Taxonomic Study of Bacillus coagulans by Deoxyribonucleic Acid-Deoxyribonucleic Acid Hybridization Technique (DNA-DNA Hybridization에 의한 Bacillus coagulans의 분류학적 연구)

  • Taxonomic study of 11 strains of Bacillus coagulans and 14 strains of 13 spccies of Bacillus by deoxyribonucleic acid (DNA)-DNA hybridization were conducted. Among the 11 strains of B. coagulans, 6 were isolated from soil and the rest were the authentic strains obtained from American Type culture collection (ATCC) or the Institute for Fermentation, Osaka (IFO). All strains were examined to confirm as they are expected species of B. coagulans by the methods of Cordon et al. according to Bergey's Manual (8th ed.). The intraspecific DNA homology indexes among the 11 strains of B. coagulans using strain ATCC 7050 as the standard ($^3$H labeled input DNA) showed 76% or, more, respectively. These findings accorded well with the results of the conventional taxonomic study according to the Bergey's Manual. The interspecific DNA homology indexes between B. coagulant strain ATCC 7050 and the type cultures of B. subtilis (168), B. licheniformis (IFO 12107), B. pumilus (IFO 12110), B. firmus (ATCC 14575), B. lentus (ATCC 10840), B. circulans (ATCC 4513), B. macelans (ATCC 8244), B. polymyxa (ATCC 842), B. sphaericus (ATCC 14577), B. brevis (ATCC 8246, IFO 12334), B. laterosporus (ATCC 64), and B. pantothenticus (ATCC 14576) respectively, showed 2 to 4%, while that of between B. coagulans ATCC 7050 and Escherichia coli K-12 was less than 1 %.

  • China is a country with a long history. Chinese culture dates back thousands of years ago. Thousand years of history left the huge quantity of archival heritage, which consists of the memory of China. From tied knots, tortoise shell, bronze, bamboo to paper, film, CD, the mankind's history is kept and continued through the evolution of the documenting media and documenting methods. In the information era, when we are immersed in the sea of information technologies, archivists, as guards of human's memory, have to look for a balance point between new and old, between unchanged and changed. On one hand, archivists should try their best to protect traditional archives in a usable, authentic way in a long term; on the other hand, they must face the challenges posed by electronic record. The information age is a stage of the social development of mankind, the digitalization of archives is an important progress of human history. The report mainly is composed of three parts of the content: first, introduce the preserving situation of Chinese archival heritage ; focus are put on "China archival heritage program" and the construction of "Special archives repository"; second, the process of digitalization of traditional archives; third, the framework of electronic record standard.

Comparison of Environment, Growth, and Management Performance of the Standard Cut Chrysanthemum 'Jinba' in Conventional and Smart Farms

  • Background and objective: This study was conducted to compare the cultivation environment, growth of cut flowers, and management performance of conventional farms and smart farms growing the standard cut chrysanthemum, 'Jinba'. Methods: Conventional and smart farms were selected, and facility information, cultivation environment, cut flower growth, and management performance were investigated. Results: The conventional and smart farms were located in Muan, Jeollanam-do, and conventional farming involved cultivating with soil culture in a plastic greenhouse, while the smart farm was cultivating with hydroponics in a plastic greenhouse. The conventional farm did not have sensors for environmental measurement such as light intensity and temperature and pH and EC sensors for fertigation, and all systems, including roof window, side window, thermal screen, and shading curtain, were operated manually. On the other hand, the smart farm was equipped with sensors for measuring the environment and nutrient solution, and was automatically controlled. The day and night mean temperatures, relative humidity, and solar radiation in the facilities of the conventional and the smart farm were managed similarly. But in the floral differentiation stage, the floral differentiation was delayed, as the night temperature of conventional farm was managed as low as 17.7℃ which was lower than smart farm. Accordingly, the harvest of cut flowers by the conventional farm was delayed to 35 days later than that of the smart farm. Also, soil moisture and EC of the conventional farm were unnecessarily kept higher than those of the smart farm in the early growth stage, and then were maintained relatively low during the period after floral differentiation, when a lot of water and nutrients were required. Therefore, growth of cut flower, cut flower length, number of leaves, flower diameter, and weight were poorer in the conventional farm than in the smart farm. In terms of management performance, yield and sales price were 10% and 38% higher for the smart farm than for the conventional farm, respectively. Also, the net income was 2,298 thousand won more for the smart farm than for the conventional farm. Conclusion: It was suggested that the improved growth of cut flowers and high management performance of the smart farm were due to precise environment management for growth by the automatic control and sensor.

