Effects of Postharvest Heat Treatment on Alleviation Chilling Injury and Improvement Storability of Oriental Melon (수확 후 고온처리가 참외의 저온장해 완화와 저장성 향상에 미치는 영향)

  • Oriental melons hold at $38^{\circ}C$ for 48 hours before storage increased their soluble solid, titratable acidity, vitamin C contents and ${\alpha}$-tocopherol activity. These heat treated oriental melons maintained lower production of carbon dioxide, ethylene and acetaldehyde and loss of fresh weight than untreated them, called control, during $3^{\circ}C$ MA storage. After 39 days in storage, the last day of storage, visual quality and internal quality, such as firmness, soluble solid, titratable acidity, vitamin C and ${\alpha}$-tocopherol activity, showed higher in heat treated oriental melons. Especially, Ion leakage of flesh, index of chilling injury, increased remarkably in control, so that heat treatment had to alleviate chilling injury in oriental melon. Moreover, while Alternaria rot was shown in control plot after 25 days in $3^{\circ}C$ MA storage, oriental melons treated heat were not appeared any decomposition after 39 days in $3^{\circ}C$ MA storage. As storage life of oriental melon was calculated by regression equation between visual quality and days in storage, that was longer 8 days in heat treated than control. Consequently, heat treatment that was mild, $38^{\circ}C$ and long period, 48 hours, executed before storage, sterilized surface alleviated chilling injury and lengthened storage life in oriental melons.

Cytotoxicity of Hyaluronic Acid Membrane Cross-linked with Lactide (락타이드로 가교시킨 히아루론산 막의 세포독성)

  • The biodegradable hyaluronic acid(HA) membranes cross-linked with lactide using the crosslinking agent, 1-ethyl-3(3-dimethyl aminopropyl) carbodiimide(EDC) were prepared as a potential biocompatible material for tissue engineering. HA membranes having different mechanical properties were synthesised by varying degree of the mole ratio of lactide to HA, EDC concentration, and crosslinking temperature. HA membranes were degradable in water solution and the degradation became slower with the increasing mole ratio of lactide to HA. HA membranes were sterilized using ethylene oxide gas and extracted with cell culture medium for 24 h at $37^{\circ}C$ and 200 rpm. Cytotoxicity of the extract was tested using NIH/3T3 mouse embryo fibroblast as a model cell. Growth inhibition was not observed in the extracts of HA membranes with the mole ratios of lactide to HA, 5 or 10, and 10% EDC concentration, however 11% of growth inhibition was observed in the extract with the mole ratio of 13. Growth inhibition was not observed in the extracts of HA membranes prepared with 5% EDC or 10% EDC and the mole ratio of lactide to HA, 10, however 12% of growth inhibition was observed in the extract with 20% EDC. Cytotoxicity was not observed in the extracts of HA membranes prepared at varying crosslinking temperatures, $15^{\circ}C,\;25^{\circ}C,\;and\;28^{\circ}C$ with the mole ratio of lactide to HA, 10 and 10% EDC.

Browning Characteristics of Ssamjang during Storage (저장기간에 따른 쌈장의 갈변 반응 특성)

  • Ssamjang samples were prepared by central composite design for five independent variables, Gochujang mash aging period $(X_1)$, Doenjang aging period $(X_2)$, Doenjang content $(X_3)$, sterilization temperature $(X_4)$ and storage temperature $(X_5)$. Browning characteristics of Ssamjang were analyzed such as water activity, free amino acids, free sugars and color during storage. Water activities of Ssamjang were ranged from 0.605 to 0.666 at the beginning of storage, and were shown to be the highest at the 8th week of storage and then decreased gradually. Among free amino acids the content of glutamic acid was the highest. Proline, leucine, phenylalanine, lysine and serine were slightly higher than the others. Larger amounts of free amino acids and free sugars were observed from the sample sterilized at $60^{\circ}C$ when compared to that at $70^{\circ}C$. Glucose, fructose and maltose as free sugars were identified from Ssamjang and glucose content was the largest among. During the storage, lightness $(L^*)$, redness $(a^*)$ and yellowness $(b^*)$ of Ssamjang were decreased and total color difference $({\Delta}E)$ was increased. The color changes in the sample surface were more affected by temperature of which storage temperature was more influenced than sterilization temperature.

  • This study was conducted to develop the method for the safe supply of Sujeonggwa (cinnamon flavored persimmon punch) in severe environments such as space, desert or deep sea, by the combined treatment of gamma irradiation with other food technologies. Commercially prepared Sujeonggwa powder could be sterilized at 4.5 kGy or above doses. However, sensory characteristics of gamma-irradiated Sujeonggwa decreased depending upon the dose. The combined treatment of vacuum packaging with the addition of vitamin C and cinnamic aldehyde in Sujeonggwa powder could minimize the change of sensory qualities induced by ionizing irradiation.

