Isolation and Identification of an Antibacterial Substance from Sea Mustard, Undaria pinnatifida, for Streptococcus mutans (미역 추출물로부터 충치 원인균, Streptococcus mutans에 대한 항균물질의 분리 및 동정)

  • An antibacterial substance to the Streptococcus mutans, a causative bacterium for decayed teeth, was isolated from the dried sea mustard, Undaria pinnatifida, and identified by GC and GC/MS. Acetone extract from the sea mustard (10.4 kg), was evaporated and partitioned to 4 fractions such as hexane, chloroform, butanol and water. The most active chloroform fraction were further purified through basic alumina, silicic acid and ODS column, successively, and finally, 3 antibacterial substances were isolated on the HPLC attached ODS column by using 95% MeOH and guided with UV detector (254 nm). Antibacterial substances (total 160mg, yield $1.5\times10^{-3}$%) had the same Rf value (0.42) on the TLC developed hexane diethyl ether acetic acid (80:30:1) and those methyl esters moved to 0.95. They were identified as the same unsaturated fatty acid, $C_{18:4,\;n-3}$ (3,6,9,12-octadecatetraenoic acid, stearidonic acid) compared relative retention times (15.5 min) with authentic fatty acid on the GC chromatogram. It was further confirmed unambiguously on the GC/MS giving molecular ion peak at m/z 290 which coincided with its methyl ester.

  • Total lipid contents were 0.58∼3.00% in 9 kinds of brown algaes, 0.47∼2.16% in 5 kinds of red algaes and 0.55∼2.99% in 2 kind of green algeas, respectively. Fatty acid compositions of the algaes were determined by gas liquid chromatography. Total polyenes and n-3 polyenes of linoleic acid(18:3), stearidonic acid(18:4), eicosapentaenoic acid(20:5) and docosahexaenoic acid(22:6) were 3.88∼57.57% and 1.46∼25.67% in the brown algaes, 5.30∼39.75% and 1.17∼21.91% in the red algaes, and 7.76∼19.27% and 3.67%∼10.61% in the green algaes, respectirely. The fatty acid contents and compositions of total polyenes and n-3 polyenes were vary different in the algae groups and sepecies.

Physicochemical Composition of Seaweed Fulvescens (Capsosiphon fulvescens) (매생이(Capsosiphon fulvescens)의 이화학적 성분)

  • Physicochemical components of dried green algae, Seaweed fulvescens (Capsosiphon fulvescens) with $96.02{\pm}0.02%$ moisture content, were investigated. Crude protein, crude lipid, ash and carbohydrate content of C. fulvescens were $31.76{\pm}0.26,\;1.01{\pm}0.21,\;13.58{\pm}0.62\;and\;53.65{\pm}0.73$, respectively. Predominant minerals were Na, Mg, Ca, K and P. Fe $(122.83{\pm}11.19mg/%)\;and\;Se\;(0.90{\pm}0.07mg%)$ contents in were higher than those of other seaweeds. Major free sugars were identified as fructose, glucose, sucrose, maltose and galactose. Organic acid including L-malic acid, citric acid, tartaric acid and d-malic acid were detected. Most abundant amino acids were glutamic acid, aspartic acid and alanine. Main fatty acids were palmitic acid, linolenic acid and stearidonic acid, with 58.37% polyenes.

Fatty Acid Compositions of Nutural Lipids and Polar Lipids in the Parts of Miyeok (Undaria pinnatifida) (미역의 부위별 중성 및 극성 지방질의 지방산 조성)

  • Fatty acid composition of frond, spororphyll and stipe of Miyeok (Undaria pinnatifida) were extensively analyzed to evaluate a nutrient significance of n-3 polyenes, n-6 polyenes, ${\gamma}$-linolenic acid (18:3, n-6) and $\alpha$-linolenic acid (18:3, n-3). Polyenes of $\alpha$-linolenic acid (22:6, n-3), stearidonic acid (18:4, n-3), eicosapentaenoic acid (20:5, n-3) and docosahexaenoic acid (22:6, n-3) were as high as 29.69%, but ${\gamma}$-linolenic acid (18:3, n-6) was as low as 0.90% when compared to total polyenes of 44.18% in frond. The n-3 polyenes of neutural lipids, glycolipids and phospholipids were 22.40%, 35.82% and 3.53% in frond, 5.26%, 9.51% and 2.37% in stipe and 8.01%, 4.49% and 2.14% in sporophyll, respectively. These results suggest that total polyenes and n-3 polyenes in frond of Miyeok may be used as resource of functional food.

미역의 부위별 중성 및 극성 지방질의 지방산 조성

  • 한국 남해안 완도지역에 자생하는 미역 (Undaria pinnatifida)의 부위별 지방산조성 특성을 계통적으로 분석코자 미역의 부위별 지방질 및 지방산 조성을 silicic acid 컬럼과 기체-액체 크로마토그래피 분석하였다. 총지방질 함량을 비교해 볼 때 엽상부 3.37%, 포자엽 4.59% 및 줄기 2.50%였으나, 엽상부는 총 포리엔산과 linolenic acid(18:3, n-3), stearidonic acid(18:4, n-3), eicosapentaenoic acid(20:5, n-3) 및 docosahexaenoic acid(22:6, n-3)와 같은 n-3계열의 포리엔산의 조성비율이 높았다. 즉 총 포리엔산과 n-3계열의 포리엔산 조성비율을 볼 때 엽상부는 44.18% 및 29.69%였고, 포자엽은 23.63% 및 5.58% 줄기는 26.66% 및 7.37%였다. 한편 중성지방질, 당지방질 및 인지방질의 n-3 포리엔산의 함량을 볼 때 엽상부는 22.40%, 35.82% 및 3.53%로 다른 부위에 비하여 현저하게 높았으나, 줄기는 5.26%, 9.51% 및 2.37%였고 포자엽은 4.49%, 8.01% 및 2.14%로 가장 낮았다.