Effect of Zero Drainage Using Drainage Zero Sensor on Root Zone Environment, Growth and Yield in Tomato Rockwool Culture (토마토 수경재배 시 배액제로 센서를 이용한 배액제로화가 근권환경, 생육 및 수량에 미치는 영향)

  • This study was carried out to investigate the effect of irrigation method adopted for reducing nutrient solution drainage on the root zone environment, growth and yield of a tomato crop grown in a rockwool medium. The irrigation control methods used were large quantity irrigation at a long interval controlled by only an integrated solar radiation sensor (standard), medium quantity irrigation at a medium interval (zero drainage 1), and small quantity irrigation at a short interval (zero drainage 2) controlled by both an integrated solar radiation sensor and a zero drainage sensor. The amount of the nutrient solution supplied and the drain percentage per plant of the standard, zero drainage 1, and zero drainage 2 were 1.4, 0.9 and 0.8 L, and 23.8, 8.6 and 3.7%, respectively. The average, minimum, and maximum water contents and EC of the standard, zero drainage 1, and zero drainage 2 were 64.5~88% and $1.5{\sim}3.5dS{\cdot}m^{-2}$, 40.3~76.0% and $2.5{\sim}4.0dS{\cdot}m^{-2}$, and 56.3~69.0% and $2.7{\sim}3.7dS{\cdot}m^{-2}$, respectively. There was no difference in leaf width, number of leaves, and stem diameter among the treatments. However, plant height and leaf length decreased in the zero drainage 1 and 2 treatments as compared to the standard. The fruit marketable yield per 10a in the zero drainage 1 and 2 treatments was about 93 and 88%, respectively, of that in the standard treatment.

Rapid Extraction of DNA using Ion Exchange Resin for Early Detection of Mycobacterium tuberculosis by the Polymerase Chain Reaction (결핵균 PCR에서 이온교환수지를 이용한 신속한 DNA 분리)

  • Background: The extraction methods of DNA from clinical samples are the major obstacle to use the PCR(polymerase Chain Reaction) in routine labortary for early detection of M. tuberculosis. We tried to improve the extraction method of DNA from sputum for establishment of the PCR in routine labortary by reducing the possibility of cross contamination and performing it easily and safely. Methods: We used the $InstaGene^{TM}$ DNA extraction kit(BioRad Co.) using Chelex 100 ion exchange resin for preparation of DNA. We compared InstaGene method in 100 cases of sputum from proteinase K method which is known as the most commonly used method for DNA purification(Experiment 1). And we compared InstaGene method in 98 cases of sputum from Microwave method developed by a company in Korea(Experiment 2). In experiment 1,245bps of IS6110 were amplified and then 188bps were amplified by nested PCR. In experiment 2,536bps in primary PCR and 276bps in nested PCR were amplified and analysed by agarose gel electrophoresis and EtBr staining. Results: When we chose AFB smear, culture, or AFB smear and culture as a standard test, PCR had low specificity and positive predictive value in both experiments. The InstaGene method has higher value in sensitivity and negative predictive value significantly than proteinase K method. The InstaGene method and the Microwave methods were similar in sensitivity, specificity, positive predictive value and negative predictive value. Conclusion: Even though both methods had lower possibility of cross contamination, shorter time requirement, simplicity, and economic advantages than Proteinase K method, the InstaGene method was a little simpler than the Microwave method. Therefore, in terms of usefulness in clinical application, the Instagene method seems to be the most useful method in DNA extraction for detection of M. tuberculosis using PCR. The reliability of this method will be clarified by further studies with enough clinical samples.

