다양한 추출방법으로 조제된 스테비아 잎 추출물의 항산화 활성 (Antioxidant Activity of Stevia Leaf Extracts Prepared by Various Extraction Methods)

  • 본 연구는 추출처리 방법별 스테비아 추출물의 항산화능을 비교하기 위하여 수행되었다. 스테비아 추출물은 열수(HWE), 발효(FHWE) 및 감압(VE)추출의 처리과정을 통하여 추출되었다. DPPH 라디칼 소거능과 환원력(FRAP) 및 지방산화 억제능을 통하여 알아본 항산화능의 평가는 감압처리되어 추출된 스테비아 추출물(VE)의 경우 그 활성이 가장 높게 관찰되었으며, 3가지 추출물 모두, 동일 농도에서 stevia의 주요한 활성성분인 stevioside와 rebaudioside보다 항산화활성이 월등히 높게 관찰되었다. 이러한 항산화능의 증가에 관한 이유를 알아보기 위하여 총 phenolic compounds의 함량을 측정한 결과 감압(VE) 추출물에서 유의적으로 높게 관찰되었다. 따라서 감압(VE) 처리 방법은 항산화 활성이 증진된 스테비아 추출물을 얻기 위한 효과적인 처리공정이라 사료된다.

  • The result of the study about the cultivation, the constituents, the sweetenity, and the development of preparations of Stevia and Hydrangea are as follows: 1. Hydrangea thrived at $200{\sim}1,400m$ above the sea-level, and the good ones were brought up at $600{\sim}1,200m$. 2. The good ones which belonged to the genera of Hydrangea were produced in the damp, dark and cold place. 3. The growth and cultivated yield of Stevia was not significantly influenced by the acidity of the soil. 4. The best season was May and June for propagation of Stevia by cutting method. 5. The growth rate of Stevia and Hydrangea was really good in the sandy fertile soil, but in the mucotic soil it was not suitable. 6. The extraction and separation of phyllodulcin by solvent had many difficulties, because it was very soluble in water, ethanol, ether, petroleum ether, acetone and benzene, etc. 7. The solubility of stevioside on the solvent was as follows: It was very soluble in water and methanol, slightly soluble in ethanol and acetone, and insoluble in ether, petroleum ether and chloroform. 8. The alkaloid reaction by Mayer reagent in Hydrangea extract was positive. 9. The ashification rate of Stevia was 8.66% to 8.72% and that of Hydrangea 17.02% to 17.04%. 10. The tannin of Stevia leaf was 7.80% to 7.88% and its of Hydrangea decreased 9.46% to 6.08% by fermentation. 11. The percent rates in minimum concentration-occurring sweetness in sugar, glycyrrhiza methanol extract, Hydrangea water extract, Stevia leaf water extract, decoction of Stevia's leaf, decoction of Stevia's stem were as follows: 1.2, 0.1, 0.1, 0.2, 0.4, 0.01, 0.1, 0.6. and sweetenity ratios of those were 1, 12, 12, 6, 3, 64, 12, 2, 12. It was very meaningful to develop preparations of stevia as stevioside, micronized powder, water extract, methanol extract and compound teas and in Hydrangea, water extract, methanol extract, single tea, and compound teas were less meaningful. 13. The genera of Hydrangea which is natural species in Korea was positive in phyllodulcin-identification test, but it was not available to make the sweet tea because of having a little content.

스테비아(Stevia. Rebaudian Bertoni)추출물 보강이 만성 알코올 섭취 흰쥐의 지질대사 및 간기능에 미치는 영향 (The Effect of Stevia (Stevia. Rebaudian Bertoni) Extract Supplementation on Lipid Metabolism and Liver Function of Rats Administered with Ethanol)

  • To investigate the effects of the supplementation of Stevia. Rebaudian Bertoni on serum and hepatic lipid levels and enzyme activities in rat administered with ethanol chronically. Sprague-Dawley male rats were orally treated with either an AIN-76 diet(C, control), a control diet plus ethanol(E, 4g/kg bw), E plus stevia extract-1(ES1, 1ml/kg bw), or E plus stevia extract-2(ES2, 2ml/kg bw) for 7 weeks. Serum triglyceride levels were increased in the E group and were decreased in the ES 1 group. Liver triglyceride levels were decreased significantly in the ES2 groups and Total-cholesterol were decreased in the ES1, ES2 groups compared with the E group. Liver $\gamma-GTP$ levels were decreased significantly in the ES1, ES2 groups compared with the E group. In addition, we have evaluated the serum or liver carnitine levels in those groups. Liver TCNE levels were increased significantly in the ES1, ES2 compared with the E group. These results may suggest that supplementation of Stevia. Rebaudian Bertoni has effects on the recovery of chronic ethanol-related diseases.

