Streptococcus mutans의 자당 대사에 미치는 과당의 영향 (THE EFFECT OF FRUCTOSE ON THE METABOLISM OF SUCROSE BY STREPTOCOCCUS MUTANS)

  • Statement of problem: Streptococcus produces energy and forms extracellular polysaccharides by metabolizing sucrose. Insoluble glucan, a kind of extracellular polysaccharide, is the important material of dental plaque. Fructose affects the metabolism of sucrose. Purpose: The purpose of this study was to evaluate the effect of fructose on the metabolism of sucrose in Streptococcus mutans. Materials and methods: To determine the effect of fructose on the formation of artificial plaque by Streptococcus mutans Ingbritt, S. mutans and fructose were placed in beakers containing M17 broth and sucrose. The wires were hung on frameworks inserted into cork stoppers, and then immersed in each of the beakers. After the incubation with gentle shaking, each wire was weighed. To analyze the effect of fructose on the sucrose metabolism by S. mutans or glucosyltransferase, S. mutans and fructose were placed in M17 broth containing sucrose. After the incubation. the remaining sucrose and polymers were analysed by thin layer chromatography. Results: The following results were obtained; 1. When Streptococcus mutans was cultured in the media containing 3% sucrose for 8 hours, the mean weight of formed artificial plaque on the wires was $124.3{\pm}3.0mg$, whereas being reduced to $20.7{\pm}10.2mg$ in the media added with 3% sucrose and 4% fructose(p<0.05) 2. When the control containing glucose was added with sucrose, the optical density of Streptococcus mutans solution cultured for 24 hours was not increased compared with the control, while being increased by adding with fructose. 3. When Streptococcus mutans was incubated in the media added with sucrose and fructose for 8 hours, the number of viable cells was increased compared with the media added with sucrose. 4. The amount of remained sucrose was increased in Streptococcus mutans culture supernatant of media added with sucrose and fructose than with sucrose only. but the amount of produced insoluble glucan was decreased. 5. The amounts of remained sucrose and produced soluble glucan were increased in the culture of glucosyltransferase-contained media added with sucrose and fructose than with sucrose only, but the amount of produced insoluble glucan was decreased . Conclusion: These results indicated that the sucrose metabolism and the production of insoluble glucan were inhibited in Streptococcus mutans by adding fructose in the media containing sucrose.

치아우식경험과 Streptococcus mutans의 관련성 (Relationship between Streptococcus mutans levels and dental caries experience)

  • Objectives : The aim of the present study was to measure the level of Streptococcus mutans(S. mutans), the major causative agent in dental caries, and to examine the relationship between S. mutans level and dental caries experience. Methods : The present study was carried out in 41 university students. S.mutans plaque was detected in the saliva using Dentocult$^{(R)}$-SM and was measured in absorbance. The data were analyzed in the relevance for number of dental caries experience. Results : The group of S. mutans value has progressive caries, which was higher than that of non-suffering caries(p<0.05). More caries experience showed higher value of S. mutans(p>0.05). According to the relationship with DMFT, level of S. mutans and absorbance, correlation with the level of salivary S. mutans is shown statistically highly significant (p<0.01). The level of S. mutans in saliva and plaque and absorbance is statistically significant(p<0.05). Conclusions : The levels of S. mutans in both group of high caries experience and progressive caries was highly detected, continuous oral health management should be carried out to prevent the possible dental caries.

Streptomyces sp. M-20 균의 대사물에 의한 Streptococcus mutans의 Glucosyltransferase 활성 억제 효과 (Inhibitory Effect of Metabolites isolated from Streptococcus mutas sp. M-20 on Glucosyltransferase Activity from Streptococcus mutans)

  • Dental caries is one of the most common oral diseases in the world. Glucosyltransferase (Gtase) of Streptococcus mutans (S. mutans) plays an important role in the develo pment of dental caries. For the purpose to develop anti- caries, we examined the effect of metabolites isolated from Streptomyces sp. M-20 on Gtase and the growth of S. mutans. Streptomyces sp. M-20 isolated from Mongolian soil showed 95~96% sequence homology with that of Streptomyces lin- colnensis. The metabolites of Streptomyces sp. M-20 were partially purified by extraction with ethyl acetate, silica gel column chromatography and preparative TLC. Partially purified metabolite, red colored component (MR-20) in ethyl acetate fraction showed potent antibacterial activitiy against S. mutans and inhibitory activity against Gtase purified from S. mutans, while another isolated yellow component (MY-20) showed no activity against S. mutans. The inhibitory activity of MR-20 against Gtase was confirmed by activity staining on SDS-polyacrylamide gel electrophoresis. The concentration of MR-20 for 50% inhibition $(IC_{50})$ against Gtase activity was $60{\mu}g/ml$. These results suggest that MR-20 can be developed for antibacterial agent and anticaries.

