Extract of Acanthopanax senticosus has recently been demonstrated to possess significant antidiabetic potential, in accordance with the traditional use of this plant as an antidiabetic natural health product. The present study evaluated the effects of fermented extract (FE) of this plant on glucose-stimulated insulin secretion, glucose uptake, and streptozotocin-induced type 1 diabetes model. A 3 h pretreatment with FE prevented $IL-1{\beta}$ and $IFN-{\gamma}$ toxicity in isolated rat islets. However, it did not affect insulin-stimulated glucose uptake in C2C12 myotubes. In addition, pretreatment of mice with FE blocked the destruction of streptozotocin-induced islets and the development of type 1 diabetes. FE reduced blood glucose level, increased insulin secretion, and improved glucose tolerance in streptozotocin-treated mice, whereas nonfermented extract (NFE) had moderate effects. Immunohistochemical staining for insulin clearly showed that pretreatment with FE blocked the STZ-induced islets destruction and restored the number of islet cells that secreted insulin to the level of the control. Although the active principles and their mechanisms of action remain to be identified, FE may nevertheless represent a novel complementary therapy and a source of novel therapeutic agents against type 1 diabetes mellitus.
This study was designed to examine the effects of fractions of ethanol extract of Benincasa hispida (wax gourd) on plasma glucose and lipid levels in streptozotocin (STZ) induced diabolic rats. sprague-Dawley rats were induced diabetes mellitus by STZ injection (45 mg/kg) into the tail vein and were divided into 5 groups: normal, STZ-control, three experimental diabetic groups. fractions of ethanol extract of Benincasa hispida were administered orally into the diabetic rats for 14 days. The food intake and body weight were monitored and plasma levels of glucose, cholesterol, HDL-cholesterol, triglyceride (TG) , free fatty acid (FFA) , and aminotransferase activity were analyzed. A significant lowering effects of plasma glucose levels were observed in the chloroform fraction and $H_2O$ fraction group compared to STZ-control group at 14 days. Administrations of each of the three fractions decreased plasma TG and FFA levels in diabetic rats. Activity of alanine aminotransferase (ALT) in $CHCl_3$ fraction and $H_2O$ fraction groups and aspartate aminotransferase (AST) in $H_2O$ fraction group significantly lower than STZ-control group. This study examined the effects of Bulnesia sarmienti ethanol extract on lipid metabolism in streptozotocin (STZ)-induced diabetic rats. Sprague Dawley rats were induced with diabetes mellitus by injecting STZ (45 mg/kg) into the tail vein, and were then divided into 5 groups: normal, STZ-control, and three experimental diabetic groups. The rats were fed a basal diet along with B. sarmienti ethanol extract for 8 weeks. According to the results, after 8 weeks of consuming the experimental diets, the body weights and food efficiency ratios of the B. sarmienti ethanol extract-fed groups were not different from those of the control group. However, the concentrations of plasma total-cholesterol, LDL-cholesterol, and triglycerides were significantly lower in the B. sarmienti ethanol extract groups as compared to those of the control group. Although a blood glucose control effect was not observed after the eight weeks of B. sarmienti ethanol extract treatment in this type-l diabetes model, it was discovered that the experimental diets were highly effective at lowering levels of serum lipids, including triglycerides, total-cholesterol, and LDL-cholesterol.
This investigation was conducted to assess the influence of Cirsium japonicum consumption on the plasma glucose and lipid profiles in streptozotocin (STZ)-induced diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats weighing 230 g by injection of STZ into the tail vein at 45 mg/kg body weight. The rats were randomly assigned into four groups: a normal and STZ-control fed an AIN-93 diet group, and diabetic groups whose diets were supplemented with 10% Cirsium japonicum powder containing leaf or root for four weeks. To observe the effects of Cirsium japonicum in the animal model, the levels of glucose, cholesterol, HDL-cholesterol, triglyceride and free fatty acid in the plasma and the levels of glycogen in the tissue were determined after four weeks. Treatment of STZ-induced diabetic rats with leaf consumption significantly lowered the diet intake when compared with STZ-control rats. The relative weights of the liver and kidney differed significantly between the normal and diabetic groups. The weights of the kidneys in the leaf group were significantly lower than those in the STZ-control group. The level of hematocrit was also significantly lower in diabetic rats whose diets were supplemented with leaves when compared to those of STZ-control rats. The plasma glucose level was found to be significantly lower in the leaf group than the STZ-control group. The effect was significant after 2 weeks. The HDL-cholesterol levels increased in all of the diabetic experimental groups when compared to the STZ-control group. These results suggest that supplementation with Circicum japonicum leaves induced considerable hypoglycemic effect in STZ-induced diabetic rats and that these leaves may be useful for the management of diabetes mellitus.
