Gelatinization Properties of Starch during Steeping Condition of Potato (감자의 수침조건에 따른 전분의 호화 특성)

  • Gelatinization properties of potato starches which were prepared by steeping at 10 ${\pm}$ 1$^{\circ}C$ or 25 ${\pm}$ 1$^{\circ}C$ for 11 days were investigated. The pasting and initial gelatinization temperatures measured by rapid visco-analyzer (RVA) were increased, but the peak and trough viscosities of potato starch were decreased by steeping. The onset temperature, peak temperature, conclusion temperature, and enthalpy of gelatinization were increased by steeping as measured by DSC. The contents of hot-water-soluble carbohydrate and amylose in potato starch were decreased by steeping.

  • The changes in physicochemical properties of potato were investigated while steeping in water for 7 days at 30${\pm}$1$^{\circ}C$. The shape of raw starch granules was round or oval, the starch granule showed birefringence distinctly under polarized light and it was kept clearly even after steeping. X-ray diffraction pattern of the starch was B-type and there was no change in the pattern after steeping. However, crystallinity was increased up to the 4th day and then decreased. Amylose contents of raw starch and the starch steeped for 7 days were 19.3% and 13.1%, respectively. When the potato starch was gelatinized in 0.15 N sodium hydroxide solution, the viscosity was decreased until the 3rd day, but increased thereafter. Gel volume of the starch in KSCN solution was decreased during steeping.

  • Gelatinization and retrogradation properties of modified starches which were prepared by steeping sweet potato at $40^{\circ}C$ for 2, 4, 7 and 10 days were investigated. The peak temperature of gelatinization and enthalpy of untreated starch by DSC were $53.9^{\circ}C\;and\;1.32\;cal/g$, respectively, but those of modified starch were increased by steeping. In gelatinization by alkali, starches with 2, 4 and 7 day steeping showed higher viscosities than untreated starch, whereas the viscosities of starches with 10 day steeping decreased. The clarities in paste decreased during storage in all starches and decreased in starches with steeping. The degrees of retrogradation by ${\alpha}-amylase-iodine$ method were higher in starches with steeping than untreated starch. The enthalpy of retrograded starches by DSC increased by steeping except 4 day steeping starch. The sweet potato extract containing sugar inhibited the retrogradation of starch paste and the degree were higher in residual starches than in untreated starch.

  • The changes in gelatinization patterns of potato were investigated while steeping in water for 7 days at 30${\pm}$1$^{\circ}C$. The transmittance of 0.1% starch suspension was increased rapidly from 60$^{\circ}C$ in raw starch and the starch steeped for 2 days, and increased rapidly from 65$^{\circ}C$ in the starch steeped for 4 and 6 days. As the steeping period increased, the transmittance was decreased at above condition. The gelatinization temperature of the starch measured by differential scanning calorimetry was increased from 62.79$^{\circ}C$ to 63.72$^{\circ}C$ as the steeping period increased. The gelatinization enthalpy reached the maximum in the starch steeped for 4 days. By amylograph, the initial gelatinization temperature was increased from 66$^{\circ}C$ to 84$^{\circ}C$ as the steeping period increased. Peak viscosity was decreased during steeping and the starch steeped for 5 days had no peak viscosity. Peak height after cooling to 50$^{\circ}C$ was increased up to the 4th day and began to decrease. As the steeping period increased, there was much loss of birefringence at higher temperature. The crystallinity by X-ray diffraction disappeared from 65$^{\circ}C$.

  • Gel characteristics of potato starches which were prepared by steeping potato at 10$\pm$ 1$\^{C}$ and 25$\^{C}$$\pm$1$\^{C}$ incubator for 11days were investigated. Results of the instrumental test of potato starch gels showed significantly different strength, hardness, gumminess, chewiness, cohesiveness by steeping time. The light microscope and scanning electron microscope of starch gels showed three-dimensional network including macroporous structure by steeping. The syneresis of potato starch gel was decreased by steeping potato at 24 for 72 hours.

  • It Is a unique dietary culture in Korea in that starch is isolated and utilized from steeped potato. In this experiment the potato was steeped in water at $30^{\circ}C$ for 7 days and the properties of starch were examined. The pH in steep water decreased, while sugar cotent (total and reducing) increased. The lager granules were diminished during steeping. Holes on the starch granules were observed from the second days of steeping The density, amylose content, phosphorus and lipid content were decreased. Relative crystallinity of starch reached the highest value at 4th day of steeping and decreased thereafter. The changes in enthalpy of gelatinization were similar. Starch was resistant to acid or enzymatic hydrolysis and showed lower values for swelling power and amylograph viscosities.

Physicochemical and Gel Properties of Starch Purified from Mealy Sweet Potato, Daeyumi (분질고구마 대유미 전분의 이화학적 및 겔 특성)

  • Purpose: The properties of starch and starch gel prepared from a newly inbred sweet potato, Daeyumi were compared to the properties of starch and starch gel prepared from Sinyulmi which is a well known mealy type sweet potato. Methods: The starch was isolated by using the alkaline steeping method. Physicochemical, pasting, and thermal properties, and crystallinity were measured. The texture properties of starch gel (10%, w/w) were examined. Results: The amylose contents of Daeyumi and Sinyulmi starches were 25.57% and 22.59%, respectively. The initial pasting temperature of Daeyumi starch was significantly higher than that of Sinyulmi starch (p<0.05), but other paste viscosities were not different. The peak and conclusion temperatures of Daeyumi starch were higher than those of Sinyulmi starch by differential scanning calorimetry. The shape of Daeyumi starch gel was more clear and rigid than the shape of Sinyulmi starch gel. The surface and the upper side of Daeyumi starch gel were smoother than the surface and the upper side of Sinyulmi starch gel. Hardness and gumminess were higher in Daeyumi starch gel than in Sinyulmi starch gel. The crystallinity types of Daeyumi and Sinyulmi starches were $C_b$ and A types, respectively, but starch gels showed an amorphous type. Conclusion: Therefore, it is suggested that Daeyumi starch would have better physicochemical properties and higher quality of starch gel than Sinyulmi starch.

Saccharification of Uncooked Starch (무증자 전분의 당화에 관한 연구)

  • For the eventual alcohol production from uncooked starch, an efficient saccharification process was examined by using the combined action of steeping, pectin depolymerase, ${\alpha}-amylase$ and glucoamylase. The total sugar content of rice, sweet potato and tapioka used were 4.53, 4.26, and 3.92 mmole/g sample. $70\;{\pm}\;10%$ of the total sugar was hydrolyzed when cooked starch was saccharified under the condition which is currently used in industry. The smaller starch particle was used, the more saccharification was obtained. Efficient saccharification was obtained by treatment with 5% $H_2SO_4$ (sample: working volume = 1:2) at $60^{\circ}C$ for 12 hr. Optimization was carried out for the saccharification of uncooked starch by the combined action of pectin depolymerase, ${\alpha}-amylase$, and glucoamylase. The conditions are: pectin depolymerase; pH 4.5, $45^{\circ}C$, 2 hr, ${\alpha}-amylase$; pH 6.0, $60^{\circ}C$, 1 hr, and glucoamylase; pH 3.5, $60^{\circ}C$, 1 hr. Simultaneous treatment of the combined action of macerating, liquifying and saccharifying enzymes yielded better result than stepwise treatment of 3 enzymes. Degrees of saccharification of uncooked tapioka, rice and sweet potato were 82, 90.5, and 84.5%, respectively on the basis of total sugar, under the optimized conditions.
