A bioassay-guided fractionation of the stem bark extract of Eucommia ulmoides Oliver (Eucommiaceae) led to the isolation of three iridoid constituents, genipin (1), geniposide (3), geniposidic acid (4) as well as (${\pm}$)-guaiacylglycerol (2) and fatty acid mixtures as active ingredients of the extract responsible for the antitumoral property. The EtOAc soluble part and BuOH soluble part of the extract demonstrated a significant inhibition on the proliferation of cultured human tumor cells such as A549 (non small cell lung), SK-OV-3 (ovary), SK-MEL-2 (melanoma), XF498 (central nerve system) and HCT-15 (colon) in vitro, whereas the remaining water soluble part exhibited a poor inhibition. Column chromatographic fractionation of the methanol and ethyl acetate extracts of the stem-bark of Anacardium occidentale led to the isolation of five compounds (1-5). Their structures were determined by spectroscopic means by comparing spectral data to be β-sitosterol (1), 2,4-dihydroxy acetophenone (2), 1-monolinolein (3), ethyl oleate (4) and β-sitosterol-3-O-β-D-glucopyranoside (5). These compounds were evaluated for cytotoxicity against human cancer cell lines: A549, SCOV3 and rat normal cell line NRK49f. Compounds 2-5 were for the first time isolated from A. occidentale.

Ulmus davidiana var. japonica is a deciduous tree used in traditional medicine. In this study, we examined the antioxidant and anti-inflammatory activity of extracts and compounds isolated from U. davidiana var. japonica stem barks for development of cosmetic phyto-materials. Phytochemical examination of the stem bark led to the isolation and characterization of three known compounds, (+)-catechin (1), (+)-catechin-7-O-${\beta}$-D-apiofuranoside (2), and procyanidin B3 ((+)-catechin-($4{\alpha}{\to}8$)-(+)-catechin) (3). Their bioactivities including antioxidant (DPPH ree radical scavenging assay) activity, anti-inflammatory (nitric oxide nhibition assay) were evaluated. Most of the crude extracts and isolates indicated significant antioxidant potential compared with BHT and ${\alpha}$-tocopherol as controls. Futhermore, all compounds showed higher inhibitory activities for NO production in Raw 264.7 cells than the L-NMMA using the positive control. Eespecially, (+)-catechin (1) and (+)-catechin-7-O-${\beta}$-D-apiofuranoside (2) which could inhibit more than 90% of the NO production at a concentration of 100 ${\mu}g/m{\ell}$, implying excellent anti-inflammatory activity.
To evaluate the quality of the crude drugs using three kinds of Cinnamomum Cortex (CC), Vietnamese CC (VCC, the stem bark of Cinnamomum obtusifolium), periderm-peeled Chinese CC (PPCC, periderm-peeled stem bark of C. cassia), Chinese CC (CCC, stem bark of C. cassia) and a Cinnamomi Ramulus (CR, the twig of C. cassia), the four essential oils were prepared by steam distillation method. Cinnamaldehdye (CAN) and an unknown substance tentatively named hydroxy-cinnamaldehdye(HCNA) were detected in the four essential oils by gas chromatography-mass spectrometry, the contents of which are significantly different one another. Vietnamese CC had the highest content of HCNA whereas CR had the highest CAN content and the lowest HCNA. Vietnamese CC exhibited the greatest cytotoxic activity against the cancer cell lines, A549, HepG-2, HL-60, P-388, U-937, and KB and CR the lowest cytotoxicity. Contents of CAN and HCNA in CCC and PPCC are positioned between VCC and CR. These results suggest that measurement of HCNA and cytotoxicity may determine the quality of CC and CR.
The optimization and microwave assisted extraction of stem bark of Terminalia arjuna, quantitative estimation of the marker compounds arjunic acid and arjunolic acid using HPTLC and the evaluation of free radical scavenging activity has been performed in this study. The central composite design was used for optimization and the values of parameters for optimized batch of microwave assisted extraction were 1000 W (Power), 3 minutes (Time) and 1/120 (Solid/solvent ratio). The solvent system to carry out the HPTLC was toluene: acetic acid: ethyl acetate (5: 5: 0.5) and quantitative estimation was done using standard equations obtained from the marker compounds. The in-vitro free radical scavenging activity was performed spectrophotometrically using ascorbic acid as standard. The value of estimated percentage yield of arjunic acid and arjunolic acid was 1.42% and 1.52% which upon experimentation was obtained as 1.38% and 1.51% respectively. The DPPH assay of the different batches of microwave assisted extraction and marker compounds taken suggested that the marker compounds arjunic acid and the arjunolic acid were responsible for the free radical scavenging activity as the batch having the maximum percentage yield of the marker compounds showed best free radical scavenging effect as compared to standard ascorbic acid. The $IC_{50}$ value of the optimized batch was found to be 24.72 while that of the standard ascorbic acid was 29.83. Hence, the yield of arjunic acid and arjunolic acid has direct correlation with the free radical scavenging activity of stem bark extract of Terminalia arjuna and have potential to serve as active lead compounds for free radical scavenging activity.