Analysis of Moisture Characteristics in Rockwool Slabs using Time Domain Reflectometry (TDR) Sensors and Their Applications to Paprika Cultivation (TDR 센서를 이용한 암면 슬라브 수분 특성 분석 및 파프리카 재배의 적용 예)

  • To investigate the characteristics of moisture content (MC), moisture distribution and starting point of drainage in a rockwool slab culture, time domain reflectometry (TDR) sensors were used in a drip irrigation system. MC values ($0{\sim}100%$) measured by TDR sensors in a slab were compared to those by loadcells. Seventy two seedlings of paprika (Capsicum annuum L.) were cultured for $5{\sim}6$ months in a green-house and the starting point of irrigation was determined by the average value of three TDR sensors which were inserted diagonally across the slabs under the plants. MCs as a standard for starting point of irrigation by TDR were determined with 40%, 50%, and 60%. Distribution of MCs in a slab measured with five TDR sensors equally spaced from two irrigation points were not much different when the MC in the slab increased from zero to saturation point. The saturated MCs in the slab were presented at $58{\sim}65%$ and the drain was started when the MC became around $50{\sim}55%$. At the saturated MC in the slab, TDR sensors presented 100% but the values from the loadcell showed 90% at the same time. However, measurement errors between two methods for MC remarkably decreased with a decrease in the MC in a slab. Especially when the MC was maintaining below 60%, the errors between TDR and loadcell methods for measuring MC in the rock-wool slab were less than 5%. There were no significant differences in number of fruits and fresh and dry weights of fruits when they were cultured under the different MC conditions with three irrigation regimes (40%, 50%, and 60%). These results indicated that the MC control by TDR sensors in a rock-wool based paprika culture can be suggested as a method to determine the starting point of irrigation for a soilless culture system.

  • Purpose : Early degeneration of articular cartilage is accompanied by a loss of glycosaminoglycan (GAG) and the consequent change of the integrity. The purpose of this study was to biochemically quantify the loss of GAG, and to evaluate the $Gd(DTPA)^{2-}$-enhanced, and T1, T2, rho relaxation map for detection of the early degeneration of cartilage. Materials and Methods : A cartilage-bone block in size of $8mm\;\times\;10mm$ was acquired from the patella in each of three pigs. Quantitative analysis of GAG of cartilage was performed at spectrophotometry by use of dimethylmethylene blue. Each of cartilage blocks was cultured in one of three different media: two different culture media (0.2 mg/ml trypsin solution, 1mM Gd $(DTPA)^{2-}$ mixed trypsin solution) and the control media (phosphate buffered saline (PBS)). The cartilage blocks were cultured for 5 hrs, during which MR images of the blocks were obtained at one hour interval (0 hr, 1 hr, 2 hr, 3 hr, 4 hr, 5 hr). And then, additional culture was done for 24 hrs and 48 hrs. Both T1-weighted image (TR/TE, 450/22 ms), and mixed-echo sequence (TR/TE, 760/21-168ms; 8 echoes) were obtained at all times using field of view 50 mm, slice thickness 2 mm, and matrix $256\times512$. The MRI data were analyzed with pixel-by-pixel comparisons. The cultured cartilage-bone blocks were microscopically observed using hematoxylin & eosin, toluidine blue, alcian blue, and trichrome stains. Results : At quantitation analysis, GAG concentration in the culture solutions was proportional to the culture durations. The T1-signal of the cartilage-bone block cultured in the $Gd(DTPA)^{2-}$ mixed solution was significantly higher ($42\%$ in average, p<0.05) than that of the cartilage-bone block cultured in the trypsin solution alone. The T1, T2, rho relaxation times of cultured tissue were not significantly correlated with culture duration (p>0.05). However the focal increase in T1 relaxation time at superficial and transitional layers of cartilage was seen in $Gd(DTPA)^{2-}$ mixed culture. Toluidine blue and alcian blue stains revealed multiple defects in whole thickness of the cartilage cultured in trypsin media. Conclusion : The quantitative analysis showed gradual loss of GAG proportional to the culture duration. Microimagings of cartilage with $Gd(DTPA)^{2-}$-enhancement, relaxation maps were available by pixel size of $97.9\times195\;{\mu}m$. Loss of GAG over time better demonstrated with $Gd(DTPA)^{2-}$-enhanced images than with T1, T2, rho relaxation maps. Therefore $Gd(DTPA)^{2-}$-enhanced T1-weighted image is superior for detection of early degeneration of cartilage.