In order to study the effect of some oriental prescriptions for polydipsia on experimental diabetes mellitus and cataract, Cheongsimyeonja-eum, Kamijowiseungki-tang and Kikukjihwang-tang were administered to streptozotocin(STZ)-induced diabetic and galactose-induced cataractous rats. The increased level of serum glucose and urea nitrogen owing to 572 were significantly decreased by administration of Cheongsimyeonja-eum, Kamijowiseungki-tang and Kikukjihwang-tang. The increased level of serum triglyceride and total cholesterol owing to STZ were significantly decreased by administration of Kamijowiseungki-tang and Kikukjihwang-tang. Reduction of body weight owing to STZ was significantly recovered in 7th day since administration of Cheongsimyeonja-eum and Kamijowiseungki-tang. Weight of liver was significantly recovered by administration of Cheongsimyeonja-eum and Kamijowiseungki-tang and weight of kidney was significantly recovered only by administration of Kikukjihwang-tang. The polydipsia of water and food were significantly decreased by administration of Cheongsimyeonja-eum, Kamijowiseungki-tang and Kikukjihwang-tang. Onset of cataract in experimental group owing to galactose was not significantly different from control group.

SANGNYANGHYOLTANG(SYT) is one of the most important prescription that has been used in oriental medicine for diabetes mellitus. The sudy was done in order to elucidate the anti-diabetic effect of SYT. After pretreatment of SYT(1,000mg/kg) for 6 weeks, the effect of of SYT was prevented on serum liver function test and hepatic lipid peroxide content in rats i.v. injected with streptozotocin(STZ, 50mg/kg, tail vein) 5 weeks after pretreatment of SYT. The hepatic microsomal cytochrome P-450 and aniline hydroxylase were significantly decreased, and aminopyrine N-demethylase activity was significantly increased in SYT-STZ group as compared with control group. Changes in aldehyde oxidase, xanthine oxidase, superoxide dismutase, catalase, epoxide hydrolase, UDP-glucuronyltransferase and sulfotransferase activities were not significantly different in any of the group. The cytosolic glutathione S-transferase activity was significantly decreased in SYT-STZ group as compared with control group. The selenium-independent glutathione peroxidase was significantly increased in SYT-STZ group as compared with control group, but there was no significant difference in selenium-dependent glutathione peroxidase in any of the groups. The hepatic glutathione concentration was significantly increased in SYT-STZ group as compared with control group, and ${\gamma}-glutamylcystein$ synthetase and glutathione reductase activities were not significantly different in any of the groups. The hepatic lipid peroxide content, serum aminotransferase and sorbitol dehydrogenase activities were slightly decreased in significantly in SYT-STZ groups.
BACKGROUND/OBJECTIVES: The primary objective of the treatment of diabetes mellitus is the attainment of glycemic control. Hyperglycemia increases oxidative stress which contributes to the progression of diabetic complications. Thus, the purpose of this study was to investigate the hypoglycemic and antioxidant effects of Daraesoon (Actinidia arguta shoot) in animal models of diabetes mellitus. MATERIALS/METHODS: Rats with streptozotocin-induced diabetes received an oral administration of a starch solution (1 g/kg) either with or without a 70% ethanol extract of Daraesoon (400 mg/kg) or acarbose (40 mg/kg) after an overnight fast and their postprandial blood glucose levels were measured. Five-week-old C57BL/6J mice were fed either a basal or high-fat/high-sucrose (HFHS) diet with or without Daraesoon extract (0.4%) or acarbose (0.04%) for 12 weeks after 1 week of adaptation to determine the effects of the chronic consumption of Daraesoon on fasting hyperglycemia and antioxidant status. RESULTS: Compared to the control group, rats that received Daraesoon extract (400 mg/kg) or acarbose (40 mg/kg) exhibited a significant reduction in the area under the postprandial glucose response curve after the oral ingestion of starch. Additionally, the long-term consumption of Daraesoon extract or acarbose significantly decreased serum glucose and insulin levels as well as small intestinal maltase activity in HFHS-fed mice. Furthermore, the consumption of Daraesoon extract significantly reduced thiobarbituric acid reactive substances and increased glutathione levels in the livers of HFHS-fed mice compared to HFHS-fed mice that did not ingest Daraesoon. CONCLUSIONS: Daraesoon effectively suppressed postprandial hyperglycemia via the inhibition of ${\alpha}$-glucosidase in STZ-induced diabetic rats. Chronic consumption of Daraesoon alleviated fasting hyperglycemia and oxidative stress in mice fed a HFHS diet.
