Genomic Fingerprinting of Staphylococcus aureus Isolated from Bovine Mastitis Milk by PCR (젖소 유방염 유래 Staphylococcus aureus의 PCR을 이용한 Genomic Fingerprinting)

  • A total of 137 strains of Staphylococcus aureus were isolated from dairy cow's milk with subclinical mastitis from 33 herds in 5 provinces and 36 strains of S aureus from clinical mastitis from 4 herds where the mastitis were severe problem. Arbitrary primed polymerase chain reactions with 10 bp oligonucleotide primer were performed and the PCR products were analysed with image analyzer, The S aureus strains were genotyped into 20 distinct DNA fingerprinting profiles. The size of PCR products ranged from 163 to 2,479 bp and PCR products of 506, 770, 784 and 2,479 bp were the most prevailing bands. Genotype 3 was founded in all 5 provinces. The various genotypes were identified in newly founded dairy herds, however, only one or two genotypes were identified in the closed herds. In clinical mastitis, only a limited number of different S aureus genotype was founded in each of the herds in comparision with subclinical mastitis. The results demonstrated that PCR-based DNA fingerprinting analysis of S aureus strain can be used to study epidemiology of mastitis, in addition, common genotype in geographic region can be useful for the development of an effective S aureus bacterin.

  • A 1-year-old, male, captive born Burmese Python (Python molurus bivittatus) presented with cloudiness of the left eye after ecdysis. Based on physical examination and history, subspectacular abscess was diagnosed. The causative microorganism was identified as a methicillin-resistant Staphylococcus aureus (MRSA). MRSA is a zoonotic problem of high concern and is a risk in public health and veterinary medicine. To our limited knowledge, this is the first reported case of MRSA infection in snakes.

  • Bacteremia occurs in a wide variety of clinical procedures in oral cavity. Reduction of the number of causative microorganisms of infective endocarditis in oral cavity by local administration of antimicrobial agents decreases the magnitude of bacteremia and possibility of infective endocarditis. The effects of chlorhexidine on Streptococcus sanguis, Streptococcus mitis, Streptococcus mutans, Streptococcus oralis, Streptococcus gordonii, Staphylococcus aureus, and Staphylococcus epidermis were investigated by measurement of turbidity. The effects of 0.1% chlorhexidine gargling for 7 days on oral bacterial flora, total streptococci, S. mutans, S. aureus, and S. epidermis in whole saliv a of 7 healthy human subjects, were investigated by measurement of Colony Forming Units (CFU). The obtained results were as follows : 1. Chlorhexidine showed significant antimicrobial effects on Streptococcus snaguis, Streptococcus mitis, Streptococcus mutans, Streptococcus oralis, Streptococcus gordonii, Staphylococcus aureus, and Staphylococcus epidermis. However, the effects on S. sanguis and S. gordonii were not apparent compared with other microorganisms. 2. Oral gargling of 0.1% chlorhexidine decreased the CFU values of normal oral bacterial flora, total streptococci, S. mutans, S. aureus, and S. epidermis in whole saliva. The antimicrobial effects were significant after 4 days of chlorhexidine gargling. 3. Local antimicrobial administration in addition to systemic antibiotic prophylaxis can be highly recommended as an effective adjunct regimen for prevention of infective endocarditis.

Growth Inhibition of Salmonella typhimurium and Staphylococcus aureus in Naengmyon-Broth by Addition of Antibacterial Dongchimi-Juice (항균성 동치미액의 첨가에 의한 냉면국물 중의 Salmonella typhimurium 및 Staphylococcus aureus 생육억제)

  • Juice of Dongchimi a Korean traditional vegetable food fermented with lactic acid bacteria has been traditionary used as broth for Naengmyon a Korean cold noodles with broth, This study was carried out to demonstrate the growth inhibition of two food poisoning bacteria Salmonella typhimurium and Staphylo-coccus aureus in Naengmyon-broth containing Dongchimi-juice fermented with high antibacterial lactic acid bacteria Lactobacillus homohiochii B21 and Leuconostoc mesenteroides C16. Naengmyon-broths were made with beef broth and Dongchimi-juice femented with lactiv acid bacteria and the changes in viable cell counts of the inoculated food poisoning bacteria in Naengmyon-broths were investigated during stor-age at 2$0^{\circ}C$ and 1$0^{\circ}C$. In Naengmyon-broth of 100% Dongchimi-juice stored at 2$0^{\circ}C$ the numbers of Sal-monella typhimurium and Staphylococcus aureus were rapidly decreased from 106CFU/ml to 106CFU/ml in 16 hours and 32 hours repectively. In Naengmyon-broth containing 10% Donchimi-juice the growths of the two food pois-oning bacteria were markedly inhibited compared with those in 100% beef broth though some growths were occurred in early phase. But in Naengmyon-broth of 100% beef broth their growths were very fast from early. Antibacterial activity of the Dongchimi-juice was more distinct at 2$0^{\circ}C$ that at 1$0^{\circ}C$ and was more active against Salmonella typhimurium than against Staphylococcus aureus.

