Secretion Capacity Limitations of the Sec Pathway in Escherichia coli

  • The secretion capacity of two E. coli strains (JM109 and AF1000) was evaluated through the expression of two human proinsulin fusion proteins using the translocation signal sequence from Staphylococcal protein A (SpA). Although a 7 to 11-fold difference in the expression levels was attained by the use of different promoters (SpA and malK promoters) and copy-number vectors (700 and 50 copies per cell), the maximum translocation rates for all the systems were around 140,000 amino acids $cell^{-1} min^{-1}$. Moreover, the secretion capacity was found to be independent of the size of the exiting peptide and its translational rate.

  • A total of 113 Staphylococcus hyicus subsp. hyicus which had been isolated from healthy and diseased pigs were examined for their ability to produce staphylococcins. For selection of staphylococcin indicator strains, 80 strains were randomly selected from 113 strains, and were tested as indicator and as donator. The 8 indicators were chosen and used to type all of the strains. The ability to produce staphylococcin could be demonstrated in 43 strains (38.1%) of 113 Staph hyicus subsp. hyicus. The frequency of activity was higher in diseased pig strains (44.4%) than in healthy pig strains(36.8%). 43 staphylococcin producer strains were differentiated into 10 reaction types. The inhibitory effects of the staphylococcin (N27 and R 3) on other staphylococci differed by the staphylococcal species tested. All the gram-negative species were completely resistant to the three producing strains.

Clinical study of empyema thoracis: a review of 110 cases (농흉의 임상적 고찰110)

  • The author made a clinical study of IIO cases of empyema thoracis who were diagnosed and treated at department of chest surgery, chosun university hospital, during the period of December 1979 through June 1983. 1. In age and sex distribution, 45 cases [41%] was under the age of 15 years, 65 cases [59%] was above the age of 15 years. The ratio of male to female was 2.6:1. 2. The predisposing factors were pneumonia 45 cases [41%] and pulmonary tuberculosis 40 cases [36.5%]. 3. The cardinal symptoms were dyspnea, chest pain, fever, cough in order. 4. Etiologic organisms were confirmed in 69% which requested in 87 cases. Staphylococcal infection were 19 cases, Streptococcal infection were 13 cases, pneumococcal infection were 11 cases. 5. In treatment of empyema, thoracentesis 4 cases, closed thoracotomy 50 cases, open drainage 29 cases, decortication 14 cases and thoracoplasty 13 cases. In children, only thoracentesis and closed thoracotomy was favorable result in treatment. 6. 103 cases were discharged with recovery and improvement but 7 cases were early discharged by their economic or personal condition without improved.

  • Several quaternary pyridinium cephalosporin analogues were prepared analogues were Prepared and evaluated in vitro for antibacterial activity against selected Gram-positive and Cram-negative organisms. Most of the synthesized analogues were either as effective or less effective against the tested bacterial organ isms than the reference com pounds, Cefpirome and Ceftazidime.

  • The complete nucleotide sequence of pKH6, a tetracycline-resistance (Tc$^{r}$) plasmid isolated from multi-drug resistant Staphylococcus aureus SA2, has been determined and compared with that of the staphylococcal Tc$^{r}$ plasmid pTl8l. The nucleotide sequences of the two plasmids are in agreement except for 7 nucleotides. All differences are caused by base pair substitutions. Among 6 substitutions, 3 occurred in coding regions. However, only two base substitutions in coding regions resulted in changes of amino acid sequences in two different ORFs of repC and Pre proteins.

  • Licochalcone E was firstly isolated from licorice root in 2005, which belongs to the retrochalcone family. Studies on the biological activities of licochalcone E were in the initial stage. In the study, we demonstrated that licochalcone E has potent antimicrobial property against Staphylococcus aureus. Furthermore, via hemolysis, Western blot, and real-time RT-PCR assays, we have shown that subinhibitory concentrations of licochalcone E dose-dependently reduces the production of ${\alpha}$-toxin in both methicillin-sensitive S. aureus (MSSA) and methicillin-resistant S. aureus (MRSA). The data suggest that licochalcone E may deserve further investigation as a potential therapeutic against S. aureus infections, or the structure of licochalcone E may be used as a basis for chemical synthesis of novel anti-S. aureus compounds.

  • Pneumatocele (PC) is a thin-walled cyst of the lung that can occur at all ages and with various etiologies. However, there is no fully accepted consensus for the management of PC in a neonatal intensive care unit. Although the management of PC is generally expectant, it is difficult to decide how long conservative management should be maintained, especially under Korea's medical care environment and the parents' worry and anxiety. We report a male neonate, born at $27^{+5}weeks$ gestation, weighing 1,000 g, who had a post infectious PC caused by methicillin-resistant Staphylococcus aureus sepsis. We treated conservatively for about 100 days (roughly 14 weeks), but unfortunately after a few days of chest retraction, acute exacerbation occurred, video assisted thoracoscopic surgery (VATS) was deemed necessary and performed. The purpose of this publication is to describe the clinical course, aggravation and relief after VATS management with a review of the literature.

Congenital swinepox of neonatal pigs in a Korean domestic farm (국내 신생 자돈에서 발생한 선천성 돈두 증례보고)

  • Three neonatal pigs from the same litter in a domestic farm were born with skin lesions. Grossly, multiple well-circumscribed, round papules distributed over the skin of the three piglets. Two piglets were submitted for a diagnosis of skin disease. Microscopically, epidermal hyperplasia with ballooning degeneration of stratum spinosum keratinocytes was observed. Some keratinocytes contained eosinophilic intracytoplasmic inclusions and a central nuclear vacuole and chromatin margination. Swinepox (SWP) virus was detected by polymerase chain reaction and nucleotide sequencing, and Staphylococcus hyicus was isolated in skin lesions. Based on the gross findings and laboratory results, these piglets were diagnosed with congenital SWP with a secondary staphylococcal infection.

  • Methicillin-resistant Staphylococcus pseudintermedius Z0118SP0130 was isolated from eye specimen of a companion dog in South Korea. The complete genome of Z0118SP0130 consisted of a 2,663,277 bp chromosome and there was no plasmid. The strain was identified as the sequence type 45 and contained a mecA gene which comprised of staphylococcal cassette chromosome mec type Vb (5C2&5). Antimicrobial resistance to erythromycin, clindamycin, quinupristin-dalfopristin, trimethoprim-sulfamethoxazole, mupirocin, oxacillin, streptomycin, and gentamicin was observed in the strain.

  • Staphylococcus epidermidis is a normal flora of human skin and is occasionally associated with pathogenic infections. We report the complete genome sequence of methicillin-resistant Staphylococcus epidermidis strain Z0118SE0272 isolated from the residential environment sharing by a companion dog and dwellers. Resistance to cefoxitin was observed in the strain, whereas it was susceptible to erythromycin, clindamycin, quinupristin-dalfopristin, trimethoprim-sulfamethoxazole, mupirocin, vancomycin, teicoplanin, linezolid, and tigecycline. The strain Z0118SE0272 identified as sequence type 130 possessed the mecA gene responsible for methicillin resistance, which composed the new type of staphylococcal cassette chromosome mec elements lacking mecRI.