Purpose: The prevalence of diabetes mellitus (DM) continues to increase worldwide, and blood glucose control may reduce mortality from diabetic complications and healthcare costs. Mulberry twig (MT) has been used as a herbal medicine in Asia, and its antidiabetic efficacy has recently been reported, but research in this area is still limited. This study examined the antidiabetic effects of water extracts of MT in diabetic animals. Methods: Six weeks old male ICR mice were divided randomly into three groups; normal control (NC, n = 10), DM control (DC, n = 10), and MT (n = 10). Streptozotocin (STZ, 50 mg/kg/day) was injected intraperitoneally into mice in the DC and MT groups for 5 consecutive days. After 10 days of the last STZ injection, the mice in the MT group were administered orally with MT water extracts (5 g/kg body weight) for 16 days. Results: The MT water extracts ameliorated the swelling of the liver in the diabetic mice and reduced the elevated levels of fasting blood and plasma glucose, total cholesterol (T-CHO), low density lipoprotein-CHO, and the ratio of high density lipotrotein (HDL)-CHO/T-CHO. The liver triglyceride (TG) and glycogen contents were also significantly lower in the MT group mice than in the DC group. The small intestine disaccharidase activity was analyzed to understand the therapeutic effects and the mechanism of MT water extracts in diabetic animals. MT group mice showed reduced lactase and sucrase activity in the proximal part of the small intestine. On the other hand, body weight, plasma insulin, TG, HDL-CHO, and hepatic T-CHO levels were similar in the DC and MT groups. Conclusion: These results suggest that MT water extracts have antidiabetic effects and can be developed as a functional source to reduce the postprandial blood glucose or to prevent DM incidence.

This study has been carried out to investigate effect of Rehmanniae Radix (RR) and pear phenolic compound (PC) on the hyperglycemic mice induced with streptozotocin (STZ). For this purpose, male mice were fed with a 0.2 mL RR extract (S group) and the pear PC (90 mg/kg/day) dissolved in a 0.2 mL RR extract (SPC group) while the control group received the same commercial diet for 6 weeks. The blood glucose contents were examined from tail vein blood once a week for 6 weeks. Samples of pancreas removed after the experimental period were processed for the immunohistochemical identification of $\beta$-cells. The levels of serum glucose were decreased significalntly (p<0.05) in the S and SPC groups compared with the control group. The BUN and creatinine levels were significantly (p<0.05) decreased in SPC group compared with the control group. Intraperitoneal glucose tolerance tests peformed at 24 hours before that period revealed that glucose tolerances in S and SPC group were ameliorated. Immunohistochemical analyses of the pancreases revealed that a lot of insulin- positive $\beta$-cells were contained in a Langerhas's islets of S and SPC groups compared with the control group, and the number of Langerhas's islets were significalntly increased in S (p<0.01) and SPC (p<0.05) groups. These results suggest that RR extract and pear PC could recover the damages induced by STZ in the hyperglycemic mice.

Fermented red ginseng (FRG) was prepared by inoculating 0.1% Lactobacillus fermentum NUC-C1 and fermenting them at $40^{\circ}C$ for 12 hours. The ginsenoside contents of FRG were increased compared with those of red ginseng (RG). Moreover, the levels of the ginsenosides Rg2, Rg3, and Rh2 in FRG increased significantly. In an oral glucose tolerance test (OGTT), blood glucose levels were lower in animals fed with RG and FRG extracts than in normal controls. In particular, FRG extracts in OGTT were superior to RG extracts. The antidiabetic effects of FRG in streptozotocin (STZ)-induced diabetic rats were investigated. Rats were divided into four groups: normal control, diabetes mellitus (DM), FRG administered at 100 mg/kg, and FRG administered at 200 mg/kg groups. FRG extracts were orally administered to each treatment group for 3 weeks, and blood glucose, insulin, and lipid levels of each group were determined. Orally administered FRG extracts significantly reduced blood glucose levels and increased plasma insulin levels in diabetic rats. Additionally, the activities of disaccharidases, including sucrase, lactase, and maltase, were decreased significantly in the FRG groups. FRG groups also had reduced triglyceride and total cholesterol levels, compared with the DM group. These results suggest that FRG may have antidiabetic effects in STZ-induced diabetic rats.
