The Anti-microbial Activity of Silk Fabrics Dyed with Chelidonium majus var. asiaticum extracts (애기똥풀 추출액으로 염색한 견직물의 항균성)

  • The purpose of this study is to scientifically proof possibility of development of natural dye which has antimicrobial activity with Chelidonium majus var. asiaticum. For that silk fabrics dyed with Chelidonium majus var. asiaticum extracts were tested for anti-microbial activity. Bacterials used for test of anti-microbial activity were Staphylococcus aureus ATCC 6538, Klebsiella pneumoniae ATCC 4352 and Trichophyton mentagrophytes IFO 5466. The results of experiment were as follows ; 1. The more the number of times of dyeing increase the more value of K/S increased. 2. Reduction of bacterium against Staphylococcus aureus ATCC 6538 of silk fabrics dyed with Chelidonium majus var. asiaticum extracts was 99.9% without reference to the number of times of dyeing. 3. Reduction of bacterium against Staphylococcus aureus ATCC 6538 of silk fabrics dyed with Chelidonium majus var. asiaticum extracts was 99.9% after irradiation of 20 hour without reference to the number of times of dyeing. 4. Reduction of bacterium against Staphylococcus aureus ATCC 6538 of silk fabrics dyed with Chelidonium majus var. asiaticum extracts was over 94.8% after dry cleaning. As above silk fabrics dyed with Chelidonium majus var. asiaticum extracts were acquired a high anti-microbial activity against Staphylococcus aureus ATCC 6538.

Biochemical Characteristics of Micrococcus varians, Staphylococcus carnosus and Staphylococcus xylosus and Their Growth on Chinese-Style Beaker Sausage

  • This study was conducted to investigate protein and carbohydrate utilization of Micrococcus varians, Staphylococcus carnosus and Staphylococcus xylosus. Sensitivity to pH, sodium chloride, potassium sorbate and sodium nitrite of these strains was also determined. In Chinese-style beaker sausage manufacturing, the growth rate of these strains during the curing period ($20^{\circ}C$ and $30^{\circ}C$) was evaluated. The results indicated that no strains could hydrolyze azo-casein and sarcoplasmic protein and only S. xylosus could hydrolyze gelatin at $30^{\circ}C$. All of these strains could oxidize and ferment fructose and mannitol. S. carnosus and S. xylosus could slightly oxidize lactose and utilize citrate. Arabinose was oxidized by S. xylosus and sorbitol was oxidized by S. carnosus. Growth of M. varians was restricted at pH 5.0 and S. carnosus and S. xylosus were restricted at pH 4.5. S. xylosus and S. carnosus were able to grow with 0.1~0.5% potassium sorbate, 50~200 ppm sodium nitrite or 1~15% sodium chloride. S. xylosus had a higher growth rate than the other strains. Staphylococcus species grew well during curing period of Chinese-style beaker sausage then followed by Micrococcaceae.

Antimicrobial Activity of Silver Ion against Salmonella typhimurium, Staphylococcus aureus and Vibrio parahaemolyticus (Salmonella typhimurium, Staphylococcus aureus, Vibrio parahaemolyticus에 대한 은 이온의 항균효과)

  • Antimicrobial activity of silver ion was tested against 3 kinds of food-borne microorganisms-Salmonella typhimurium, Staphylococcus aureus and Vibrio parahaemolyticus-using paper disk and broth medium methods. In paper disk method, silver ion showed antimicrobial activity against S. typhimurium and V. parahaemolyticus at the concentration above 2 ppm and 10 ppm, respectively, where as it was not detected in S. aureus with 20 ppm of silver ion concentration. In broth medium, the growth of S. typhimurium and V. Porahaemolyticus could be retarded at 0.3 ppm and 0.5 ppm of silver ion concentration respectively. In the presence of 1.0 ppm of silver ion, the growth of S. typhimurium was inhibited completely. In S. aureus, the growth was retarded at 5 ppm and was inhibited at l5 ppm completely.