마와 꿀풀 추출물에 의한 Streptococcus mutans의 산 생성 및 Glucosyltransferase 저해효과 (Glucosyltransferase Inhibition of Streptococcus mutans by Dioscorea batatas and Prunella vulgaris extract)

  • 마, 꿀풀 추출물의 용매 분획별 구강병인균에 대한 항균 및 항우식 효과를 알아본 결과 다음과 같은 결과를 얻었다. S. mutans에 대한 마, 꿀풀 헥산 추출물의 상대성장률과 산 생성 억제 및 GTase 활성 저해효과을 알아본 결과, 마 헥산 추출물의 0.25 mg/ml 농도에서 100%, 꿀풀헥산 추출물의 0.5 mg/ml 농도에서 100% 성장을 억제하였다. 따라서 농도가 증가함에 따라 성장이 억제되는 것을 볼 수 있었다. 2.0 mg/ml 농도에서 마가 pH 7.2, 꿀풀이 pH 7.0로 나타나 pH 5.6, pH 5.2인 대조군과 비교하여 볼 때 마, 꿀풀 헥산 추출물을 첨가물이 산 생성을 억제하여 중성으로 보여주고 있으며, GTase 활성 저해효과는 마와 꿀풀의 헥산 추출물의 2.0 mg/ml농도에서 35%, 25%로 각각 활성을 저해하였다.

천연에센셜오일의 Streptococcus mutans에 의한 치석형성 억제 활성을 이용한 구강관리제품 개발 가능성에 대한 융합연구 (Convergence research on the possibility of development of oral care products using the anti-plaque activity of natural essential oils against Streptococcus mutans)

  • 본 논문은 Lavender, Tea tree, Eucalyptus, Lemongrass 4종의 천연에센셜오일의 Streptpccous mutans 에 의한 치석형성억제 효과에 대하여 연구해 보고자 하였다. Streptpccous mutans 에 의한 치석형성 억제 활성 측정결과인 Lavender 에센셜 오일의 MAC (Minium anti-adhesive concentration, 최소부착저지 농도)가 1.0 % 이고 Tea tree 에센셜 오일, Eucaliptus 에센셜 오일, lemongrass 에센셜 오일의 MAC는 0.5 % 로 나타났다. 에센셜오일의 농도를 달리하여 Streptpccous mutans와 혼합한 배양액을 항온조에서 배양한 후 용액의 pH를 측정하여 최소부착 저지농도 이하에서 산의 생성으로 pH가 낮아진 것을 확인할 수 있었고, 이 결과는 에센셜 오일이 Streptpccous mutans 에 의한 산의 생성을 억제하는 효과가 있다는 것이다. 이러한 결과들로부터 천연에센셜오일 중 특히 Tea tree, Eucalyptus, Lemongrass는 치석생성을 억제하는 천연소재로 구강관리 제품개발에 활용할 수 있는 가능성을 확인할 수 있었다.

Monoclonal Antibody를 이용한 Streptococcus mutans 검출 방법의 임상적 적용에 관한 연구 (DETECTION SYSTEM OF STREPTOCOCCUS MUTANS IN SALIVA USING MONOCLONAL ANTIBODY)

  • Monoclonal antibody를 이용한 Saliva-$check^{TM}$ Mutans키트의 타액 Streptococcus mutans검출방법으로서의 활용도와 임상적 우식지수 및 기존의 세균배양방법과의 상관관계를 알아보기 위해 2008년 2월에서 5월 중 평촌키즈웰치과에 내원한 만 2세에서 만 8세 사이의 92명의 아동을 대상으로 Streptococcus mutans 검출검사를 실시하였으며 또한 우식에 영향을 미치는 다른 요인인 치면세균막 pH와 타액완충능력검사를 실시하여 다음과 같은 결론을 얻었다. 1. Saliva-$check^{TM}$ Mutans 검사결과 양성을 나타낸 아동은 27명으로 29.3%이었고, 음성을 나타낸 아동은 65명으로 70.65%이었다. 우식경험유치면률은 음성 아동 13.89%, 양성 아동 25.23%으로 나타났다. 2. Monoclonal antibody를 이용한 검사방법 인 Saliva-$check^{TM}$ Mutans와 기존의 세균 배양방법인 $Dentocult^{(R)}$-SM은 각각 상이한 검사방법을 이용함에도 불구하고 검사결과 높은 상관관계를 보였다(p<0.01). 3. Saliva-$check^{TM}$ Mutans검사와 치면세균막 pH검사와는 역상관관계를 나타내었으나(p<0.01),타액완충능 검사와는 상관관계가 나타나지 않았다(p>0.05). 이상의 결과로 보았을 때 Monoclonal antibody를 이용한 검사방법인 Saliva-$check^{TM}$ Mutans는 구강 내 Streptococcus mutans를 측정하는 방법으로 적당하며 또한 검사에 필요한 시간을 대폭 줄일 수 있고 사용방법도 매우 간편하게 개발되어 환자들에게 적용함에 있어 효과적이라고 사료되었다.

