Strawberry mild yellow edge potexvirus from Strawberry in Korea

  • Symptoms induced in the leaves of strawberry (Fragaria x ananassa Duch.), 'Seolhyang' and 'Eyeberry', were mosaic, distortion and black colored edge on leaves at Nonsan area, one of the important production areas in Korea. Electron microscopy by quick-dip revealed the flexuous rod-shape particles having about 550-600 nm length. Cytoplasmic inclusion bodies composed of aggregated virus particles were observed frequently in mesophyll parenchyma and epidermal cells for the leaves of strawberry. The specific primers amplifying products of 635 bp and 729 bp were developed for RT-PCR detection of Strawberry mild yellow edge virus (SMYEV). Nucleotide identity of the CP gene of SMYEV was 92.8-99.2% with those of other SMYEV isolates from Gen-Bank database.

A successful regeneration from shoot tips of Fragaria x ananassa DUCH following cryopreservation by droplet-vitrification (작은방울유리화법을 이용한 딸기 생장점 초저온동결보존)

  • This study describes an efficient and widely applicable droplet-vitrification following cryopreservation for shoot tips of (Fragaria x ananassa DUCH. cvs. 'Derunoka' and 'Jumbo pure berry'. The shoot tips of strawberry were precultured in Murashige and Skoog (MS) liquid medium supplemented with sucrose (0.3-0.7M). Precultured explants were treated with loading solution (LS, C4) containing glycerol 17.5% and sucrose 17.5% for 40 min and exposed to dehydration solution (B1) containing 50% of glycerol and 50% of sucrose for 60 min at $25^{\circ}C$, and then transferred onto droplets containing $2.5{\mu}l$ PVS3 on sterilized aluminum foils ($4cm{\times}0.5cm$) prior to direct immersion in liquid nitrogen (LN) for 1 h. The highest regeneration rate (%) was obtained when shoot tips were precultured with treatment-2 (exposing of shoot tips to MS + 0.3M Sucrose for 30 h and then treated with MS+0.5 M sucrose for 16 h) at $25^{\circ}C$ in both the cultivars. The viability of cooled samples, followed by culturing on MS medium for 4 weeks was 77.8% and 60.0% for 'Derunoka' and 'Jumbo pure berry', respectively. This result shows droplet-vitrification would be a promising method for cryobanking strawberry germplasm.

Isolation of a cDNA Encoding a Chloroplast Triosephosphate Isomerase from Strawberry

  • A cDNA clone encoding chloroplast triosephosphate isomerase (TPI-cp) was isolated from strawberry fruit cDNA library. Sequence analyses indicated that the cDNA contains an open reading frame of 314 amino acids (33.5 kDa) composed of a transit peptide (59 amino acids) in amino terminal region and mature protein (255 amino acids). The existence of transit peptide in the deduced amino acid sequence implies that it encodes a chloroplast isoform. The protein sequence is more similar to other plant chloroplast isoforms than cytosolic isoforms. RNA blot analysis indicated that its expression is ubiquitous in examined five tissues, flowers, leaves, petioles, roots and fruits, and shows differential pattern according to fruit ripening. Genomic DNA blot analysis showed that TPI-cp is encoded by multiple genes in strawberry. Through sequence comparison and phylogenetic tree construction, TPI-cp is distinctively grouped into dicot and chloroplast isoforms.

Growth and Nutrient Uptake of 'Seolhyang' Strawberry (Fragaria × ananassa Duch) Responded to Elevated Nitrogen Concentrations in Nutrient Solution (질소 시비농도가 '설향' 딸기(Fragaria × ananassa Duch)의 생장과 무기원소 흡수에 미치는 영향)

  • This research tested five modified Hoagland solutions for proper fertility management of nitrogen in 'Seolhyang' strawberry cultivations. The changes of nutrient solutions were in total nitrogen concentrations ($meq{\cdot}L^{-1}$) such as 0 (${NO^-}_3$), 2.5 (${NO_3}^-$), 5 (${NO_3}^-$), 10 ($7.5{NO_3}^-$ + $2.5{NH_4}^+$) and 15 ($10{NO_3}^-$ + $5{NH_4}^+$). The fresh weight and $NO_3$-N concentrations in petiole sap responded to the elevated nitrogen concentrations in fertilizer solution in quadratic (y=7.10+2.668x-$0.115x^2$, $R^2=0.7983^{***}$) and linear (y=26.14+5.245x, $R^2=0.6083^{***}$), respectively. The dry weight and N content of the above ground plant tissue responded to the elevated nitrogen in quadratic (y=2.140+0.492x-$0.022x^2$, $R^2=0.6110^{***}$) and linear (y=0.569+0.033x, $R^2=0.6952^{***}$), respectively. In our experiment the solution with $5meq{\cdot}L^{-1}$ of ${NO_3}^-$ showed positive results in both dry and fresh weight productions. However, a future research about modification of this solution and growth and nutrient uptake response is necessary to achieve better growth of 'Seolhyang' strawberry.