Preparation and Characterization of Canned Salmon Frame (연어 frame 통조림의 제조 및 특성)

  • This study was conducted to prepare canned salmon frame and to characterize its food components. In the preparation of high-quality canned foods, the boiling water generated in the pre-heating process should be removed, and then the pre-treated canned salmon frame should be sterilized for an $F_0$ value of 12 min. The proximate composition of the canned salmon frame prepared under optimal conditions (CSFP) was 58.4% moisture, 15.7% protein, 21.4% lipid, and 3.5% ash. Based on the results of volatile basic nitrogen and microbiological tests, the CSFP was acceptable. The sensory score for the color of CSFP was 4.1 points, which was higher than that of commercial canned salmon frame (CCSF). However, there were no significant differences in the sensory scores for flavor and taste between CSFP and CCSF. The total amino acid content of CSFP was 14.58 g/100 g, which was 4.9% lower than that of CCSF. The major amino acids in CSFP were aspartic acid (11.0%), glutamic acid (14.8%), and lysine (10.6%), which accounted for 36.4% of the total amino acid content. The CSFP was high in calcium and phosphorus, while it was low in magnesium and zinc. The major fatty acids in CSFP were 16:0 (15.2%), 18:1n-9 (17.0%), 18:2n-6 (16.7%), 20:5n-3 (9.3%), and 22:6n-3 (8.8%). Based on the results, CSFP is a high-quality canned food in terms of hygiene and nutrition.

The Frequency of Detecting Prevotella intermedia and Prevotella nigrescens in Korean Adult Periodontitis Patients (한국인 치주 감염 환자에서의 Prevotella intermedia와 Prevotella nigrescens의 발현빈도)

  • Prevotella intermedia has been implicated as a potent pathogen in many kinds of periodontal, pulpal and periapical diseases. However, it has been isolated from periodontally healthy adults and from edentulous children as well. The intraspecies heterogeneity of Prevotella intermedia has been demonstrated in early studies and finally Shah & Gharbia confirmed the existence of 2 DNA homology groups and proposed dividing Prevotella intermedia into 2 species, Prevotella intermedia and Prevotella nigrescens. This study was designed to examine the frequency of Prevotella intermedia and Prevotella nigrescens in diseased periodontal pockets and healthy gingival sulcus of Korean people by PCR based on 16s ribosomal DNA sequence. One hundred adults who had adult periodontitis but not taken any periodontal treatment or antibiotics during previous 6 months and 50 adults who had healthy periodontal tissue were selected for this study. The sulcular fluid was collected into VMGA by sterilized paper point and diluted to 1,000 times in anaerobic chamber. $100{\mu}{\ell}$ of sample was cultured in $37^{\circ}C$ for 10 days. Among the bacterial colonies, BPB were selected and cultured in BHI broth and then Prevotella intermedia was identified through Gram staining and biochemical test. Identified Prevotella intermedia was cultured again and centrifuged. DNA was extracted from the pellet using several reagents. PCR was performed by previously designed primer. The results were followed. 1. BPB were isolated from 39 of 100 samples of diseased periodontal pockets(39%). 2. Prevotella intermedia was identified from 24 of 39 BPB samples. 3. Among 24 Prevotella intermedia, 21 were confirmed as Prevotella inter - media(87.5) and 2 were confirmed as Prevotella nigrescens(8.33%). 4. BPB were isolated from 9 of 50 samples of periodontally healthy patients. Among them only two were identified as Prevotella intermedia, that is, one was confirmed as Prevotella intermedia and the other was Prevotella nigrescens.

In Vitro Study Evaluating the Antimicrobial Activity of Vancomycin-Impregnated Cement Stored at Room Temperature in Methicillin-Resistant Staphylococcus aureus (상온에서 보관한 반코마이신 함유 항생제 시멘트의 메티실린 내성 포도상구균에 대한 항균력 조사)

  • Purpose: Treatment of diabetic foot infection due to methicillin-resistant Staphylococcus aureus (MRSA) remains challenging. Applying vancomycin-impregnated cement is one of the best methods of treatment. Vancomycin-impregnated cement has been used worldwide; however, to date, there is a limited number of studies regarding its use. We evaluated the duration of antimicrobial activity of vancomycin-impregnated cement stored at room temperature after manufacturing. Materials and Methods: The vancomycin-impregnated cement was manufactured by mixing 1 g of vancomycin with 40 g of polymer and adding 17.90 g of liquid monomer. The cement dough was shaped into flat cylinders with diameter and height of 6 mm and 2 mm, respectively. Another cement of the same shape without mixing vancomycin was prepared as the negative control. All manufactured cements were sterilized with ethylene oxide gas and stored at room temperature. Each cement was placed on Mueller Hinton agar plate lawned with standard MRSA strain. Standard vancomycin disk and gentamicin disk were placed together. After 24 hours, the diameter of inhibition zone was measured, and if the diameter was less than 15 mm, vancomycin-impregnated cement was regarded as a loss of antimicrobial activity. The study was repeated every 2 weeks until vancomycin-impregnated cements lost their antimicrobial activity. Results: Vancomycin-impregnated cement stored for a duration of 16 weeks created a 14 mm inhibition zone, while vancomycin disk created a 15 mm inhibition zone. Vancomycin-impregnated cement stored for a duration of 17 weeks created 7 mm and 9 mm inhibition zones, while vancomycin disk created 16 mm and 15 mm inhibition zones, respectively. Conclusion: We found a decrease of antimicrobial activity in vancomycin-impregnated cements after 16 weeks. After 17 weeks, they showed definite loss of antimicrobial activity. Therefore, we recommend not using vancomycin-impregnated cement spacers that has been stored for more than 16 weeks at room temperature.