Process Development of Concentration of n-3 PUFAs from Fish Oil by Means of Lipase (리파제의 아실 체인 특이성을 이용한 물고기 기름에서 n-3 다중불포화지방산의 농축공정개발)

  • Experiments on the process development for the concentration of polyunsaturated fatty acid (PUFAs) such as eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) from fish oil by using acyl chain specificity of Candida cylindracea lipase were performed. Five kinds of oils were hydrolyzed with Candida cylindracea lipase. Among then, Candida cylindracea lipase just had low activity on the PUFAs-rich fish oil. After the hydrolysis of fish oil, free fatty acid was removed and fatty acid components of glyceride mixtures were analyzed. When the hydrolysis was about 70%, the DHA content in the glyceride mixture was about three times more than that in the original fish oil. The EPA and stearidonic acid contents in the glyceride mixtures, however, were similar to that of the original fish oil. In this work, these results showed that the concentration process of PUFAs by using the acyl chain specificity of Candida cylindreacea lipase was effective in producing glycerides that contained a high concentration of PUFAs in good yield.

Fermentation Process Development of Recombinant Hansenula polymorpha for Gamma-Linolenic Acid Production

  • Development of the strain and the fermentation process of Hansenula polymorpha was implemented for the production of ${\gamma}$-linolenic acid ($GLA,\;C18:3{\Delta}^{6,9,12}$), an n-6 polyunsaturated fatty acid (PUFA) that has been reported to possess a number of health benefits. The mutated ${\Delta}^6$-desaturase (S213A) gene of Mucor rouxii was expressed in H. polymorpha under the control of the methanol oxidase (MOX) promoter. Without the utilization of methanol, a high-cell-density culture of the yeast recombinant carrying the ${\Delta}^6$-desaturase gene was then achieved by fed-batch fermentation under glycerol-limited conditions. As a result, high levels of the ${\Delta}^6$-desaturated products, octadecadienoic acid ($C18:2{\Delta}^{6,9}$), GLA, and stearidonic acid ($C18:4{\Delta}^{6,9,12,15}$), were accumulated under the derepression conditions. The GLA production was also optimized by adjusting the specific growth rate. The results show that the specific growth rate affected both the lipid content and the fatty acid composition of the GLA-producing recombinant. Among the various specific growth rates tested, the highest GLA concentration of 697 mg/l was obtained in the culture with a specific growth rate of 0.08 /h. Interestingly, the fatty acid profile of the yeast recombinant bearing the Mucor ${\Delta}^6$-desaturase gene was similar to that of blackcurrant oil, with both containing similar proportions of n-3 and n-6 essential fatty acids.

Comparison of Aquacultured and Wild Undaria pinnatifida Thalli for Evaluation of Eight Quality Standards (미역의 8가지 품질기준 설정을 위한 양식산 및 자연산 미역의 비교)

  • Eight quality standards of the brown seaweed, Undaria pinnatifida were designed and evaluated with dry products collected from aquacultured and wild areas. Contents of n-3 fatty acids (stearidonic acid and eicosapentaenoic acid), anti-edema activity, and total carbohydrate were measured as functional criteria. Total bacteria and molds were counted for safety conditions. Amounts of a brown pigment fucoxanthin, debris, and moisture were measured to evaluate the visible status of the products. When the seaweeds were tested in 1 g dry weight basis, they showed n-3 fatty acids in the range of approximately 0.2-2.8 mg, total carbohydrate of 0.39-0.71 g, total bacteria of $10^{4-6}$ cells, mold of $10^2$ cells, fucoxanthin of 0.4-1.5 mg, and debris of 10-24 mg. Anti-edema activity was 0.03-0.47 AU at 40 mg/mL extract. Moisture content was 11.4-12.6%.

Fatty Acid Compositions of Sea Algaes in the Southern Sea Coast of Korea (남해안 해조류의 지방산 조성)

  • Lipid contents and fatty acid compositions of 16 kind sea algaes in the southern sea coast of Korea were analyzed to evaluate nutrient significance of total polyene, n-3 polyenes and n-6 polyene fatty acids. Total lipid contents were 0.58 ∼ 3.00% in 9 kinds of brown algaes, 0.47 ∼ 2.16% in 5 kinds of red algaes and 0.55 ∼2..99% in 2 kinds of green algeas. N-3 polyenes and n-6 polyenes of the fatty acid compositions were 1.46∼25.67% and 0∼48.51% in the brown algaes, 1.17∼21.91% and 0.28 ∼29.35% in the red algaes, and 3.67%∼ 10.61% and 4.10 ∼ 8.66% in the green algaes, respectirely. The fatty acid contents and compositions of n-3 polyenes and n-6 polyenes, monoenes and saturates were vary different in the algae groups and sepecies.