  • The Presence and quantity of protein A in Staphylococcus hyicus subsp hyicus isolates from pigs were determined by indirect hemagglutination test and enzyme-linked immunosorbent assay(ELISA). Cell-bound and extracellular protein A was demonstrated in 87.7% and 36.0% of 489 isolates, respectively, by indirect hemagglutination test. When contents of the protein A were estimated by ELISA method, all of the isolates that were positive to protein A in indirect hemagglutination test produced extracellular protein A of less than 1ng/ml. Most of the isolates produced cell-bound protein A of less than 1ng/ml, whereas 28 isolates produced 1 to 35ng/ml and 11 isolates produced 25 to 108ng/ml.

  • Isolation of Staphylococcus hyicus subsp. hyicus from piglets suffering from exudative epidermitis and healthy pigs was performed, and some properties of the isolates were examined. Twentry-six litters of exudative epidermitis observed in the field were sucking piglets with ages ranging from 1 to 5 weeks and most (73.1%) of these cases occurred in piglets between 2 and 3 weeks of age. Staphylococcus hyicus subsp. hyicus was isolated front 192(38.9%) of 494 healthy pigs. The rate of isolation of these organism from healthy pigs was found to vary greatly among farms at an isolation rate of 33.3% to 45.5% and this organism was isolated more frequently front sucking piglets below the age of 8 weeks than adult pigs. All of the isolates originated from the diseased and healthy pigs were positive in heat-stable DNase, Tween 80 hydrolysis, gelatinase, protease, but negative in clumping factor and hemelysin on sheep blood agar. There was no difference related to origin in the production of extracellular active substances. Typical lesions of the classic exudative epidermitis were produced by inoculation of Staphylococcus hyicus subsp. hyicus isolated from the diseased and healthy pigs. The pathogenic potentials of isolates from healthy pigs were no different from that of the isolates obtained from diseased pigs.

Antimicrobial Effect of Ulmus davidiana var. japonica Extracts on Food-Borne Pathogens (식중독 유발 세균의 증식에 미치는 느릅나무 뿌리껍질 추출물의 영향)

  • This study was performed to investigate the antimicrobial effect of Ulmus davidiana var. japonica extracts against food-borne pathogens. First, Ulmus davidiana var. japonica was extracted with methanol at room temperature and the fractionation of the methanol extracts was carried out by using petroleum ether, chloroform, and ethyl acetate, and methanol respectively. The antimicrobial activity of Ulmus davidiana var. japonica extracts was determined by using a paper disc method against food-borne pathogens and food spoilage bacteria. The methanol extracts of Ulmus davidiana var. japonica showed the highest antimicrobial activity against Staphylococcus epidermidis and Escherichia coli. Finally, the growth inhibition curves were determined by using methanol extracts of Ulmus davidiana var. japonica against Staphylococcus epidermidis and Salmonella typhimurium. The methanol extract of Ulmus davidiana var. japonica had strong antimicrobial activity against Staphylococcus epidermidis at the concentration of 4,000 ppm. At this concentration, the growth of Salmonella typhimurium was retarded more than 48 hours and up to 12 hours for Staphylococcus epidermidis. In conclusion, the methanol extracts of Ulmus davidiana var. japonica inhibit efficiently Staphylococcus epidermidis and Salmonella typhimurium.

Antibacterial Activity of Ethanol Extract of Saussurea lappa (목향(木香) 에탄올추출물의 항균효과(抗菌效果))

  • Objective : The purpose of this study is to investigate the antibacterial activity of Saussurea lappa for the applications of herb-based extracts to both cosmetic and medicinal industries. Methods : Antibacterial activity of ethanol extract of Saussutes lappa (ESL) was assessed using agar diffusion and broth dilution methods, and determined by whether clear zone was formed around paper disc and in terms of the size (mm) of clear zone, Results : ESL provided activity against Staphylococcus aureus KCTC 1927 in concentration of 1 mg/ml with an clear zone of 16 mm, and showed an activity against Staphylococcus epidermids KCTC 1917 in concentration of 1 mg/ml with an clear zone of 18mm. Moreover, the minimum inhibitory concentration (MIC) of ESL against S. sureus and S. epidermidls were 1 mg/ml for both bacterial species. However, ESL showed no growth inhibition against Pseudomonas aeruginosa KCTC 12513, Enterobactet aerogenes KCTC 2190, Escherichia coli KCTC 2571, Salmonella typhimurium KCTC 1925 and Propionibacterium acnes KCCM 41747. Conclusions : Antibacterial activity of ESL against Staphylococcus sureus and Staphylococcus epidermids causing eye and skin diseases was proved. The result suggest that ESL may be useful as a natural preservative on behalf of synthetic preservatives.