In this study, we investigated antioxidant capacity and determined the phenolic and flavonoid contents using each of various solvent conditions from fermented Rhus verniciflua stem bark (F-RVS). Each extracts displayed markedly similar content of extraction yield. However, M80 extract showed a significantly higher antioxidant activity in comparison to other extract investigated. M80 exhibited strong DPPH radical scavenging activity with $RC_{50}$ value of $10.5{\pm}1.4{\mu}g/m{\ell}$, reducing power value 1.04 Abs (concentration of 1 mg/$m{\ell}$), and anti-lipid peroxidation activity value of 94.6% (concentration of 10 mg/$m{\ell}$). M80 extract showed the high content of total phenolic (319.7 mg GAE/$m{\ell}$ extract) and total flavonoid (111.6 mg QE/$m{\ell}$ extract). Phenolic and flavonoid compounds showed significant relationship in DPPH radical scavenging ($R^2$=0.911 and 0.912, each extract) and reducing power ($R^2$=0.903 and 0.837, each extract) from fermented R. verniciflua stem bark. However, antilipid peroxidation activity ($R^2$=0.589 and 0.441, each extract) was not significant like DPPH radical scavenging and reducing power. Therefor the result indicated that the potential antioxidant activities and functional values were ovserved significantly at M80 solvent condition from the fermented R. verniciflua stem bark.
This study was performed to determine whether stem bark extract containing saponin of Albizzia julibrissin in the diet influences gonadal maturation and spawning in medaka (Oryzias latipes). The crude extraction containing saponin (HaBC) was partially purified from n-BuOH extraction of A. julibrissin stem bark by Diaion HP-20, Silica gel and Sephadex LH-20 chromatographies. We fed diets supplemented with HaBC to medaka. We then studied the effects of the HaBC supplement on the suppression of gonadal maturation and spawning in female medaka that were reared in aquaria with recirculation systems. In the experiment with immature female medaka, the periods of initiation of gonadal maturation and spawning were delayed in the fish that were fed diets supplemented with at least HaBC 20 mg/g-feed. In the experiment with mature female medaka, the fish that were fed diets supplemented with at least HaBC 20 mg/g-feed had lower GSIs than the control diet group did. The results showed that the growth of the immature medaka was not correlated with the amount of supplementation of HaBC in the diet. However, the condition factors (CF) in the medaka that were fed diets supplemented with at least HaBC 20 mg/g-feed were higher than in the medaka fed on the control diet. We concluded that extracts containing saponin from the stem bark of A. julibrissin have the potential to inhibit gonadal maturation in female medaka, but they did not act as growth stimulation. Further studies are required to determine the mechanism of the action.
In order to develop as a natural source of anticancer materials of Cudrania tricuspidata, the cytotoxicity of methanol extracts by harvesting parts and times against 8 cell lines including 293 (normal kidney cells) and A-431 (epidermoid carcinoma cells) were investigated using MTT assay. All harvesting parts had hardly cytotoxicity against 293. And methanol extracts of stem bark and root bark showed very high cytotoxicities against 7 cancer cell lines. The cytotoxicity was the highest against HeLa (cervix adenocarcinoma cells) and followed by MCF-7 (breast adenocarcinoma cells), AGS (stomach adenocarcinoma cells), HT-29 (colon adenocarcinoma cells), HepG2 (hepatoblastoma cells), A549 (lung carcinoma cells) and A-431. By the way, leaf extract had a cytotoxicity against only AGS and ripe fruit extract had no cytotoxicity. Among harvesting times, the cytotoxicity of root bark were high from April to September but that of stem bark showed a little difference. These results showed that anticancer activities of Cudrania tricuspidata extracts were eventful changes by harvesting parts and times.