  • 관중의 근경, 감초의 뿌리, 고삼의 뿌리, 진피의 과피로 부터 에탄올로 추출물을 분리하였다. 이들 4종류의 에탄올 추출물을 이용하여 Streptococcus mutans에 대한 항균활성을 조사하였다. 그 결과 관중의 근경, 감초의 뿌리, 고삼의 뿌리는 Streptococcus mutans에 대하여 항균활성을 나타냈다. 그 중 관중과 고삼의 추출물 300ug/disk 농도는 Streptococcus mutans에 대한 뚜렷한 항균활성을 나타냈다.l 이 결과 관중의 추출물은 Streptococcus mutans에 대한 황균 활성물질로 잠재성이 있음을 알 수 있었다.

Streptococcus mutans와 Lactobacillus acidophilus 에 대한 Polylysine의 항균활성 (Antimicrobial Activity of Polylysine against Streptococcus mutans and lactobacillus acidophilus)

  • To evaluate antidental caries activity of polylysine cell growth and acid production of Streptococcus mutans and lactobacillus acidophilus were microbiologically monitored in anaerobic broth system containing various concentration of polylysine. The pH and heat stability of polylysine having antimicrobial activity were also examined. Two tested microbes were fairly well grown in broth containing polylysine 0.1mg/ml however inhibited at 1 and 2mg/ml of polylysine concentration. Especially lag times of Strep-tococcus mutans and Lactobacillus acidophilus were prolonged to about hour at 1.0 and 2.0 mg/ml of poly-lysine. acid production of Streptococcus mutans and lactobacillus acidophilus was also decreased by poly-lysine. Antimicrobial activity of polylysine was not affected by the change of pH and the heat treat-ment.

Streptococcus mutans NCTC 10449 에서 생산된 Glucosyltransferase 의 정제 및 특성 (Purification and Characterization of Glucosyltransferase from Streptococcus mutans)

  • Streptococcus mutans NCTC 10449 가 생산하는 Glucosyltransferase 를 순수 분리하고 그 특성을 조사하였다. 이 효소는 충치 형성 과정중 insoluble glucan 의 형성과 관련되어 있다. 이 효소의 분자량은 152,000 이며 최적 pH 와 온도는 각각 6.5 와 $35^{\circ}C$ 이며 중성이상의 pH 에서 산성에서보다 다소 안정하였다. 이 효소의 활성에 대한 Km 값은 48 mM 정도였고, 이 효소에 의해 생산된 insoluble glucan 은 타액으로 부터 분리된 균에 의해 생산된 insoluble glucan 보다 dextranase 에 의한 분해도가 높았다.

키토산을 첨가한 개상용 레진의 세균부착에 관한 연구 (ADHERENCE OF ORAL BACTERIA ON VARIOUS DENTURE RELINING MATERIALS WITH CHITOSAN)

  • The purpose of this study was to evaluate the rate of adherence of bacteria on various denture relining materials and to find out the effects of chitosan, when it was added to denture relining materials. Denture relining materials such as Tokuso rebase normal $set^{(R)}$, Mild $rebaron^{(R)}$, $Kooliner^{TM}$, and New $truliner^{TM}$ were used in this study. The adherence of Streptococcus mutans was studied on the surfaces in the denture relining materials with chitosan and in those without chitosan. When chitosan was added to M17 broth and MRS broth, the viable cell count of Streptococcus mutans was reduced. The viable cell count of Streptococcus mutans on the specimens decreased in the following order : Mild rebaron, Tokuso rebase normal set, and Newtruliner and Kooliner. The deture relining materials with chitosan showed a lower rate of adherence of Streptococcus mutans than those without chitosan.
