Research on Ultrasonic Sterilization Effect of Treatment Equipments (초음파 처리조의 구조에 따른 살균효과 연구)

  • In the process of sterilization during the manufacture of beverages, both the treatment time and the temperature influence the beverage taste and flavor. Therefore, to improve the beverage quality, a nonthermal sterilization method is preferred to a high-temperature one. A nonthermal method, ultrasonic treatment by shock waves from cavitation, has been known to have a sterilizing effect. In this study, to observe the effect of ultrasonic frequency and treatment time on beverage quality, batch type ultrasonic equipment was fabricated and tested. Further, to evaluate the applicability of this ultrasonic method to an in-situ process, flow type ultrasonic heptagonal equipment was fabricated and then used to perform a sterilization experiment. Subsequently, the sterilizing effect was measured under ultrasonic and low-temperature ($55^{\circ}C$) treatments. Based on the obtained results, ultrasonic sterilization technology that can be applied for treatments at flow rates of up to 3 L/min was developed.

Determination of Optimum Sterilization Condition for the Production of Retort Pouched Curry Sauce (레토르트 카레 소스 생산을 위한 최적살균 조건의 설정)

  • In order to optimize sterilization conditions of retort pouched curry sauce, sterilization processes for eighteen conditions by varying temperature, time and method were conducted through $3^{2}{\times}2^{1}$ experimental factorial design. Quality evaluations before and after sterilization included measurements of vitamin (niacin) retention, pH and color differences, and organoleptic test(taste, color and viscosity). $F_{o}$ values were also measured at each condition. Product qualities were mainly affected by sterilization temperature and time, whereas sterilization method had no significant effect. Effect of sterilization time on product qualities was higher than that of sterilization temperature. From the response surface analysis, an optimum range of sterilization condition simultaneously satisfying desired specifications was determined to be $123.5^{\circ}C$, 21.5 min to $127.5^{\circ}C$, 17.0 min. In this range, the sterility($F_{o}$ value) at a cold point during sterilization was approximately 15.0 min.

  • Yeung Nam University Medical Center, Department Of Central medical Supply Background: The adverse effect of intravenous therapy combined with various complications. Because sterilization technique and appropriate nursing care can prevent various complications, it is important to use appropriate sponge in intravenous therapy. The purpose of study was to identy sterilization effect and detection of bacteria and to provide basic data for use of appropriate disinfectants. Methods: From May 15 1995 to Aug. 3. 1995, disinfectants that were used in Yeung Nam University Medical Center were tested by bacteria culture. To test sterilization effect of disinfectants of intravenous injection sites after disinfection, 10 subjects were used and were tested by bacteria culture for the study. Results: 1) By sterilization effect of disinfectants, bacteria were increased from 103 to 10 from 48 hours in both 2% Zephanon and 2% zephanon that was sterilized by steam, from 10 to 10 from 72 hours and 10 from 48 hours in 70% lsoprophyl alchol. Also, bacterias were detected in 70% lsoprophyl alchol on 48 hours. 2) By stenlization effect of sponge that were used in nursing unit, bacterias were detected in 2% Zephanon on 2 hours, 70% lsoprophyl alchol on 2 hours, 70% lsoprophyl alchol on 8 hours and 70% lsoprophyl alchol on 48 hours. 3) By sterilization effect and detection of bacteria of intravenous site after disinfection, bacterias were detected in 10 of 10 control groups, 8 of 10 sites that were disinfected by steam sterilized 2% zephanon sponge, 6 of 10 sites that were disinfected by 70% lsoprophyl alchol and 4 of 10 sites that were disinfected by 10% Batadine. Conclusions : it is conclued that 70% lsoprophyl alchol is appropriate for intravenous therapy and 10% Betadine is appropriate in ward that were polluted the air and in immunodeficient patients.

