Incidence of Plant-Parasitic Nematodes in Strawberry Nursery and Nematode Dispersal by Daughter Plant (딸기 육묘장 토양 내 식물기생선충의 감염현황과 자묘를 통한 선충의 분산)

  • To survey the incidence of plant-parasitic nematode in strawberry nursery, 117 soil samples were collected from main producing area of strawberry nursery plants in 2016. The incidences of two major problematic nematodes such as root-lesion nematodes (Pratylenchus spp.) and root-knot nematodes (Meloidogyne spp.) were 11% and 3%, respectively. Strawberry nurseries using upland soil either in fields or in plastic-houses showed were higher infection rates of Pratylenchus spp. or Meloidogyne spp. than others. On the other hand, a strawberry nursery using pots filled with bed soil in plastic-house was the lowest infection rate with 0%. Phylogenetic relationships and distance analysis were performed for identification of Pratylenchus spp. and Meloidogyne spp. detected from strawberry nurseries. As the results, they were identified as Pratylenchus penetrans, Pratylenchus vulnus and Meloidogyne hapla. Meanwhile, when nursery plants of strawberry infested with P. vulnus transplanted to pots filled with sterilized soil, P. vulnus was detected from soil in pots as 33% frequency. Thus, P. vulnus can be spread through nursery plants of strawberry infested with that. These studies suggested that Pratylenchus spp. and Meloidogyne spp. were ploblematic nematodes in major areas of strawberry nursery plants and P. vulnus could be spread through nursery plants of strawberry.

Emission of NO2 Gas Causing Damage to Plants in an Acid Soil under Conditions Favorable for Denitrification

  • Nitrogen dioxide ($NO_2$) gas damage on vegetable crops commonly occurs in plastic film houses where relatively large amounts of $NO_3{^-}$ are applied in acid soils. In acid soils, $HNO_2$ can be formed from the $NO_2{^-}$ accumulated during denitrification, and $NO_2$ can be evolved from the chemical self-decomposition of $HNO_2$. In this study, $NO_2$ gas production and its detrimental effects on plants were investigated in soils of various conditions to elucidate the mechanisms involved in the gas production. A silty loam soil was amended with $NO_3{^-}$ (500 mg N $kg^{-1}$) and glucose, and pH and moisture of the soil were adjusted respectively to 5.0 and 34.6% water holding capacity (WHC) with 0.01 M phosphate buffer. The soil was placed in a 0.5-L glass jar with strawberry leaf or $NO_2$ gas absorption badge in air space of the jar, and the jar was incubated at $30^{\circ}C$. After 4-5 days of incubation, dark burning was observed along the outside edge of strawberry leaf and $NO_2$ production was confirmed in the air space of jar. However, when the soil was sterilized, $NO_2$ emission was minimal and any visible damage was not found in strawberry leaf. In the soil where water or $NO_3{^-}$ content was reduced to 17.3% WHC or 250 mg N $kg^{-1}$, $NO_2$ production was greatly reduced and toxicity symptom was not found in strawberry leaf. Also in the soil where glucose was not amended, $NO_2$ production was significantly reduced. In soil with pH of 6.5, $NO_2$ was evolved to the level causing damage to strawberry leaf when the soil conditions were favorable for denitrification. However, compared to the soil of pH 5.0, the $NO_2$ production and its damage to plants were much less serious in pH 6.5. Therefore, the production of $NO_2$ damaging plants might be occurred in acid soils when the conditions are favorable for denitrification.

Inhibitory Effect of Dioscorea Bulbifera MeOH Extract on Pro-inflammatory Mediator In Vitro and In Vivo (황약자(黃藥子) 메탄올 추출물의 염증억제 효과)