A High-Efficiency Direct Somatic Embryogenesis System for Strawberry(Fragaria x ananassa Duch.) Cultivar Chandler

  • A high-efficiency, reproducible somatic embryogenesis system for strawberry cultivar Chandler was developed. Thirty-one somatic embryos per explant(max no.) were recorded in leaf discs which were cultured on medium containing MS salts+$B_5$ vitamins+2% glucose+4.0 mg $1^{-1}$TDZ(Thidiazuron) and incubated at $10{\pm}1^{\circ}C$ under darkness for one week followed by three weeks under 16-h photoperiod. The scanning electron microscopic(SEM) ontogeny revealed the normal development of somatic embryos from globular to heart-shaped and dissection microscopy from torpedo-shaped to cotyledonary-stage embryos. The maximum germination percentage of 48% could be obtained on MS medium containing kinetin(1.0 mg $1^{-1}$) and the maximum survival percentage(79%) of plantlets after four weeks was found to be in the mixture of vermiculite, peatmoss, and soilrite(1:1:1).

Authentication and classification of strawberry varieties by analysis of their leaves using near infrared spectroscopy.

  • It is well known now that near infrared spectroscopy (NIRS) is a fast, no destructive, and inexpensive analytical technique that could be used to classify, identify, and authenticate a wide range of foods and food items. Therefore, the main aims of this study were to provide a new insight into the authentication of five strawberry (Fragaria x ananassa) varieties and to correlate them with geographical zones and the propagating methods used. Three weeks plants of five different strawberry varieties (F. x ananassa Duch. cv Camarosa, Seascape, Chandler, F. Chiloensis, and F. Virginiana) were cultivated in vitro first and then transferred to pots with special soil, and grown in a greenhouse at CINVESTAV, all varieties were acquired from California (USA). After 18 months, ten leaves from each variety were collected. Transmission spectra from each leave were recorded over a range of 10, 000-4, 000 cm$-^{1}$, 32 scans of each strawberry leave were collected using a resolution of 4 cm$-^{1}$ with a Paragon IdentiCheck FT-NIR System Spectrometer. Triplicates of each strawberry leave were used. All spectra were analyzed using principal component analysis (PCA) and soft independent modeling class analogy (SIMCA). The optimum number of components to be used in the regression was automatically determined by the software. Camarosa was the only variety grown from the same shoot but propagated by a different method (direct or in vitro). Five different classes (varieties) or clusters were observed among samples, however, larger inter class distances were presented by the two wildtype samples (F. Chiloensis and F. Virginiana). Camarosa direct and Camarosa in vitro displayed a small overlapping region between them. On the other hand, Seascape variety presented the smallest rejection percentage among all varieties (more similarities with the rest of the samples). Therefore, it can be concluded that the application of NIRS technique allowed the authentication of all strawberry varieties and geographical origin as well. It was also possible to form subclasses of the same materials. The results presented here demonstrate that NIRS is a very powerful and promising analytical tool since all materials were authenticated and classified based on their variety, origin, and treatment. This is of a tremendous relevance since the variety and origin of a plant material can be established even before it gives its typical fruit or flower.

Humidification and Shading Affect Growth and Development of Cutting Propagated 'Maehyang' Strawberry (Fragaria × ananassa Duch.) at Propagation Stage (삽목번식 시 가습과 차광 처리에 따른 '매향' 딸기의 생육)

  • This study was conducted to examine the effect of humidification and shading during cutting propagation on growth and development of strawberry (Fragaria x ananassa Duch.) 'Maehyang' plants at a propagation stage. The runner cuttings were stuck on Nov. 23, 2017 in propagation benches set in a Venlo-type glasshouse. Four shading treatments, no shading (control, C), 55% shading with white lawn (W55), 55% black shading net (B55), or 100% black plastic film (B100) with either an intermittent fog system (H) or without fog system. The shading and fog systems were removed 2 weeks after sticking of strawberry cuttings. A nutrient solution for strawberry, which was developed by Yamazaki, was supplied once a day with electrical conductivity (EC) $1.6dS{\cdot}m^{-1}$ and pH 5.8. Growth parameters such as plant height, longest root, crown diameter, leaf chlorophyll, leaf area and fresh and dry weight were measured at 7 days and 26 days after sticking. There was no significant difference in growth of above-aerial part of strawberry. The overall growth of the strawberry roots was better grew by providing fog than that not provide fog. The root fresh weight and root dry weight after 26 days after sticking of strawberry cutting was the best in the treatment that provided fog system without shading (CH). The longest root after 26 days after sticking of strawberry cutting was the best in the treatments that provided fog system with either 55% white lawn (W55H) and 55% black shading net (B55H). These results suggest that morphogenesis of these plants were affected by humidification and shading types. In a broader perspective, these results can be used to optimize studies of other crops grown from cuttings.