Antioxidant and Antimelanogenic Effects of Stevia rebaudiana Flower Extract

  • Stevia rebaudiana (Asteraceae), a perennial plant, has been used as a low-calorie sweetener and is being developed as a therapeutic agent for diabetes, hypertension, myocardial diseases, and microbial infections. Despite the common use of its leaves and stem, the bioavailability of the components present in S. rebaudiana flowers, when used as ingredients of cosmetics, has not been well investigated. Herein, we investigated the antioxidative and antimelanogenic effects of an aqueous extract of S. rebaudiana flowers (Stevia-F). Total flavonoid and phenolic content in Stevia-F were determined to be $8.64{\pm}0.23mg$ of quercetin equivalents/100 g and $631.5{\pm}2.01mg$ of gallic acid equivalents/100 g, respectively. The $IC_{50}$ values of Stevia-F for reducing power, and 2,2-diphenyl-1-picryl-hydrazyl-hydrate radical, hydrogen peroxide, and nitric oxide scavenging activities were 5541.96, 131.39, 466.34, and $10.44{\mu}g/mL$, respectively. Stevia-F showed inhibitory effects on the tyrosinase ($IC_{50}=134.74{\mu}g/mL$) and ${\alpha}$-glucosidase ($IC_{50}=114.81{\mu}g/mL$) activities. No significant cytotoxicity of Stevia-F was observed in B16F10 cells, treated with up to $100{\mu}g/mL$ of the extract for 24 and 48 h (p > 0.05). Stevia-F ($1-100{\mu}g/mL$) suppressed ${\alpha}$-melanocyte stimulating hormone-induced melanin production in B16F10 cells (p < 0.05) and also inhibited the cellular tyrosinase activity (p < 0.05). Overall, our results show that Stevia-F possesses potential for inhibiting tyrosinase and ${\alpha}$-glucosidase activities and has significant antioxidant capacity. The antimelanogenic potential of Stevia-F should extend the usage of S. rebaudiana flowers in the development of skin-whitening products.

스테비아 추출물 발효액에서 분리된 유효 미생물들의 동정 및 항미생물 활성 (Identification of Effective Microorganisms Isolated from Fermented Stevia Extract and Their Antimicrobial Activity)

  • 스테비아(Stevia rebaudiana Bertoni)는 남미가 원산지인 국화과의 감미식물이다. 스테비아추출물 발효액으로부터 세균 23균주와 효모 10균주를 분리하여 일반적인 분류학적 방법과 분자유전학적 방법으로 동정하였다. 스테비아추출물 발효액에서 분리된 균주들은 5속 10종의 세균과 1종의 효모균에 속하는 것으로 나타났다.16S와 18S rDNA 염기서열 분석에 근거하여 계통수를 작성하였다. 분리균들의 항미생물 활성을 여러 세균과 식물병원성 진균들에 대해 조사하였다. 분리균들 중에서는 Lactobacillus paracasei SB13이 광범위한 세균들에 대해서 강한 항균활성을 나타내었다. 이들 결과는 토양개량을 위한 친환경적 미생물 제제를 개발하는데 도움을 줄 것이다.

스테비아 잎추출물을 첨가한 그릭스타일 요거트의 발효특성 및 저장기간 중 품질특성 (Fermentation and Quality Characteristics during the Storage of Greek-style Yogurt Supplemented with Stevia Leaf Extract)

  • In this study, the fermentation and quality characteristics of Greek-style yogurt supplemented with stevia leaf extract during storage were examined. Stevia leaf extract was extracted from dried leaves with hot water and 70% ethanol. The pH decreased significantly in all samples over time during fermentation (p<0.05). In particular, the sharpest decrease was detected in the group in which the hot water extract of stevia was added. Acidity increased significantly over time during fermentation (p<0.05). Lactic acid bacteria counts increased in all samples up to 9 h after the start of fermentation, but decreased after 12 h. The sugar content decreased over time, and there were no major differences between samples. Based on the results of a sensory evaluation, the group treated with stevia extract was rated high in sweetness, but was rated somewhat low in color and flavor. Accordingly, it was rated low in overall acceptability. In a storability experiment, as the days of storage increased, the pH gradually decreased, and acidity showed a tendency to increase. The sugar content showed a tendency to decrease and then increase.

신(新) 감미(甘味) 자원(資源) Stevioside의 안전성(安全性)에 관(關)한 연구(硏究) (A Study on the Safety of Stevioside as a New Sweetening Source)

  • 감미(甘味) 식물(植物) stevia의 추출물(抽出物)(주(主) 감미(甘味) 성분(成分),stevioside)을 흰쥐에 투여함으로써 본 감미 성분의 안전성(安全性)을 검토하였다. 복강내(腹腔內) 주사(注射)로 치사(致死)를 $LD_{50}$를 측정한 결과 추출물(抽出物)로 보아서는 3,400 mg/Kg이고 stevioside로 보면 1,700 mg/Kg이었으며, stevia 추출물의 56일간에 걸친 다량(多量) 구광(口腔) 투여(投與)$(2.5{\sim}5.0\;g/Kg{\cdot}day)$에도 실험 동물의 성장(成長)에 아무 장애(障碍)가 초래되지 않았다. 56일간 구강 투여 후의 전혈(全血) 검사(檢査)(RBC, WBC, Hb 및 Hct), 혈청(血淸) 성분(成分) 분석(分析)(총 단백질, 혈당(血糖), 콜레스테롤 및, GOT 등 17개 조사 항목) 및 간(肝) 조직(組織) 검사(檢査)(간 세포 핵퇴폐(核頹廢)), Kupffer 세포의 증식(增殖), 문맥성(門脈城) 섬유화(纖維化) 등 11개 소견(所見))는 총 33개 항목인데, 그 중 lactate dehydrogenase (LDH) 활성을 제외하고는 모두 대조군(對照群)과 시험군(試驗群) 사이에 유의차(有意差)를 볼수 없었다. 상기한 실험 결과에 입각하여 stevia 추출물 및 stevioside는 흰쥐에 대하여 급성(急性) 및 아급성(亞急性) 독성(毒性)을 발현(發現)하지 아니하는 것으로 사료(思料)된다.