  • Dioscorea bulbifera is one of the traditional medicinal herb. It commonly used in the treatment of hematemesis, epistaxis, tuberculous cervical lymphadenitis, laryngitis, acute infectious disease in East Asia. In the present study, we have demonstrated the anti-inflammatory effects of Dioscorea bulbifera MeOH extract (DBME) in macrophage cell line. To investigate mechanism of the anti-inflammatory activity, we examined the effects of the lipopolysaccaride (LPS)-induced production of nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), pro-inflammatory cytokines and expression of inducible NO synthase (iNOS), cyclooxygenase-2 (COX-2), p-inhibitory ${\kappa}B{\alpha}$ (p-$I{\kappa}B{\alpha}$), and nuclear factor-${\kappa}B$ (NF-${\kappa}B$) in a murine macrophage cell line RAW 264.7. The RAW 264.7 cells were cultured in DMEM + serum medium for 24 hrs. After serum starvation for 24 hrs, the cells were treated with DBME 0.03, 0.10, 0.30 mg/$m{\ell}$ for 1 h, followed by stimulation with LPS (1 ${\mu}g/m{\ell}$) for activation of immune response. After treatment, cell viability was measured by MTT assay, and NO production was monitored by measuring the nitrite content in culture medium. The protein band of iNOS, COX-2, p-$I{\kappa}B{\alpha}$, and NF-${\kappa}B$ was determined by immunoblot analysis and levels of cytokine were analyzed by sandwich immunoassays. There were three experimental groups: carrageenan, DBME 0.3, 1.0 g/kg. Rats were administrated either carrageenan (40% PEG) or carrageenan + DBME (0.3, 1.0 g/kg body weight) for 4 days (p.o.). To induce acute paw edema, rats were injected 1% carrageenan (100 ${\mu}{\ell}$/rat, dissolved in sterilized saline). The effect of DBME in the carrageenan-induced rat paw edema. As results, DBME has an inhibitory effect on the production of NO, PGE2, TNF-${\alpha}$, IL-$1{\beta}$ and IL-6 and on the expression of iNOS, COX-2, p-$I{\kappa}B{\alpha}$ and translocation of NF-${\kappa}B$ to nuclear from cytosol. In addition, DBME effectively inhibited the increases of paw edema induced by carrageenan treatment in vivo. These results suggest that DBME can inhibit production of pro-inflammatory mediators and might be a useful source for treatment of acute inflammatory disease.

  • FAO standard for heat sterilization of wood, International standards for phytosanitary measures (ISPM) No.15, must meet heat-treated wood core temperature to be higher than $56^{\circ}C$ and keep the temperature for more than 30 minutes. This study was carried out to analyze the heat treatment characteristics of domestic pinewood sterilized with the FAO standard. To enhance the effectiveness of heat treatment process in mountainous district energy consumption and time required to reach target temperature were evaluated at various temperature and relative humidity conditions and moisture contents of wood. Heat-treatment of high temperature and high humidity reduced the required heating time. Lower humidity levels at same temperature reduced energy consumption per unit time. However, lower humidity levels could not reduce total energy consumption greatly because longer treatment time was required at that condition. It is necessary to estimate energy consumption and predict treatment time in dynamic heating and cooling situations, because it frequently happens not to meet optimum treatment condition due to poor surrounding climates and operation performance of heat treatment facility in real field.

  • We investigated springs in Seoul in 2009 to know the change of water quality according to storage method of spring water, the concentration of chemical compounds and their correlation. Even spring water that originally satisfied national standard for drinking water could be exceeded national standard for drinking water by storage method such as storage bottle, temperature and period; especially used PET bottles could affect the increase of total colony counts. Therefore, spring water is desirable to be consumed on the spot, or to be stored in sterilized bottles in refrigerator rather than room temperature at home, and also to be consumed shortly not exceeding 24 hours. Total colony counts, coliform, yersinia, $F^-$, $Cl^-$, $NO_3^--N$, hardness, total Solids, pH, color and Al were exceeded national standard for drinking water at some springs. The result of correlation analysis shows that hardness and total solids, which are caused by several ionic compounds, had relatively high correlations with other chemical compounds.

Detection Rate of Periodontopathogens Associated with Cardiovascular Diseases in Denture. (의치 표면에서 심혈관질환과 관련된 치주질환 원인 세균의 검출)

  • The aim of this study is to investigate the detection rate of putative periodontopathogens, Porphyromonas gingivalis, Tannerella forsythia, and Actiobacillus actinomycetemcomitans, related to cardiovascular diseases(CVD). Plaques were sampled from 15 subjects (4 sites of denture base and/or tooth) with sterilized explorers and were transported in IX PBS. The detection of periodontopathogens was performed by polymerase chain reaction with species-specific primers based on 16S rDNA. The PCR products were cloned into pGEM-T easy vector and its nucleotides were sequenced in order to confirm the specificity. Our data showed that the detection rate of P. gingivalis and T forsythia in denture base of edentulous patients was 25% and 75%, respectively. And the detection rate of P. gingivalis and T.forsythia in denture base of patient having one more tooth was 91%. The results indicate that plaque of denture base may serve as reservoirs of oral bacteria related to CVD.