A Survey of Disinfection Methods for Formula Bottle (국내 젖병소독법의 실태조사)

  • Purpose: There has been a number of reports on elution of bisphenol-A, an endocrine disrupting chemical, from milk bottles, all concerning the potential health effect of the substance. In this study, we measured the elution of bisphenol-A from milk bottles during boiling-water sterilization, to suggest a safe sterilization method. Method: Through the survey of 200 mothers, 78.5% of them expressed their concern on the possible harmful effect of bisphenol-A, which might be eluted from the bottles. And it was found that most mothers use boiling-water to sterilize bottles; duration of boiling bottles could be divided into three groups of 3, 5, and 10 minutes. We measured the elution of bisphenol- A from three different brands of bottles, subject to various mode of sterilization found in the survey. Result: In all the measurements, the concentration of the eluted bisphenol-A was in the range of 0.3 - 0.7 ppb, far below 2.5ppm, the level set up by Korea Food %amp; Drug Administration(KFDA) for a potential harmful effect. Conclusion: Although trace amount of bisphenol -A can be eluted from polycarbonate milk bottles during sterilization, and bottle feeding, the amount is found to be too small to cause any health related impact for infants.

Influence of Sterilization Methods on Atrazine Assimilation (멸균법이 atrazine의 분해과정에 미치는 영향)

  • We investigated the influence of various known sterilization methods on atrazine assimilation. The present study was designed to investigate the effect of autoclaving, sodium azide and mercuric chloride treatment on the assimilation of atrazine in soil and sediment. The sterilization reactor treated with sodium azide resulted in $^{14}CO_2$ generation and atrazine was rapidly disappeared from reactor through chemical reaction with sodium azide. These findings seem to indicate that sodium azide sterilization is not recommended for atrazine studies. In sample reactors autoclaved or treated with mercuric chloride, $^{14}CO_2$ generation was not detected and most of the disappeared atrazine was found to exist as hydroxyatrazine. These results suggested that autoclaving or mercuric chloride treatment could be effective sterilization methods. However, chemical properties(pH and redox potential) of soil and sediment were altered by any of the sterilization methods applied. So it was suspected that these altered properties could affect distribution and mineralization of atrazine in soil and sediment. In addition, both autoclaving and mercuric chloride treatment have altered $K_d$ values of hydroxyatrazine more significantly than those of atrazine. Consequently, although autoclaving and mercuric chloride treatment are effective sterilization methods, one must be careful in using them in practice as these methods may cause chemical degradation of both of atrazine and its metabolites and changes in chemical properties of soil and sediment. In conclusion, careful assessment of sterilization methods must be made for the degradation studies of chemicals in soil and sediment in order to minimize possible undesirable chemical degradation of sample and/or changes in physico-chemical properties of soil and sediment by the selected sterilization methods.

Reuse of Oyster Shell Waste as Antimicrobial Water Treatment Agent by Silver Ion Exchange

  • A water treatment agent with antimicrobial activity(Ag-Os) was created by exchanging silver ion($Ag^{+}$) on calcined oyster shell powder. The desorption of the exchanged silver ion was negligible, thereby indicating a stable antimicrobial water treatment agent. The sterilization effect of Ag-Os on underwater microorganisms was then investigated. An MIC (Minimum Inhibitory Concentration) test result indicated that Ag-Os had an excellent sterilization effect on G-germs, such as Escherichia coli and Pseudomonas aeruginosa. Most germs were annihilated with an Ag-Os concentration of 200 ppm and contact time of 60 minutes. The sterilization effect was mainly dependent on the contact time. The zeta potential of the Ag-Os powder adsorbed on sand was measured relative to the concentration of exchanged silver ion. As the concentration of the exchanged silver ion increased, the surface charge density of the anions on the surface of the Ag-Os powder adsorbed on sand also increased. Accordingly, this result indicated that a higher silver ion than ion exchange capacity was present on the particle surface due to adsorption. Consequently, this increased concentration of exchanged silver ion would appear to significantly enhance the sterilization power.