Standard Chemotherapy with Excluding Isoniazid in a Murine Model of Tuberculosis (마우스 결핵 모델에서 Isoniazid를 제외한 표준치료의 예비 연구)

  • Background: Isoniazid (INH, H) is a key drug of the standard first-line regimen for the treatment of tuberculosis (TB), yet some reports have suggested that treatment efficacy was maintained even though INH was omitted from the treatment regimen. Methods: One hundred forty C57BL/6 mice were infected with the H37Rv strain of M. tuberculosis with using a Glas-Col aerosol generation device, and this resulted in depositing about 100 bacilli in the lung. Four weeks after infection, anti-TB treatment was initiated with varying regimens for 4-8 weeks; Group 1: no treatment (control), Group 2 (4HREZ): 4 weeks of INH, rifampicin (R), pyrazinamide (Z) and ethambutol (E), Group 3: 1HREZ/3REZ, Group 4: 4REZ, Group 5: 4HREZ/4HRE, Group 6: 1HREZ/3REZ/4RE, and Group 7: 4REZ/4RE. The lungs and spleens were harvested at several time points until 28 weeks after infection, and the colony-forming unit (CFU) counts were determined. Results: The CFU counts increased steadily after infection in the control group. In the 4-week treatment groups (Group 2-4), even though the culture was negative at treatment completion, the bacilli grew again at the 12-week and 20-week time points after completion of treatment. In the 8-week treatment groups (Groups 5-7), the bacilli did not grow in the lung at 4 weeks after treatment initiation and thereafter. In the spleens of Group 7 in which INH was omitted from the treatment regimen, the culture was negative at 4-weeks after treatment initiation and thereafter. However, in Groups 5 and 6 in which INH was taken continuously or intermittently, the bacilli grew in the spleen at some time points after completion of treatment. Conclusion: TThe exclusion of INH from the standard first-line regimen did not affect the treatment outcome in a murine model of TB in the early stage of disease. Further studies using a murine model of chronic TB are necessary to clarify the role of INH in the standard first-line regimen for treating TB.

Improvement of Straw Loading Method on Survival of Mouse IVF/IVC Blastocysts Cryopreserved by Vitrification (체외수정과 체외배양에서 생산된 생쥐 배반포기배의 초자화 동결에 있어서 Straw Loading 방법의 개선)

  • This study was carried out to investigate the effect of straw loading method and thawing protocol on the in vitro development of in vitro produced mouse blastocysts cryopreserved by vitrification. Three loading types of straw I, Il and III on loading and sealing method were made for vitrification. The ability of the solution on straw loading methods to remain vitreous during warming was tested by exposed in air for 1 to 10 s sec. and then plunged the vitrified straws into water bath at 25°C. Embryos to be vitrified were equilibrated to the 20% EG for 5min. and exposed in EFS 40 for 1min. The plug ends of Straw I and Straw II were sealed with straw powder and Straw III was treated straw powder, followed by heat sealing and then plunged into LN$_2$. The results obtained in these experiments were summarized as follows; 1) Straw I embryo column mostly changed from transparent to opaque upon thawing without exposure in air for 3-6 sec. Straw II embryo column was I improved partially but was not remained completely vitreous during warming. However, when Straw lll loading method was used, the embryo column was remained vitreous completely. 2) High survival rates and development rates of each groups (middle blastocysts and hatching blastocysts) of vitrified embryos were obtained by using Straw III loading method than Straw I method (P<0.05). And the range of s standard error was low in Straw lll method. 3) When the embryos vitrified-frozen were placed in air for 3, 5 and l0sec. and then warmed rapidly in water bath at 25$^{\circ}C$, the survival rates after 24h of culture were 72.7-87.1% and the development rates to hatching stage after 48h of culture were 34.0-48.4%. There were no significantly differences according to exposure time in air during warming. In conclusion, the present results showed that highly survival and low standard error of vitrified-frozen mouse bIastocysts were obtained by using straw lll loading, double sealing and appropriate 2 step warming method.