  • Effects of irradiation on the physicochmical properties and its quality of powdered hot pepper paste which was used as a minor matrial of meat products, were investigated during the three months storage. Proximate components of powdered hot pepper paste were not remarkably changed according to the irradiation dose except the slight increase of total and reducing sugars. Capsanthin was insignificantly decreased by the increase of irradiation dose. Color difference according to the irradiation dose could not be distinguished by naked eye but was slightly changed by mechanical measurement. Total bacteria and E.coli were sterilized by the irradiation of 10kGy and 5kGy, respectively and no microorganisms were grown up after three months storage at $37^{\circ}C$. Sterilization of powered hot pepper paste by irradiation was expected to be superior to the traditional methods in the wholsomeness and sterilizing efficacy.

Efficacy of Electrolyzed Water and Aqueous Chlorine Dioxide for Reducing Pathogenic Microorganism on Chinese Cabbage (전기분해수 및 이산화염소수 처리에 따른 배추의 미생물 제어 효과)

  • This study evaluated the efficacy of strong acidic electrolyzed water (SAcEW), low alkaline electrolyzed water (LAlEW) and aqueous chlorine dioxide (ACD) for reducing pathogenic bacteria($Escherichia$ $coli$, $Bacillus$ $cereus$, $Salmonella$ Typhimurium, $Stapylococcus$ $aureus$) on Chinese cabbage. Artificially inoculated Chinese cabbage was immersed for 1, 5 and 10 min with TW, NaClO, EW and ACD. Generally, leaves showed more effective reduction than stems. Regarding the inhibitory effect, ACD treatment showed the highest effects rather than other treatments. When Chinese cabbage was immersed for 3 min in sterilized water, it was reduced to a minimum of 1.33 log CFU/g at LAlEW and a maximum of 4.70 log CFU/g at ACD. Compared to NaClO, ACD and LAlEW which showed a reduction of 3.2 log CFU/g ($Sal.$ Typhimurium) and 2.7 log CFU/g($B.$ $cereus$), respectively. Furthermore, the others had similar inhibitory effects compared to NaClO.

Physicochemical Characteristics of Cheonnyuncho Fruit (Opuntia humifusa) Fermented by Leuconostoc mesenteroides SM (Leuconostoc mesenteroides SM을 이용한 천년초(Opuntia humifusa) 열매 발효물의 물리·화학적 특성)

  • Functional properties of Cheonnyuncho fruit paste (CFP) were enhanced by fermentation, using Leuconostoc mesenteroides SM. The stability of CFP pigment was maintained and microorganisms in crude CFP were sterilized by heating at $80^{\circ}C$ for 30 min. The production of dextran in CFP was increased by increasing the fermentation time and sugar content, resulting in an increase of consistency. The CFP fermented with 20% sucrose contained 4.8% mucilage after fermentation for 2 days. In particular, the production of mucilage was greatly enhanced by fortification with 3% sodium citrate in CFP, resulting in mucilage production of 7%. In addition, the viscous modulus of fermented CFP showed a greater increase than that of the elastic modulus. Thus, it was concluded that CFP fermented by L. mesenteroides SM can be fortified with functional ingredients, such as mucilage and probiotics, with enhanced rheological properties. Therefore fermented CFP can be utilized as the functional ingredients for functional foods.

Sensory Evaluation of Various Gouda Cheeses Produced from Raw Milk (비살균 원유로 제조된 다양한 Gouda 치즈의 관능평가)

  • Since 2018, the Korean government has permitted the production and sale of any cheese made from raw milk and aged for at least 60 days. The present study aimed to investigate the sensory characteristics of various types of Gouda cheese produced from raw milk during 60-day aging. The average pH of the raw milk used in this experiment was $6.7{\pm}0.4$, similar to that of the sterilized milk. The pH of raw-milk Gouda cheese was $5.2{\pm}0.5$ a day after the 60-day aging period, wherein the pH was $5.5{\pm}0.3$. In total, 5 samples were used for sensory evaluation in this experiment: Cheeses 1~5. With respect to flavor, Cheeses 1 and 5 received the highest and lowest scores, respectively. Raw-milk Gouda cheese produced using Salmonella spp.-contaminated raw milk was not greatly influenced by flavor; however, it had a negative effect on its appearance. In the future, it is imperative to carry out sensory evaluation of cheese produced with raw milk contaminated with various food-contaminating bacteria.