Sterilization Test of Microorganisms of Slow-released ClO2 Gas Gel-Pack (서방출형 이산화염소 가스 젤팩의 미생물 살균 시험)

  • Even though chlorine dioxide ($ClO_2$) is utilized in a pre-treatment due to its effective sterilizing activity for microorganisms and its safety for food, it has a limitation in maintaining freshness of the food product. In this study, a low-concentration $ClO_2$ gas was produced in a packaging form of air-permeable gel pack so that it could be released continuously over several days. The amount of $ClO_2$ gas emission and microbial inactivation effect against foodborne pathogens were measured during the release of $ClO_2$ gas. As a result of measuring the change of color in order to confirm whether the chlorine dioxide gas was eluted in the form of a sustained release, the yellowness was significantly higher at higher gel pack concentration and higher value during storage periods. The slow-released $ClO_2$ gel-pack showed clear inactivation effect against Escherichia coli and Staphylococcus aureus with 99.9% inactivation efficiency. As a result of measuring the sterilization effect of Listeria monocytogenes by the concentration of chlorine dioxide gas, the sterilization effect was increased as the concentration was increased. Therefore, the slow-released $ClO_2$ gel-pack is feasible to apply for industry usages.

Treatment of Ar/O2 Atmospheric Pressure Plasma for Sterilization (아르곤과 산소 대기압 플라즈마 방전 효과를 이용한 살균처리)

  • The sterilization effects of atmospheric pressure plasma with the mixture of argon and oxygen were analyzed. The plasma reactor with the shape of dielectric barrier discharge produced the uniform distribution of glow discharge and generated ozone gas effectively according to the various process parameters. The sterilization for E. coli was affected by power, oxygen ratio in the mixture gas, treatment time and distance between reactor and sample. The concentration of ozone was a major source for the sterilization of E. coli, which was enhanced by the increase of power and oxygen ratio. In this study, the effect of atmospheric pressure plasma treatment for the sterilization was confirmed and its result can deliver the atmospheric pressure plasma treatment as the novel sterilization method instead of conventional methods.

Sterilization effect and fatty acid composition of silkworm powder (Bombyx mori L.) by heat treatment

  • The effect of heat treatment on the sterilization and fatty acid compositions of silkworm powder was carried out under the guidance of Ministry of Food and Drug Safety. Food borne pathogens or microorganisms including E. coli, Salmonella spp., Listeria monocytogenes, Staphylococus aureus, Clostridium perfringens and aflatoxin, were not detected. The fatty acid composition was hardly changed after the silkworm powder was treated to a high temperature of $121^{\circ}C$. The low temperature sterilization of silkworm powder at $63^{\circ}C$ decreased the concentration of E. coli while high temperature sterilization at $121^{\circ}C$ has found no traces of microorganism.

  • To determine the sterility of the prepared allogeneic bone of the human, culture of the allografts prior to implantation was performed on fresh-frozen and freeze-dried bone. Before the use of allografts to the patients, it must be confirmed about the sterility, cellular cytotoxicity, immune reaction, and osteoinductive potential as a biomaterials. Oral and maxillofacial surgeons demand for allograft bone will be increased in the future. Wonkwang Bone Bank attempted to meet this demand, has performed series of experimental study on the allograft bone of the Koreans to evaluate the physical and chemical suitability of the bone since the surgeons applications will have broadened from benign cystic lesions to fracture malunions and non-unions, large segmental defects, and whole-bone allorgrafts after tumor surgery. The results obtained were as follows: 1. Freeze-drying(FD) only shoed some bactericidal effects of the normal and osteo bone but in cases of performing EO gas sterilization, the FD effects was not clear. 2. The fact that FD has little effect than the EO gas sterilization on normal bone postulated that the presence of microbiota may be due to an operation and bone processing procedure. 3. FD and EO gas sterilization had a remarkable effect on the osteo bone. 4. The sterilization effect were EO gas, Freeze-drying, Fresh-Frozen with descending order. But all sterilization method were not complete to preserve and implant allograft bone. We are now performing further continuous study on the radiation and chemical sterilization procedure to make safe and complete allograft bone.
