  • The aim of the present study was to survey the frequency of inducible and constitutive phenotypes and inducible cross-resistant genes by regulating the methylation of 23S rRNA (ermA, ermB, and ermC) and macrolide efflux-related msrA gene in Staphylococcus aureus and S. epidermidis strains. A total of 172 bacterial isolates (identified based on standard tests), were examined in this study. Antibiotic susceptibility was determined by the disk diffusion method, and all isolates were evaluated with respect to inducible and constitutive phenotypes. The presence of ermA, ermB, ermC, and msrA genes was investigated by a PCR assay. The constitutive resistance phenotypes showed a higher distribution among the isolates. R phenotype was detected more among S. epidermidis isolates (46.25%). ermB, ermC, and msrA genes were detected more in methicillin-resistant S. aureus (MRSA) and methicillin-resistant S. epidermidis (MRSE) isolates that had R and HD phenotypes (>77% strains). The ermA gene had the lowest frequency among MRSA, MRSE, MSSA, and MSSE strains (<14% isolates). Distribution of inducible resistance genes in MRSA and MRSE strains, and possibly other species, leads to increased constitutive resistance to erythromycin, clindamycin, and other similar antibiotics. Therefore, it can be challenging to treat infections caused by these resistant strains.

  • Methicillin resistant Staphylococcus epidermidis Z0117SE0041 was isolated from nasal mucosa of human who raised companion dog. The complete genome of strain Z0117SE0041 consists of a 2.5 Mb chromosome and three circular plasmids with about 47, 36, and 11 kb in size, respectively. Since strain Z0117SE0041 raises concerns due to its potential to cause a disease and disseminate antibiotic resistance, further genome analysis is required in detail.

  • Methicillin resistant Staphylococcus epidermidis Z0117SE0042 was isolated from nasal mucosa of veterinarian. The complete genome of strain Z0117SE0042 contains a 2.5 Mb chromosome and two circular plasmids of about 24 kb and 23 kb. • We evaluated the antibiotic susceptibilities, hemolytic activities, and technological properties of 36 Staphylococcus xylosus strains and 49 S. pseudoxylosus strains predominantly isolated from fermented soybean foods from Korea. Most of the strains were sensitive to chloramphenicol, erythromycin, gentamycin, kanamycin, lincomycin, oxacillin, tetracycline, and trimethoprim. However, 23 strains exhibited potential phenotypic acquired resistance to erythromycin, lincomycin, and tetracycline. Based on breakpoint values for staphylococci from the Clinical and Laboratory Standards Institute, >30% of the isolates were resistant to ampicillin and penicillin G, but the population distributions in minimum inhibitory concentration tests were clearly different from those expected for acquired resistance. None of the strains exhibited clear α- or β-hemolytic activity. S. xylosus and S. pseudoxylosus exhibited salt tolerance on agar medium containing 20% and 22% (w/v) NaCl, respectively. S. xylosus and S. pseudoxylosus strains possessed protease and lipase activities, which were affected by the NaCl concentration. Protease activity of S. pseudoxylosus was strain-specific, but lipase activity might be a characteristic of both species. This study confirms the potential of both species for use in high-salt soybean fermentation, but the safety and technological properties of strains must be determined to select suitable starter candidates.

  • Antibiotic resistance patterns were determined for 14 strains of Staphylococcus aureus isolated at a hospital in Pusan during summer in 1989. Resistance to chloramphenicol or clindamycin was recorded in 100% of strains. Resistance to the other compounds tested was as follows: tetracycline 86%, gentamicin 79%, tobramycin 71%, kanamycin 71%, erythromycin 57%, ampicillin 57%, methicillin 50%, streptomycin 29%, cephalothin 29%, and trimethoprim 21%. All strains were sensitive to vancomycin and rifampicin. All strains showed multiple resistance to more than 3 antibiotics.

  • The complete nucleotide sequence of pKH4, a small multidrug resistance (smr) plasmid isolated from multidrug resistant Staphylococcus aureus SA5, was determined. Sequence analysis has revealed that pKH4 has two open reading frames for Rep and Smr proteins. The comparison of the amino acid sequence of Smr protein of pKH4 with those of other Smr proteins of various Staphylococcus showed that Smr protein of pKH4 is a new member of the SMR family.

  • Puerperal sepsis is one of the leading causes of maternal morbidity and mortality worldwide. Postpartum pelvic infections can cause various complications, including wound infections and necrotizing fasciitis. Several microorganisms are known to cause such infections; however, no study has reported on Staphylococcus caprae, a coagulase-negative staphylococcus that is isolated frequently from animals and infrequently from human specimens, as a causative agent. Here, we report a rare case of septic shock complicated by necrotizing fasciitis after a cesarean section. This is the first report of a human isolate of S. caprae in association with puerperal sepsis and necrotizing fasciitis.

  • Effects of the commercial laundry detergents, fabric softeners, and bleaches on bactericidal activities were studied. Bactericidal activities under various conditions during washing, rinsing, and bleaching were evaluated in terms of minimum inhibitory concentration(MIC) by a broth dilution method. The experiments were carried out using Staphylococcus aureus and Escherichia coli which were usually employed in antibacterial activity tests. The results are summarized as follows : 1. All samples of Powder detergents(10 samples), soaps(7 samples) and fabric softeners(4 samples) were inactive against the Staphylococcus aureus and Escherichia coli. 2. One sample in the liquid detergents(6 samples) showed a very low MIC for Staphylococcus aureus and Escherichia coli., and three samples showed a selective effect for two bacterial strains. 3. Bleaching agent(containing peroxide) did not have bactericidal effect for Staphylococcus aureus and Escherichia coli. but the other(containing chlorine) had the effect for every bacterial strain in standard concentration. 4. All samples showed bactericidal effect above 60$^{\circ}C$.

  • To produce the recombinant fibronectin-binding protein(FnBP) for development of subunit vaccine against Staphylococcus aureus. The fnbp gene was amplified from the chromosomal DNA of S aureus KNU 196 strain using the polymerase chain reaction, and cloned into pGEX-4T-2. Then, the recombinant FnBP fused with glutathione-S-transferase was produced in E coli, purified by affinity chromatography, and identified its antigenicity and immunogenicity by Western blot. The recombinant FnBP produced in this study is considered to have the same property of native FnBP purified from S aureus, and is expected to be useful as a candidate for S aureus subunit vaccine.

  • When tetracycline resistancy were cured by ethidium bromide treatment, some of the cured strains lost the tetracycline resistance plasmid while other strains kept the plasmids. Both strains of lost and remained plasmids were digested with restriction endonuclease Hind III and these cleaved plasmids were compared with that of parent strains, two plasmid remained strains showed same cleavage patterns between parent and cured strains, however, one plasmid lost strain showed dissimilarity with parent strain, but in the other one strain, among 4 plasmid lost colonies, 2 showed same but other 2 showed different patterns compared to parent strain. Tetracycline was transfered by conjugation in on set(Staphylococcus aureus donor versus Staphylococcus epidermidis, recipient) with relative high frequency but the other 2 sets showed a low degree of frequency and the other 2 sets exhibited no transfer.
