) compared to a control group. The results in this study suggest that the dietary soyoligosaccharides has strong on reduction of reduction of blood glucose and lipid concentrations in streptozotoicin-induced diabetic rats. Thus, the use of soyoligosaccharides at a diet for IDDM patients may help to improve diabetes mellitus symptoms.(Korean J Nutrition 34(1) : 3-13, 2001)

Improvement of PENS on Peripheral Nerve Conduction Function in STZ-Induced Diabetic Rats (당뇨유발백서에서 피하신경전기자극의 말초신경기능 개선효과)

  • Purpose: This study aimed the effects of percutaneous electric nerve stimulation (PENS) applied to different parts of the streptozotocin-induced diabetic rats on the change of glucose and nerve. Methods: rats (ten weeks old) were selected as the subjects; the normal group was five rats, and the diabetes induction group II, III and IV were five rats, respectively, which were randomly sampled from the twenty-five streptozotocin-administered rats with more than $240\;d{\ell}/m{\ell}$ of blood sugar. For PENS, electric current with 2 Hz of stimulation frequency and $200\;{\mu}s$ of pulse duration was applied to the subjects for fifteen minutes a day, six days a week, for three weeks. Calculation of glucose and weight, and nerve conduction test were conducted forty-eight hours and three weeks after streptozotocin administration, respectively. Results: As for change of glucose and weight, the group III with stimulation to the acupoints and the group IV with stimulation to non-acupoints showed significant differences from the control group II (p<0.05). As for MNCV (motor nerve conduction velocity), the group III with stimulation to the acupoints showed significant differences from the group IV with stimulation to non-acupoints and the control group II (p<0.05). Conclusion: PENS had the effects of inhibiting increase of glucose, change of weight and decrease of nerve conductive function between the distal and proximal ends of the peripheral nerve in the STZ-induced diabetic rats.

Galactolipids from Mori Folium and their Hypoglycemic Effect (혈당강하효과를 나타내는 상엽의 당지질 성분)

  • In order to find out hypoglycemic constituents from natural resources, Korean medicinal plants being used for the treatments of diabetes mellitus were tested with streptozotocin-induced diabetic rats. From the above screening, the $CH_2Cl_2$ fraction of 80% methanol extracts of Morus alba leaves showed hypoglycemic activity. The $CH_2Cl_2$fraction was chromatographed on silica gel, resulting in 9 subfractions, and tested hypoglycemic activity. The subfraction which had a strong hypoglycemic activity was rechromatographed and two galactolipids (compounds 1 and 2) were isolated along with a triterpene (compound 3) from other fractions. On the basis of spectroscopic data, acid hydrolysis, alkaline hydrolysis and enzyme hydrolysis results, compounds 1, 2 and 3 were identified as $1-O-(9Z,12Z,15Z-octatrienoyl)-2-O-(9Z,12Z,15Z-octatrienoyl)-3-O-{\beta}-D-galactopyranosyl$ glycerol, $2(R)-1-O-octadecanoyl-2-O-(9Z,12Z,15Z-octatrienoyl)-3-O-{\beta}-D-galactopyranosyl$ glycerol, and $3{\beta}-hydroxylup-20(29)-ene$ acetate, respectively. Compound 2 (1mg/kg p.o.) has approximately 16% hypoglycemic activity in streptozotocin-induced diabetic rats.

Effects of Bombycis corpus on Streptozotocin-induced Diabetic Rats (Streptozotocin으로 유발된 당뇨쥐에 대한 백강잠(白?蠶)의 혈당 및 당대사 효소활성에 관한 효과)

  • Objective : This study was undertaken to investigate how Bombycis corpus (BC) effects the development and progress of complications occurring in Diabetes Mellitus (DM). Methods : Laboratory rats were seperated into three groups; normal, rats with DM and treated with BC, and rats with DM and not treated. In this study DM was experimentally induced through injection of streptozotocin. The BC treated group was given BC extract p.o. for 15 days. Then, The activities of glucose phosphatic enzymes and polyol pathway channels were observed. Results : The blood glucose level greatly increased in the DM groups after injection of streptoztocin, but it significantly decreased in the BC treated group. Significantly enhanced levels of serum insulin levels were seen in the BC treated group, while supressed levels were seen in the untreated DM group. Weight was recovered by the BC treated group, matching the normal group. Decreased enzyme activity of aldose reductase, sorbitol dehydrogenase and glucose-6-phosphatase were seen in BC treated diabetic rats. Increased enzyme activity, of the glucokinase and hexokinase were seen in BC treated diabetic rats. Conclusions : This study suggests that BC normalized the blood glucose and serum insulin levels destablized by DM. Because increased activity of glucose phosphatic enzymes, glucokinase and hexokinase, and decreased glucose-6-phosphatase activity, and suppression of polyol pathway enzymes, aldose reductase and sorbitol dehydrogenase, were all seen, these observations suggest that BC suppresses blood glucose levels and prevents complications due to DM.