  • 본 연구는 열수 추출과 에탄올 추출법으로 추출한 스테비아 추출물을 그릭 요거트에 당으로 0.5%, 1% 비율로 첨가하여 DPPH, ABTS 라디칼 소거능과 FRAP 측정법을 통해 환원력을 측정하여 항산화능을 비교하고 총 폴리페놀 함량을 측정하기 위하여 수행되었다. 대조군에 비해 스테비아 잎 추출물을 첨가하였을 때 항산화능이 증가하였고 열수 추출물보다 에탄올 추출물을 첨가하였을 때 더 항산화능이 증가하는 경향을 보였으며, 총 폴리페놀 함량 또한 증가하는 경향을 보였다. 따라서 그릭 요거트 에스테비아 잎 추출물을 첨가하여 항산화 기능성이 함유된 발효유로서 에탄올 추출법이 효과적이라 사료된다.

스테비아 추출액을 엽면시비 하여 재배한 딸기의 품질특성 (Quality Characteristics of Strawberry Cultivated with Foliar Application of Stevia (Stevia rebaudiana B.) Extract)

  • 본 연구에서는 스테비아 추출액의 엽면시비가 딸기의 품질에 미치는 영향을 조사하였다. 당도는 무처리 딸기와 스테비아 추출액 엽면시비 딸기가 각각 $7.32^{\circ}Brix,\; 7.93^{\circ}Brix$로 스테비아 추출액을 엽면시비 한 딸기가 높게 나타났으며 산도는 무처리 딸기(0.96%)가 스테비아 추출액 엽면시비 딸기(0.83%)보다 높았다. 저장 중 조직의 강도는 감소하였으며 무처리 딸기와 스테비아 추출액 엽면시비 딸기의 유의적 차이는 없었다. 또한 저장 중 L값, a값, b값은 감소하는 경향을 보였으나 스테비아 추출액 엽면시비 딸기가 무처리 딸기보다 높게 나타났다. 유리당은 fructose, glucose, sucrose로 구성되어 있었으며 스테비아 추출액을 엽면시비 한 딸기의 유리당 함량이 전반적으로 무처리 딸기보다 높았다. 유기산의 함량은 citric acid, malic acid, oxalic acid, succinic acid, fumaric acid 순이었으며 무처리 딸기가 스테비아 추출액 엽면시비 딸기보다 높은 함량을 나타냈다. 관능검사결과 색상과 향은 무처리 딸기와 스테비아 추출액 엽면시비 딸기가 유의적 차이가 없었으나 단맛, 신맛, 전반적인 기호도는 스테비아 추출액을 엽면시비 한 딸기의 선호도가 높은 것으로 나타났다.

Stevia rebaudiana의 항산화 효과 (Anti-oxidant Effect on Stevia rebaudiana)

  • Stevia rebaudiana is a traditional herb used as a sweetener in Brazil and Paraguay as well as Korea and China. This study investigated the efficacy of Stevia rebaudiana methanol extract (SRE) to protect cells against the mitochondrial dysfunction and apoptosis in hepatocyte. To determine the effects of SRE on oxidative stress, we used the human derived hepatocyte cell line, HepG2 cell. Treatment of arachidonic acid (AA)+iron in HepG2 cells synergistically amplified cytotoxicity, as indicated by the excess reactive oxygen species (ROS) and mitochondrial permeability transition by fluorescence activated cell sorter (FACS) and immunoblot analysis. Treatment with SRE protected hepatocytes from AA+iron-induced cellular toxicity, as shown by alterations in the protein levels related with cell viability such as procaspase-3. SRE also prevented the mitochondrial dysfunction induced by AA+iron, and showed anti-oxidant effects as inhibition of $H_2O_2$ production and GSH depletion. Moreover, we measured the effects of SRE on AMP-activated protein kinase (AMPK), a key regulator in determining cell survival or death. Acetyl-CoA Carboxylase (ACC), a direct downstream target of AMPK. SRE increased phosphorylation of ACC, and prevented the inhibition of ACC phosphorylation by AA+iron. These results indicated that SRE has the ability to protect cells against AA+iron-induced $H_2O_2$ production and mitochondrial impairment, which may be mediated with AMPK-ACC pathway.