Screening of Antimicrobial Activity from the Marine-Derived Fungus (해양균류의 항균활성 검색)

  • Acetone extracts of 301 strains of marine-derived fungus were tested for antimicrobial activity against three strains of bacteria. The bacteria consisted of three pathogens, Staphylococcus aureus, methicillin-resistant S. Aureus, and multidrug-resistant S. aureus. The acetone extracts of 10 strains (MFA117, MFA130, MFA134, MFA206, MFA217, MFA268, MFA277, MFA291, MFA292, MFA301) showed strong activity, inhibiting 100% of the bacterial growth. These antimicrobial active strains were cultlued in SWS medium on a 1 L scale and the resulting broth and mycelium were extracted to afford mycelium extract (000M) and broth extract (000B), respectively. Antimicrobial activity for all extracts has been tested as the results, the mycelium extract of one strain (217M) and the broth extracts of 9 strains (117B,130B, 134B, 206B, 268B, 277B, 291B, 292B, 301B) exhibited relatively high levels of activity at minimal inhibitory concentration (MIC) values of $500-125\;{\mu}g/mL$ range. Among them, the extracts, 277B, 291B, 292B and 301B showed the most significant antimicrobial activity with $IC_{50}$ values of $125\;{\mu}g/mL$.

In Vitro Activity of Taurine-5-Bromosalicylaldehyde Schiff Base Against Planktonic and Biofilm Cultures of Methicillin-Resistant Staphylococcus aureus

  • Staphylococcus aureus is a major human pathogen, implicated in both community and hospital acquired infections. The therapy for methicillin-resistant Staphylococcus aureus (MRSA) infections is becoming more difficult because of multidrug resistance and strong biofilm-forming properties. Schiff bases have attracted attention as promising antibacterial agents. In this study, we investigated the in vitro activity of taurine-5-bromosalicylaldehyde Schiff base (TBSSB) against MRSA. The minimum inhibitory concentrations (MIC) and minimum bactericidal concentration (MBC) were determined using a microtiter broth dilution method. TBSSB effectively inhibited planktonic MRSA, with an MIC of $32{\mu}g/ml$. The time-kill curve confirmed that TBSSB exhibited bactericidal activity against MRSA. TBSSB was also found to significantly inhibit MRSA biofilm formation at 24 h, especially at $1{\times}MIC$ and sub-MIC levels. Furthermore, scanning electron microscopy and transmission electron microscopy showed remarkable morphological and ultrastructural changes on the MRSA cell surface, due to exposure to TBSSB. This study indicated that TBSSB may be an effective bactericidal agent against MRSA.

Preparation of Ag-PS and Ag-PSS Particles by ${\gamma}$-Irradiation and Their Antimicrobial Efficiency against Staphylococcus aureus ATCC 6538 and Klebsiella pneumoniae ATCC 4352

  • Polystyrene, PS, particles of 450 nm diameter and poly(styrene-co-styrene sulfonate), PSS, particles of 140-160 nm diameter were prepared by emulsifier-free emulsion polymerization. The surfaces of the PS and PSS particles were coated with Ag nanoparticles for the application of antimicrobial agents by reduction of Ag ions using ${\gamma}$-irradiation. The Ag-PS and Ag-PSS were characterized by High-Resolution Transmittance Electron Microscopy (HR-TEM), Field-Emission Scanning Electron Microscopy (FE-SEM), and Energy Dispersive X-ray Spectroscopy (EDXS). The HR-TEM and EDXS data showed that the Ag nanoparticles were loaded on the surface of the PS and PSS particles, respectively. The antimicrobial efficiency of the Ag-PS and Ag-PSS particles (0.4 g) with ca. 100 ppm Ag, which was coated onto yam (KS K 0905-1996 rule), was tested against Staphylococcus aureus ATCC 6538 and Klebsiella pneumoniae ATCC 4352 after 100 washing cycles (KS K 0432-1999 rule). The antimicrobial efficiency of the Ag-PS particles against Staphylococcus aureus ATCC 6538 and Klebsiella pneumoniae ATCC 4352 was 99.9% after 100 cycles washing., confirming that the Ag-PS particles can be used as antimicrobial agents.