Development of Detachable IORT Table for Colorectal Cancer (장착-탈거 및 경사각 조절이 가능한 대장직장암의 수술 중 방사선 치료대의 개발)

  • In spite of remarkable improvement of surgical skills and anesthesia, local failure still occurred in 36-45$ \% $ of locally advanced colorectal cancer after curative resection with or without pre-or post-operative irradiation. Intraoperative radiation therapy(IORT) is the ideal modality which resectable lesions are removed surgically 3nd the remaining cancer nests are sterilized by irradiation during a surgical procedure. Therefore, the excellent local control without the damage of the adjacent normal tissues can be achieved. In IORT, judicious set up of the treatment cone on the treatment surface of the patient is required for accurate and homogenous dose distribution within treatment field, especially on the slopping surface of sacrum and pelvic sidewall which are the common sites of the local recurrence in rectal cancer. For this purpose, adequate co-ordination of gantry rotation and table tilting are essential. Adjusting gantry rotation is not difficult but tilting of the table is impossible inconventional treatment couch. Department of Therapeutic Radiology in Yeungnam University Medical Center developed the IORT table for colorectal cancer which is easy to set up and detach on the Linac treatment couch within 5 minutes. The range of tilting with head-up and head-down is about 30 degree which is efficient and easy-to-use, not only for IORT but also for colorectal surgery. So far, authors performed IORT with newly developed treatment table in 2 patients with rectal cancer and we found that this newly developed table could contribute in improving the dose distribution of IORT and surgical procedure for colorectal cancer.

Development of Reference Material Using Natural Seawater for Nutrient Analysis in Seawater (자연해수를 이용한 해수 중 영양염 분석용 표준물질 개발)

  • Dissolved nutrients in seawater are recognized as an essential biogeochemical factor for detecting global environmental changes. The importance of nutrient reference material for seawater has been increased greatly for the comparison of nutrient data, measured in different time and space in global ocean by various researchers with different levels in nutrient analysis skill. In this study, we described the homogeneity and stability of nutrient reference material for seawater using natural seawater, collected at a station of Shihwa Lake, at a coastal station near Uljin (surface water), and at a station over the Ulleung Basin (surface water and 1500 m depth water) and sterilized. Based on the homogeneity data, the nutrient reference materials has similar homogeneity compared to other nutrient reference materials. During 3-13 month period, there was no unidirectional trend of increase or decrease in nutrient concentration of newly developed nutrient reference material for seawater. However, a sustained measurement is required to check stability for longer period.

Incidence of Plant-Parasitic Nematodes in Strawberry Nursery and Nematode Dispersal by Daughter Plant (딸기 육묘장 토양 내 식물기생선충의 감염현황과 자묘를 통한 선충의 분산)

  • To survey the incidence of plant-parasitic nematode in strawberry nursery, 117 soil samples were collected from main producing area of strawberry nursery plants in 2016. The incidences of two major problematic nematodes such as root-lesion nematodes (Pratylenchus spp.) and root-knot nematodes (Meloidogyne spp.) were 11% and 3%, respectively. Strawberry nurseries using upland soil either in fields or in plastic-houses showed were higher infection rates of Pratylenchus spp. or Meloidogyne spp. than others. On the other hand, a strawberry nursery using pots filled with bed soil in plastic-house was the lowest infection rate with 0%. Phylogenetic relationships and distance analysis were performed for identification of Pratylenchus spp. and Meloidogyne spp. detected from strawberry nurseries. As the results, they were identified as Pratylenchus penetrans, Pratylenchus vulnus and Meloidogyne hapla. Meanwhile, when nursery plants of strawberry infested with P. vulnus transplanted to pots filled with sterilized soil, P. vulnus was detected from soil in pots as 33% frequency. Thus, P. vulnus can be spread through nursery plants of strawberry infested with that. These studies suggested that Pratylenchus spp. and Meloidogyne spp. were ploblematic nematodes in major areas of strawberry nursery plants and P. vulnus could be spread through nursery plants of strawberry.