The antidiabetic properties of fractions of Lycopus lucidic Turcz in streptozotocin diabetic rats (택란 분획물이 당뇨유발 흰쥐에서의 혈당강하에 미치는 영향)

  • This study was carried out to evaluate the effects of fractions of methanol(MeOH) extracts of Lycopus lucidic Turcz on hyperglycemia and energy metabolites in streptozotocin(STZ) diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats weighing 200-220 g by an injection of STZ dissolved in a citrate buffer into the tail vein at a dose of 45 mg/kg of body weight, and the rats were divided into 7 groups, that is, one normal group and 6 diabetic groups: STZ-control, hexane, chloroform(CHCl$\sub$3/). ethylacetate(EtOAc), butanol(BuOH) and H$\sub$2/O fraction-fed groups. All groups were fed an AIN-93 diet and the fractions of Lycopus lucidic Turcz were administered orally with 2 % Tween 80 for 14 days after the STZ injection. Body weight, diet intake and organ weights were monitored. The plasma levels of blood glucose, insulin and protein were determined. The plasma concentrations of cholesterol, HDL-cholesterol, triglycerides and free fatty acid were assayed. The plasma activities of aspartate aminotransferase (AST) and alanine aminotransferase(ALT) were also measured. Body weight losses were observed by feeding the fractions of Lycopus lucidic Turcz in STZ experimental groups, and the kidney weight was increased. The extent of blood glucose decrement was significantly greater in the hexane and BuOH fraction-fed groups than STZ-control group. The plasma protein level was significantly lower in the H$\sub$2/O fraction-fed group. The plasma cholesterol level was decreased in BuOH and H$\sub$2/O fraction-fed groups compared with the STZ-control group. The levels of free fatty acids in the CHC1$\sub$3/ and H$\sub$2/O fraction-fed groups were significantly decreased(p<0.05). ALT activitiy of BuOH fraction-fed group was lower than control but it was not significantly different. These results suggest that the fractions of Lycopus lucidic Turcz are capable of lowering blood glucose and fat metabolites concentrations when administered to STZ-treated rats, and AST/ALT activity and insulin levels show the possibility of therapeutic use to diabetes mellitus.

Effect of CHCl$_3$fraction of Alisma canaliculatum with selenium on the plasma glucose and lipids levels in streptozotocin-induced diabetic rats (택사 CHCl$_3$분획물과 Selenium 보충이 당뇨 흰쥐의 혈당과 지질함량에 미치는 영향)

  • The purpose of this study was to investigate the effect of chloroform(CHC1$_3$) fraction of Alisma canaliculatum All. Braun et Bouche(Ac) with selenium(Se) on the plasma glucose and lipid levels in streptozotocin(STZ)-induced diabetic rats. Male Sprague-Dawley rats were divided into five groups. The normal and diabetic rats were separated into four groups: the STZ-control group, the Ac group, the Ac-Se group and the Se group. Diabetes was induced in the male rats by an injection of STZ into the tail vein at a dose of 45 mg/kg. The CHCl$_3$fraction of Ac(250 mg/kg) was administered orally for 14 days. The supplementation was achieved with the AIN-93 recommended diet by adding 2 mg/kg diet of selenium as Na$_2$SeO$_3$. which was prepared freshly everyday. The body weight, hematocrit(Hct), glucose, insulin, cholesterol, HDL-cholesterol, triglyceride(TG) and free fatty acids(FFA) concentrations in plasma were measured. The aminotransferase activities were also analyzed. The changes of body weight in the experimental groups were not significantly different from that of the STZ-control group, but diabetes hyperphagia accompanied changes with body weight loss in Ac-Se group. The levels of Plasma cholesterol were not significantly different among the experimental groups. The concentrations of FFA in the Ac-Se group increased significantly compared with the STZ-control group. The effect of Se alone significantly increased aspartate aminotransferase activity and alanine aminotransferase activity. The results showed that the treatment of CHCl$_3$fraction of Ac in combination with Se has no synergistic effect. There was a tendency for the plasma glucose levels to decrease when Se was administered into diabetic rats. Supplementation of Se in diabetic rats did not elicit a significant increase in plasma insulin levels and exhibited hypotriglyceridemic effect.