Epidemiological Investigation of Staphylococcus aureus Isolated from Bovine Mastitis Based on the Polymorphism of Coagulase Gene (젖소 유방염 유래 Staphylococcus aureus의 Coagulase Gene 유전형 분석에 의한 감염경로 규명)

  • Because Staphylococcus aureus (S. aureus) has variable number of short sequence repeat region in coagulase gene, it has been used to investigate the relatedness of S. aureus isolates. In this study, we isolated S. aureus strains from 20 dairy farms with bovine mastitis from September 2000 to August 2001. PCR-RFLP analysis of coagulase gene revealed 10 different patterns. Most of the S. aureus isolates showed only one coagulase gene RFLP pattern per farm. However, there were several S. aureus clones spreading between dairy farms. All the farms showed poor management conditions of milking machine and milker, indicating that managements for mastitis control program include use of proper milking matching, premilking sanitation, and segregation in the S. aureus infection herd. Our data suggest that PCR-RFLP analysis of coagulase gene might be applicable for the epidemiological investigations of S. aureus isolated from bovine mastitis cows.

A Longitudinal Study on Salivary Microorgnisms and Immunoglobulin A after Delivery of Fixed Orthodontic Appliances (고정식 교정 장치물 장착후 타액내 미생물과 면역 글로블린 A의 변화)

  • Fixed orthodontic appliances for the treatment of malocclusion has iatrogenic side effect such as demineralization of enamel, gingivitix and gingival hyperplasia. The purpose of this study is to longitudinally investigate the salivary microorganisms and immunoglobulin A after delivery of fixed orthodontic appliances for 10 months. Eight orthodontic patients were included in this study and the author has investigated the numbers of general bacteria, Streptococcus mutans Staphylococcus aureus and concentration of immunoglobulin A from unstimulated whole saliva. The author examined these parameters at prebracketing, 1 month after, 4 months after, 7 months after and 10 months after delivery of fixed orthodontic appliances. The obtained results were as follows : There were significant increases in the number of salivary general bacteria, Streptococcus mutans and Staphylococcus aureus after delivery of fixed orthodontic appliances The numbers of general bacteria were significantly increased at 1 month after (p<005), 4 months after (p<0.05), 7 months after (p<0.01), compared with prebracketing. However it showed no difference at 10 month after compared with 7 months after bracketing. The Numbers of Staphylococcus aureus were significantly increased at 1 month after (p<0.05), 4 months after(p<0.01), 7 month(p<0.01), compared with prebracketing. However it showed decreasing pattern at 10 months after compared with 7 months after bracketing. There was no significant difference in the concentration of immunoglobulin A after delivery of fixed orthodontic appliances.

  • Manuka oil sometime named New Zealand's tea tree oil is soluble in oil and come from nature. The $\alpha$-pinene extracted from Manuka oil and R-limonene which is one of the component of extracted Citrex from Grapefruit were used to estimate the antimicrobial activity and to improve the capability of antiseptic. Disk diffusion and broth dilution methods were used to measure the antimicrobial activity. Escherichia coli which is gram-negative bacteria and Staphylococcus aureus which is gram-positive bacteria were used as strain. The antimicrobial activity of Manuka oil and $\alpha$-pinene for Escherichia coli, Staphylococcus aureus is similar when the concentration of Manuka oil and $\alpha$-pinene is $10{\mu}l$. However, Antimicrobial activity of Manuka oil for Escherichia coli, Staphylococcus aureus is better than that of $\alpha$-pinene when the concentration of Manuka oil and $\alpha$-pinene is low. Antimicrobial activity of Citrex is superior to that of R-limonene. The proper ratio of Maunka oil and Citrex can improve the antimicrobial activity. The proper ratio obtained from studies was 75% of Maunka oil and 25% Citrex for Escherichia coli, 25% of Maunka oil and 75% Citrex for Staphylococcus aureus.