Characterization of Symptom and Determination of Tissue Critical Concentration for Diagnostic Criteria in 'Maehyang' Strawberry (Fragaria$\times$ananassa Duch.) as Influenced by Magnesium Concentrations in the Fertigation Solution ('매향' 딸기의 마그네슘 영양진단을 위한 결핍증상 및 식물체 내 한계농도)

  • This study was carried out to investigate the effect of magnesium (Mg) concentrations in nutrient solution on growth of and nutrient uptake by 'Maehyang' strawberry. Tissue analysis based on dry weight and petiole sap were also conducted to determine the threshold level in plants when Mg deficiency disorders developed in strawberry plants. In the Mg deficient plants, the spotted yellowing or yellowing area developed on the interveinal area of mature leaves. The dark brown color also developed on the interveinal area of mature leaves with marginal browning or marginal necrosis. The response in dry weight production of 'Maehyang' strawberry to elevated Mg concentrations in nutrient solution was quadratic and the equation is y=6.84+1.7533x-$0.9278x^2$ $(R^2=0.1081^{***})$. But the Mg contents in tissue increased lineally with the equation of y=0.1764+0.1275x $(R^2=0.8307^{***})$. The trends of fresh weight production and Mg concentrations in petiole sap were also quadratic (y=24.127+7.3565x-$1.125x^2$, $R^2=0.2314^{^***}$) and linear (11.954+5.793x, $R^2=0.6869^{***}$), respectively. To prevent growth suppression, the Mg concentrations based on dry weight of above ground tissue and in petiole sap should be in the range of 0.30 to 0.65% and 19 to $40mg{\cdot}kg^{-1}$, respectively, fer the commercial production of 'Maehyang' strawberry.

  • Objective of this research was to investigate the effect of calcium concentrations in the fertilizer solution on growth and development of Ca deficiency in 'Maehyang' strawberry (Fragaria $\times$ ananassa Duch.). The margins of the youngest leaves were scorched and they developed a cupped shape as they were expanded in Ca deficient plants. The vein area of the youngest leaves became brown when the deficiency became severe. The quadratic responses were observed in dry weight production to elevated Ca concentrations in fertilizer solutions with the highest growth in 4.5 mM treatment. The regression equation was y=2.4026+1.0209x-$0.098x^2$ $(R^2=0.3546^{***})$. However, tissue Ca contents increased lineally as the Ca concentrations in fertilizer solutions were elevated (y=1.2108+0.1333x, $R^2=0.9189^{***}$). In changes of the fresh weight and Ca concentrations in petiole sap, fresh weight production showed quadratic responses to elevated Ca concentrations in fertilizer solution, but Ca concentration increased lineally. The equations in changes of fresh weight and Ca concentrations were y=9.273+4.882x-$0.4245x^2$ $(R^2=0.4935^{***})$ and y=52.311+3.2917x $(R^2=0.6918^{***})$, respectively. When the concentration of calcium at which plant growth was retarded by 10% is regarded as critical concentration level, the calcium contents based on dry weight of above ground plant tissue and in petiole sap should be in the range between 1.6 to 2.25% and 63 to $79mg{\cdot}kg^{-1}$, respectively.

Breeding of New Ever-bearing Strawberry "Doha" Variety

  • "Doha" is a new strawberry (Fragaria x ananassa Duch.) variety, which was released by the Highland Agriculture Research Institute in 2021. The "Doha" variety originates from a 2014 cross between "Saebong No. 3" and "Yeolha," both of which exhibited excellent ever-bearing characteristics, including continuous flowering and large fruits under long-day and high temperature conditions. This new cultivar was initially named "Saebong No. 13" after examining its characteristics and productivity during summer cultivation between 2015 and 2018. After regional adaptability tests, "Doha" was selected from "Saebong No. 13" as an elite cultivar. The general characteristics of "Doha" include spreading, elliptic leaves, and strong growth. The fruits are long and conical and of a red color. The plant height of "Doha" was similar to that of "Goha," but the number of leaves was lower. The number of flower clusters of "Doha" was 8.6, which was 2.8 fewer than that of the control variety, "Goha," with 11.4. The average fruit weight of "Doha" was 13.9 g, which was 4.9 g heavier than that of "Goha." The fruit hardness of "Doha" was 35.5 g·mm-2, which was 9.4 g·mm-2 harder than that of "Goha." The marketable yield of "Doha" was 26,971 kg·ha-1, 125% more than that of "Goha" with 21,479 kg·ha-1. The findings of this study suggest that "Doha" is a hard fruit and high-yielding variety of ever-bearing strawberries that could increase farming income when distributing to farmers.