Antimicrobial Resistance of Clinically Important Bacteria Isolated from Burn Wound Infections in Children (화상감염 소아환자에서 분리된 주요 균종에 대한 항생제의 내성률)

  • Background & Objectives: Burn injury mortality and septic complication are frequent and well-known in burned pediatric patients. The overuse of antibiotics is the base for development of wound infection by resistant microorganisms as well as opportunist agents. Methods: We have carried out a study of the bacterial profile and antimicrobial resistance clinically important bacteria isolated from burn wound infections in children patients. The most common isolate from burn wound cultures was Pseudomonas aeruginosa (26.8%), followed by Staphylococcus aureus (25.4%), Acinetobacter baumannii (12.7%), coagulase negative staphylococcus (12.0%), Enterococcus faecium (7.7%), Escherichia coli (4.9%), Enterococcus faecalis (3.7%), Burkholderia cepacia (3.0%), Enterobacter cloacae (2.3%) and Klebsiella pneumonia (2.3%). Colistin was very significantly effective drug in gram negative organism, such as Pseudomonas aeruginosa and Acinetobacter baumannii. Results & Conclusion: The resistance rates were 65% and 98% to piperacillin, 63% and 97% to ceftazidime, 28% and 50% to levofloxacin. The most effective antibiotic in gram positive organism, such as Staphylococcus aureus, coagulase negative staphylococcus were moxifloxacin. The resistance rates were 83% and 64% to ciprofloxacin, 80% and 17% to clindamycin.

A Peptide Antibiotic AMRSA1 Active against Multidrug-resistant Staphylococcus aureus Produced by Streptomyces sp. HW-003

  • The antibiotic-producing strain HW-003 was screened from soil and found to be effective against the multidrug-resistant Staphylococcus aureus. The spore chain of HW-003 was retinaculiaperti, and the spore surface was spiny. Strain HW-003 has a LL-diaminopimelic acid isoform in the cell wall. The aerial mass color of the strain was gray, and the reverse side was yellow-brown. The strain produced melanin, but did not produce soluble pigments. According to the Taxon program, HW-003 showed best match with Streptomyces cyaneus. Antibiotic production reached a maximum after 72-h cultivation. The antibiotic was purified with silica gel column chromatography, octadecylsilyl column chromatography, and HPLC. The purified antibiotic, AMRSA1, showed strong inhibitory activity against multidrug-resistant Staphylococcus aureus and gram-positive bacteria. The molecular weight of AMRSA1 was about 1, 100. AMRSA1 was a peptide antibiotic containing alanine and serine.

  • This study was conducted to evaluate the modified drug sensitivity test for the shortening of detection time and clinical application. The results obtained were summarized as follows. 1. Inhibitory zone in 6 strains of Staphylococcus and Escherichia coli tested to sensitivity teat were reached at 100% between 2 and 6 hours. 2. The sensitivity test of Staphylococcus A, B and E. coli No. 2, No. 3 to antibacterial ointment drugs were strong positive(+++), but Staphylococcus A. A. and E. coli No. 1 were mild resistant(+). 3. The sensitivity test of Staphylococcus A. B and E. coli No. 2, No. 3 to antibacterial injection drugs in 6 hours after treatment were strong positive(+++), and Staphylococcus A. A. and E. coli No. 1 were moderate positive(++). 4. This modified method was detected 86% degree of sensitive between 7 to 12 hours after treatment. 5. We think this modified method was more practical compare to other methods.