Chardin's Genre Paintings of Child Education: The Enlightenment Views on Children of the French Bourgeois Class in the 18th Century (샤르댕의 아동 교육 장르화 - 18세기 프랑스 부르주아의 계몽주의적 아동관)

  • This paper examines four genre paintings on the subject of child education by Jean-Baptiste-Sim${\'{e}}$on Chardin(1699-1779). The Governess, The Diligent Mother, Saying Grace, and The Morning Toilette garnered critical attention after they were exhibited in the Salon from 1739 to 1741. After the exhibition, the paintings were made into prints and frequently sold to members of the bourgeois class in Paris. The iconographical details of Chardin's genre paintings have, thus far, been compared to Dutch genre pictures of the seventeenth century. Further, most studies conducted on Chardin's paintings focus on formal analysis rather than the historical and social contexts. Through attempting social-contextual readings of Chardin's educational series, this paper argues that the significance of Chardin's painting series of child education lies in his representation of the ideal French bourgeois family and the standard of early childhood education in the eighteenth-century French Enlightenment period. In each of the four child education paintings, Chardin depicted a mother with children in a domestic space. Even though this theme derives from traditional Dutch genre paintings in the seventeenth century, the visual motifs, the pictorial atmosphere and the painting techniques of Chardin all project the social culture of eighteenth century France. Each painting in the child education series exemplifies respectively the attire of a French gentlemen, the social view on womanhood and the education of girls, newly established table manners, and the dressing up culture in a 'toilette' in eighteenth century France. Distinct from other educational scenes in previous genre paintings, Chardin accentuated the naive and innocent characteristics of a child and exemplified the mother's warmth toward that child in her tender facial expressions and gesturing. These kinds of expressions illustrate the newly structured standard of education in the French Enlightenment period. Whereas medieval people viewed children as immature and useless, people in the eighteenth century began to recognize children for their more positive features. They compared children to a blank piece of paper (tabula rasa), which signified children's innocence, and suggested that children possess neither good nor bad virtues. This positive perspective on children slowly transformed the pedagogical methods. Teaching manuals instructed governesses and mothers to respect each child's personality rather than be strict and harsh to them. Children were also allotted more playtimes, which explains the display of various toys in the backgrounds of Chardin's series of four paintings. Concurrently, the interior, where this exemplary education was executed, alludes to the virtue of the bourgeois's moderate and thrifty daily life in eighteenth century France. While other contemporary painters preferred to depict the extravagant living space of a French bourgeoisie, Chardin portrayed a rather modest and cozy home interior. In contrast to the highly decorated living space of aristocrats, he presented the realistic, humble domestic space of a bourgeois, filled with modern household objects. In addition, the mother is exceptionally clad in working clothes instead of fashionable dresses of the moment. Fit to take care of household affairs and children, the mother represents the ideal virtues of a bourgeois family. It can be concluded that the four genre paintings of child education by Chardin articulate the new standards of juvenile education in eighteenth century France as well as the highly recognized social virtues between French bourgeois families. Thus, Chardin's series of child education would have functioned as a demonstration of the ideal living standards of the bourgeois class and their emphasis on early childhood education in the French Enlightenment period.
